1Adenovirus-mediated gene delivery:Potential applications for gene and cell-based therapies in the new era of personalized medicine显示文摘With rapid advances in understanding molecular pathogenesis of human diseases in the era of genome sciences and systems biology,it is anticipated that increasing numbers of therapeutic genes or targets will become available for targeted therapies.Despite numerous setbacks,efficacious gene and/or cell-based therapies still hold the great promise to revolutionize the clinical management of human diseases.It is wildly recognized that poor gene delivery is the limiting factor for most in vivo gene therapies.There has been a long-lasting interest in using viral vectors,especially adenoviral vectors,to deliver therapeutic genes for the past two decades.Among all currently available viral vectors,adenovirus is the most efficient gene delivery system in a broad range of cell and tissue types.The applications of adenoviral vectors in gene delivery have greatly increased in number and efficiency since their initial development.In fact,among over 2000 gene therapy clinical trials approved worldwide since 1989,a significant portion of the trials have utilized adenoviral vectors.This review aims to provide a comprehensive overview on the characteristics of adenoviral vectors,including adenoviral biology,approaches to engineering adenoviral vectors,and their applications in clinical and preclinical studies with an emphasis in the areas of cancer treatment,vaccination and regenerative medicine.Current challenges and future directions regarding the use of adenoviral vectors are also discussed.It is expected that the continued improvements in adenoviral vectors should provide great opportunities for cell and gene therapies to live up to its enormous potential in personalized medicine.Cody S.Lee Elliot S.Bishop Ruyi Zhang Xinyi Yu Evan M.Farina Shujuan Yan Chen Zhao Zongyue Zeng Yi Shu Xingye Wu Jiayan Lei Yasha Li Wenwen Zhang Chao Yang Ke Wu Ying Wu Sherwin Ho Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Russell R.Reid Tong-Chuan He 2017Genes & Diseases2017,4,2:19

3The origin and geological significance of lunar ridges显示文摘Lunar ridges are a kind of familiar linear structures developed on the lunar surface. The distribution pattern, formation mechanism and research significance of lunar ridges are discussed in this paper. Single lunar ridges are usually distributed in the form of broken lineation, and, as whole, lunar ridges are trapezoidal or annular in shape around the maria. As to the formation mechanism, only volcanism or tectonism was emphasized in the past, but the two processes are seldom taken into combined consideration. On the basis of detailed analyses, the authors thought that tectonism is a prerequisite for the formation of lunar ridges, while volcanism is the key factor controlling their particular shapes. 4Process optimization of neodymium chloride solutions precipitated by magnesium bicarbonate显示文摘Rare earths(REs) are of vital importance for the development of new materials and green energy.Magnesium bicarbonate is one of the most recyclable and environmental-friendly precipitant for REs recovery from leaching solutions. Nd_2(CO_3)_3 has difficulties in industrial production. So in this study,the precipitation of neodymium from chloride solution by magnesium bicarbonate are investigated. The effects of feeding method, [HCO_3^-]/[Nd^(3+)] mole ratio, feeding speed and reaction temperature on yield and impurity(magnesia) content are systematically studied. Results show that the impurity(magnesia)content decreases to 0.010 wt% with a yield approaching to 100% obtained under the conditions of[HCO_3^-]/[Nd^(3+)] = 3.00 by parallel flow addition at 50 ℃. The major impurity(magnesia) in rare earth carbonates mainly presents in the form of physical absorption, which can be easily removed by scrubbing. 5An Overview of Finite/Fixed-Time Control and Its Application in Engineering Systems显示文摘The finite/fixed-time stabilization and tracking control is currently a hot field in various systems since the faster convergence can be obtained. By contrast to the asymptotic stability,the finite-time stability possesses the better control performance and disturbance rejection property. Different from the finite-time stability, the fixed-time stability has a faster convergence speed and the upper bound of the settling time can be estimated. Moreover, the convergent time does not rely on the initial information.This work aims at presenting an overview of the finite/fixed-time stabilization and tracking control and its applications in engineering systems. Firstly, several fundamental definitions on the finite/fixed-time stability are recalled. Then, the research results on the finite/fixed-time stabilization and tracking control are reviewed in detail and categorized via diverse input signal structures and engineering applications. 