1人参皂苷Rg1对糖尿病大鼠心肌氧化应激及细胞凋亡的影响(英文)显示文摘目的:探讨人参皂苷Rg1对高血糖所致心肌损害的保护作用及其机制。创新点:使用糖尿病大鼠为实验对象,探讨三种浓度的人参皂苷Rg1对糖尿病心肌损伤的保护作用及其机制,检测其是否具有浓度依赖性。方法:将60只Wistar大鼠随机分组,其中空白对照组10只,另50只给予高脂高糖饲养,4周后腹腔注射40 mg/kg链脲佐菌素(STZ)。成功制备糖尿病大鼠模型40只,再随机分为糖尿病模型组,糖尿病大鼠+低剂量人参皂苷Rg1(10 mg/(kg·d)),糖尿病大鼠+中剂量人参皂苷Rg1(15 mg/(kg·d)),糖尿病大鼠+高剂量人参皂苷Rg1(20 mg/(kg·d))。12周后处死大鼠,取血测定空腹血糖、总胆固醇(TC)、甘油三酯(TG)、心肌酶及氧化应激水平,留取心肌组织使用透射电镜观察心肌细胞超微结构改变,应用TUNEL法检测心肌细胞凋亡,免疫组化检测细胞凋亡相关蛋白半胱氨酸天冬氨酸蛋白酶3(CASP3)和Bcl-x L的表达。结论:人参皂苷Rg1对糖尿病大鼠糖脂代谢无明显影响,人参皂苷Rg1可降低糖尿病大鼠血清肌钙蛋白(c Tn I)和肌酸激酶同工酶(CK-MB)水平,改善心肌细胞超微结构,减少心肌细胞凋亡,降低大鼠血清和心肌组织中丙二醛(MDA)含量,提高超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH)水平,降低凋亡蛋白CASP3的表达,同时提高Bcl-x L蛋白表达。总之,人参皂苷Rg1能显著保护糖尿病大鼠心肌损伤,其机制可能与其抗氧化及抗细胞凋亡作用有关。Hai-tao YU Juan ZHEN Bo PANG Jin-ning GU Sui-sheng WU 2015Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2015,16,5:23

4Sub-Lethal Effects of Fenvalerate on the Development, Fecundity, and Juvenile Hormone Esterase Activity of Diamondback Moth, Plutella xylostella (L.)显示文摘The toxicities of fenvalerate (20% EC) to the 3rd instar larvae of diamondback moth (DBM), Plutella xylostella (L.), reared on three host plants viz., radish, oilseed rape, and cabbage were tested. The LC50 values of fenvalerate to the 3rd instar larvae of DBM varied with host plants, however, there wasn’t any significant difference among them (P > 0.05). Similarly, DBM fed on three host plants had same pupal weight, pupal period, pupation rate, adult emergence rate, female ratio, and fecundity. The activity of juvenile hormone esterase (JHE, EC 3.1.1.1) in the 3rd instar larvae of DMB did not significantly vary with host plants, either. These suggested that DBM had similar fitness on the three host plant species. When fed on the host plants pretreated with fenvalerate at the concentrations equivalent to LC20, LC50 and LC80, the pupation rate, pupal weight, adult emergence rate, female ratio, fecundity, and JHE activity of the tested insects were declined as compared with insects in control treatments fed on the same host plant species. Furthermore, the pupal period of the tested insects was extended after fenvalerate treatment. The decrease in JHE activity after fenvalerate treatment in the tested insects could partly explain the changes in the mentioned growth parameters. 5Liver Enzymes Concentrations Are Closely Related to Pre-diabetes:Findings of the Shanghai Diabetes Study Ⅱ (SHDS Ⅱ)显示文摘Objective To investigate the relationship of liver enzymes with hyperglycemia in a large population in Shanghai and identify the association between liver enzymes and insulin resistance.Methods A total of 3 756 participants were enrolled.Each participant underwent an oral glucose tolerance test and completed a questionnaire.Anthropometric indices were recorded and serum samples were collected for measurement.Results Liver enzymes concentrations were independently associated with i-IGT,IFG+IGT,and diabetes.With the increase of ALT and GGT concentrations,ORs for i-IGT,IFG+IGT,and diabetes increased gradually.By comparing patients in the highest quartile of GGT concentrations or ALT concentrations with those in the lowest quartile (Q1),ORs for i-IGT,IFG+IGT,or diabetes was significant after adjustment.Both ALT and GGT concentrations were linearly correlated with HOMA-IR and independently associated with HOMA-IR [ALT OR (95% CI):2.56 (1.51-4.34) P=0.00;GGT OR (95% CI):2.66 (1.53-4.65) P=0.00].Conclusion Serum ALT and GGT concentrations were closely related to pre-diabetes and diabetes in the Shanghai population and positively associated with insulin resistance.GAO Fei PAN Jie Min HOU Xu Hong FANG Qi Chen LU Hui Juan TANG Jun Ling GU Hui Lin PAN Zhi Jian YAO You Hua SHEN Wei Zhen JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:15
