1From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. 2Vulnerability of Water Resources and Its Spatial Heterogeneity in Haihe River Basin, China显示文摘To manage water resources effectively, a multiscale assessment of the vulnerability of water resources on the basis of political boundaries and watersheds is necessary. This study addressed issues on the vulnerability of water resources and provided a multiscale comparison of spatial heterogeneity under a climate change background. Using improved quantitative evaluation methods of vulnerability, the Theil index and the Shannon-Weaver index, we evaluated the vulnerability of water resources and its spatial heterogeneity in the Haihe River Basin in four scales, namely, second-class water resource regions(Class ⅡWRRs), third-class water resource regions(Class Ⅲ WRRs), Province-Class ⅡWRRs, and Province-Class Ⅲ WRRs. Results show that vulnerability enhances from the north to south in the different scales, and shows obvious spatial heterogeneity instead of moving toward convergence in multiscale assessment results. Among the Class Ⅱ WRRs, the Tuhai-Majia River is the most vulnerable area, and the vulnerability of the Luanhe River is lower than that of the north of the Haihe River Basin, which in turn is lower than that of the south of the Haihe River Basin. In the scales of Class Ⅲ WRRs and Province-Class Ⅲ WRRs, the vulnerability shows obvious spatial heterogeneity and diversity measured by the Theil index and the Shannon-Weaver index. 4Characterization of VOC emissions from construction machinery and river ships in the Pearl River Delta of China显示文摘Speciated characterization of Volatile Organic Compounds (VOCs),including oxygenated VOCs (OVOCs),from construction machinery and river ships in China is currently lacking.In this regard,we conducted field measurement on speciated VOC (including OVOC) emissions from six construction machinery and five river ships in the Pearl River Delta (PRD) region to identify VOC emission characteristics.We noticed that OVOC emissions from construction machinery and ships accounted for more than 50%of the total VOC emissions,followed by alkenes,aromatics and alkanes.Formaldehyde and acetaldehyde were the most emission species,accounting for 61.8%-83.2%of OVOCs.For construction machinery,the fuel-based emission factors of roller,grader and pile driver were 3.12,3.12 and 7.36 g/kg,respectively.With the rigorous restraint by the national emission standards,VOC emissions of construction machinery had decreased considerably,especially during stageⅢ.Ozone formation potential was also significantly reduced due to the significant decrease in emissions of OVOCs and alkenes with higher reactivity.For river ships,the fuel-based emission factors of cargo ships and speedboat were 1.46 and 0.44 g/kg,respectively.VOC emissions from construction machinery and river ships in Guangdong Province in 2017 were 8851.0 and 4361.0 ton,respectively.This study filled the knowledge gaps of reactive gas emissions from different kinds of non-road mobile sources over the PRD,and more importantly,highlighted the necessity in adding OVOC measurement to give a complete and accurate depiction of reactive gas emissions from non-road mobile sources.Richao Wang Zibing Yuan Junyu Zheng Cheng Li Zhijiong Huang Wenshi Li Yan Xie Yiran Wang Kaiyang Yu Lejun Duan 2020Journal of Environmental Sciences2020,32,10:5
5Metamorphic, deformation, fluids and geological significance of low-temperature retrograde mylonites of Diancangshan metamorphic massif along Ailaoshan-Red River strike-slip fault zone, Yunnan, China显示文摘Diancangshan metamorphic massif is one of the four metamorphic massifs developed along the Ailaoshan-Red River strike-slip fault zone, Yunnan, China. It has experienced multi-stage metamorphism and deformation, especially since the late Oligocene it widely suffered high-temperature ductile shear deformation and exhumation of the metamorphic rocks from the deep crust to the shallow surface. Based on the previous research and geological field work, this paper presents a detailed study on deformation and metamorphism, and exhumation of deep metamorphic rocks within the Diancangshan metamorphic massif,especially focusing on the low-temperature overprinted retrogression metamorphism and deformation of mylonitic rocks. With the combinated experimental techniques of optical microscope, electron backscatter diffraction attachmented on field-emission scanning electron microscopy and cathodoluminescence, our contribution reports the microstructure, lattice preferred orientations of the deformed minerals, and the changes of