1Molecular detection of epithelial-mesenchymal transition markers in circulating tumor cells from pancreatic cancer patients:Potential role in clinical practice显示文摘AIM To evaluate the clinical properties of three subpopulations of circulating tumor cells(CTCs) undergoing epithelial-mesenchymal transition(EMT) in pancreatic ductal adenocarcinoma(PDAC) patients.METHODS We identified CTCs for expression of the epithelial cell marker cytokeratin or epithelial cell adhesion molecule(EpCAM)(E-CTC), the mesenchymal cell markers vimentin and twist(M-CTC), or both(E/M-CTC) using the CanPatrol system. Between July 2014 and July 2016, 107 patients with PDAC were enrolled for CTC evaluation. CTC enumeration and classification were correlated with patient clinicopathological features and outcomes.RESULTS CTCs were detected in 78.5% of PDAC patients. The number of total CTCs ranged from 0 to 26 across all 107 patients, with a median value of six. CTC status correlated with lymph node metastasis, TNM stage, distant metastasis, blood lymphocyte counts, and neutrophil-to-lymphocyte ratio(NLR). Kaplan-Meier survival analysis showed that patients with ≥ 6 total CTCs had significantly decreased overall survival and progression-free survival compared with patients with < 6 total CTCs. The presence of M-CTCs was positively correlated with TNM stage(P < 0.01) and distant metastasis(P < 0.01). 2LC-MS determination and pharmacokinetic study of salidroside in rat plasma after oral administration of suspensions of traditional Chinese medicine Erzhi Wan and Fructus Ligustri lucidi显示文摘A simple,rapid and sensitive liquid chromatography-mass spectrometry(LC-MS)method was developed for the determination of salidroside in rat plasma and study of its pharmacokinetics after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi into Wistar rats.Plasma sample of 200 μL was extracted with acetic ether-isopropanol(2∶1)and the extraction was performed on a Kromasil C18 column(150 mm×4.6 mm,5 μm)with the mobile phase of methanol-water(41∶59,v/v)within a run time of 6.0 min.The analyte was monitored with positive electrospray ionization(ESI)by selected ion monitoring(SIM)mode.The target ions were m/z 323.05 for salidroside and m/z 411.05 for internal standard(IS)geniposide.A good linear relationship was obtained over the range of 5.0-500.0 ng/mL and the lower limit of quantification was 5.0 ng/mL.The validated method was successfully applied to the pharmacokinetic study of salidroside in rat plasma after oral administration of suspension of Erzhi Wan and Fructus Ligustri lucidi.Ze-Yun Li Qing Li Jiang Lu Jun-Hong Ling Xi-Hua Yu Xiao-Hui Chen Kai-Shun Bi 2011Journal of Pharmaceutical Analysis2011,1,1:10
3Subtyping Animal Influenza Virus with General Multiplex RT-PCR and Liquichip High Throughput (GMPLex)显示文摘This study developed a multiplex RT-PCR integrated with luminex technology to rapidly subtype simultaneously multiple influenza viruses. Primers and probes were designed to amplify NS and M genes of influenza A viruses HA gene of H1, H3, H5, H7, H9 subtypes, and NA gene of the N1 and N2 subtypes. Universal super primers were introduced to establish a multiplex RT-PCR (GM RT-PCR). It included three stages of RT-PCR amplification, and then the RT-PCR products were further tested by LiquiChip probe, combined to give an influenza virus (IV) rapid high throughput subtyping test, designated as GMPLex. The IV GMPLex rapid high throughput subtyping test presents the following features: high throughput, able to determine the subtypes of 9 target genes in H1, H3, H5, H7, H9, N1, and N2 subtypes of the influenza A virus at one time; rapid, completing the influenza subtyping within 6 hours; high specificity, ensured the specificity of the different subtypes by using two nested degenerate primers and one probe, no cross reaction occurring between the subtypes, no non-specific reactions with other pathogens and high sensitivity. When used separately to detect the product of single GM RT-PCR for single H5 or N1 gene, the GMPLex test showed a sensitivity of 10-5(= 280ELD50) forboth tests and the Luminex qualitative ratio results were 3.08 and 3.12, respectively. When used to detect the product of GM RT-PCR for H5N1 strain at the same time, both showed a sensitivity of 10-4(=2800 ELD50). 4Organic photovoltaic cell with 17% efficiency and superior processability显示文摘The development of organic photoactive materials,especially the newly emerging non-fullerene electron acceptors(NFAs),has enabled rapid progress in organic photovoltaic(OPV)cells in recent years.Although the power conversion efficiencies(PCEs)of the top-performance OPV cells have surpassed 16%,the devices are usually fabricated via a spin-coating method and are not suitable for large-area production.Here,we demonstrate that the fine-modification of the flexible side chains of NFAs can yield 17%PCE for OPV cells.More crucially,as the optimal NFA has a suitable solubility and thus a desirable morphology,the high efficiencies of spin-coated devices can be maintained when using scalable blade-coating processing technology.Our results suggest that optimization of the chemical structures of the OPV materials can improve device performance.This has great significance in larger-area production technologies that provide important scientific insights for the commercialization of OPV cells.Yong Cui Huifeng Yao Ling Hong Tao Zhang Yabing Tang Baojun Lin Kaihu Xian Bowei Gao Cunbin An Pengqing Bi Wei Ma Jianhui Hou 2020National Science Review2020,7,7:7
