2Replacement of Dietary Fish Oil with Vegetable Oils Improves the Growth and Flesh Quality of Large Yellow Croaker(Larmichthys crocea)显示文摘We investigated the effect of the replacement of dietary fish oil with vegetable oils on the growth and flesh quality of large yellow croaker(Larmichthys crocea). The basal diet(FO) was formulated to contain 66.5% fish meal and 6.4% menhaden fish oil; whereas the other 3 experimental diets were formulated by replacing the fish oil with 50% soybean oil(SO50), 100% soybean oil(SO100) and 100% palm oil(PO100), respectively. The 4 diets were randomly assigned to 4 floating sea cages(3.0 m × 3.0 m × 3.0 m), and each was stocked with 250 fish individuals with an initial average weight of 245.29 g ± 7.45 g. The fish were fed to apparent satiation twice a day at 5:00 and 17:00, respectively, for 12 weeks. Experimental analysis showed that the specific growth rate of fish fed SO50 or PO100 were significantly higher than that of fish fed FO or SO100(P<0.05), and crude lipid contents of ventral muscle and viscera were significantly lower in fish fed FO than in those fed the other 3 diets(P<0.05). No significant differences in condition factor, viscerosomatic index, hepatosomatic index, gutted yield and colorimetric values of fish among the dietary treatments were observed(P>0.05). Compared to FO diet, SO50, SO100 and PO100 diets led to substantial decreases in the liquid loss and water loss from fresh fillets(1 d, 4℃)(P<0.05). Similarly, thiobarbituric acid reactive substance(TBARS) values of fillets under different storage conditions(1 d, 4℃; 7 d, 4℃; 4 weeks,-20℃; 8 weeks,-20℃) decreased significantly after partial or complete replacement of fish oil with vegetable oils. 3Direct generation of an ultrafast vortex beam in a CVD-graphene-based passively mode-locked Pr:LiYF4 visible laser显示文摘We report on the direct generation of passively mode-locked vortex lasers in the visible spectral region, for the first time to the best of our knowledge, using a Pr :Li YF4(Pr:YLF) crystal as the gain medium. A stable mode-locked TEM00 mode has been achieved with a maximum average output power of 75 m W using a graphene saturable absorber mirror. The mode-locked pulse width is measured to be as short as about 73.4 ps at a repetition rate of about 140 MHz, and the laser wavelength is at about 721 nm with spectral width of about 0.5 nm. By slightly misaligning the laser resonator, a first-order Laguerre-Gaussian mode(LG0,1) has also been obtained with output power reduced to about 22 m W. The achieved LG0,1 mode has been verified via a home made improved Fizeau interferometer. 5The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy.Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu 2021Cancer Communications2021,41,1:5
64-Formyl-2-(2H-benzotriazol-2-yl)-phenol: an ESIPT chromophore显示文摘In the present work, excited state intramolecular proton transfer(ESIPT) emission properties of a new benzotriazole derivative 4-formyl-2-(2H-benzotriazol-2-yl)-phenol(C1) were studied. 4-Formyl-2-(2H-benzotriazol-2-yl)-methoxy-benzene(C2), 4-formyl-2-(2H-benzotriazol-2-yl)-phenyl acetate(C3) and 4-methyl-2-(2H-benzotriazol-2-yl)-phenol(C4) were used as the reference molecules.1H NMR chemical shift of hydroxy group in C1 was located at more down field than that of C4 or p-hydroxy-benzaldehyde(C5), respectively. C1 showed two absorption bands in the range of 260–400 nm zones in various solvents, while C2 and C3 exhibited single absorption band. The equal molar mixtures of C2/C5 or C3/C5 showed single absorption band.C2 and C3 displayed single fluorescence emission band in various solvents, while C1 exhibited dual emission bands in some strong polar solvents. Furthermore, the second emission band in these strong polar solvents showed the large Stokes shift. The results show that the second emission band of C1 was produced by ESIPT. C2 and C3 could not undergo ESIPT due to no hydroxy group. 8PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. 17Size and composition dependence of melting temperature of binary nanoparticles显示文摘Based on the ideal solution approximation, the model for size-dependent melting temperature of pure metal nanoparticles is extended to binary alloy systems. The developed model, free of any adjustable parameter, demonstrates that the melting temperature is related to the size and composition of alloy nanoparticles. The melting temperature of CuNi, PbBi and SnIn binary alloy nanocrystals is found to be consistent with the experiments and molecular dynamics simulations. 18Investigating the Effect of Methylcyclopentadienyl Manganese Tricarbonyl on the Properties of Unleaded Motor Gasoline (1): Induction Period显示文摘In this paper, the effect of MMT on the induction period of unleaded motor gasoline was studied, the manganese concentration, storage period of MMT-blended gasoline and environmental variables such as temperature and radiation intensity were considered to be main factors affecting the induction period of gasoline,when MMT-blended gasoline was exposed to light. It is found from experiments that the addition of MMT can improve the induction period of gasoline that is shielded from light, and reduce the induction period remarkably,when the gasoline is exposed to light. 20Microbiome-metabolome analysis reveals cervical lesion alterations显示文摘Cervical cancer(CC)continues to be one of the most common cancers among females worldwide.It takes a few years or even decades for CC to arise in a minority of women with cervical precancers.An increasing corpus of studies today indicates that local microecology and carcinogenesis are intimately related.To investigate the changes in cericovaginal microecology with the development of cervical cancer,we performed 16S rDNA sequencing and metabolomic analysis in cericovaginal fluid from 10 LSIL patients,10 HSIL patients,10 CC patients and 10 healthy controls to reveal the differential flora and metabolites during cervical carcinogenesis.Carcinogenesis is associated with alterations in microbiome diversity,individual taxa,and functions with notable changes in Lactobacillus,Prevotella and Aquabacterium,as well as in cervicovaginal metabolites that correlate with cervicovaginal microbial patterns.Increased bacterial diversity and a decline in the relative abundance of Lactobacillus,the dominant species in the cericovaginal flora,are observed when cervical lesions advance.According to KEGG pathway enrichment analysis,lipids and organic acids change as cervical cancer progresses,and the phenylalanine,tyrosine,and tryptophan biosynthesis pathway is essential for the development of cervical cancer.Our results reveal that microbic and metabolomic profiling is capable of distinguishing CC from precancer and highlights potential biomarkers for the early detection of cervical dysplasia.These differential microorganisms and metabolites are expected to become a potential tool to assist in the diagnosis of cervical cancer.Hanjie Xu Lou Liu Feng Xu Min Liu Yuexiao Song Jiale Chen Huiying Zhan Ye Zhang Dexiang Xu Yu Chen Mudan Lu Daozhen Chen 2022Acta Biochimica et Biophysica Sinica2022,54,10:0