6Gut-lymph-lung pathway mediates sepsis-induced acute lung injury显示文摘This review attempts to unveil the possible mechanisms underlying how gut lymph affects lung and further gives rise to acute respiratory distress syndrome,as well as potential interventional targets under the condition of ischemia-reperfusion injury.We searched electronic databases including PubMed,MEDLINE,Cochrane Central Register of Controlled Trials,Google Scholar,Web of Science,and Embase to identify relevant literatures published up to December 2019.We enrolled the literatures including the Mesh Terms of'gut lymph or intestinal lymph and acute lung injury or acute respiratory distress syndrome.'Gut is considered to be the origin of systemic inflammation and the engine of multiple organ distress syndrome in the field of critical care medicine,whereas gut lymph plays a pivotal role in initiation of ischemia-reperfusion injury-induced acute respiratory distress syndrome.In fact,in the having been established pathologic model of sepsis leading to multiple organ dysfunction named by Gut Lymph theory,a variety of literatures showed the position and role of changes in gut lymph components in the initiation of systemic inflammatory response,which allows us to screen out potential intervention targets to pave the way for future clinic and basic research.Can Jin Jie Chen Juan Gu Wei Zhang 2020Chinese Medical Journal2020,,18:14
7Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 8The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 9Climate and soil moisture environment during develop-ment of the fifth palaeosol in Guanzhong Plain显示文摘Based on weathering characteristics of the fifth palaeosol layer (S5) of four sections in Guanzhong Plain, the thickness of the weathered profile of the paleosol is determined to be greater than the ordi- nary soil, a weathered and leached loess layer thicker than 2 m. The distribution depth of the red argil- lans, the weathered and leached loess layer, Fe2O3, CaCO3 and Sr content under the S5 all indicate that the precipitation in Guanzhong Plain was over 900 mm at that time. The distribution depth of gravity water zone reached 4.2 m at least, and the soil moisture content was generally more than 20% within the range of 4.2 m. At that time there was sufficient soil moisture and no dried earth layer developed in Guanzhong Plain, suitable for the forest to develop. When this soil developed, the mean annual pre- cipitation was more than the annual soil moisture evaporation. The value of soil moisture balance was positive and the atmospheric precipitation could supply the underground water normally. Soil water was weak acidic in the middle and late stages when S5 developed in Guanzhong Plain. It was a kind of subtropical climate and even more humid and warmer than the northern edge of the subtropical climate zone in Guanzhong Plain when the S5 developed. At that time the subtropical climate was prevailing over the northern side and southern side of Qingling Mountains, showing the Mountains no longer to be the boundary between the subtropical zone and the temperate zone in China. 10Rapid Detection of Haemophilus influenzae and Haemophilus parainfluenzae in Nasopharyngeal Swabs by Multiplex PCR显示文摘Objective To establish multiplex PCR-based assays for detecting H.influenzae and H.parainfluenzae.And the PCR-based assays were applied to detect the carriage rates of H.influenzae and H.parainfluenzae in nasopharyngeal swab specimens which were collected from healthy children.Methods Multiplex primers for species-specific PCR were designed by using DNAstar soft based on the sequences of 16S rRNA genes from genus Haemophilus to detect H.influenzae and H.parainfluenzae.Results The sensitivity of the 16S rRNA PCR assay for detecting H.influenzae and H.parainfluenzae was 97.53% and 100% respectively,and the specificity was 95.89% and 96.63% respectively.Youden's Index on the ability to detect H.influenzae and H.parainfluenzae was 0.9342 and 0.9663 respectively.666 nasopharyngeal swab specimens were collected from healthy children.The detection rates of H.influenzae and H.parainfluenzae were 14.11% and 16.07% respectively by using