mineral composition phases of the superposition low-temperature retrograde mylonites. All these results indicate that:(1) Diancangshan deep metamorphic rock has experienced early high-temperature leftlateral shear deformation and late extension with rapid exhumation, the low-temperature retrogression metamorphism and deformation overprinted the high-temperature metamorphism, and the high-temperature microstructure and texture are in part or entirely altered by subsequent low-temperature shearing;(2) the superposition of low-temperature deformation-metamorphism occurs at the ductile-brittle transition; and(3) the fluid is quite active during the syn-tectonic shearing overprinted lowtemperature deformation and metamorphism. The dynamic recrystallization and/or fractures to micro-fractures result in the strongly fine-grained of the main minerals, and present strain localization in micro-domians, such as micro-shear zones in the mylonites. 6Neuroligins Differentially Mediate Subtype-Specific Synapse Formation in Pyramidal Neurons and Interneurons显示文摘Neuroligins(NLs) are postsynaptic cell-adhesion proteins that play important roles in synapse formation and the excitatory-inhibitory balance. They have been associated with autism in both human genetic and animal model studies, and affect synaptic connections and synaptic plasticity in several brain regions. Yet current research mainly focuses on pyramidal neurons, while the function of NLs in interneurons remains to be understood. To explore the functional difference among NLs in the subtypespecific synapse formation of both pyramidal neurons and interneurons, we performed viral-mediated shRNA knockdown of NLs in cultured rat cortical neurons and examined the synapses in the two major types of neurons. Our results showed that in both types of neurons, NL1 and NL3 were involved in excitatory synapse formation, and NL2 in GABAergic synapse formation. Interestingly, NL1 affectedGABAergic synapse formation more specifically than NL3,and NL2 affected excitatory synapse density preferentially in pyramidal neurons. 7Numerical Simulation of Water and Sand Blowouts When Penetrating Through Shallow Water Flow Formations in Deep Water Drilling显示文摘In this study, we applied a two-phase flow model to simulate water and sand blowout processes when penetrating shallow water flow(SWF) formations during deepwater drilling. We define ‘sand' as a pseudo-component with high density and viscosity, which can begin to flow with water when a critical pressure difference is attained. We calculated the water and sand blowout rates and analyzed the influencing factors from them, including overpressure of the SWF formation, as well as its zone size, porosity and permeability, and drilling speed(penetration rate). The obtained data can be used for the quantitative assessment of the potential severity of SWF hazards. The results indicate that overpressure of the SWF formation and its zone size have significant effects on SWF blowout. A 10% increase in the SWF formation overpressure can result in a more than 90% increase in the cumulative water blowout and a 150% increase in the sand blowout when a typical SWF sediment is drilled. Along with the conventional methods of well flow and pressure control, chemical plugging, and the application of multi-layer casing, water and sand blowouts can be effectively reduced by increasing the penetration rate. 9Efficiency of cotton bollworm (Helicoverpa armigera Hübner) control of different Bt cotton varieties in North China显示文摘Background: The cotton bollworm(Helicoverpa armigera) is one of cotton's most destructive insect pests in terms of yield and quality. Since 1997, China has grown commercially available transgenic Bacillus thuringiensis(Bt) cotton. We aimed to investigate the variation in resistance of transgenic Bt cotton varieties to cotton bollworm in North China.Methods: Populations of cotton bollworm were monitored from 2008 to 2015 in environments where Bt cotton was planted adjacent to other non-Bt crops. The study included 197 Bt cotton varieties planted in 42 counties/locations in three provinces(Hebei, Shandong and Henan) of North China, which were evaluated through field investigations,bioassays, and enzyme-linked immunosorbent assays(ELISA).Results: The average number of cotton bollworms never exceeded the action threshold(10 larvae per 100 cotton plants), however, their number reached 19.55 per 100 cotton plants in 2011. The ratios of damaged plants to total Bt cotton stem-tips, buds, and bolls was low except in 2010, when the ratios reached 1.82%, 2.09%, and 10.63%,respectively. The results of bioassay showed that the corrected mortality were higher at the second generation cotton bollworm stage than the third and fourth germination stages. 