6Alterations in gut microbiota and metabolites associated with altitude-induced cardiac hypertrophy in rats during hypobaric hypoxia challenge显示文摘The gut microbiota is involved in host responses to high altitude.However,the dynamics of intestinal microecology and their association with altitude-related illness are poorly understood.Here,we used a rat model of hypobaric hypoxia challenge to mimic plateau exposure and monitored the gut microbiome,short-chain fatty acids(SCFAs),and bile acids(BAs)over 28 d.We identified weight loss,polycythemia,and pathological cardiac hypertrophy in hypoxic rats,accompanied by a large compositional shift in the gut microbiota,which is mainly driven by the bacterial families of Prevotellaceae,Porphyromonadaceae,and Streptococcaceae.The aberrant gut microbiota was characterized by increased abundance of the Parabacteroides,Alistipes,and Lactococcus genera and a larger Bacteroides to Prevotella ratio.Trans-omics analyses showed that the gut microbiome was significantly correlated with the metabolic abnormalities of SCFAs and BAs in feces,suggesting an interaction network remodeling of the microbiome-metabolome after the hypobaric hypoxia challenge.Interestingly,the transplantation of fecal microbiota significantly increased the diversity of the gut microbiota,partially inhibited the increased abundance of the Bacteroides and Alistipes genera,restored the decrease of plasma propionate,and moderately ameliorated cardiac hypertrophy in hypoxic rats.Our results provide an insight into the longitudinal changes in intestinal microecology during the hypobaric hypoxia challenge.Abnormalities in the gut microbiota and microbial metabolites contribute to the development of high-altitude heart disease in rats.Zhiyuan Pan Yichen Hu Zongyu Huang Ni Han Yan Li Xiaomei Zhuang Jiye Yin Hui Peng Quansheng Gao Wenpeng Zhang Yong Huang Yujun Cui Yujing Bi Zhenjiang Zech Xu Ruifu Yang 2022Science China(Life Sciences)2022,65,10:5
7Ultrafast nonlinear absorption and nonlinear refraction in few-layer oxidized black phosphorus显示文摘We experimentally investigated the nonlinear optical response in few-layer oxidized black phosphorus(OBP) by the femtosecond Z-scan measurement technique, and found that OBP not only possesses strong ultrafast saturable absorption but also a nonlinear self-defocusing effect that is absent in black phosphorus(BP). The saturable absorption property originates mainly from the direct band structure, which is still maintained in OBP. The emergence of self-defocusing might originate from the combined consequences of the oxygen-induced defects in BP. Our experimental findings might constitute the first experimental evidence on how to dynamically tune its nonlinear property, offering an inroad in tailoring its optical properties through chemical modification(oxidation, introducing defects, etc.). 8Investigation on full distribution CNC system based on SERCOS bus显示文摘A full distribution CNC system based on SERCOS bus is studied in accordance with the limitations of traditional PC-based motion card. The conventional PC-based motion control card is dispersed into several autonomous intelligent servo-control units with the function of servo driver. The autonomous intelligent servo-control units realize the loop control of position, velocity and current. Interpolation computation is completed in PC and the computational results are transferred to every autonomous intelligent servo-control unit by high speed SERCOS bus. Software or hardware synchronization technology is used to ensure all servomotors are successive and synchronously running. The communication and synchronization technology of SERCOS are also researched and the autonomous intelligent servo-control card is developed byself. 10Toxicity assessment and histopathological analysis of nano-ZnO against marine fish(Mugilogobius chulae) embryos显示文摘The toxicity of nano-materials has received increasing attention in recent years.Nevertheless,relatively few studies have focused on their oceanic distributions and toxicities.In this study,we assessed nano-ZnO toxicity in marine organisms using the yellowstriped goby(Mugilogobius chulae).The relative differences in nano-ZnO dissolution and dispersal in seawater and fresh water were also investigated.The effects of nano-ZnO on embryonic development,deformity,hatching,mortality,and histopathology were analyzed.In addition,the effects of the Zn^(2+) concentration on M.chulae hatching and mortality were compared.The results showed that nano-ZnO had higher solubility in seawater than in fresh water.Nano-ZnO significantly inhibited hatching.By the fifth day of exposure,the LC_(50) of nano-ZnO was 45.40 mg/L,and the mortality rate spiked.Hatching inhibition and lethality