1Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control.Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu 2021Science China(Life Sciences)2021,64,5:46
2Novel and potent inhibitors targeting DHODH are broad-spectrum antivirals against RNA viruses including newly-emerged coronavirus SARS-CoV-2显示文摘Emerging and re-emerging RNA viruses occasionally cause epidemics and pandemics worldwide,such as the on-going outbreak of the novel coronavirus SARS-CoV-2.Herein,we identified two potent inhibitors of human DHODH,S312 and S416,with favorable drug-likeness and pharmacokinetic profiles,which all showed broad-spectrum antiviral effects against various RNA viruses,including influenza A virus,Zika virus,Ebola virus,and particularly against SARS-CoV-2.Notably,S416 is reported to be the most potent inhibitor so far with an EC5o of 17 nmol/L and an SI value of 10,505.88 in infec-ted cells.Our results are the first to validate that DHODH is an attractive host target through high antiviral efficacy in vivo and low virus replication in DHODH knock-out cells.This work demonstrates that both S312/S416 and old drugs(Leflunomide/Teriflunomide)with dual actions of antiviral and immuno-regulation may have clinical potentials to cure SARS-CoV-2 or other RNA viruses circulating worldwide,no matter such viruses are mutated or not.Rui Xiong Leike Zhang Shiliang Li Yuan Sun Minyi Ding Yong Wang Yongliang Zhao Yan Wu Weijuan Shang Xiaming Jiang Jiwei Shan Zihao Shen Yi Tong Liuxin Xu Yu Chen Yingle Liu Gang Zou Dimitri Lavillete Zhenjiang Zhao Rui Wang Lili Zhu Gengfu Xiao Ke Lan Honglin Li Ke Xu 2020Protein & Cell2020,11,10:14
3Functional analysis of the larval serum protein gene promoter from silkworm,Bombyx mori.显示文摘The regulation region of larval serum protein gene, Bombyx mori. (BmLSP), consisting of the first intron, the first exon, the central promoter region and 5′-upstream region, is cloned from genomic DNA from the silkworm va-riety of Suju譓inghu. Using PCR and restriction endonu-clease methods, a series of luciferase reporter plasmids, driven by different length of BmLSP promoters, are con-structed. Via the transient expression system in BmN cells, the effects of the regulation elements and foreign insect hor-mones on the BmLSP promoter activity are investigated. The results demonstrate that the promoter activity of BmLSP is 5.8- or 4.4-fold higher than that of BmLSPs whose first in-tron or the element in 5′-upstream region harboring the homologous sequence with the first intron of light-chain fib-roin gene (EHIF) is deleted, respectively, suggesting that both the first intron and EHIF contain the main positive cis-acting elements. However, the inactive mariner transposable ele-ment (MTE) in 5′-upstream region presents a negative effect. Furthermore, the effects of juvenile hormone analogue (JHA) on the BmLSP promoter activity show a typical dose-dependent manner, that is, low concentration treat-ments increase the BmLSP promoter activity and high con-centration treatments decrease it. 4Three-dimensional bioprinting of multicell-laden scaffolds containing bone morphogenic protein-4 for promoting M2 macrophage polarization and accelerating bone defect repair in diabetes mellitus显示文摘Critical-sized bone defect repair in patients with diabetes mellitus remains a challenge in clinical treatment because of dysfunction of macrophage polarization and the inflammatory microenvironment in the bone defect region.Three-dimensional(3D)bioprinted scaffolds loaded with live cells and bioactive factors can improve cell viability and the inflammatory microenvironment and further accelerating bone repair.Here,we used modified bioinks comprising gelatin,gelatin methacryloyl(GelMA),and 4-arm poly(ethylene glycol)acrylate(PEG)to fabricate 3D bioprinted scaffolds containing BMSCs,RAW264.7 macrophages,and BMP-4-loaded mesoporous silica nanoparticles(MSNs).Addition of MSNs effectively improved the mechanical strength of GelMA/gelatin/PEG scaffolds.Moreover,MSNs sustainably released BMP-4 for long-term effectiveness.In 3D bioprinted scaffolds,BMP-4 promoted the polarization of RAW264.7 to M2 macrophages,which secrete anti-inflammatory factors and