2Effects of Long-Term Winter Planted Green Manure on Physical Properties of Reddish Paddy Soil Under a Double-Rice Cropping System显示文摘Soil physical properties are important indicators of the potential for agricultural production.Our objective was to evaluate the effects of long-term inputs of green manures on physical properties of a reddish paddy soil(Fe-Typic Hapli-Stagnic Anthrosols) under a double cropping system.The common cropping pattern before the study was early-late rice-fallow(winter).The field treatments included rice-rice-fallow(R-R-WF),rice-rice-rape(R-R-RP),rice-rice-Chinese milk vetch(RR-MV),and rice-rice-ryegrass(R-R-RG).The rape,Chinese milk vetch and ryegrass were all incorporated as green manures 15 d before early rice transplanting during the following year.The soil bulk density in all green manure treatments was significantly reduced compared with the winter fallow treatment.Soil porosity with green manure applications was significantly higher than that under the winter fallow.The green manure treatments had higher 0.25-5 mm water stable aggregates and aggregates stabilities in the plow layer(0-15 cm depth) compared with the fallow treatment.The mean weight diameter(MWD) and normalized mean weight diameter(NMWD) of aggregates in the green manure treatment were larger than that with the winter fallow.Soil given green manure retained both a higher water holding capacity in the plow layer soil,and a larger volume of moisture at all matric potentials(-10,-33 and-100 kPa).We conclude that the management of double-rice fields in southern central China should be encouraged to use green manures along with chemical fertilizers to increase SOC content,improve soil physical properties and soil fertility.YANG Zeng-ping XU Ming-gang ZHENG Sheng-xian NIE Jun GAO Ju-sheng LIAO Yu-lin XIE Jian 2012Journal of Integrative Agriculture2012,11,4:40
3Extract of Naotaifang, a compound Chinese herbal medicine, protects neuron ferroptosis induced by acute cerebral ischemia in rats显示文摘Objective:Our previous research showed that Naotaifang(a compound traditional Chinese herbal medicine)extract(NTE)has clinically beneficial effects on neurological improvement of patients with acute cerebral ischemia.In this study,we investigated whether NTE protected acute brain injury in rats and whether its effects on ferroptosis could be linked to the dysfunction of glutathione peroxidase 4(GPX4)and iron metabolism.Methods:We established an acute brain injury model of middle cerebral artery occlusion(MCAO)in rats,in which we could observe the accumulation of iron in neurons,as detected by Perl’s staining.Using assay kits,we measured expression levels of ferroptosis biomarkers,such as iron,glutathione(GSH),reactive oxygen species(ROS)and malonaldehyde(MDA);further the expression levels of transferrin receptor1(TFR1),divalent metal transporter 1(DMT1),solute carrier family 7 member 11(SLC7 A11)and GPX4 were determined using immunohistochemical analysis,real-time quantitative polymerase chain reaction and Western blot assays.Results:We found that treatment with NTE reduced the expression levels of TFR1 and DMT1,reduced ROS,MDA and iron accumulation and reduced neurobehavioral scores,relative to untreated MCAO rats.Treatment with NTE increased the expression levels of SLC7 A11,GPX4 and GSH,and the number of Nissl bodies in the MCAO rats.Conclusion:Taken together,our data suggest that acute cerebral ischemia induces neuronal ferroptosis and the effects of treating MCAO rats with NTE involved inhibition of ferroptosis through the TFR1/DMT1 and SCL7 A11/GPX4 pathways.Bin Lan Jin-wen Ge Shao-wu Cheng Xi-long Zheng Jun Liao Chao He Zheng-qing Rao Guo-zuo Wang 2020Journal of Integrative Medicine2020,18,4:37

5A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. 6Effects of Long-Term Winter Planted Green Manure on Distribution and Storage of Organic Carbon and Nitrogen in Water-Stable Aggregates of Reddish Paddy Soil Under a Double-Rice Cropping System显示文摘In agricultural systems, maintenance of soil organic matter has long been recognized as a strategy to reduce soil degradation. Manure amendments and green manures are management practices that can increase some nutrient contents and improve soil aggregation. We investigated the effects of 28 yr of winter planted green manure on soil aggregate-size distribution and aggregateassociated carbon(C) and nitrogen(N). The study was a randomized completed block design with three replicates. The treatments included rice-rice-fallow, rice-rice-rape, rice-rice-Chinese milk vetch and rice-rice-ryegrass. The experiment was established in 1982 on a silty light clayey paddy soil derived from Quaternary red clay(classified as Fe-Accumuli-Stagnic Anthrosols) with continuous early and late rice. In 