| 1 | Individualized prediction of perineural invasion in colorectal cancer: development and validation of a radiomics prediction model显示文摘Objective: To develop and validate a radiomics prediction model for individualized prediction of perineural invasion(PNI) in colorectal cancer(CRC).Methods: After computed tomography(CT) radiomics features extraction, a radiomics signature was constructed in derivation cohort(346 CRC patients). A prediction model was developed to integrate the radiomics signature and clinical candidate predictors [age, sex, tumor location, and carcinoembryonic antigen(CEA) level]. Apparent prediction performance was assessed. After internal validation, independent temporal validation(separate from the cohort used to build the model) was then conducted in 217 CRC patients. The final model was converted to an easy-to-use nomogram.Results: The developed radiomics nomogram that integrated the radiomics signature and CEA level showed good calibration and discrimination performance [Harrell's concordance index(c-index): 0.817; 95% confidence interval(95% CI): 0.811–0.823]. Application of the nomogram in validation cohort gave a comparable calibration and discrimination(c-index: 0.803; 95% CI: 0.794–0.812).Conclusions: Integrating the radiomics signature and CEA level into a radiomics prediction model enables easy and effective risk assessment of PNI in CRC. | 4 | Aluminium is essential for root growth and development of tea plants(Camellia sinensis)^∞显示文摘On acid soils,the trivalent aluminium ion(Al3+)predominates and is very rhizotoxic to most plant species.For some native plant species adapted to acid soils including tea(Camellia sinensis),Al3+has been regarded as a beneficial mineral element.In this study,we discovered that Al3+is actually essential for tea root growth and development in all the tested varieties.Aluminum ion promoted new root growth in five representative tea varieties with dose-dependent responses to Al3+availability.In the absence of Al3+,the tea plants failed to generate new roots,and the root tips were damaged within 1 d of Al deprivation.Struc-tural analysis of root tips demonstrated that Al was required for root meristem development and activity.In situ morin@staining of Al3+in roots revealed that Al mainly localized to nuclei in root meristem cells,but then gradually moved to the cytosol when Al3+was subsequently withdrawn.This movement of Al3+from nuclei to cytosols was accompanied by exacerbated DNA damage,which suggests that the nuclear-targeted Al primarily acts to maintain DNA integrity.Taken together,these results provide novel evidence that Al3+is essential for root growth in tea plants through maintenance of DNA integrity in meristematic cells. | Lili Sun Mengshi Zhang Xiaomei Liu Qianzhuo Mao Chen Shi Leon VKochian Hong Liao | 2020 | Journal of Integrative Plant Biology2020,62,7: | 12 |
| 5 | The development of MOFs-based nanomaterials in heterogeneous organocatalysis显示文摘Metal-organic framework(MOF) is a class of inorganic-organic hybrid material assembled periodically with metal ions and organic ligands. MOFs have always been the focuses in a variety of frontier fields owing to the advantageous properties, such as large BET surface areas, tunable porosity and easyfunctionalized surface structure. Among the various application areas, catalysis is one of the earliest application fields of MOFs-based materials and is one of the fastest-growing topics. In this review, the main roles of MOFs in heterogeneous organocatalysis have been systematically summarized, including used as support materials(or hosts), independent catalysts, and sacrificial templates. Moreover, the application prospects of MOFs in photocatalysis and electrocatalysis frontiers were also mentioned.Finally, the key issues that should be conquered in future were briefly sketched in the final parts of each item. | 6 | Vertical response of microbial community and degrading genes to petroleum hydrocarbon contamination in saline alkaline soil显示文摘A column microcosm was conducted by amending crude oil into Dagang Oilfield soil to simulate the bioremediation process. The dynamic change of microbial communities and metabolic genes in vertical depth soil from 0 to 80 cm were characterized to evaluate the petroleum degradation potential of indigenous microorganism. The influence of environmental variables on the microbial responds to petroleum contamination were analyzed. Degradation extent of 42.45% of n-alkanes(C8–C40) and 34.61% of 16ΣPAH were reached after 22 weeks. Relative abundance of alkB, nah, and phe gene showed about 10-fold increment in different depth of soil layers. Result of HTS profiles demonstrated that Pseudomonas, Marinobacter and Lactococcus were the major