3PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 4Passive-source ocean bottom seismograph(OBS) array experiment in South China Sea and data quality analyses显示文摘Long-term passive source ocean bottom seismograph(OBS) observatory is challenging due to various technical difficulties. In order to gain experience in this field, and to reveal the lithospheric structure beneath the extinct ridge in the central South China Sea(SCS), we carried out a passive source OBS array experiment, which includes 18 OBSs, in the deep portion of SCS. Here we present the instrumentation, the OBS deployment and recovery of this experiment, and more importantly, the data quality evaluated by a number of approaches. Through processing and inspecting waveforms from global, regional and local earthquakes, we find that most of recovered OBSs have good data quality with discernible main phases. The ambient noise analyses of OBS recordings show that their noise is higher than the global average, and the horizontal component is noisier than the vertical, indicating current impacts on horizontal components are more severe. In the period range of 5–10 s, there is a noise notch for the SCS OBSs, and noise levels of horizontal components are comparable to the vertical. This feature, which is not seen at OBS stations in open ocean, suggests the distant sources for double frequency microseism in this marginal sea are not significant. In addition, we successfully determined the orientations for 7 OBSs by investigating their Rayleigh wave polarizations; and we demonstrated the dispersion feature of Rayleigh waves through the frequency-time analysis. 5Modified k-ω model using kinematic vorticity for corner separation in compressor cascades显示文摘A new method of modifying the conventional k-ω turbulence model for corner separation is proposed in this paper. The production term in the ω equation is modified using kinematic vorticity considering fluid rotation and deformation in complex geometric boundary conditions. The corner separation flow in linear compressor cascades is calculated using the original k-ω model, the modified k-ω model and the Reynolds stress model(RSM). The numerical results of the modified model are compared with the available experimental data, as well as the corresponding results of the original k-ω model and RSM. In terms of accuracy, the modified model, which significantly improves the performance of the original k-ω model for predicting corner separation, is quite competitive with the RSM. 8Systematic identification of endogenous RNA polymeraseⅢpromoters for efficient RNA guidebased genome editing technologies in maize显示文摘Single-guide RNA(sg RNA) is one of the two core components of the CRISPR(clustered regularly interspaced short palindromic repeat)/Cas(CRISPR-associated) genome-editing technology. We established an in vitro Traffic Light Reporter(TLR) system, which is designated as the same colors as traffic lights such as green, red and yellow were produced in cells. The TLR can be readily used in maize mesophyll protoplast for a quick test of promoter activity. The TLR assay indicates the variation in transcription activities of the seven Pol III promoters, from 3.4%(U6-1) to over 21.0%(U6-6). The U6-2 promoter, which was constructed to drive sg RNA expression targeting the Zm Wx1 gene, yielded mutation efficiencies ranging from 48.5% to 97.1%. Based on the reported and unpublished data, the in vitro TLR assay results were confirmed to be a readily system and may be extended to other plant species amenable to efficient genome editing via CRISPR/Cas. 9Anatomical and functional outcomes after Densiron 68 heavy silicone oil tamponade for complicated retinal detachment in Chinese eyes显示文摘AIM:To evaluate the safety and efficacy of Densiron 68 heavy silicone oil (HSO) tamponade for complicated retinal detachment(RD)in Chinese eyes.METHODS:Twenty-one eyes of 21 patients with complicated RD were included in this retrospective study.All patients underwent pars plana vitrectomy with an internal tamponade using Densiron 68 HSO.Anatomical and functional results and complications were evaluated,including retinal status,visual acuity(VA),intraocular pressure(IOP),intraocular inflammation,lens opacity,and HSO emulsification.RESULTS:Allthepatients were followed up for 3mo to1y(5.8±1.16mo).Retinal reattachment was achieved in 19of 21 patients(90.5%).VA improved in 18 of 21 patients(85.7%),from 1.93 logMAR(±0.48)to 1.52 logMAR(±0.45)(P=0.001).Postoperative complications included early dispersion of HSO in 7 eyes(38.8%),cataract in 10 of 18phakic eyes(55.5%),moderate postoperative inflammation reaction in 10 eyes(47.6%),and elevated IOP in 5 eyes(23.8%),all of which were controlled by medication or by surgery.CONCLUSION:Highanatomical and functional success rates can be achieved with primary vitrectomy for complicated RD by using Densiron 68 HSO;however,it should not be ignored that Densiron 68 HSO can cause some complications in the eye.Fang Liu Hui Li Le Feng Fang Wang 2014International Journal of Ophthalmology(English edition)2014,7,3:7