isolation and culture methods.The detection rates of H.influenzae and H.parainfluenzae were 43.54% and 57.96% respectively by 16S rRNA PCR assays.The carriage rates of serotypes a,b,c,d,e,f and non-typeable isolates were 0% (0/666),0.15% (1/666),1.20% (8/666),0.15% (1/666),1.20% (8/666),1.80% (12/666),95.50% (636/666) respectively.Conclusion The multiplex PCR assays were very rapid,reliable and feasible methods for detection of H.influenzae and H.parainfluenzae in pharyngeal swab specimens which were compared to conventional isolation and culture methods.95.5% of H.influenzae strains in healthy children were nontypeable.The encapsulated or typable strains were mainly three serotypes which was c,e,and f serotype.TIAN Guo Zhong ZHANG Li Juan WANG Xiao Lei ZHANG Li LI Shu Feng GU Chang Mei SUN Jian CUI Bu Yun 2012Biomedical and Environmental Sciences2012,25,3:8
11Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
12Integrated transcriptomic and metabolomic analyses of a wax deficient citrus mutant exhibiting jasmonic acid-mediated defense against fungal pathogens显示文摘Naturally,resistant crop germplasms are important resources for managing the issues of agricultural product safety and environment deterioration.We found a spontaneous mutant of‘Newhall’navel orange(Citrus sinensis Osbeck)(MT)with broad-spectrum protections against fungal pathogens in the orchard,postharvest-storage,and artificial inoculation conditions.To understand the defense mechanism of MT fruit,we constructed a genome-scale metabolic network that integrated metabolome and transcriptome datasets.The coordinated transcriptomic and metabolic data were enriched in two sub-networks,showing the decrease in very long chain fatty acid(by 41.53%)and cuticular wax synthesis(by 81.34%),and increase in the synthesis of jasmonic acid(JA)(by 95.23%)and JA-induced metabolites such as 5-dimethylnobietin(by 28.37%)in MT.Furthermore,cytological and biochemical analyses confirmed that the response to fungal infection in MT was independent of wax deficiency and was correlated with the levels of jasmonates,and the expression of plant defensin gene PDF1.2.Results of exogenous application of MeJA and JA inhibitors such as propyl gallate proved that JA-mediated defense contributes to the strong tolerance against pathogens in MT.Our results indicated that jasmonate biosynthesis and signaling are stimulated by the fatty acid redirection of MT,and participate in the tolerance of pathogenic fungi.Yizhong He Jingwen Han Runsheng Liu Yuduan Ding Jinqiu Wang Li Sun Xiaoming Yang Yunliu Zeng Weiwei Wen Juan Xu Hongming Zhang Xiang Yan Zhaoxing Chen Zuliang Gu Hong Chen Huanqing Tang Xiuxin Deng Yunjiang Cheng 2018Horticulture Research2018,5,1:8

14A detailed investigation of low latitude tweek atmospherics observed by the WHU ELF/VLF receiver:2.Occurrence features and associated ionospheric parameters显示文摘As a companion paper to Zhou RX et al.(2020),this study describes application of the automatic detection and analysis module to identify all the tweek atmospherics detectible in the WHU ELF/VLF receiver data collected at Suizhou station during the period of 3 February through 29 February 2016.Detailed analysis of the identified low-latitude tweek events reveals that the occurrence rate varies considerably—from 800 to 6000 tweeks per day,and exhibits a strong diurnal and local time dependence,the peak occurring before local midnight.The diurnal variation of identified tweeks was similar to that of the lightning data obtained by the World-Wide Lightning Location Network(WWLLN)..Estimates of the propagation distance and ionospheric reflection height of tweek atmospherics suggest that the majority(~92%)of the low latitude tweeks originate from the lightning activity within a radius of 4000 km and that they are very likely to reflect from the lower ionospheric D-region at the height range of 75–85 km.At these lower ionospheric reflection altitudes,~74%of the corresponding electron densities from the tweek spectral measurements are within 24.5–27.5 cm^-3.The daily variation of estimated D-region electron densities in the considered period(February 2016)also exhibits a small overall increasing trend from early to later in the month.Juan Yi XuDong Gu Wen Cheng XinYue Tang Long Chen BinBin Ni RuoXian Zhou ZhengYu Zhao Qi Wang LiQing Zhou 2020Earth and Planetary Physics2020,4,3:8