thereby reduce the levels of pro-inflammatory factors.BMP-4 released from MSNs and BMP-2 secreted from M2 macrophages collectively stimulated the osteogenic differentiation of BMSCs in the 3D bioprinted scaffolds.Furthermore,in calvarial critical-size defect models of diabetic rats,3D bioprinted scaffolds loaded with MSNs/BMP-4 induced M2 macrophage polarization and improved the inflammatory microenvironment.And 3D bioprinted scaffolds with MSNs/BMP-4,BMSCs,and RAW264.7 cells significantly accelerated bone repair.In conclusion,our results indicated that implanting 3D bioprinted scaffolds containing MSNs/BMP-4,BMSCs,and RAW264.7 cells in bone defects may be an effective method for improving diabetic bone repair,owing to the direct effects of BMP-4 on promoting osteogenesis of BMSCs and regulating M2 type macrophage polarization to improve the inflammatory microenvironment and secrete BMP-2.Xin Sun Zhenjiang Ma Xue Zhao Wenjie Jin Chenyu Zhang Jie Ma Lei Qiang Wenhao Wang Qian Deng Han Yang Jinzhong Zhao Qianqian Liang Xiaojun Zhou Tao Li Jinwu Wang 2021Bioactive Materials2021,6,3:12
5Spatial and Seasonal Patterns of Ichthyoplankton Assemblages in the Haizhou Bay and Its Adjacent Waters of China显示文摘Surveys were conducted in five voyages in Haizhou Bay and its adjacent coastal area from March to December 2011 during full moon spring tides. The ichthyoplankton assemblages and the environmental factors that affect their spatial and seasonal patterns were determined. Totally 35 and 12 fish egg and larvae taxa were identified, respectively. Over the past several decades, the egg and larval species composition has significantly changed in Haizhou Bay and its adjacent waters, most likely corresponding with the alteration of fishery resources, which are strongly affected by anthropogenic activities and climate change. The Bray-Curtis dissimilarity index identified four assemblages: near-shore bay assemblage, middle bay assemblage and two closely related assemblages(near-shore/middle bay assemblage and middle/edge of bay assemblage). The primary species of each assemblage principally reflected the spawning strategies of adult fish. The near-shore bay assemblage generally occurred in near-shore bay, with depths measuring <20 m, and the middle bay assemblage generally occurred in the middle of bay, with depths measuring 20 to 40 m. Spatial and seasonal variations in ichthyoplankton in each assemblage were determined by interactions between biological behavioral traits and oceanographic features, particularly the variation of local conditions within the constraint of a general reproductive strategy. 6Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10

8A reduced order aerothermodynamic modeling framework for hypersonic vehicles based on surrogate and POD显示文摘Aerothermoelasticity is one of the key technologies for hypersonic vehicles. Accurate and efficient computation of the aerothermodynamics is one of the primary challenges for hypersonic aerothermoelastic analysis. Aimed at solving the shortcomings of engineering calculation, computation fluid dynamics(CFD) and experimental investigation, a reduced order modeling(ROM)framework for aerothermodynamics based on CFD predictions using an enhanced algorithm of fast maximin Latin hypercube design is developed. Both proper orthogonal decomposition(POD) and surrogate are considered and compared to construct ROMs. Two surrogate approaches named Kriging and optimized radial basis function(ORBF) are utilized to construct ROMs. Furthermore,an enhanced algorithm of fast maximin Latin hypercube design is proposed, which proves to be helpful to improve the precisions of ROMs. Test results for the three-dimensional aerothermodynamic over a hypersonic surface indicate that: the ROMs precision based on Kriging is better than that by ORBF, ROMs based on Kriging are marginally more accurate than ROMs based on PODKriging. 