10Development and validation of a machine learning-based nomogram for prediction of intrahepatic cholangiocarcinoma in patients with intrahepatic lithiasis显示文摘Background:Accurate diagnosis of intrahepatic cholangiocarcinoma(ICC)caused by intrahepatic lithiasis(IHL)is crucial for timely and effective surgical intervention.The aim of the present study was to develop a nomogram to identify ICC associated with IHL(IHL-ICC).Methods:The study included 2,269 patients with IHL,who received pathological diagnosis after hepatectomy or diagnostic biopsy.Machine learning algorithms including Lasso regression and random forest were used to identify important features out of the available features.Univariate and multivariate logistic regression analyses were used to reconfirm the features and develop the nomogram.The nomogram was externally validated in two independent cohorts.Results:The seven potential predictors were revealed for IHL-ICC,including age,abdominal pain,vomiting,comprehensive radiological diagnosis,alkaline phosphatase(ALK),carcinoembryonic antigen(CEA),and cancer antigen(CA)19-9.The optimal cutoff value was 2.05μg/L for serum CEA and 133.65 U/mL for serum CA 19-9.The accuracy of the nomogram in predicting ICC was 82.6%.The area under the curve(AUC)of nomogram in training cohort was 0.867.The AUC for the validation set was 0.881 from The Second Affiliated Hospital of Wenzhou Medical University,and 0.938 from The First Affiliated Hospital of Fujian Medical University.Conclusions:The nomogram holds promise as a novel and accurate tool to predict IHL-ICC,which can identify lesions in IHL in time for hepatectomy or avoid unnecessary surgical resection.Xian Shen Huanhu Zhao Xing Jin Junyu Chen Zhengping Yu Kuvaneshan Ramen Xiangwu Zheng Xiuling Wu Yunfeng Shan Jianling Bai Qiyu Zhang Qiqiang Zeng 2021Hepatobiliary Surgery and Nutrition2021,10,6:1

13Distal axis sulfide mineralization on the ultraslow-spreading Southwest Indian Ridge:an LA-ICP-MS study of pyrite from the East Longjing-2 hydrothermal field显示文摘The newly discovered East Longjing-2 hydrothermal field(ELHF-2)is located on the Dragon Horn oceanic core complex of the ultraslow-spreading Southwest Indian Ridge,approximately 12 km from the ridge axis.This study measured the chemical compositions of pyrite from ELHF-2 using a laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS)to investigate the genesis of the field.Three generations of pyrite were classified,and found that:Py1 and Py2,rich in V,Mn,U,and Se,occur in altered basalt debris and the silica alteration matrix,respectively.Py3 was mainly intergrown with chalcopyrite in quartz veins and had higher Cu,In,Ag,Sb,and Au contents than Py1 and Py2.Some elements,such as Au,Se,and Pb,are likely presented as direct substitution with Fe^(2+)in pyrite,while Cu,Zn,Co,Ni,and Ag probably occur both as direct substitution with Fe and as distributed micro-to nanoparticle-sized sulfides.Meanwhile,the occurrence of V,Mn,and U is likely presented as oxide inclusions.Trace element geochemistry suggested that the pyrite was formed under hightemperature conditions,and the ore forming elements were likely derived from ultramafic rocks.In addition,Py1and Py2 were formed under higher water/rock ratio and higher temperature conditions,with more seawater involvement compared with Py3.The formation of ELHF-2 was probably driven by exothermic serpentinization reactions with an additional magmatic heat.This study shows that high-temperature hydrothermal circulation driven by magmatic activity can be developed on distal rift flank areas of magma-starved ultraslow-spreading ridges.Shili Liao Chuanwei Zhu Jianping Zhou Weiyong Liu Junyu Yu Jin Liang Weifang Yang Wei Li Jia Liu Chunhui Tao 2021Acta Oceanologica Sinica2021,40,5:1
14Wear resistance performance of high entropy alloy–ceramic coating composites synthesized via a novel combined process显示文摘Titanium nitride(TiN), characterized by its high hardness and strength, was widely used as ceramic coating to improve the wear resistance of matrix materials. In this work, AlCrFeNiTi_(x) high-entropy alloy(HEA) powders were synthesized by direct electrochemical reduction in molten salt from the mixed metal oxides. Then,TiN ceramic coating on the AlCrFeNiTi_x bulk HEA containing the topologically close-packed(TCP) phase(σphase, Laves phase, and Ti_(3)Al phase) was prepared by vacuum hot pressing sintering, where nitride element come from boron nitride parting agent sprayed on the