petroleum-degrading bacteria in0–30 and 30–60 cm depth of soils. Fusarium and Aspergillus were the dominant oil-degrading fungi in the 0–60 cm depth of soils. Visceral hypersensitivity is closely associated with irritable bowel syndrome. Mild moxibustion can relieve chronic visceral hyperalgesia in rats with irritable bowel syndrome. We hypothesized that prokineticin-1 and prokineticin receptor-1 is the key target in the mechanism. This study established chronic visceral hyperalgesia rat models by colorectal distention. Protein and mRNA expression of prokineticin-1 and prokineticin receptor-1 were determined by immunohisto-chemical method and fluorescence quantitative-PCR, respectively, and were found to be signifi-cantly increased in visceral hyperalgesic rats. | 11 | Overexpression of C-terminal fragment of glutamate receptor 6 prevents neuronal injury in kainate-induced seizure via disassembly of Glu R6-PSD95-MLK3 signaling module显示文摘Our previous study showed that when glutamate receptor(Glu R)6 C terminus-containing peptide conjugated with the human immunodeficiency virus Tat protein(Glu R6)-9c is delivered into hippocampal neurons in a brain ischemic model, the activation of mixed lineage kinase 3(MLK3) and c-Jun NH2-terminal kinase(JNK) is inhibited via Glu R6-postsynaptic density protein 95(PSD95). In the present study, we investigated whether the recombinant adenovirus(Ad) carrying Glu R6 c could suppress the assembly of the Glu R6-PSD95-MLK3 signaling module and decrease neuronal cell death induced by kainate in hippocampal CA1 subregion. A seizure model in Sprague-Dawley rats was induced by intraperitoneal injections of kainate. The effect of AdGlur6-9c on the phosphorylation of JNK, MLK3 and mitogen-activated kinase kinase 7(MKK7) was observed with western immunoblots and immunohistochemistry. Our findings revealed that overexpression of Glu R6 c inhibited the interaction of Glu R6 with PSD95 and prevented the kainate-induced activation of JNK, MLK3 and MKK7. Furthermore, kainate-mediated neuronal cell death was significantly suppressed by Glu R6 c. | 12 | Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. | 13 | Progressive loss of striatal dopamine terminals in MPTP-induced acute parkinsonism in cynomolgus monkeys using vesicular monoamine transporter type 2 PET imaging([^(18)F]AV-133)显示文摘The 1-methyl-4-phenyl-1,2,3,4-tetrahydropyridine(MPTP)-induced parkinsonism model, particularly in non-human primates, remains the gold-standard for studying the pathogenesis and assessing novel therapies for Parkinson's disease. However, whether the loss of dopaminergic neurons in this model is progressive remains controversial, mostly due to the lack of objective in vivo assessment of changes in the integrity of these neurons. In the present study, parkinsonism was induced in cynomolgus monkeys by intravenous administration of MPTP(0.2 mg/kg) for 15 days; stable parkinsonism developed over 90 days, when the symptoms were stable. Noninvasive positron emission tomographic neuroimaging of vesicular monoamine transporter 2 with 9-[18F]fluoropropyl-(+)-dihydrotetrabenazine([18F]AV-133) was used before, and 15 and 90 days after the beginning of acute MPTP treatment. The imaging showed evident progressive loss of striatal uptake of [18F]AV-133. The dopaminergic denervation severity had a significant linear correlation with the clinical rating scores and the bradykinesia subscores. | 14 | Follistatin-like protein 1 plays a tumor suppressor role in clear-cell renal cell carcinoma显示文摘Background:We previously showed that the expression of follistatin-like protein 1(FSTL1)was significantly down-regulated in metastatic clear-cell renal cell carcinoma(ccRCC).In this study,we aimed to characterize the role of FSTL1 in the development of ccRCC.Methods:The effects of FSTL1 on cell activity and cell cycle were investigated in ccRCC cell lines with altered FSTL1 expression.Gene expression microarray assays were performed to identify the major signaling pathways affected by FSTL1 knockdown.The expression of FSTL1 in ccRCC and its effect on postoperative prognosis were estimated in a cohort with 89 patients.Results:FSTL1 knockdown promoted anchorage-independent growth,migration,invasion,and cell cycle of ccRCC cell lines,whereas FSTL1 overexpression attenuated cell migration.FSTL1 knockdown up-regulated nuclear factor-κB(NF-κB)and hypoxia-inducible factor(HIF)signaling pathways,increased epithelial-to-mesenchymal transition,up-regulated interleukin-6 expression,and promoted tumor necrosis factor-α-induced degradation of NF-κB inhibitor(IκBα)in ccRCC cell lines.FSTL1 immunostaining was selectively positive in epithelial cytoplasm in the loop of Henle,and positive rate of FSTL1 was significantly lower in ccRCC