9Metabolomic and transcriptomic study to understand changes in metabolic and immune responses in steers under heat stress显示文摘Heat stress(HS)damages livestock by adversely affecting physiological and immunological functions.However,fundamental understanding of the metabolic and immunological mechanisms in animals under HS remains elusive,particularly in steers.To understand the changes on metabolic and immune responses in steers under HS condition,we performed RNA-sequencing and proton nuclear magnetic resonance spectroscopy-based metabolomics on HS-free(temperature humidity index[THI]value:64.92±0.56)and HS-exposed(THI value:79.13±0.56)Jersey steer(n=8,body weight:559.67±32.72 kg).This study clarifies the metabolic changes in 3 biofluids(rumen fluid,serum,and urine)and the immune responses observed in the peripheral blood mononuclear cells of HS-exposed steers.This integrated approach allowed the discovery of HS-sensitive metabolic and immunological pathways.The metabolomic analysis indicated that HS-exposed steers showed potential HS biomarkers such as isocitrate,formate,creatine,and riboflavin(P<0.05).Among them,there were several integrative metabolic pathways between rumen fluid and serum.Furthermore,HS altered mRNA expression and immune-related signaling pathways.A meta-analysis revealed that HS decreased riboflavin metabolism and the expression of glyoxylate and dicarboxylate metabolism-related genes.Moreover,metabolic pathways,such as the hypoxia-inducible factor-1 signaling pathway,were downregulated in immune cells by HS(P<0.05).These findings,along with the datasets of pathways and phenotypic differences as potential biomarkers in steers,can support more in-depth research to elucidate the interrelated metabolic and immunological pathways.This would help suggest new strategies to ameliorate the effects of HS,including disease susceptibility and metabolic disorders,in Jersey steers.Jun Sik Eom Da Som Park Sang Jin Lee Bon-Hee Gu Shin Ja Lee Sang-Suk Lee Seon-Ho Kim Byeong-Woo Kim Sung Sill Lee Myunghoo Kim 2022Animal Nutrition2022,,4:1

17Light-stabilized FHA2 suppresses miRNA biogenesis through interactions with DCL1 and HYL1显示文摘The precise regulation of microRNA(miRNA)biogenesis is crucial for plant development,which requires core microprocessors and many fine tuners to coordinate their miRNA processing activity/specificity in fluctuating cellular environments.During de-etiolation,light triggers a dramatic accumulation of core microprocessors and primary miRNAs(pri-miRNAs)but decreases pri-miRNA processing activity,resulting in relatively constant miRNA levels.The mechanisms underlying these seemingly contradictory regulatory changes remain unclear.In this study,we identified forkhead-associated domain 2(FHA2)as a light-stabilized suppressor of miRNA biogenesis.We found that FHA2 deficiency increased the level of mature miRNAs,accompanied by a reduction in pri-miRNAs and target mRNAs.Biochemical assays showed that FHA2 associates with the core microprocessors DCL1,HYL1,and SE,forming a complex to suppress their pri-miRNA processing activity.Further analyses revealed that FHA2 promotes HYL1 binding but inhibits the binding of DCL1-PAZ-RNase-RNA-binding domains(DCL1-PRR)to miRNAs,whereas FHA2 does not directly bind to these RNAs.Interestingly,we found that FHA2 protein is unstable in the dark but stabilized by light during de-etiolation.Consistently,disruption of FH A led to defects in light-triggered changes in miRNA expression and reduced the survival rate of deetiolated seedlings after prolonged light deprivation.Collectively,these data suggest that FHA2 is a novel light-stabilized suppressor of miRNA biogenesis and plays a role in fine-tuning miRNA processing during de-etiolation.Seung Jun Park Suk Won Choi Gu Min Kim Christian Moller Hyun-Sook Pai Seong Wook Yang 2021Molecular Plant2021,14,4:1

20在初始PSA和Gleason评分较高的前列腺癌病人中前列腺活检术后分期MRI对T3疾病的预测力会降低显示文摘为提高预测囊外扩展(ECE)和精囊浸润(SVI)的准确性,我们评估了影响MRI(磁共振成像)预测的变量,并估计了这些变量对术前MRI分期和病理结果之间准确性的影响。121名行机器手前列腺癌根治术(RALP)的局限性或局限性晚期前列腺癌病人,经直肠活检后,所有入选病人都接受了MRI的检查进行分期诊断。经过RALP后,只有43.8%(53/121)的病人与MRI的预测分期相匹配。与在ECE预测中的匹配组相比,非匹配组的初始前列腺特异抗原(PSA)明显高于匹配组(非匹配组12.8ng mL-1,匹配组8.1ng mL-1,P=0.048).在对SVI的预测中,非匹配组中,初始PSA8.1w.17.3ng mL-1,P=0.009和穿刺活检Gleason评分6.5w.7.6,P=-0.035均显著高于匹配组。如果把10ng mL-1和20ng mL-1的PSA水平作为临床临界值,预测ECE时,对于PSA水平高于20ng mL-1的病人,MRJ准确性会下降(75.6%,64.5%,37.5%,P=0.01);对这组病人预测SVI时,MRI的准确性也会明显下降(91.5%,77.4%,37.5%,P〈0.01)。如果把Gleason评分为7分作为临床临界值,对Gleason评分高于7分的病人,预测SvI时MRI的准确性也会显著下降(93.9%,82.1%,62.9%,P=0.01)。因此,对于这些患者,为了获得前列腺癌根治术过程中的切缘阴性,能提供大量信息的是手术发现而不是活检后的MRI图像,暗示在前列腺活检以前进行MRI检查在临床上更有优势。Young Hwii Ko Deuk Jae Sung Sung Gu Kang Seok Ho Kang Jeong Gu Lee Je Jong Kim Jun Cheon 2011Asian Journal of Andrology2011,13,3:0
