2The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. 3The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 4Characterization of the positive SARP family regulator PieR for improving piericidin A1 production in Streptomyces piomogeues var.Hangzhouwanensis显示文摘Piericidin A1,a member ofɑ-pyridone antibiotic,exhibits various biological activities such as antimicrobial,antifungal,and antitumor properties and possesses potent respiration-inhibitory activity against insects due to its competitive binding capacity to mitochondrial complex I.The biosynthetic pathway of piericidin A1 has been reported in Streptomyces piomogeues var.Hangzhouwanensis,while the regulatory mechanism remains poorly understood.In this study,a Streptomyces antibiotic regulatory protein(SARP)family transcriptional regulator PieR was characterized.Genetic disruption and complementation manipulations revealed that PieR positively regulated the production of piericidin A1.Moreover,the overexpression of pieR contributed to the improvement of piericidin A1 productivity.The real-time quantitative PCR(RT-qPCR)was carried out and the data showed that pieR stimulated the transcription of all the biosynthesis-related genes for piericidin A1.In order to explore the regulatory mechanism,electrophoresis mobility shift assays(EMSA)and DNase I footprinting experiments have been conducted.A protected region covering 50 nucleotides within the upstream region of pieR was identified and two 5-nt direct repeat sequences(5′-CCGGA-3′)in the protected region were found.These findings,taken together,set stage for transcriptional control engineering in the view of optimizing piericidin A1 production and thus provide a viable potent route for the construction of strains with high productivity.Yan Li Lingxin Kong Jufang Shen Qing Wang Qian Liu Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:4
5Spatial Structure,Hierarchy and Formation Mechanisms of Scientific Collaboration Networks:Evidence of the Belt and Road Regions显示文摘Scientific collaboration has become an important part of the people-to-people exchanges in the Belt and Road initiative,and remarkable progress has been made since 2013.Taking the 65 countries along the Belt and Road(BRI countries)as the research areas and using collaborated Web of Science(WOS)core collection papers to construct an international scientific collaboration matrix,the paper explores the spatial structure,hierarchy and formation mechanisms of scientific collaboration networks of 65 countries along the Belt and Road.The results show that:1)Beyond the Belt and Road regions(BRI regions),Central&Eastern Europe,China and West Asia&North Africa have formed a situation in which they all have the most external links with other countries beyond BRI regions.China has the dominant role over other BRI countries in generating scientific links.The overall spatial structure has changed to a skeleton structure consisting of many dense regions,such as Europe,North America,East Asia and Oceania.2)Within the Belt and Road regions,Central&Eastern Europe has become the largest collaboration partner with other sub-regions in BRI countries.The spatial structure of scientific collaboration networks has transformed from the‘dual core’composed of China and the Central&Eastern Europe region,to the‘multi-polarization’composed of‘one zone and multi-points’.3)The hierarchical structure of scientific collaboration networks presents a typical‘core-periphery’structure,and changes from‘single core’to‘double cores’.4)Among the formation mechanisms of scientific collaboration networks,scientific research strength and social proximity play the most important roles,while geographical distance gradually weakens the hindrance to scientific collaboration.GU Weinan LIU Hui 2020Chinese Geographical Science2020,30,6:3