15A detailed investigation of low latitude tweek atmospherics observed by the WHU ELF/VLF receiver:Ⅰ. Automatic detection and analysis method显示文摘As a dispersive wave mode produced by lightning strokes, tweek atmospherics provide important hints of lower ionospheric(i.e., D-region) electron density. Based on data accumulation from the WHU ELF/VLF receiver system, we develop an automatic detection module in terms of the maximum-entropy-spectral-estimation(MESE) method to identify unambiguous instances of low latitude tweeks.We justify the feasibility of our procedure through a detailed analysis of the data observed at the Suizhou Station(31.57°N, 113.32°E) on17 February 2016. A total of 3961 tweeks were registered by visual inspection;the automatic detection method captured 4342 tweeks, of which 3361 were correct ones, producing a correctness percentage of 77.4%(= 3361/4342) and a false alarm rate of 22.6%(= 981/4342).A Short-Time Fourier Transformation(STFT) was also applied to trace the power spectral profiles of identified tweeks and to evaluate the tweek propagation distance. It is found that the fitting accuracy of the frequency–time curve and the relative difference of propagation distance between the two methods through the slope and through the intercept can be used to further improve the accuracy of automatic tweek identification. 16Optimization of variables for maximizing efficacy and efficiency in aerial spray application to cotton using unmanned aerial systems显示文摘Aerial spraying can support efficient defoliation without crop contact.With the recent introduction to unmanned aerial system(UAS)for aerial spraying in China,there is a need to determine the optimum application variables to achieve high efficacy and efficiency with low costs.The present research involved field studies across two annual cotton production seasons in North Xinjiang,China.Four factors,including volume rate(A),tank mix including spray adjuvants(B),flight altitude(C),flight speed(D)and three levels of L9(3^(4))orthogonal arrays were carried out to optimize the application parameters for three types of UASs.These included different numbers of rotors as follows:four-rotors,six-rotors and eight-rotors.Spray coverage,distribution uniformity(coefficient of variation(CV)of droplet coverage),rates of cotton defoliation and boll opening,application efficiency and cost were measured and assessed.Results showed that:(1)the rates of defoliation and boll opening by aerial cotton defoliant application could meet the requirement of cotton mechanized harvesting;(2)the optimal scenario for the three UASs was A_(3)B_(2)C_(1)D_(3),Volume rate(A3):48 L/hm^(2);Tank mix and concentration(B_(2)):(Tuotulong 225+Sujie 750+Ethephon 2250)mL/hm^(2),Flight altitude(C_(1)):1.5 m,and Flight speeds(D_(3))for unmanned helicopters with four-rotors,six-rotors and eight-rotors were 3.12 m/s,2.51 m/s and 3.76 m/s,respectively.These results can provide guidance for cotton defoliant aerial spraying in China using UAS.Juan Liao Ying Zang Xiwen Luo Zhiyan Zhou Yubin Lan Yu Zang Xiuyan Gu Weiming Xu Andrew John Hewitt 2019International Journal of Agricultural and Biological Engineering2019,12,2:6