graphite mold. The effect of titanium content on the crystal structure, microstructure, hardness, and wear resistance of the products were investigated by X-ray diffraction, field emission scanning electron microscope, field emission electron-probe microanalysis,Vickers hardness tester, and friction–abrasion testing machine. The bulk HEAs exhibit excellent hardness and its hardness increases significantly with the increase of titanium content. The wear mechanism changes from both of predominantly delamination and accompanied oxidative wear to single delamination wear,which is due to ultra-high melting point and high hot hardness of TiN, that can effectively prevent the oxidation and deformation of the worn surface. 15Impact of Accumulated Temperature on Wetland Vegetation Area in Poyang Lake显示文摘Accumulated temperature,which is now widely used in agronomy,is an important ecological factor to the growth of plants,but few relative studies have been found on the vegetation area of floodplain grasslands in Poyang Lake.This research used the classification and regression tree(CART)to classify normalized vegetation area index derived from MODIS LAI(Moderate Resolution Imaging Spectroradiometer Leaf Area Index)images from 2008 to 2014,according to different climate indexes,such as mean daily air temperature(n),accumulated temperature(jw),daily maximum temperature(g),daily minimum temperature(d),accumulative precipitation(j),water level(s)and average water level for 20 days preceding(a).The results showed that:(1)The accumulated temperature and the 20-day average WL(water level)were found to have the greatest impact on variation in wetland vegetation area,and they both dominated the classification process twice;(2)Two classification thresholds of accumulated temperature were 790°C and 1784°C,approximately corresponding to the beginning of April and midMay;(3)790°C could also be used as a threshold to select remote sensing images to analysis the annual variability of vegetation,i.e.while accumulated temperature is lower than 790°C,remote sensing images of similar accumulated temperature rather than similar date should be selected from different years for comparison.We also found that,effects of different hydrological factors on area of floodplain grasslands showed stage characteristics:(1)From January to March,water level changes slowly with less rainfall,as a result,the 20-day average WL which can interpret the hydrologic characteristics smoothly showed significant importance in this stage;(2)While entering April,intense rainfall make accumulative precipitation to be the dominating factor of classification;(3)From late April to mid-May,in condition of accumulative precipitation higher than 405 mm,daily water level is of most importance,because to the flood recession process as well as rapid water level fluctuations.Xin Yao Junyu Zhu Hong Zeng Wenzheng Yu Hanxiaoya Zhang 2022Computers, Materials & Continua2022,,10:0
16Functional characterization of novel NPRL3 mutations identified in three families with focal epilepsy显示文摘Focal epilepsy accounts for 60% of all forms of epilepsy, but the pathogenic mechanism is not well understood. In this study,three novel mutations in NPRL3(nitrogen permease regulator-like 3), c.937_945del, c.1514dup C and 6,706-bp genomic DNA(g DNA) deletion, were identified in three families with focal epilepsy by linkage analysis, whole exome sequencing(WES) and Sanger sequencing. NPRL3 protein is a component of the GATOR1 complex, a major inhibitor of m TOR signaling. These mutations led to truncation of the NPRL3 protein and hampered the binding between NPRL3 and DEPDC5, which is another component of the GATOR1 complex. Consequently, the mutant proteins enhanced m TOR signaling in cultured cells, possibly due to impaired inhibition of m TORC1 by GATOR1. Knockdown of nprl3 in Drosophila resulted in epilepsy-like behavior and abnormal synaptic development. 17VOC emission caps constrained by air quality targets based on response surface model:A case study in the Pearl River Delta Region,China显示文摘Because of the recent growth in ground-level ozone and increased emission of volatile organic compounds(VOCs),VOC emission control has become a major concern in China.In response,emission caps to control VOC have been stipulated in recent policies,but few of them were constrained by the co-control target of PM_(2.5)and ozone,and discussed the factor that influence the emission cap formulation.Herein,we proposed a framework for quantification of VOC emission caps constrained by targets for PM_(2.5)and ozone via a new response surface modeling(RSM)technique,achieving 