14A TCP Performance Enhancement Scheme in Infrastructure based Vehicular Networks显示文摘Transmission Control Protocol(TCP) in infrastructure based vehicular networks is dedicated to support reliable Internet services for mobile users.However,an end-toend TCP flow not only experiences some common challenges in wireless mobile networks,such as high packet loss rate,medium access competition,unstable wireless bandwidth,and dynamic topology,etc.,but also suffers from performance degradation due to traffic congestion at the Road-Side Units(RSUs) that connect the wireline and wireless networks.In order to address the challenging issues related to reliable TCP transmissions in infrastructure based vehicular networks,we propose an RSU based TCP(R-TCP) scheme.For wireline source nodes,R-TCP adopts a novel flow control mechanism to adjust transmission rates according to the status of bottleneck link.Specifically,during the short wireless connection time in Infrastructure based vehicular networks,R-TCP quickly chooses an ideal transmission rate for data transmissions instead of activating the slow start algorithm after the connection is established,and successfully avoids the oscillation of the transmission rate.Simulation results show that R-TCP achieves great advantages than some relate proposals in terms of throughput,end-to-end delay,and packet loss rate.HA Le FANG Lijin BI Yuanguo LIU Wei 2015China Communications2015,12,6:5

16DTI and pathological changes in a rabbit model of radiation injury to the spinal cord after ^(125)I radioactive seed implantation显示文摘Excessive radiation exposure may lead to edema of the spinal cord and deterioration of the nervous system. Magnetic resonance imaging can be used to judge and assess the extent of edema and to evaluate pathological changes and thus may be used for the evaluation of spinal cord injuries caused by radiation therapy. Radioactive ^(125)I seeds to irradiate 90% of the spinal cord tissue at doses of 40–100 Gy(D90) were implanted in rabbits at T_(10) to induce radiation injury, and we evaluated their safety for use in the spinal cord. Diffusion tensor imaging showed that with increased D90, the apparent diffusion coefficient and fractional anisotropy values were increased. Moreover, pathological damage of neurons and microvessels in the gray matter and white matter was aggravated. At 2 months after implantation, obvious pathological injury was visible in the spinal cords of each group. 17Fruit and vegetable consumption and its relation to risk of asthenopia among Chinese college students显示文摘AIM: To investigate the associations between fruit and vegetable consumption and risk of asthenopia among Chinese college students.METHODS: A total of 1022 students were selected from five universities by a multi-stage stratified cluster sampling method. They were surveyed via a self-administered questionnaire including socio-demographic features,dietary and lifestyle habits, eye-related symptoms, eye care habits and history of diseases. Ascertainment of asthenopia was based on participants' subjectively reported symptoms. The associations between fruit and vegetable intake with asthenopia risk were assessed using multivariate logistic regression analysis.RESULTS: There were no significant associations between total fruit and vegetable, total vegetable, or fruit and the risk of asthenopia. Higher intake of dark-green leafy vegetable was likely to be inversely associated with asthenopia risk [odd ratio(OR): 0.60; 95%CI: 0.37-0.97;P_(trend)=0.21] after controlling for nondietary and dietary risk factors. 20NF-κB and FosB mediate inflammation and oxidative stress in the blast lung injury of rats exposed to shock waves显示文摘Blast lung injury(BLI)is the major cause of death in explosion-derived shock waves;however,the mechanisms of BLI are not well understood.To identify the time-dependent manner of BLI,a model of lung injury of rats induced by shock waves was established by a fuel air explosive.The model was evaluated by hematoxylin and eosin staining and pathological score.The inflammation and oxidative stress of lung injury were also investigated.The pathological scores of rats’lung injury at 2 h,24 h,3 days,and 7 days post-blast were 9.75±2.96,13.00±1.85,8.50±1.51,and 4.00±1.41,respectively,which were significantly increased compared with those in the control group(1.13±0.64;P<0.05).The respiratory frequency and pause were increased significantly,while minute expiratory volume,inspiratory time,and inspiratory peak flow rate were decreased in a time-dependent manner at 2 and 24 h post-blast compared with those in the control group.In addition,the expressions of inflammatory factors such as interleukin(IL)-6,IL-8,FosB,and NF-κB were increased significantly at 2 h and peaked at 24 h,which gradually decreased after 3 days and returned to normal in 2 weeks.The levels of total antioxidant capacity,total superoxide dismutase,and glutathione peroxidase were significantly decreased 24 h after the shock wave blast.Conversely,the malondialdehyde level reached the peak at 24 h.These results indicated that inflammatory and oxidative stress induced by shock waves changed significantly in a time-dependent manner,which may be the important factors and novel therapeutic targets for the treatment of BLI.Hong Wang Wenjuan Zhang Jinren Liu Junhong Gao Le Fang Zhiyong Liu Baoqing Xia Xiaolin Fan Cunzhi Li Qing Lu Airong Qian 2021Acta Biochimica et Biophysica Sinica2021,53,3:4