6CypA Regulates AIP4-Mediated M1 Ubiquitination of Influenza A Virus显示文摘Cyclophilin A (CypA) is a peptidyl-prolyl cis/trans isomerase that interacts with the matrix protein (M1) of influenza A virus (IAV) and restricts virus replication by regulating the ubiquitin–proteasome-mediated degradation of M1. However,the mechanism by which CypA regulates M1 ubiquitination remains unknown. In this study, we reported that E3 ubiquitin ligase AIP4 promoted K48-linked ubiquitination of M1 at K102 and K104, and accelerated ubiquitin–proteasome-mediated degradation of M1. The recombinant IAV with mutant M1 (K102 R/K104 R) could not be rescued, suggesting that the ubiquitination of M1 at K102/K104 was essential for IAV replication. Furthermore, CypA inhibited AIP4-mediated M1 ubiquitination by impairing the interaction between AIP4 and M1. 7A syntactic path-based hybrid neural network for negation scope detection显示文摘The automatic detection of negation is a crucial task in a wide-range of natural language processing(NLP)applications,including medical data mining,relation extraction,question answering,and sentiment analysis.In this paper,we present a syntactic path-based hybrid neural network architecture,a novel approach to identify the scope of negation in a sentence.Our hybrid architecture has the particularity to capture salient information to determine whether a token is in the scope or not,without relying on any human intervention.This approach combines a bidirectional long shortterm memory(Bi-LSTM)network and a convolutional neural network(CNN).The CNN model captures relevant syntactic features between the token and the cue within the shortest syntactic path in both constituency and dependency parse trees.The Bi-LSTM learns the context representation along the sentence in both forward and backward directions.We evaluate our model on the Bioscope corpus,and get 90.82%F-score(78.31%PCS)on the abstract sub-corpus,outperforming features-dependent approaches.Lydia LAZIB Bing QIN Yanyan ZHAO Weinan ZHANG Ting LIU 2020Frontiers of Computer Science2020,14,1:2
8Clinical Application of X-ray,B-scan,and CT in the Diagnosis of Ocular Foreign Bodies显示文摘Purpose:To analyze the sensitivity and specificity of X-ray, B-ultrasound, and CT scan in diagnosing ocular foreign bodies, and to compare the accuracy of these three imaging tools in locating these foreign bodies. Methods:A retrospective review was conducted of radio graphic and clinical eye examination data from 62 patients (66 eyes),suspected of having ocular foreign bodies at time of first presentation to the Zhongshan Ophthalmic Center (Guangzhou, China) between August 2007 and October 2011. The sensitivity and specificity of X-ray, B-ultrasound and CT scan in the diagnosis of ocular foreign bodies were investigated and their accuracy in locating these foreign bodies was compared. Results: A total of 75.8% (n=47) subjects had ocular foreign bodies; 66% of these were metallic. Patients with intraocular, eye wall,and extraocular foreign bodies accounted for 46, 28, and 26% of the total, respectively. The sensitivities of X-ray,CT scan,and B-ultrasound in the diagnosis of ocular foreign bodies were 58.3%, 61.7%, and 75%, the specificityies were 63.3%, 100%, and 87.5%, and the accuracy in locating foreign bodies was 73.4%, 94.7%, and 86.5%, respectively. The properties of foreign bodies affected the diagnosistic accuracy of X-ray,but exerted little impact upon B-ultrasound and CT scan. Conclusion:A fairly high incidence of ocular foreign bodies was noted in patients suspected with ocular foreign bodies at their first presentations.X-ray combined with CT scan or X-ray in combination with B-ultrasound showed a relatively high sensitivity and specificity in diagnosing ocular foreign bodies. 9Different discretization method used in coupled water and heat transport mode for soil under freezing conditions显示文摘A coupled water and heat transport mode is established based on the Richards equation to study water flow and heat transport in soil during freezing process. Both the finite difference and finite element method are used in the discretization, respectively. Two different computer programs are written and used to simulate an indoor unidirectional frozen test. The freezing depth, freezing rate and temperature variation are compared among lab tests, finite difference calculation simulation and finite element calculation simulation. 10Tanshinone IIa antagonizes spinal ischemia/reperfusion injury by interfering with upstream glutamic acid factors显示文摘Based on the hypothesis that upstream factor inhibition results in better treatment effects than downstream factor inhibition, the present study interfered with glutamic acid (Glu)-released upstream factors, such as Glu transporter function and Na+-K+-adenosine triphosphatases (ATPase) activity relativly. Rats with spinal cord ischemia/reperfusion injury received intraperitoneal injections of tanshinone IIa and Glu uptake and Na+-K+-ATPase activity were increased. 