50%computational cost savings of the quantification.In the Pearl River Delta(PRD)region,the VOC emission caps constrained by air quality targets varied greatly with the NOxemission reduction level.If control measures in the surrounding areas of the PRD region were not considered,there could be two feasible strategies for VOC emission caps to meet air quality targets(160μg/m^(3)for the maximum 8-hr-average 90th-percentile(MDA8-90%)ozone and 25μg/m^(3)for the annual average of PM_(2.5)):a moderate VOC emission cap with<20%NOxemission reductions or a notable VOC emission cap with>60%NOxemission reductions.If the ozone concentration target were reduced to 155μg/m^(3),deep NOxemission reductions is the only feasible ozone control measure in PRD.Optimization of seasonal VOC emission caps based on the Monte Carlo simulation could allow us to gain higher ozone benefits or greater VOC emission reductions.If VOC emissions were further reduced in autumn,MDA8-90%ozone could be lowered by 0.3-1.5μg/m^(3),equaling the ozone benefits of 10%VOC emission reduction measures.The method for VOC emission cap quantification and optimization proposed in this study could provide scientific guidance for coordinated control of regional PM_(2.5)and O_(3)pollution in China.Ya’nan Hu Bowen Shi Xin Yuan Chuanzeng Zheng Qing’e Sha Yufan Yu Zhijiong Huang Junyu Zheng 2023Journal of Environmental Sciences2023,,1:0
18Van der Waals epitaxial growth and optoelectronics of a vertical MoS_(2)/WSe_(2)p-n junction显示文摘Two-dimensional(2D)transition metal dichalcogenides(TMDs)have attracted extensive attention due to their unique electronic and optical properties.In particular,TMDs can be fexibly combined to form diverse vertical van der Waals(vdWs)heterostructures without the limitation of lattice matching,which creates vast opportunities for fundamental investigation of novel optoelectronic applications.Here,we report an atomically thin vertical p-n junction WSe_(2)/MoS_(2)produced by a chemical vapor deposition method.Transmission electron microscopy and steady-state photoluminescence experiments reveal its high quality and excellent optical properties.Back gate feld efect transistor(FET)constructed using this p-n junction exhibits bipolar behaviors and a mobility of 9 cm^(2)/(V·s).In addition,the photodetector based on MoS_(2)/WSe_(2)heterostructures displays outstanding optoelectronic properties(R=8 A/W,D^(*)=2.93×10^(11)Jones,on/of ratio of 10^(4)),which benefted from the built-in electric feld across the interface.The direct growth of TMDs p-n vertical heterostructures may ofer a novel platform for future optoelectronic applications.Yu Xiao Junyu Qu Ziyu Luo Ying Chen Xin Yang Danliang Zhang Honglai Li Biyuan Zheng Jiali Yi Rong Wu Wenxia You Bo Liu Shula Chen Anlian Pan 2022Frontiers of Optoelectronics2022,15,4:0

20Nuclear translocation of OsMADS25 facilitated by OsNAR2.1 in reponse to nitrate signals promotes rice root growth by targeting OsMADS27 and OsARF7显示文摘Nitrate is an important nitrogen source and signaling molecule that regulates plant growth and development.Although several components of the nitrate signaling pathway have been identified,the detailed mechanisms are still unclear.Our previous results showed that OsMADS25 can regulate root development in response to nitrate signals,but the mechanism is still unknown.Here,we try to answer two key questions:how does OsMADS25 move from the cytoplasm to the nucleus,and what are the direct target genes activated by OsMADS25 to regulate root growth after it moves to the nucleus in response to nitrate?Our results demonstrated that OsMADS25 moves from the cytoplasm to the nucleus in the presence of nitrate in an OsNAR2.1-dependentmanner.Chromatin immunoprecipitation sequencing,chromatin immunoprecipitation qPCR,yeast one-hybrid,and luciferase experiments showed that OsMADS25 directly activates the expression of OsMADS27 and OsARF7,which are reported to be associated with root growth.Finally,OsMADS25-RNAi lines,the Osnar2.1 mutant,and OsMADS25-RNAi Osnar2.1 lines exhibited significantly reduced root growth compared with the wild type in response to nitrate supply,and expression of OsMADS27 and OsARF7 was significantly suppressed in these lines.Collectively,these results reveal a new mechanismby which OsMADS25 interacts with OsNAR2.1.This interaction is required for nuclear accumulation of OsMADS25,which promotes OsMADS27 and OsARF7 expression and root growth in a nitratedependent manner.Junyu Wu Shuaiqi Yang Nana Chen Qining Jiang Linli Huang Jiaxuan Qi Guohua Xu Lisha Shen Hao Yu Xiaorong Fan Yinbo Gan 2023Plant Communications2023,4,6:0