2009, soil samples were collected(0-15 cm depth) from the field treatment plots and separated into water-stable aggregates of different sizes(i.e., >5, 2-5, 1-2, 0.5-1, 0.25-0.5 and <0.25 mm) by wet sieving. The long-term winter planted green manure significantly increased total C and N, and the formation of the 2-5-mm water-stable aggregate fraction. Compared with rice-rice-rape, rice-rice-Chinese milk vetch and rice-rice-ryegrass, the rice-rice-fallow significantly reduced 2-5-mm water-stable aggregates, with a significant redistribution of aggregates into micro-aggregates. Long-term winter planted green manure obviously improved C/N ratio and macro-aggregate-associated C and N. 7Long-Term Effect of Fertilizer and Rice Straw on Mineral Composition and Potassium Adsorption in a Reddish Paddy Soil显示文摘Increasing K+ adsorption can be an effective alternative in building an available K pool in soils to optimize crop recovery and minimize losses into the environment. We hypothesized that long-term fertilization might change K+ adsorption because of changes in the chemical and mineralogical properties of a rice (Oryza sativa L.). The aims of this study were (i) to determine clay minerals in paddy soil clay size fractions using X-ray diffraction methods and a numerical diagram-decomposition method; (ii) to measure K+ adsorption isotherms before and after H 2 O 2 oxidation of organic matter, and (iii) to investigate whether K+ adsorption is correlated with changes in soil chemical and mineral properties. The 30-yr long-term fertilization treatments caused little change in soil organic C (SOC) but a large variation in soil mineral composition. The whole-clay fraction (<5 μm) corresponded more to the fertilization treatment than the fine-clay fraction (<1 μm) in terms of percentage of illite peak area. The total percentage of vermiculite-chlorite peak area was significantly negatively correlated with the total percentage of illite peak area in the <5 μm soil particles (R=-0.946, P<0.0006). Different fertilization treatments gave significantly different results in K+ adsorption. The SOC oxidation test showed positive effects of SOC on K+ adsorption at lower K+ concentration ( 120 mg L-1) and negative effects at higher K+ concentration (240 mg L-1). The K+ adsorption by soil clay minerals after SOC oxidization accounted for 60-158% of that by unoxidized soils, suggesting a more important role of soil minerals than SOC on K+ adsorption. The K+ adsorption potential was significantly correlated to the amount of poorly crystallized illite present (R=0.879, P=0.012). 9High-resolution remote sensing mapping of global land water显示文摘Land water, one of the important components of land cover, is the indispensable and important basic information for climate change studies, ecological environment assessment, macro-control analysis, etc. This article describes the overall study on land water in the program of global land cover remote sensing mapping. Through collection and processing of Landsat TM/ETM+, China's HJ-1 satellite image, etc., the program achieves an effective overlay of global multi-spectral image of 30 m resolution for two base years, namely, 2000 and 2010, with the image rectification accuracy meeting the requirements of 1:200000 mapping and the error in registration of images for the two periods being controlled within 1 pixel. The indexes were designed and selected reasonably based on spectral features and geometric shapes of water on the scale of 30 m resolution, the water information was extracted in an elaborate way by combining a simple and easy operation through pixel-based classification method with a comprehensive utilization of various rules and knowledge through the object-oriented classification method, and finally the classification results were further optimized and improved by the human-computer interaction, thus realizing high-resolution remote sensing mapping of global water. The completed global land water data results, including Global Land 30-water 2000 and Global Land 30-water 2010, are the classification results featuring the highest resolution on a global scale, and the overall accuracy of self-assessment is 96%. 