tissues than in adjacent renal tissues(P<0.001).The mul-tivariate Cox regression analysis showed that the intratumoral FSTL1 expression conferred a favorable independent prognosis with a hazard ratio of 0.325(95%confidence interval 0.118-0.894).HIF-2αexpression was negatively cor-related with FSTL1 expression in ccRCC specimens(r=−0.229,P=0.044).Intratumoral expression of HIF-2α,rather than HIF-1α,significantly predicted an unfavorable prognosis in ccRCC(log-rank,P=0.038).Conclusions:FSTL1 plays a tumor suppression role possibly via repressing the NF-κB and HIF-2αsignaling pathways.To increase FSTL1 expression might be a candidate therapeutic strategy for metastatic ccRCC. | Yan Liu Xiaojie Tan Wenbin Liu Xi Chen Xiaomei Hou Dan Shen Yibo Ding Jianhua Yin Ling Wang Hongwei Zhang Yongwei Yu Jianguo Hou Timothy C.Thompson Guangwen Cao | 2018 | Cancer Communications2018,38,1: | 5 |
| 15 | Proto-Tethys ophiolitic mélange in SW Yunnan: Constraints from zircon U-Pb geochronology and geochemistry显示文摘An early Paleozoic Proto-Tethys ocean in western Yunnan has long been postulated although no robust geological evidence has been identified.Here we investigated the recently-identified Mayidui and Wanhe ophiolitic mélanges in SW Yunnan,which occurs in a N-S trending belt east of the late Paleozoic Changning-Menglian suture zone.The ophiolites consist mainly of meta-basalts(amphibole schists),meta-(cumulate)gabbros and gabbroic diorites,and meta-chert-shale,representing ancient oceanic crust and pelagic and hemipelagic sediments,respectively.Six samples of gabbros and gabbroic diorites from 3 profiles(Mayidui,Kongjiao and Yinchanghe)yielded zircon U-Pb ages between 462±6 Ma and 447±9 Ma,constraining the formation of the Mayidui and Wanhe ophiolites to Middle Ordovician.Gabbros from the Mayidui and Kongjiao profiles share similar geochemical characteristics with affinities to tholeiitic series,and are characterized by depleted to slightly enriched LREEs relative to HREEs with(La/Sm)N=0.69-1.87,(La/Yb)N=0.66-4.72.These,along with their predominantly positive wholerock eNd(t)and zircon eHf(t)values,indicate a MORB-like magma source.By contrast,the meta-mafic rocks from the Yinchanghe profile show significantly enriched LREEs((La/Sm)N=0.97-3.33,(La/Yb)N=1.19-14.93),as well as positive whole-rock eNd(t)and positive to negative zircon eHf(t)values,indicating an E-MORB-type mantle source.These geochemical features are consistent with an intra-oceanic setting for the formation of the Mayidui-Wanhe ophiolites.Our data,integrated with available geological evidence,provide robust constraints on the timing and nature of the Mayidui-Wanhe ophiolitic mélange,and suggest that the ophiolites represent remnants of the Proto-Tethys Ocean,which opened through separation of the Indochina and Simao blocks from the northern margin of Gondwana before the Early Cambrian,and evolved through to the Silurian. | Guichun Liu Zaibo Sun Jianwei Zi M.Santosh Tianyu Zhao Qinglai Feng Guangyan Chen Xiaomei Nie Jing Li Shitao Zhang | 2021 | Geoscience Frontiers2021,12,5: | 5 |

The effects of grain-refined by yttrium on the morphology and the grain size of the primary α-Al in semi-solid Al7Si0.3Mg alloy are researched. The results indicate that semi-solid Al7Si0.3Mg alloy with particle-like and rosette-like primary α-Al can be prepared by low temperature pouring from liquid Al7Si0.3Mg alloy grain-refined by yttrium. The grain size and particle morphology of primary α-Al in Al7Si0.3Mg alloy are markedly improved by 0.5 wt pct Y. | 17 | IL-17-triggered downregulation of miR-497 results in high HIF-1α expression and consequent IL-1β and IL-6 production by astrocytes in EAE mice显示文摘Interleukin 17(IL-17)is increasingly recognized as a key factor that contributes to the pathogenesis of multiple sclerosis(MS)and its experimental mouse autoimmune encephalomyelitis(EAE)model.However,the roles and regulatory mechanisms of IL-17-induced pro-inflammatory cytokine production in EAE mice remain largely unclear.In this study,the expression of IL-17,hypoxia inducible factor-1α(HIF-1α),IL-1β,IL-6 and microRNA-497(miR-497),as well as their intrinsic associations,was investigated using EAE model mice and cultured astrocytes exposed to IL-17 in vitro.We observed markedly increased production of IL-17,HIF-1α,IL-1βand IL-6 in the brain tissues of EAE mice,while the expression and secretion of HIF-1α,IL-1βand IL-6 were also significantly increased when cultured primary astrocytes from mice were stimulated with IL-17.Meanwhile,the expression of miR-497 was downregulated both in vivo and in vitro.Subsequent in vitro experiments revealed that IL-17 induced the production of IL-1βand IL-6 in astrocytes through the upregulation of HIF-1αas a transcriptional