| 1 | Effect of magnesium ions/Type I collagen promote the biological behavior of osteoblasts and its mechanism显示文摘Type I collagen(Col I)is a main component of extracellular matrix(ECM).Its safety,biocompatibility,hydrophilicity and pyrogen immunogenicity make it suitable for tissues engineering applications.Mg2t also control a myriad of cellular processes,including the bone development by enhancing the attachment and differentiation of osteoblasts and accelerating mineralization to enhance bone healing.In our studies,Mg2t bind collagen to promote the proliferation and differentiation of osteoblasts through the expression of integrins and downstream signaling pathways.In order to clarify the biological behavior effect of 10mM Mg2t/Col I coating,we performed 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT),alkaline phosphatase(ALP),406-diamidino-2-phenylindole(DAPI),Alizarin red staining and Rhodamine B-isothiocyanate(RITC)-labeled phalloidin experiments and found that 10mM Mg2t group,Col I-coating group,10mM Mg2t/Col I-coating group,respectively,promoted the proliferation and differentiation of osteoblasts,especially 10mM Mg2t/Col I-coating group.We detected the mRNA expression of osteogenic-related genes(Runx2,ALP and OCN,OPN and BMP-2)and the protein expression of signaling pathway(integrin a2,integrin b1,FAK and ERK1/2),these results indicated that 10mM Mg2t/Col I coating play an critical role in up-regulating the MC3T3-E1 cells activity.The potential mechanisms of this specific performance may be through activating via integrin a2b1-FAK-ERK1/2 protein-coupled receptor pathway. | Xiaojing Nie Xirao Sun Chengyue Wang Jingxin Yang | 2020 | Regenerative Biomaterials2020,7,1: | 8 |
| 2 | Determination of rare earth elements in plant protoplasts by MAA显示文摘A preliminary study on the speciation of rare earth elements in plant cells has been carried out by molecular activation analysis (MAA). Mesophyll protoplasts of Brassica napus were isolated by enzymatic digestion. After being washed with isosmotic solution containing EDTA for several times, the protoplasts were purified by gradient centrifugation. Then the concentration of rare earth elements (REEs) in the protoplasts was determined by neutron activation analysis. | 3 | Hd1,Ghd7,and DTH8 synergistically determine the rice heading date and yield-related agronomic traits显示文摘Heading date determines the seasonal and regional adaptation of rice(Oryza sativa L.)varieties and is mainly controlled by photoperiod sensitivity(PS).The core heading date genes Hd1,Ghd7,DTH8,and PRR37 act synergistically in regulating the PS.In this study,we systematically analyze the heading date,PS,and agronomic traits of eight homozygous lines with various combinations of Hd1,Ghd7,and DTH8 alleles in the prr37 background under long-day(LD)and short-day(SD)conditions,respectively.We find that Hd1 alone promotes heading,regardless of the day length.However,under LDs,Hd1 suppresses flowering,in coordination with functional Ghd7 or with Ghd7 and DTH8.These loci cooperate to negatively regulate the Ehd1-Hd3 a/RFT1 pathway and delay heading.Under SDs,Hd1 competes with various heading suppressors to promote heading.Therefore,the dual function of Hd1 is vital for PS.The lines carrying Hd1 alone show reduced plant height with fewer primary and secondary branches in panicles.Lines carrying Ghd7 and DTH8(with hd1)show delayed heading and improve agronomic traits.Overall,our results reveal the regulation of rice PS flowering by the core heading date genes and their effects on agronomic traits,providing valuable information for the selection of rice varieties for adaptation to different light and temperature conditions. | Kangli Sun Minghui Huang Wubei Zong Dongdong Xiao Chen Lei Yanqiu Luo Yingang Song Shengting Li Yu Hao Wanni Luo Bingqun Xu Xiaotong Guo Guangliang Wei Letian Chen Yao-Guang Liu Jingxin Guo | 2022 | Journal of Genetics and Genomics2022,49,5: | 4 |

| 5 | Epithelial-mesenchymal transition status of circulating tumor cells in breast cancer and its clinical relevance显示文摘Objective:Circulating tumor cells(CTCs)play a critical role in cancer metastasis,but their prevalence and significance remain unclear.This study attempted to track the epithelial-mesenchymal transition(EMT)status of CTCs in breast cancer patients and investigate their clinical relevance.Methods:In this study,the established negFACS-IF:E/M platform was applied to isolate rare CTCs and characterize their EMT status in breast cancer.A total of 89 breast cancer patients were recruited,including stage 0–III(n=60)and late stage(n=29)cases.Results:Using the negFACS-IF:E/M platform,it was found that in human epidermal growth factor receptor 2(HER2)+patients,mesenchymal CTCs usually exhibited a high percentage of HER2+cells.Stage IV breast cancer patients had considerably more CTCs than stage 0–III patients.Among stage 0–III breast cancers,the HER2 subtype included a significantly higher percentage of mesenchymal and biphenotypic(epithelial and mesenchymal)CTCs than the luminal A or B subtypes.Among stage IV patients,CTCs were predominantly epithelial in cases with local recurrence and were more mesenchymal in cases with distant metastasis.By applying a support vector machine(SVM)algorithm,the EMT status of CTCs could distinguish between breast cancer cases with metastasis/local recurrence and those without recurrence.Conclusions:The negFACS-IF:E/M platform provides a flexible and generally acceptable method for the highly sensitive and specific detection of CTCs and their EMT traits in breast cancer.This study demonstrated that the EMT status of CTCs had high clinical relevance in breast cancer,especially in predicting the distant metastasis or local recurrence of breast cancer. | Jiaojiao Zhou Xuan Zhu Shijie Wu Jingxin Guo Kun Zhang Chunjing Xu Huihui Chen Yuxi Jin Yuting Sun Shu Zheng Yiding Chen | 2020 | Cancer Biology & Medicine2020,17,1: | 3 |