10The Epidemiological Characteristics of Pulmonary Tuberculosis——Kashgar Prefecture,Xinjiang Uygur Autonomous Region,China,2011-2020显示文摘Introduction:National Notifiable Disease Reporting System(NNDRS)plays an important role in the early detection and control of tuberculosis(TB)in China.This study analyzed the epidemiological characteristics of pulmonary tuberculosis(PTB)in Kashgar Prefecture,Xinjiang Uygur Autnomous Region,China from 2011 to 2020 to provide a scientific basis for developing TB control strategies and measures in Kashgar.Methods:The data were collected from the NNDRS,which included the geographical distribution,age,sex,occupation,and pathogenic classification of reported PTB cases in 12 counties/cities of Kashgar Prefecture from 2011 to 2020.Descriptive statistics were used to describe the characteristic of PTB epidemic in Kashgar.Results:There were 189,416 PTB cases reported during 2011–2020,with a mean annual PTB case notification rate(CNR)of 451.29/100,000.A rising trend in the rate of reported PTB between 2011 and 2017(χ2 trend=26.09,P<0.01)and a declining trend between 2018 and 2020(χ2 trend=314.44,P<0.01)were observed.The months with the highest reported number of PTB cases were March to May and November to December.The mean annual rate of reported PTB was 451.88/100,000 for males and 450.67/100,000 for females.In addition,19.76%of patients were bacteriologically-confirmed(Bac+)cases(37,425/189,416),and the mean annual Bac+CNR was 89.17/100,000,rising from 64.76/100,000 in 2011 to 139.12/100,000 in 2020(χ^(2)_(trend)=74.44,P<0.01).Conclusions:The CNR of reported PTB in Kashgar showed a significant declining trend in the past three years.Males,elderly population,winter and spring,and farmers as an occupation were the main factors associated with high incidence of tuberculosis in Kashgar.Targeted prevention and treatment of TB should be strengthened in key groups in this region.Diermulati Tusun Maiweilanjiang Abulimiti Xirizhati Mamuti Zhenjiang Liu Da Xu Guanzhen Li Xiaowang Peng Tuerhong Abudureyimu Lijie Zhang Yanlin Zhao Xichao Ou 2021China CDC weekly2021,3,26:6
11Adding-Point Strategy for Reduced-Order Hypersonic Aerothermodynamics Modeling Based on Fuzzy Clustering显示文摘Reduced order models-ROMs) based on the snapshots on the CFD high-fidelity simulations have been paid great attention recently due to their capability of capturing the features of the complex geometries and flow configurations. To improve the efficiency and precision of the ROMs, it is indispensable to add extra sampling points to the initial snapshots, since the number of sampling points to achieve an adequately accurate ROM is generally unknown in prior, but a large number of initial sampling points reduces the parsimony of the ROMs. A fuzzy-clustering-based adding-point strategy is proposed and the fuzzy clustering acts an indicator of the region in which the precision of ROMs is relatively low. The proposed method is applied to construct the ROMs for the benchmark mathematical examples and a numerical example of hypersonic aerothermodynamics prediction for a typical control surface. 12Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats.Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang 2022Science China(Life Sciences)2022,65,10:5
13Boosted electromagnetic wave absorption performance from synergistic induced polarization of SiC_(NWs)@MnO_(2)@PPy heterostructures显示文摘In the last decade,electromagnetic pollution has caused people’s considerable attention.Developing absorbing material with low cost,lightweight,simple preparation,and high electromagnetic attenuation efficiency has become a feasible means to deal with this problem.In this work,core–shell SiC_(NWs)@MnO_(2)@PPy(NWs:nanowires,PPy:polypyrrole)heterostructures composed of SiC nanowires core,MnO_(2)nanosheets inter-layer,and PPy coating were successfully prepared through chemical vapor deposition and two-step electrodeposition process.Taking advantage of the interfacial polarization and dipole polarization,the obtained product displays excellent electromagnetic wave absorption performances with the minimum reflection loss(RLmin)of−50.59 dB when the matching thickness is 2.41 mm,and the optimal effective absorption bandwidth(EAB)value reaches to 6.64 GHz at a matching thickness of 2.46 mm,revealing that the SiC_(NWs)@MnO_(2)@PPy nanocomposite could be served as a promising electromagnetic wave absorbing material.On the basis of systematic analysis concerning the electromagnetic parameters,the dissipation process of the incident electromagnetic wave was demonstrated reasonably,which may provide a referable preparation strategy for novel heterostructures,especially nonmagnetic lightweight absorbing material.Meng Zhang Laibin Zhao Wenxin Zhao Ting Wang Liying Yuan Yuying Guo Yuxin Xie Tingting Cheng Alan Meng Zhenjiang Li 2023Nano Research2023,16,2:4
