1Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 2A human circulating immune cell landscape in aging and COVID-19显示文摘Age-associated changes in immune cells have been linked to an increased risk for infection.However,a global and detailed characterization of the changes that human circulating immune cells undergo with age is lacking.Here,we combined scRNA-seq,mass cytometry and sCATAC-seq to compare immune cell types in peripheral blood collected from young and old subjects and patients with COVID-19.We found that the immune cell landscape was reprogrammed with age and was characterized by T cell polarization from naive and memory cells to effector,cytotoxic,exhausted and reg-ulatory cells,along with increased late natural killer cells,age-associated B cells,inflammatory monocytes and age-associated dendritic cells.In addition,the expression of genes,which were implicated in coron-avirus susceptibility,was upregulated in a cell subtype-specific manner with age.Notably,COVID-19 promoted age-induced immune cell polarization and gene expression related to inflammation and cellular senes-cence.Therefore,these findings suggest that a dysreg-ulated immune system and increased gene expression associated with SARS-CoV-2 susceptibility may at least partially account for COVID-19 vulnerability in the elderly.Yingfeng Zheng Xiuxing Liu Wenqing Le Lihui Xie He Li Wen Wen Si Wang Shuai Ma Zhaohao Huang Jinguo Ye Wen Shi Yanxia Ye Zunpeng Liu Moshi Song Weiqi Zhang Jing-Dong J.Han Juan Carlos lzpisua Belmonte Chuanle Xiao Jing Qu Hongyang Wang Guang-Hui Liu Wenru Su 2020Protein & Cell2020,11,10:10

5Evaluation on Fracture Toughness at Dynamic Loading for Welded Joint Based on the Local Approach显示文摘The changes in mechanical properties and fracture toughness by dynamic loading were investigated with experiments.The parameter R, which can reflect the effect of the loading rate and the temperature rising during the high loading rate, could be employed to describe the constituent relation for the typical structure steel and its weld metal. The dynamic loading effect on the stress/strain fields and the temperature variation in the vicinity of the crack tip was analyzed by the finite element method, the dynamic fracture behavior was evaluated based on the local approach. 6Fusion boundary evolution, precipitation behaviour, and interaction with dislocations in an Fe–22Cr–15Ni steel weldment during long-term creep显示文摘A tungsten inert gas welded joint between a novel heat-resistant austenitic steel and ERNiCrCoMo-1 weld metal was investigated before and after creep in this study. The evolution of the microstructures in the base and weld metals was discussed based on the electron back-scatter diffraction(EBSD) and transmission electron microscopy(TEM) analyses. The preferred orientations of the fusion boundary after creep revealed the influence of the applied stress on creep deformation mechanism. A cooperative nucleation process of M23C6 carbides in the base metal was proposed. The finely distributed Cu-rich phase was cut off by the dislocations during creep, leading to increased mean size and reduced amount of the nano-Cu phase. A modified triple-precipitate hardening model was constructed based on TEM observations of the interactions between the particles and the dislocations in the base metal after creep at 200 MPa. 7Study on the Basic Mechanical Behaviors of High Density Polyethylene Electrofusion Welded Joints at Different Temperature显示文摘The basic mechanical behaviors of high density polyethylene electrofusion welded joint at different temperatures were studied by using differently designed specimens in this paper. The results show that the strength of weld bonding plane is higher than that of the pipe and socket materials at room temperature. In order to get the shear strength of electrofusion welded joint,the effective bond lengths were reduced by cutting artificial groove through the socket. The effective bonding length of welded joint to get the shear strength is decreased with decreasing testing temperature. 20Understanding auditory neuropathy spectrum disorder: a system- atic review in transgenic mouse models显示文摘Auditory neuropathy spectrum disorder is a unique group of hearing dysfunctions characterized by preserved outer hair cell function and abnormal neural conduction of the auditory pathway. However, the pathogenic mechanism underlying this disorder is not clear. We therefore performed a systematic review of genetic mouse models with different gene mutations to provide a valuable tool for better understanding of the process and the possible molecular mechanisms. Of the 18 articles retrieved, nine met the required criteria. All biochemical, histological, and electrophysiological results were recorded for each of the mouse models, as was the transgenic technology.