factor,indicating that IL-17-mediated downregulation of miR-497 enhanced HIF-1αexpression.Furthermore,astrocytespecific knockdown of IL-17RA and HIF-1αor astrocyte-specific overexpression of miR-497 by infection with different lentiviral vectors containing an astrocyte-specific promotor markedly decreased IL-1βand IL-6 production in brain tissues and alleviated the pathological changes and score of EAE mice.Collectively,these findings indicate that decreased miR-497 expression is responsible for IL-17-triggered high HIF-1αexpression and consequent IL-1βand IL-6 production by astrocytes in EAE mice. | Kai Shan Rongrong Pang Chenhui Zhao Xiaomei Liu Wenxing Gao Jing Zhang Dan Zhao Yingwei Wang Wen Qiu | 2017 | Cellular & Molecular Immunology2017,14,11: | 5 |

| 19 | Aquaculture area extraction and vulnerability assessment in Sanduao based on richer convolutional features network model显示文摘Sanduao is an important sea-breeding bay in Fujian,South China and holds a high economic status in aquaculture.Quickly and accurately obtaining information including the distribution area,quantity,and aquaculture area is important for breeding area planning,production value estimation,ecological survey,and storm surge prevention.However,as the aquaculture area expands,the seawater background becomes increasingly complex and spectral characteristics differ dramatically,making it difficult to determine the aquaculture area.In this study,we used a high-resolution remote-sensing satellite GF-2 image to introduce a deep-learning Richer Convolutional Features(RCF)network model to extract the aquaculture area.Then we used the density of aquaculture as an assessment index to assess the vulnerability of aquaculture areas in Sanduao.The results demonstrate that this method does not require land and water separation of the area in advance,and good extraction can be achieved in the areas with more sediment and waves,with an extraction accuracy>93%,which is suitable for large-scale aquaculture area extraction.Vulnerability assessment results indicate that the density of aquaculture in the eastern part of Sanduao is considerably high,reaching a higher vulnerability level than other parts. | LIU Yueming YANG Xiaomei WANG Zhihua LU Chen LI Zhi YANG Fengshuo | 2019 | Journal of Oceanology and Limnology2019,37,6: | 4 |
| 20 | Effects of electroacupuncture on c-Fos expression in the spinal cord and brain of rats with chronic visceral hypersensitivity显示文摘BACKGROUND:Visceral hypersensitivity is the main cause of irritable bowel syndrome.c-Fos is a marker of visceral hypersensitivity in the central nervous system.Electroacupuncture can relieve chronic visceral hypersensitivity in rats,but the mechanism is still unknown. OBJECTIVE:To identify c-Fos expression in the spinal cord and cerebral cortex of rats with chronic visceral hypersensitivity,and to test the effects of electroacupuncture on pain sensitivity in rats with chronic visceral hypersensitivity. DESIGN,TIME AND SETTING:A randomized controlled animal experiment was performed at the Animal Experimental Center,Shanghai University of Traditional Chinese Medicine,from January to April,2007. MATERIALS:A total of 24 neonatal,male,Sprague Dawley rats,aged five days old,were equally and randomly assigned into a normal group,a model group,and an electroacupuncture group. Rabbit anti-rat c-Fos antibody and Evision secondary antibody kits(Sigma,USA),diaminobenzidine kit(Dako,Denmark),and an LD202H electroacupuncture apparatus(Huawei,Beijing,China) were used in this study. METHODS:Neonatal rats from the model and electroacupuncture groups were used to establish rat models of chronic visceral hypersensitivity by the saccule stimulation method.After model establishment,0.25 mm diameter electric needles were inserted into Tianshu(ST 25) and Shangjuxu(ST37) at a depth of approximately 0.5 cm,with an square wave(alternating current frequency at 100/20 Hz,amplitude ranged 0.2-0.6 ms,intensity at 1 mA) once for 20 minutes,once a day,for seven days.Rats in the normal and model groups were not treated. MAIN OUTCOME MEASURES:Following 7 days of treatment,c-Fos expression in the spinal cord and cerebral cortex was detected by immunohistochemistry.After the first electroacupuncture treatment,abdominal withdrawal reflex scores were investigated to evaluate the pain threshold for chronic visceral hypersensitivity in rats. RESULTS:Visceral hypersensitivity increased c-Fos staining(P<0.05),and electroacupuncture significantly decreased the number of these cells to near normal levels(P>0.05).Abdominal withdrawal reflex scores were significantly lower in the electroacupuncture and normal groups than in the model group(P<0.05) and were similar between the electroacupuncture and normal groups (P>0.05).