14Suppress voltage decay of lithium-rich materials by coating layers with different crystalline states显示文摘Li-rich oxides are considered as the most commercial potential cathode materials due to the high theoretical specific discharge capacity. Here, ZrO_(2) in different crystalline states are applied as the coating layers to enhance the electrochemical performance of hollow Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_(2) materials.Meanwhile, a series of characterizations(XRD, SEM, TEM, EDX etc.) are conducted to compare the effects of ZrO_(2) coating layer with different crystalline states on the host material. 15Application of Spatial and Temporal Entropy Based on Multivariate Data for Measuring the Degree of Urban Function Mix显示文摘Quantifying the degree of urban function mix has great signifi cance to both urban planning practices and studies. This paper, fi rst of all, based on the data of points of interest(POIs), analyzes the spatial distribution patterns of urban functions in Beijing. Then according to the taxi GPS data with the origin and destination points(taxi-ODs), we identify the spatial and temporal pattern of the degree of urban function mix in Beijing. Finally, based on the information entropy model of the POIs and taxi-ODs, we establish a spatial entropy and a temporal entropy to quantitatively assess the degree of urban function mix, confirming by virtue of correlation analysis and regression analysis that the spatial entropy of POIs is closely associated with the temporal entropy of taxi-ODs, which proves that it is reasonable to assess the degree of urban function mix via these two entropies. As a result, the degree of urban function mix in Beijing is characterized by slowly decreasing from its core area along the 3rd and 4th Ring Roads to its periphery. In detail, there is the highest degree of urban function mix in Beijing's traditional core areas, including Zhongguancun, CBD(Central Business District), the Old Dongcheng District, the Old Xicheng District, and the area within the 2nd Ring Road; a higher degree in Wangjing area; and a low degree in Tiantongyuan and Huilongguan communities. 163D urchin like V-doped CoP in situ grown on nickel foam as bifunctional electrocatalyst for efficient overall water-splitting显示文摘Cobalt phosphide (CoP) is considered to be a potential candidate in the field of electrocatalysis due to its low-cost, abundant resources and high electrochemical stability. However, there is a great space for further improvement of its electrocatalytic performance since its charge transfer rate and catalytic activity have not reached a satisfactory level. Herein, we design and fabricate a three dimensional urchins like V-doped CoP with different amounts of V-doping on nickel foam electrode. The V-doped CoP/NF electrode with optimized amounts of V-doping (10%) exhibits outstanding hydrogen evolution reaction (HER) performance under universal-pH conditions and preeminent oxygen evolution reaction (OER) performance in alkaline media. Notably, the assembled water-splitting cell displays a cell voltage of only 1.53 V at 10 mA·cm−2 and has excellent durability, much better than many reported related bifunctional catalysts. The experiment results and theoretical analysis revealed that vanadium atoms replace cobalt atoms in CoP lattice. Vanadium doping can not only raise the density of electronic states near the Fermi level enhancing the conductivity of the catalyst, but can also optimize the free energy of hydrogen and oxygen-containing intermediates adsorption over CoP, thus promoting its catalytic activity. Moreover, the unique nanostructure of the catalyst provides the various shortened channels for charge transfer and reactant/electrolyte diffusion, which accelerates the electrocatalytic process. 