| 5 | Elimination of a Retrotransposon for Quenching Genome Instability in Modern Rice显示文摘Transposable elements(TEs)constitute the most abundant portions of plant genomes and can dramatically shape host genomes during plant evolution.They also play important roles in crop domestication.However,whether TEs themselves are also selected during crop domestication has remained unknown.Here,we identify an active long terminal repeat(LTR)retrotransposon,HUO,as a potential target of selection during rice domestication and breeding.HUO is a low-copy-number LTR retrotransposon,and is active under natural growth conditions and transmitted through male gametogenesis,preferentially inserting into genomic regions capable of transcription.HUO exists in all wild rice accessions and about half of the archaeological rice grains(1200–7000 years ago)and landraces surveyed,but is absent in almost all modern varieties,indicating its gradual elimination during rice domestication and breeding.Further analyses showed that HUO is subjected to strict gene silencing through the RNA-directed DNA methylation pathway.Our results also suggest that multiple HUO copies may trigger genomic instability through altering genome-wide DNA methylation and small RNA biogenesis and changing global gene expression,resulting in decreased disease resistance and yield,coinciding with its elimination during rice breeding.Together,our study suggests that negative selection of an active retrotransposon might be important for genome stability during crop domestication and breeding. | Yu Peng Yingying Zhang Yijie Gui Dong An Junzhong Liu Xun Xu Qun Li Junmin Wang Wen Wang Chunhai Shi Longjiang Fan Baorong Lu Yiwen Deng Sheng Teng Zuhua He | 2019 | Molecular Plant2019,12,10: | 2 |
| 6 | Assessing the genetic diversity of the critically endangered Chinese sturgeon Acipenser sinensis using mitochondrial markers and genome-wide single-nucleotide polymorphisms from RAD-seq显示文摘As a living fossil, the endangered Chinese sturgeon(Acipenser sinensis) has been considered a national treasure in China. Here,the famous Gezhouba Dam and Three Gorges Dam on the Yangtze River were built in 1988 and 2006, for economic purposes.The natural population of Chinese sturgeon has declined since then, as these dams block its migratory route to the original spawning grounds in the middle reaches of the Yangtze River. In 2013 and 2014, there was an absence of spawning where it typically happened near the Gezhouba Dam. Nevertheless, from April to June in 2015, over 1,000 larvae with different body lengths(10–35 cm) were detected along the Shanghai Yangtze Estuary; but only little is currently known about the population genetic structure of the Chinese sturgeon. Herein, we inferred population genetic parameters from 462 available Chinese sturgeon specimens based on a 421-bp fragment of the mitochondrial DNA(mtDNA) D-loop region and 1,481,620 SNPs(singlenucleotide polymorphisms) generated by restriction site-associated DNA sequencing(RAD-seq). For the D-loop dataset, 15 haplotypes were determined. Randomly picked 23 individuals, representing the 15 D-loop haplotype groups, were subsequently used for further RAD-seq validation. The average nucleotide diversity calculated from the mtDNA and RAD datasets was 0.0086 and 0.000478, respectively. The overall effective female population size was calculated to be 1,255 to 2,607, and the long-term effective population size was estimated to range from 11,950 to 119,500. We observed that the genetic variability and the effective female population size of the current population in the Yangtze River are severely low, which are similar to the data reported over 10 years ago. | 10 | Colon hydrotherapy plus Traditional Chinese Medicine to treat non-alcoholic fatty liver disease:a pilot study显示文摘OBJECTIVE: To demonstrate the potential to treat non-alcoholic fatty liver disease(NAFLD) with colon hydrotherapy(CHT) plus Traditional Chinese Medicine(TCM).METHODS: A total of 20 patients were enrolled into the study and received CHT with TCM for 2 weeks.Body mass index(BMI) and levels of serum triglycerides(TG) and total cholesterol(TC) were compared between pre-treatment and post-treatment.RESULTS: Two-week treatment with CHT plus TCM significantly lowered BMI and reduced blood lipids.BMI decreased from 29.5 ± 4.3 to 25.4 ± 1.0, while mean TG levels decreased by 0.70 mmol/L on average from baseline and mean TC levels decreased by0.37 mmol/L. Forty-five percent of patients exhibited TC decreasing by more than 10% from baseline and25%ofpatientsexhibited TCdecreasingbymore than 20%. Sixty percent of patients exhibited TG decreasing by more than 20% and 20% of patients exhibited TG decreasing by more than 40%. However,high-density and low-density lipoprotein cholester-ol levels did not change significantly after intervention. | 14 | Electrohydrodynamic lithography of metallic mesh for optically transparent flexible and conformal antennas显示文摘The optically transparent antenna is becoming a very attractive proposition for various applications, such as wearable devices and vehicle radars. The fabrication of transparent flexible/conformal antennas is a long-lasting interest in academia and industry.However, the preparation of radio-frequency radiators with excellent conductivity and optical transmittance is still quite challenging. Herein, we introduce a facile approach to directly fabricate optically transparent flexible and conformal coplanar waveguide-fed antennas using programmable electrohydrodynamic lithography. Metallic meshes with transmittance above 90%have been successfully created based on the conformal electrohydrodynamic printing of high-viscosity photoresist masks, and the corresponding sheet resistance can be tuned down to ~2 Ω/□. Then, the geometrical structure of the proposed transparent antenna has been systematically optimized because of the basic radio frequency components, including the radiator, feeder line,ground plane, and size of metallic meshes. Optically transparent flexible and conformal antennas are finally obtained, presenting an optical transmittance of 92% and 55%, respectively. The simulated and measured results demonstrate that the transparent antennas with a good optoelectronic performance indeed exhibit a nice electromagnetic behavior.