10Increased brachial-ankle pulse wave velocity is associated with impaired endothelial function in patients with coronary artery disease显示文摘Background Pulse wave velocity and flow-mediated vasodilation (FMD) are widely used as noninvasive modalities for evaluating atherosclerosis. However, it is not known whether pulse wave velocity is related to FMD in patients with coronary artery disease (CAD). Therefore, the present study was designed to investigate the alteration in brachial-ankle pulse wave velocity (baPWV) and endothelial function in CAD patients. Methods Thirty-three patients with CAD and thirty control subjects were recruited for this study. baPWV was measured non-invasively using a VP 1000 automated PWV/ABI analyzer (PWV/ABI, Colin Co. Ltd., Komaki, Japan). Endothelial function as reflected by FMD in the brachial artery was assessed with a high-resolution ultrasound device. Results baPWV was increased in CAD patients compared with control subjects [(1756.1±253.1) cm/s vs (1495.3±202.3) cm/s, P<0.01]. FMD was significantly reduced in CAD patients compared with control subjects [(5.2±2.1) % vs (11.1±4.4) %, P<0.01]. baPWV correlated with FMD (r =-0.68, P<0.001). The endothelium-independent vasodilation induced by sublingual nitroglycerin in the brachial artery was similar in the CAD group compared with the control group. Conclusions CAD is associated with increased baPWV and endothelial dysfunction. Increased baPWV parallels diminished endothelial function. 11Effect of notoginsenoside R1 on hepatic microcirculation disturbance induced by gut ischemia and reperfusion显示文摘AIM: To assess the effect of notoginsenoside R1 on hepatic microcirculatory disturbance induced by gut ischemia/reperfusion (I/R) in mice.METHODS: The superior mesenteric artery (SMA) of C57/BL mice was ligated for 15 min to induce gut ischemia followed by 30-min reperfusion. In another set of experiments, R1 was continuously infused (10 mg/kg per hour) from 10 min before I/R until the end of the investigation to study the influence of R1 on hepatic microcirculatory disturbance induced by gut I/R. Hepatic microcirculation was observed by inverted microscopy, and the vascular diameter, red blood cell (RBC) velocity and sinusoid perfusion were estimated. Leukocyte rolling and adhesion were observed under a laser confocal microscope. Thirty and 60 min after reperfusion, lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate transaminase (AST) in peripheral blood were determined. The expression of adhesion molecules CD11b/CD18 in neutrophils and tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) and monocyte chemotactic protein-1 (MCP-1) in plasma were evaluated by flow cytometry. E-selectin and intercellular adhesion molecule-1 (ICAM-1) in hepatic tissue were examined by immunofluorescence. RESULTS: After gut I/R, the diameters of terminal portal venules and central veins, RBC velocity and the number of perfused sinusoids were decreased, while the leukocyte rolling and adhesion, the expression of E-selectin in hepatic vessels and CD18 in neutrophils, IL-6, MCP-1, LDH, ALT and AST were increased. R1 treatment attenuated these alterations except for IL-6 and MCP-1.CONCLUSION: R1 prevents I/R-induced hepatic microcirculation disturbance and hepatocyte injury. 12Total salvianolic acid improves ischemia-reperfusion-induced microcirculatory disturbance in rat mesentery显示文摘AIM:To investigate the effect of total salvianolic acid(TSA) on ischemia-reperfusion(I/R)-induced rat mesenteric microcirculatory dysfunctions.METHODS:Male Wistar rats were randomly distributed into 5 groups(n = 6 each):Sham group and I/R group(infused with saline),TSA group,TSA + I/R group and I/R + TSA group(infused with TSA,5 mg/kg per hour).Mesenteric I/R were conducted by a ligation of the mesenteric artery and vein(10 min) and subsequent release of the occlusion.TSA was continuously infused either starting from 10 min before the ischemia or 10 min after reperfusion.Changes in mesenteric microcirculatory variables,including diameter of venule,velocity of red blood cells in venule,leukocyte adhesion,free radicals released from venule,albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope.Meanwhile,the expression of adhesion molecules CD11b/CD18 on neutrophils was evaluated by flow cytometry.Ultrastructural evidence of mesenteric venules damage was assessed after microcirculation observation.RESULTS:I/R led to multiple responses in mesenteric post-capillary venules,including a significant increase in the adhesion of leukocytes,production of oxygen radicals in the venular wall,albumin efflux and enhanced mast cell degranulation in vivo.All the I/R-induced manifestations were significantly reduced by pre-or post-treatment with TSA,with the exception that the I/R-induced increase in mast cell degranulation was inhibited only by pre-treatment with TSA.Moreover,preor post-treatment with TSA significantly attenuated the expression of CD11b/CD18 on neutrophils,reducing the increase in the number of caveolae in the endothelial cells of mesentery post-capillary venules induced by I/R.CONCLUSION:The results demonstrated that TSA protects from and ameliorates the microcirculation disturbance induced by I/R,which was associated with TSA inhibiting the production of oxygen-free radicals in the venular wall and the expression of CD11b/CD18 on neutrophils.Ming-Xia Wang Yu-Ying Liu Bai-He Hu Xiao-Hong Wei Xin Chang Kai Sun Jing-Yu Fan Fu-Long Liao ChuanShe Wang Jun Zheng Jing-Yan Han Ming-Xia Wang 2010World Journal of Gastroenterology2010,16,42:15
