1The Post-meiotic Deficicent Anther1 (PDA1) gene is required for post-meiotic anther development in rice显示文摘To understand the molecular mechanism of male reproductive development in the model crop rice,we isolated a complete male sterile mutant post-meiotic deficient anther1 (pda1) from a γ-ray-treated rice mutant library.Genetic analysis revealed that the pda1 mutant was controlled by a recessive nucleus gene.The pda1 mutant anther seemed smaller with white appearance.Histological analysis demonstrated that the pda1 mutant anther undergoes normal early tapetum development without obvious altered meiosis.However,the pda1 mutant displayed obvious defects in postmeiotic tapetal development,abnormal degeneration occurred in the tapetal cells at stage 9 of anther development.Also we observed abnormal lipidic Ubisch bodies from the tapetal layer of the pda1 mutant,causing no obvious pollen exine formation.RT-PCR analysis indicated that the expression of genes involved in anther development including GAMYB,OsC4 and Wax-deficient anther1 (WDA1) was greatly reduced in the pda1 mutant anther.Using map-based cloning approach,the PDA1 gene was finely mapped between two markers HLF610 and HLF627 on chromosome 6 using 3,883 individuals of F2 population.The physical distance between HLF610 and HLF627 was about 194 kb.This work suggests that PDA1 is required for post-meiotic tapetal development and pollen/microspore formation in rice.Lifang Hu Hexin Tan Wanqi Liang Dabing Zhang 2010Journal of Genetics and Genomics2010,37,1:8

13In/ex-situ Raman spectra combined with EIS for observing interface reactions between Ni-rich layered oxide cathode and sulfide electrolyte显示文摘The interfacial instability between Ni-rich layered oxide cathodes and sulfide electrolytes is a serious problem,leading to poor electrochemical properties of all-solid-state lithium batteries(ASSLB).The chemical/electrochemical side reactions are considered to be the origin of the interfacial deterioration.However,the influence of chemical and electrochemical side reactions on the interfacial deterioration is rarely studied specifically.In this work,the deterioration mechanism of the interface between LiNi0.85-xCo0.15AlxO2 and Li10GeP2S12 is investigated in detail by combining in/ex-situ Raman spectra and Electrochemical Impedance Spectroscopy(EIS).It can be determined that chemical side reaction between LiNi0.8Co0.15Al0.05O2 and Li10GeP2S12 will occur immediately once contacted,and the interfacial deterioration becomes more serious after charge-discharge process under the dual effects of chemical and electrochemical side reactions.Moreover,our research reveals that the interfacial stability and the cycle performance of ASSLB can be greatly enhanced by increasing Al-substitution for Ni in LiNi0.85-xCo0.15AlxO2.In particular,the capacity retention of LiNi0.6Co0.15Al0.25O2 cathode after 200 cycles can reach 81.9%,much higher than that of LiNi0.8Co0.15Al0.05O2 cathode(12.5%@200 cycles).This work gives an insight to study the interfacial issues between Ni-rich layered oxide cathode and sulfide electrolyte for ASSLBs.Xuelei Li Huilan Guan Zhijie Ma Ming Liang Dawei Song Hongzhou Zhang Xixi Shi Chunliang Li Lifang Jiao Lianqi Zhang 2020Journal of Energy Chemistry2020,29,9:1
14Mechanoresponsive MiR-138-5p Targets MACF1 to Inhibit Bone Formation显示文摘Mechanical stimuli play an essential role in maintaining bone remodeling and skeletal integrity.Meanwhile,bone can respond to the changes of mechanical condition to adjust its mass and architecture.Clinical studies discover that bedridden patients showed osteoporotic T-scores and low bone mineral density,and long-term immobilized patients presented reduced markers of bone formation.However,as bone formation mediated by osteoblast differentiation is a complex process,the underlying molecular mechanism of mechanical stimuli regulating bone formation is still unclear.Recent evidences show that microRNAs(miRNAs)are involved in mechanical stimuli regulating bone formation or osteoblast differentiation.Nevertheless,no direct evidence identifies mechanoresponsive miRNA in both human and animal bones,and clarifies its mechanoresponsive role under different mechanical conditions(e.g.mechanical unloading,reloading,loading).In the current study,we screened for differentially expressed miRNAs in bone specimens of bedridden patients with fractures,then identified that the expression of miR-138-5p,but not the other miRNAs,altered withbedridden time and was negatively correlated with the expression of the bone formation marker genes Alp(alkaline phosphatase).Moreover,miR-138-5p was up-regulated with reduced bone formation during unloading and down-regulated with increased bone formation during reloading in hind4imb unloaded mice.In addition,miR-138-5p was verified to be responsive to different mechanical unloading condition and cyclic mechanical stretch condition in primary osteogenic cells,respectively.Further in vitro data suggested that mechanoresponsive miR-138-5p directly targeted microtubule actin crosslinking factor 1(MACF1)to inhibit