11Late Quaternary terrigenous sedimentation in the Western Arctic Ocean as exemplified by a sedimentary record from the Alpha Ridge显示文摘Terrigenous components in sediment core B84A from the Alpha Ridge,Western Arctic Ocean,have been investigated to reconstruct Mid to Late Quaternary variations in sedimentation,provenance,and related climate changes. The core stratigraphy,evaluated by a combination of variations in Mn content,color cycles,foraminiferal abundance,and lithological correlation,extends back to estimated Marine Isotope Stage 12. Twelve Ice Rafted Detritus(IRD,>250 μm) events were identified and interpreted to mostly occur during deglaciation.The Canadian Arctic,which was covered by ice sheets during glacial periods,is suggested to be the major source region. The IRD events likely indicate the collapses of ice sheets,possibly in response to abrupt climate changes.Grain size analysis of B84A indicates sedimentologically sensitive components in core B84A in the 4-9 μm and 19-53 μm silt subfractions,which are inferred to be mainly transported by currents and sea ice,respectively. Down core variability of these two fractions may indicate changes in ice drift and current strength. 15Breaking or following the membrane-targeting mechanism:Exploring the antibacterial mechanism of host defense peptide mimicking poly(2-oxazoline)s显示文摘Peptides exert important biological functions but their application is hindered by their susceptibility to proteolysis and poor stability in vivo.Thus,functional peptide mimics have drawn a great deal of attention to address this challenge.Poly(2-oxazoline)s,a class of biocompatible and proteolysis-resistant polymer,can work as host defense peptide mimics without following the general membrane-targeting mechanism as shown in our previous work.This observation encouraged us to figure out if poly(2-oxazoline)s are special and break the general membrane-targeting mechanism of host defense peptides and their mimics.In this study,we aimed at the connection between structure and antibacterial mechanism of poly(2-oxazoline)s.A new γ-aminobutyric acid(GABA)-pendent poly(2-oxazoline)was synthesized and investigated to compare with glycine-pendent poly(2-oxazoline)in our previous study,with the former polymer has two extra CH2 groups in the sidechain to increase the hydrophobicity and amphiphilicity.Membrane depolarization assay suggested that incorporating two more CH2 groups into the sidechain of poly(2-oxazoline)resulted in a mechanism switch from DNA-targeting to membrane-targeting,which was supported by the slow time-kill kinetics and slightly distorted and sunken membrane morphology.Besides,GABA-pendent poly(2-oxazoline)showed potent activity against methicillin-resistant S.aureus and low hemolysis on human red blood cells.Moreover,repeated use of the antimicrobial poly(2-oxazoline)did not stimulate bacteria to obtain resistance,which was an obvious advantage of membrane-targeting antimicrobial agents.Chengzhi Dai Min Zhou Weinan Jiang Ximian Xiao Jingcheng Zou Yuxin Qian Zihao Cong Zhemin Ji Longqiang Liu Jiayang Xie Zhongqian Qiao Runhui Liu 2020Journal of Materials Science & Technology2020,59,24:1

18Gated recurrent unit model for a sequence tagging problem显示文摘Combinatory categorial grammer(CCG) supertagging is an important subtask that takes place before full parsing and can benefit many natural language processing(NLP) tasks like question answering and machine translation. CCG supertagging can be regarded as a sequence labeling problem that remains a challenging problem where each word is assigned to a CCG lexical category and the number of the probably associated CCG supertags to each word is large. To address this, recently recurrent neural networks(RNNs), as extremely powerful sequential models, have been proposed for CCG supertagging and achieved good performances. In this paper, a variant of recurrent networks is proposed whose design makes it much easier to train and memorize information for long range dependencies based on gated recurrent units(GRUs), which have been recently introduced on some but not all tasks.