17Numerical Simulation of Characteristics of CEX Ions in Ion Thruster Optical System显示文摘Charge exchange(CEX) ions could inflict severe damages on the ion thruster optical system.This article is aimed at investigating the characteristics of the CEX ions and their influences upon the optical system by means of particle-incell(PIC) ion simulation and Monte Carlo collision(MCC) methods.The results from numerical simulation indicate that despite the fact that CEX ions appear in the entire beamlet region near the ion optical system,the ones that present themselves downstream of the accelerator grid have good reason for attracting more attention.As their trajectories are significantly affected by the local electric field,a great number of CEX ions are accelerated toward grids resulting in sputtering erosion.When the influences of the CEX ions are considered in the numerical simulation,there could hardly be observed augments in the screen grid current,but the accelerator grid current increases from zero to 1.4% of the beamlet current.It can be understood from the numerical simulation that the CEX ions formed in the region far downstream of the accelerator grid should be blamed for the erosion on the downstream surface of the accelerator grid.Zhong Lingwei Liu Yu Li Juan Gu Zuo Jiang Haocheng Wang Haixing Tang Haibin 2010Chinese Journal of Aeronautics2010,23,1:5

19Effect of cyclic wetting-drying on tensile mechanical behavior and microstructure of clay-bearing sandstone显示文摘The understanding of the weakening mechanism of tensile strength of rock subjected to cyclic wetting-drying is critical for rock engineering.Tensile strength tests were conducted on a total of 35 sandstone specimens with different wetting-drying cycles.The crack propagation process and acoustic emission characteristics of the tested samples were obtained through a high-speed camera and acoustic emission system.The results indicate that the tensile strength is observably reduced after cyclic wetting-drying,and the extent of the reduction is not only related to the number of wettingdrying cycle,but also closely related to the clay mineral content of the sample.In addition,as the cycles of wetting-drying increase,the effect of each single cycle on tensile strength get reduced until it becomes constant.Moreover,the crack initiation and penetration time is prolonged as the number of wetting-drying cycle increases,which indicates that cyclic wetting-drying weakens the rock stiffness and enhances the ductility of sandstone.Meanwhile,the acoustic emission characteristics of the tested samples further confirmed the ductile behaviour of the sandstone samples with increasing wetting-drying cycle.Furthermore,through the analysis of the microstructure and mineral composition of the samples with different wetting-drying cycles,it is concluded that the main weakening mechanisms of sandstones containing clay minerals are frictional reduction,chemical and corrosive deterioration.Pingye Guo Juan Gu Yi Su Jiong Wang Zhanwen Ding 2021International Journal of Coal Science & Technology2021,8,5:4
20Influence of substrate metals on the crystal growth of AlN films显示文摘AlN films were deposited by reactive radio frequency (RF) sputtering on various bottom electrodes,such as Al,Ti,Mo,Au/Ti,and Pt/Ti.The effects of substrate metals on the orientation of AlN thin films were investigated.The results of X-ray diffraction,atomic force microscopy,and field emission scanning electron microscopy show that the orientation of AlN films depends on the kinds of substrate metals evidently.The differences of AlN films deposited on various metal electrodes are attributed to the differences in lattice mismatch and thermal expansion coefficient between the AlN material and substrate metals.The AlN film deposited on the Pt/Ti electrode reveals highly the c-axis orientation with well-textured columnar structure.The positive role of the Pt/Ti electrode in achieving the high-quality AlN films and high-performance film bulk acoustic resonator (FBAR) may be attributed to the smaller lattice mismatch as well as the similarity of thermal expansion coefficient between the deposited AlN material and the Pt/Ti electrode substrate.Juan Xiong Hao-shuang Gu Kuan Hu Ming-zhe Hu 2010International Journal of Minerals,Metallurgy and Materials2010,17,1:4
