1新城疫病毒某水禽分离株经鸡体传代后由非致病型转变为速发型的研究显示文摘最近 ,一株无毒力的新城疫病毒 (NDV)在鸡体内繁殖时 ,变成了强毒株。但至今尚未能证实 ,经鸡体传代的野生水禽新城疫病毒是否也具有变成速发型毒株的能力。为了通过实验证明从水禽中分离的非致病型NDV可以转变为速发型病毒 ,我们通过在鸡体内传播鹅源性无毒力株 ,经气囊接种连续传代 9次 ,随后再在鸡脑内传代 5次。结果显示 ,该病毒的毒力变得很强 ,致死率可达 1 0 0 %。通过致病性试验证实 ,其具有典型的速发型病毒特征 ;融合蛋白裂解位点的序列分析表明 ,原始的分离株含有无毒力型毒株共有的裂解序列 :E_R_Q_E_R/L,而通过鸡体反复传代后 ,该序列变为致病性毒株共有的序列 :K_R_Q_K_R/F。这些结果表明 ,野生水禽中自然存在的无毒力毒株 ,具有无毒力株相应的序列 ,但当其在鸡群中传播时 ,则具有变成高致病性病毒的能力。同时研究表明 ,鸡体提供了该病毒从非致病型向致病型转变的选择机制。于圣青 丁铲 NORIKO KISHIDA HIROSHI ITO HIROSHI KIDA KOICHI OTSUKI YOSHIHIRO KAWAOKA TOSHIHIRO ITO 2003中国预防兽医学报2003,25,1:32

5Targeting tight junctions during epithelial to mesenchymal transition in human pancreatic cancer显示文摘Pancreatic cancer continues to be a leading cause of cancer-related death worldwide and there is an urgent need to develop novel diagnostic and therapeutic strategies to reduce the mortality of patients with this disease. In pancreatic cancer, some tight junction proteins, including claudins, are abnormally regulated and therefore are promising molecular targets for diagnosis, prognosis and therapy. Claudin-4 and-18 are overexpressed in human pancreatic cancer and its precursor lesions. Claudin-4 is a high affinity receptor of Clostridium perfringens enterotoxin(CPE). The cytotoxic effects of CPE and monoclonal antibodies against claudin-4 are useful as novel therapeutic tools for pancreatic cancer. Claudin-18 could be a putative marker and therapeutic target with prognostic implications for patients with pancreatic cancer. Claudin-1,-7, tricellulin and marvelD3 are involved in epithelial to mesenchymal transition(EMT) of pancreatic cancer cells and thus might be useful as biomarkers during disease. Protein kinase C is closely related to EMT of pancreatic cancer and regulates tight junctions of normal human pancreatic duct epithelial cells and the cancer cells. 6Serum Mac-2 binding protein is a novel biomarker for chronic pancreatitis显示文摘AIM: To determine the efficacy of Mac-2 binding protein(Mac-2bp) for diagnosis of chronic pancreatitis.METHODS: Fifty-nine healthy volunteers(HV), 162 patients with chronic pancreatitis(CP), and 94 patients with pancreatic ductal adenocarcinoma(PDAC) were enrolled in this study. We measured serum Mac-2bp using our developed enzyme-linked immunosorbent assay kit. Additional biochemical variables were measured using an automated analyzer(including aminotransferase, alanine aminotransferase, γ-glutamyltransferase, alkaline phosphatase, triglyceride, C-reactive protein, and amylase levels) or chemiluminescent enzyme immunoassay(carbohydrate antigen 19-9 and carcinoembryonic antigen). The ability of Mac-2bp to predict CP diagnosis accurately was assessed using receiver operating characteristic(ROC) analyses.RESULTS: Serum Mac-2bp levels were significantly increased in CP patients compared to HV(P < 0.0001) and PDAC patients(P < 0.0001). Area under the ROC curve values of Mac-2bp for the discrimination of CP from HV and PDAC were 0.727 and 0.784, respectively. Multivariate analyses demonstrated that serum Mac-2bp levels were independent determinants for CP diagnosis from HV and PDAC patients. Immunohistological staining showed that Mac-2bp was expressed faintly in the pancreas tissues of both CP and PDAC patients. Serum aspartate aminotransferase, alanine aminotransferase, γ-glutamyltransferase, alkaline phosphatase, and triglyceride levels were significantly higher in patients with CP or PDAC. Serum Mac-2bp levels were highly correlated with protein levels of alanine