were dose-dependent over a range of1–25 mg/L nano-ZnO.Zn^(2+) inhibited hatching and increased lethality,but its effects were weaker than those of nano-ZnO at the same concentrations.Nano-ZnO also induced spinal bending,oedema,hypoplasia,and other deformities in M.chulae embryos and larvae.Histopathology revealed vacuolar degeneration,hepatocyte and enterocyte enlargement,and morphological abnormalities of the vertebrae.Therefore,nano-ZnO caused malformations in M.chulae by affecting embryonic growth and development.We conclude that nano-ZnO toxicity in seawater was significantly positively correlated with the associated Zn^(2+) concentration and sedimentary behaviour.The toxicity of nano-ZnO was cumulative and showed a critical point,beyond which embryonic and developmental toxicity in marine fish was observed.Jianjun Li Zhanming Chen Ren Huang Zongyu Miao Lei Cai Qingping Du 2018Journal of Environmental Sciences2018,30,11:4

13Platoon control of connected vehicles with heterogeneous model structuresconsidering external disturbances显示文摘This article studies the distributed cooperative control problem with the heterogeneous model structures and external disturbances for the connected vehicle(CV)platoon.We propose a hierarchical framework to separate information flow topology from local dynamics control,which aims to deal with the heterogeneous model structures of CV platoon.This hierarchical framework splits the control scheme into two layers,which include an observer in the upper-level layer and an integral sliding mode(ISM)controller in the lower-level layer.Then,the conditions for the asymptotic stability of the CV platoon are derived and the effectiveness of the ISM controller is demonstrated through the Lyapunov method.The research shows that compared with the traditional methods,the hierarchical framework does not need to specify the topology structure as a commonly used topology.Finally,numerical simulation results are performed to test the effectiveness and superiority of the developed controller.Yongfu Li Zongyu Qin Hao Zhu Srinivas Peeta Xinbo Gao 2022Green Energy and Intelligent Transportation2022,1,3:1

15Physical, Antioxidant and Thermal Shock Properties of Cu/Ti_2AlC Conductive Composites显示文摘Cu/Ti2AlC composites were fabricated by vacuum hot-pressing technique. Phase composition was analyzed by XRD and morphology of fracture was observed by SEM. Physical performance such as density, resistivity, hardness and friction coefficient with different volume fraction of Cu/Ti2AlC composites were studied. When the content of Ti2AlC increased from 10% to 70%, the relative density reduced from 99.38% to 90.56% and the resistivity increased significantly. Hardness reached the maximum value when Ti2AlC was at 60% and friction coefficient declined with the increasing of Ti2AlC. Cu/Ti2AlC composites, showing good conductivity properties and friction performance. Oxidation resistance enhanced obviously with the content of Ti2AlC increasing. 16rransmembrane domain dependent inhibitory unction of FcyRIIB显示文摘FcyRIIB,the only inhibitory IgG Fc receptor,functions to suppress the hyper-activation of immune cells.Numerous studies have illustrated its inhibitory function through the ITIM motif in the cytoplasmic tail of FcyRIIB. 17R158Q and G212S,novel pathogenic compound heterozygous variants in SLC12A3 of Gitelman syndrome显示文摘The dysfunction of Na^(+)-Cl^(−)cotransporter(NCC)caused by mutations in solute carrier family12,member 3 gene(SLC12A3)primarily causes Gitelman syndrome(GS).In identifying the pathogenicity of R158Q and G212S variants of SLC12A3,we evaluated the pathogenicity by bioinformatic,expression,and localization analysis of two variants from a patient in our cohort.The prediction of mutant protein showed that p.R158Q and p.G212S could alter protein’s three-dimensional structure.Western blot showed a decrease of mutant Ncc.Immunofluorescence of the two mutations revealed a diffuse positive staining below the plasma membrane.Meanwhile,we conducted a compound heterozygous model—Ncc^(R156Q/G210S)mice corresponding to human NCC R158Q/G212S.Ncc^(R156Q/G210S)mice clearly exhibited typical GS features,including hypokalemia,hypomagnesemia,and increased fractional excretion of K^(+)and Mg^(2+)with a normal blood pressure level,which made Ncc^(R156Q/G210S)mice an optimal mouse model for further study of GS.A dramatic decrease and abnormal localization of the mutant Ncc in distal convoluted tubules contributed to the phenotype.The hydrochlorothiazide test showed a loss of function of mutant Ncc in Ncc^(R156Q/G210S)mice.These findings indicated that R158Q and G212S variants of SLC12A3 were pathogenic variants of GS.Zongyue Li Huixiao Wu Shuoshuo Wei Moke Liu Yingzhou Shi Mengzhu Li Ning Wang Li Fang Bo Xiang Ling Gao Chao Xu Jiajun Zhao 2022Frontiers of Medicine2022,16,6:1