osteoblast differentiation.In vivo,we constructed an osteoblastic miR-138-5p transgenic mice model(TG138)with the Runx2promoter,and found that overexpression miR-138-5p supressed bone formation.Moreover,osteoblast-targeted inhibition of miR-138-5p sensitized bone anabolic response to mechanical loading in TG138 mice.Predominantly,the osteoblast-targeted inhibition of miR-138-5p could counteract bone formation reduction induced by hind limb unloading.Taken together,the mechanoresponsive miR-138-5p inhibited bone anabolic response for developing a novel bone anabolic sensitization strategy.Zhihao Chen Zhao Fan Liang Chao Lifang Hu Chen Lei Zhang Yan Yin Chong Dijie Li Tian Ye Wuxia Qiu Kewen Zhang Chaofei Yang Xiaona Li Li Yu Weiyi Chen Zhang Ge Qian Airong 2019医用生物力学2019,34,A01:1

17The gene expression level and prognosis analysis of membrane protein AP2M1 based on the protein spectrum of exosomes in hepatocellular carcinoma cells显示文摘Objective:Adopting mass spectrometry to analyze the protein spectrum of the exosomes in human hepatocellular carcinoma cells and screening the target membrane protein in order to analyze the relationship between its gene expression level and prognosis.Methods:The exosomes of hepatocellular carcinoma cells HepG2 and 7721 were isolated by ultracentrifugation,and the extracted exosomes were verified by transmission electron microscope, western blotting,and nanosight.The relative quantitative proteomics analysis was used to analyze the protein spectrum of HepG2 and 7721 cells in exosomes.The bioinformatics was used to collect the signal pathway,and the target protein AP2M1 was verified by qRTPCR.Gene Expression Profiling Interactive Analysis(GEPIA),OncoLnc,and The Cancer Genome Atlas(TCGA)database were used to analyze the expression level of AP2M1 and the survival rate of patients with liver cancer.Results:The extracellular exosomes of HepG2 and 7721 cells were isolated by ultracentrifugation.High-purity exosomes were obtained and verified by transmission electron microscopy,western blotting,and nanosight.There were 836 proteins co-expressed by the exosomes of HepG2 and 7721.By the analysis of Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway,a total of 34 pathways that were related to liver cancer were collected,and the target membrane protein AP2M1 was screened.AP2M1 highly expressed in HepG2 cells,HepG2 and Huh7 cells' exosomes were detected by qRT-PCR(P<0.05).The data of GEPIA showed that comparing to the adjacent tissues,gene AP2M1 was highly expressed in hepatocarcinoma tissues.Kaplan-Meier survival curve analysis was performed by OncoLnc.It was found that the survival rate of patients with liver cancer and high expression of AP2M1 was significantly lower than patients with liver cancer and low expression of AP2M1(P<0.01).Conclusion:The high expression of membrane protein AP2M1 in the exosomes of hepatoma cells and the high expression of its gene in liver cancer tissues predicted the low survival rate.Lifang Liang Chunmeng Wei Yasi Li Fengjie Wan Xuejing Huang Min He 2018广西医科大学学报2018,35,11:1
18Scores of amino acid 0D-3D information as applied in cleavage site prediction and better specificity elucidation for human immunodeficiency virus type 1 protease显示文摘A new set of descriptors,namely score vectors of the zero dimension,one dimension,two dimensions and three dimensions(SZOTT),was derived from principle component analysis of a matrix of 1369 structural variables including 0D,1D,2D and 3D information for the 20 coded amino acids. SZOTT scales were then used in cleavage site prediction of human immunodeficiency virus type 1 protease. Linear discriminant analysis(LDA) and support vector machines(SVM) were applied to developing models to predict the cleavage sites. The results obtained by linear discriminant analysis(LDA) and support vector machines(SVM) are as follows. The Matthews correlation coefficients(MCC) by the resubstitution test,leave-one-out cross validation(LOOCV) and external validation are 0.879 and 0.911,0.849 and 0.901,0.822 and 0.846,respectively. The receiver operating characteristic(ROC) analysis showed that the SVM model possesses better simulative and predictive ability in comparison with the LDA model. 20A Detection Method of WLAN Security Mechanisms Based on MAC Frame Resolution显示文摘Security mechanism detection is not only an important content of vulnerabilities evaluation but also the foundation of key strength test for wireless local area network(WLAN). This paper analyzes the specifications of WLAN security mechanisms and points out the defects in design of security mechanisms detection algorithm based on the standards. By capturing and analyzing a large number of Beacon frames from different vendor's access points(APs),we summarize the relevant fields and information elements in a Beacon frame,and present their values or status when an AP is set to every specific security mechanism. A detection algorithm of WLAN security mechanisms is proposed based on the experimental study result and the pseudo code of a reference implementation for the algorithm is designed.