5Genome-wide identification and expression analyses of cytochrome P450 genes in mulberry(Morus notabilis)显示文摘Cytochrome P450 S play critical roles in the biosynthesis of physiologically important compounds in plants.These compounds often act as defense toxins to prevent herbivory.In the present study,a total of 174 P450 genes of mulberry(Morus notabilis C.K.Schn) were identified based on bioinformatics analyses.These mulberry P450 genes were divided into nine clans and 47 families and were found to be expressed in a tissue-preferential manner.These genes were compared to the P450 genes in Arabidopsis thaliana.Families CYP80,CYP92,CYP728,CYP733,CYP736,and CYP749 were found to exist in mulberry,and they may play important roles in the biosynthesis of mulberry secondary metabolites.Analyses of the functional and metabolic pathways of these genes indicated that mulberry P450 genes may participate in the metabolism of lipids,other secondary metabolites,xenobiotics,amino acids,cofactors,vitamins,terpenoids,and poiyketides.These results provide a foundation for understanding of the structures and biological functions of mulberry P450 genes.Bi Ma Yiwei Luo Ling Jia Xiwu Qi Qiwei Zeng Zhonghuai Xiang Ningjia He 2014Journal of Integrative Plant Biology2014,56,9:5
6Efficacy and economic analysis of Ex- PRESS implantation versus trabeculectomy in uncontrolled glaucoma: a systematic review and Meta-analysis显示文摘AIM:To systematically review the current evidence based on the efficacy and cost of Ex-PRESS implantation and trabeculectomy(Trab)for uncontrolled glaucoma.·METHODS:Clinical trials were identified by electronic databases(Pub Med,EMBASE,ISI Web of science and Cochrane library),and data,such as intraocular pressure(IOP),the complete and qualified success rate,the postoperative complications and the cost,were exacted from these relevant studies.Weighted mean difference(WMD),odds ratio(OR)and 95%confidence intervals(CIs)were calculated and were pooled using a randomeffects model.·RESULTS:Eleven relevant publications and two abstracts met the inclusion criteria.The efficacy of ExPRESS was similar to that of Trab in the percentage of IOP reduction(IOPR%)at 1,2y(WMD:-2.01;95%CI:-7.92-3.90;P=0.50 and WMD:2.89;95%CI:-8.05-13.83;P=0.60,respectively).Ex-PRESS possessed a significant higher complete and qualified success rate(OR:1.59;95%CI:1.07-2.35;P=0.02 and OR:1.74;95%CI:1.06-2.86;P=0.03,respectively).Moreover,Ex-PRESS exerted a significantly lower frequency of hypotony and hyphema than Trab(OR:0.39;95%CI:0.21-0.72;P=0.003 and OR:0.27;95%CI:0.10-0.69;P=0.003,respectively).However,there was no consistent result on the cost between the two groups according to the previous three studies.·CONCLUSION:Both Trab and Ex-PRESS have equivalent efficacy in lowering IOP,yet Ex-PRESS had a lower risk of hypotony and hyphema than Trab.Nevertheless,whether the cost of Ex-PRESS was less than that of Trab should be further investigated to ensure evidence-based conclusion in the long run.Ling Wang Fang Sha Da-Dong Guo Hong-Sheng Bi Jun-Kang Si Yu-Xiang Du Kai Tang 2016International Journal of Ophthalmology(English edition)2016,9,1:5
7Development of A Super-Sensitive Diagnostic Method for African Swine Fever Using CRISPR Techniques显示文摘African swine fever(ASF)is an infectious disease caused by African swine fever virus(ASFV)with clinical symptoms of high fever,hemorrhages and high mortality rate,posing a threat to the global swine industry and food security.Quarantine and control of ASFV is crucial for preventing swine industry from ASFV infection.In this study,a recombinase polymerase amplification(RPA)-CRISPR-based nucleic acid detection method was developed for diagnosing ASF.As a highly sensitive method,RPA-CRISPR can detect even a single copy of ASFV plasmid and genomic DNA by determining fluorescence signal induced by collateral cleavage of CRISPR-lw Cas13 a(previously known as C2c2)through quantitative real-time PCR(q PCR)and has the same or even higher sensitivity than the traditional q PCR method.A lateral flow strip was developed and used in combination with RPA-CRISPR for ASFV detection with the same level of sensitivity of Taq Man q PCR.Likewise,RPA-CRISPR is capable of distinguishing ASFV genomic DNA from viral DNA/RNA of other porcine viruses without any cross-reactivity.This diagnostic method is also available for diagnosing ASFV clinical DNA samples with coincidence rate of 100%for both ASFV positive and negative samples.RPA-CRISPR has great potential for clinical quarantine of ASFV in swine industry and food security.Meishen Ren Hong Mei Ming Zhou Zhen F.Fu Heyou Han Dingren Bi Fuhu Peng Ling Zhao 2021Virologica Sinica2021,36,2:5