aminotransferase, γ-glutamyltransferase, and C-reactive protein, but not amylase, suggesting that the damaged liver produces Mac-2bp. 10mPGES-1 expression in non-cancerous liver tissue impacts on postoperative recurrence of HCC显示文摘AIM:To investigate whether microsomal prostaglandin E synthase-1 (mPGES-1) expression in hepatocellular carcinoma (HCC) and in non-cancerous liver affects HCC prognosis after hepatectomy. METHODS: The relationship between patient clinical prof iles, tumor factors, surgical determinants, and mPGES-1 expression and the recurrence-free survival rate were examined in 64 patients who underwent curative hepatectomy between March 2003 and December 2006. RESULTS: The scores for mPGES-1 expression were higher in well differentiated and moderately differentiated HCC tissues than in poorly differentiated HCC tissues (well differentiated, 5.1 ± 2.7; moderately differentiated, 5.1 ± 1.7; poorly differentiated, 3.0 ± 1.8). In noncancerous liver tissues, the mPGES-1 levels were higher in injured liver tissues than in normal tissues. Cirrhotic livers had higher mPGES-1 levels than livers with chronic hepatitis (normal livers, 3.3 ± 0.7; chronic hepatitic livers, 5.4 ± 1.9; cirrhotic livers, 6.4 ± 1.6). A univariate analysis revealed that the recurrence-free survival rate was signif icantly lower in patients with vascular invasion,a higher mPGES-1 level in non-cancerous liver tissue,a larger tumor diameter (≥5 cm), and a lower serum albumin level (≤3.7 g/dL). The mPGES-1 expression in HCC tissues did not correlate well with postoperative recurrence. 14Kidney GATA3^(+) regulatory T cells play roles in the convalescence stage after antibody-mediated renal injury显示文摘FoxP3^(+)regulatory T cells(Tregs)play crucial roles in peripheral immune tolerance.In addition,Tregs that reside or accumulate in nonlymphoid tissues,called tissue Tregs,exhibit tissue-specific functions and contribute to the maintenance of tissue homeostasis and repair.In an experimental mouse model of crescentic glomerulonephritis induced by an anti-glomerular basement membrane antibody,Tregs started to accumulate in the kidney on day 10 of disease onset and remained at high levels(~30–35%of CD4^(+)T cells)during the late stage(days 21–90),which correlated with stable disease control.Treg depletion on day 21 resulted in the relapse of renal dysfunction and an increase in Th1 cells,suggesting that Tregs are essential for disease control during the convalescence stage.The Tregs that accumulated in the kidney showed tissue Treg phenotypes,including high expression of GATA3,ST2(the IL33 receptor subunit),amphiregulin(Areg),and PPARγ.Although T-bet^(+) Tregs and RORγt^(+) Tregs were observed in the kidney,GATA3^(+) Tregs were predominant during the convalescence stage,and a PPARγagonist enhanced the accumulation of GATA3^(+) Tregs in the kidney.To understand the function of specific genes in kidney Tregs,we developed a novel T cell transfer system to T cell-deficient mice.This experiment demonstrates that ST2,Areg,and CCR4 in Tregs play important roles in the accumulation of GATA3^(+) Tregs in the kidney and in the amelioration of renal injury.Our data suggest that GATA3 is important for the recruitment of Tregs into the kidney,which is necessary for convalescence after renal tissue destruction.Ryota Sakai Minako Ito Kyoko Komai Mana Iizuka-Koga Kazuhiko Matsuo Takashi Nakayama Osamu Yoshie Koichi Amano Hiroshi Nishimasu Osamu Nureki Masato Kubo Akihiko Yoshimura 2021Cellular & Molecular Immunology2021,18,5:2