17Identification of five Pampus species from the coast of China based on sagittal otolith morphology analysis显示文摘Sagittal otoliths morphology were compared to identify five Pampus species of the Chinese coast(Chinese pomfret Pampus chinensis, grey pomfret P. cinereus, southern lesser pomfret P. minor, plumage pomfret P.punctatissmus, and Pampus sp.). Otoliths demonstrated species-specific characteristics with noticeable differences in overall shape, margin and anterior region among species. With species divergence, otoliths get thinner following further developed rostrum and anti-rostrum. Interspecies variation in otolith could associate with experienced different temperature and water column. 18An Equivalent Substitute Strategy for Constructing 3D Ordered Porous Carbon Foams and Their Electromagnetic Attenuation Mechanism显示文摘Three-dimensional(3D)ordered porous carbon is generally believed to be a promising electromagnetic wave(EMW)absorbing material.However,most research works targeted performance improvement of 3D ordered porous carbon,and the specific attenuation mechanism is still ambiguous.Therefore,in this work,a novel ultra-light egg-derived porous carbon foam(EDCF)structure has been successfully constructed by a simple carbonization combined with the silica microsphere template-etching process.Based on an equivalent substitute strategy,the influence of pore volume and specific surface area on the electromagnetic parameters and EMW absorption properties of the EDCF products was confirmed respectively by adjusting the addition content and diameter of silica microspheres.As a primary attenuation mode,the dielectric loss originates from the comprehensive effect of conduction loss and polarization loss in S-band and C band,and the value is dominated by polarization loss in X band and Ku band,which is obviously greater than that of conduction loss.Furthermore,in all samples,the largest effective absorption bandwidth of EDCF-3 is 7.12 GHz under the thickness of 2.13 mm with the filling content of approximately 5 wt%,covering the whole Ku band.Meanwhile,the EDCF-7 sample with optimized pore volume and specific surface area achieves minimum reflection loss(RL_(min))of−58.08 dB at 16.86 GHz while the thickness is 1.27 mm.The outstanding research results not only provide a novel insight into enhancement of EMW absorption properties but also clarify the dominant dissipation mechanism for the porous carbon-based absorber from the perspective of objective experiments.Meng Zhang Hailong Ling Ting Wang Yingjing Jiang Guanying Song Wen Zhao Laibin Zhao Tingting Cheng Yuxin Xie Yuying Guo Wenxin Zhao Liying Yuan Alan Meng Zhenjiang Li 2022Nano-Micro Letters2022,14,10:3
19Sunlight responsive photocatalysts:AgBr/ZnO hybridna nomaterial显示文摘A novel high-effective sunlight-induced AgBr/ZnO hybrid nanophotocatalyst has been synthesized and it was characterized by different techniques such as X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), scanning electron microscopy (SEM) and ultraviolet-visible spectrophotometry (UV-vis). The AgBr/ZnO hybrid nanophotocatalyst has excellent photocatalytic activity for photodegradation of methyl orange (MO) under sunlight irradiation. The MO degradation efficiency for AgBr/ZnO is about 98% after 1 hour under sunlight irradiation. These results suggested that AgBr/ZnO is a promising candidate for the development of highly efficient sunlight photocatalysts. 20Markerless human motion capture by Markov random field and dynamic graph cuts with color constraints显示文摘Currently,many vision-based motion capture systems require passive markers attached to key loca-tions on the human body. However,such systems are intrusive with limited application. The algorithm that we use for human motion capture in this paper is based on Markov random field (MRF) and dynamic graph cuts. It takes full account of the impact of 3D reconstruction error and integrates human motion capture and 3D reconstruction into MRF-MAP framework. For more accurate and robust performance,we extend our algorithm by incorporating color constraints into the pose estimation process.