13Effects of daily telephone-based re-education before taking medicine on Helicobacter pylori eradication: A prospective single-center study from China显示文摘AIM: To investigate the effects of daily telephonebased re-education(TRE) before taking medicine for the eradication of Helicobacter pylori(H. pylori) on the compliance and the eradication rate in a Chinese patient population.METHODS : Aprospective, physician-blind e d, randomized, controlled clinical study was conducted. The patients were randomly assigned to receive TRE every day before taking medicine(TRE group) or no TRE(control group). The patients in the TRE group received regular instructions before taking medicine for the eradication of H. pylori during the entire courseof treatment through telephone calls. The patients in the control group received detailed instructions at the time of seeing a doctor for the guidance. The primary outcome was the H. pylori eradication rate after treatment. The secondary outcomes included the clinical remissions after treatment, adverse events, compliance, and patients' satisfaction.RESULTS: A total of 140 patients were randomized, 70 to the TRE group and 70 to the control group. As the primary outcome, the H. pylori eradication rates in the TRE and control groups were 62.7% and 71.2% in per protocol analysis(P = 0.230), and 52.9% and 52.9% in intention-to-treat analysis(P = 0.567), respectively. As the secondary outcomes, there were no significant differences in the patients' satisfaction between the two groups(good, 79.7% vs 76.9%; fair, 13.6% vs 19.2%; poor, 6.7% vs 3.9%, for the TRE group and control group, respectively; P > 0.05 for all); the rates of adverse effects were 15.2% and 63.5% in the TRE and control groups, respectively(P < 0.001); the compliance rates in the TRE and control groups were 85.7% and 74.3%, respectively(P = 0.069).CONCLUSION: Daily TRE before taking medicine had no significant impact on the patients' compliance, satisfaction, or H. pylori eradication, but reduced the rate of adverse events.Chun-Hua Wang Sheng-Tao Liao Jun Yang Chun-Xia Li Ying-Ying Yang Ran Han Dong-Feng Chen Chun-Hui Lan 2015World Journal of Gastroenterology2015,21,39:15
14Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 15Role of gut microbiota via the gut-liver-brain axis in digestive diseases显示文摘The gut-brain axis is a bidirectional information interaction system between the central nervous system(CNS) and the gastrointestinal tract, in which gut microbiota plays a key role. The gut microbiota forms a complex network with the enteric nervous system, the autonomic nervous system, and the neuroendocrine and neuroimmunity of the CNS, which is called the microbiota-gut-brain axis. Due to the close anatomical and functional interaction of the gut-liver axis, the microbiota-gut-liver-brain axis has attracted increased attention in recent years. The microbiota-gut-liver-brain axis mediates the occurrence and development of many diseases, and it offers a direction for the research of disease treatment. 17Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. 19Electroacupuncture inhibits annulus fibrosis cell apoptosis in vivo via TNF-α-TNFR1-caspase-8 and integrin β1/Akt signaling pathways显示文摘OBJECTIVE: To examine whether electroacupuncture(EA) treatment inhibited cell apoptosis of intervertebral annulus fibrosis(AF) via tumor necrosis factor-α(TNF-α)-tumor necrosis factor receptor 1(TNFR1)-caspase-8 and integrin β1/Akt signaling pathways in a rat model of cervical intervertebral disc degeneration caused by unbalanced dynamic and static forces.METHODS: Thirty-two Sprague-Dawley rats were included in this study, of which 24 rats underwent surgery to induce cervical intervertebral disc degeneration, while eight rats received EA treatment at Dazhui(GV 14). Immunohistochemical staining was used to detect TNF-α, TNFR1, and caspase-8Apoptosis of AF cells was examined with terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The m RNA and protein expression levels of integrin β1 andAkt were evaluated with real-time polymerase chain reaction and western blot analysis, respectively.RESULTS: Treatment with EA decreased TUNEL-positive AF cells and lowered TNF-α, TNFR1 and caspase-8 positive cells compared with control groups. EA treatment also increased integrin β1and Akt m RNA and protein levels compared with controls.CONCLUSION: Treatment with EA inhibits AF cell apoptosis through suppression of the TNF-α-TNFR1-caspase-8 signal pathway and increases the expression of integrin β1 and Akt. 20Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