8Effects of intensive control of blood glucose and blood pressure on microvascular complications in patients with type Ⅱ diabetes mellitus显示文摘AIM: To evaluate the effects of intensive control of blood glucose and blood pressure on microvascular complications in patients with type Ⅱ diabetes by comparing the therapeutic effects of intensive and standard treatment in patients with type Ⅱ diabetes. METHODS: A total of 107 patients with type Ⅱ diabetes were randomly assigned into intensive and standard treatment groups. Patients in the intensive treatment group received preterax (perindopril/ indapamide) to control blood pressure, and gliclazide (diamicron) MR to control blood glucose. Patients in the standard treatment group received routine medications or placebo. Urinary microalbumin (UMA), urinary creatinine (UCR), the UMA/ UCR ratio, and visual acuity were monitored according to the study design of the ADVANCE trial. Direct ophthalmoscopy and seven-field stereoscopic retinal photography were used to examine the fundi at baseline,and repeated after 5 years of treatment. RESULTS: The characteristics of patients in both groups were well balanced at baseline. After 5 years of treatment, visual acuity was found to be decreased in the standard group (P=0.04), but remained stable in the intensive group. The severity of diabetic retinopathy had not progressed in patients in the intensive group, but had deteriorated in the standard group (P=0.0006). The UMA/UCR ratio was not obviously changed in patients in the intensive group, whereas it was significantly increased in the standard group (P=0.00). 10An introduction to the novel vacuum plume effects experimental system显示文摘This paper introduces a newly developed vacuum Plume effects Experimental System(PES) used for plume effect tests of rocket engines and vacuum heat tests of satellites. The design level, manufacturing technique, and testing capabilities of the PES have reached a highly advanced level at home and abroad. The PES mainly consists of a vacuum chamber, vacuum acquisition system, nitrogen system, helium system, and parameter measurement system. A breakthrough was obtained on the Large Scale Cryo-Pumping System, which was based on a combined liquid nitrogen and liquid helium heat sink. An internal cryopump with a limiting temperature of 4.2 K and an efficient absorption area of 305 m2 was developed. The absorption capability of the cryopump was above 7×107 L/s. Vacuum plume tests were performed in the temperature ranges of ambient temperature, liquid nitrogen, and liquid helium. The experimental results showed that the plume test capability of PES is higher than that of similar foreign equipment STG and CHAFF-4. 14Hepatitis B Immunoprophylactic Failure and Characteristics of the Hepatitis B Virus Gene in Mother-Infant Pairs in Parts of China显示文摘Objective To determine the hepatitis B immunoprophylactic failure rate in infants born to hepatitis B virus(HBV) infected mothers and to characterize HBV genes. Methods HBV-serological testing was conducted for pregnant women and infants. The complete genomes of 30 HBV isolates were sequenced, and genetic characteristics were analyzed using MEGA 5 software. Results The immunoprophylactic failure rate for infants who had completed the scheduled hepatitis B vaccination program was 5.76%(32/556). High sequence homology(99.8%-100%) was observed in 8 of the 10 mother-infant pairs. We identified 19 subgenotype C2 strains, 9 subgenotype B2 strains, and 2 subgenotype C1 strains. Three serotypes were detected: adr(19/30), adw(9/30), and ayw(2/30). The frequency of amino acid mutation of the ‘a' determinant region was 16.67%(5/30), including that of Q129 H, F134 Y, S136 Y, and G145 E. We detected 67 amino acid mutations in the basal core promoter, precore, and core regions of the genome. Conclusion The immunoprophylactic failure rate in infants born to HBV-infected mothers is low in the regions of China examined during this study. 15Genetic profiling of cancer with circulating tumor DNA analysis显示文摘Circulating cell-free tumor DNA(ctDNA) in the blood is DNA released from apoptotic, circulating, and living tumor cells. ctDNA is about 140 nt in length and has a half-life of about 1.5 h. ctDNA analysis provides a noninvasive means to assess the genetic profile of cancer in real time. With the advent of molecular technologies, including digital PCR and massively parallel sequencing(MPS), ctDNA analysis has shown promise as a highly sensitive and specific alternative to conventional tissue biopsy in cancer detection, longitudinal monitoring, and precision therapy. 18Improved method for analyzing the degradation of estrogens in water by solid-phase extraction coupled with ultra performance liquid chromatography-ultraviolet detection显示文摘We established an improved method for the determination of four estrogens including estriol (E3), 17β-estradiol (E2), 17α-ethynyl-estrodiol (EE2) and estrone (E1) in water. The method consisted of solid-phase extraction (0.5 L water) and subsequent analysis of analytes by ultra-performance liquid chromatography (UPLC) with an ultraviolet detector (UVD). Base-line separation was achieved for all studied estrogens using a column (50 mm × 2.1 mm) packed with 1.7 μm particle size stationary phase. Recovery was higher than 88% and detection limits ranged between 12.5-23.7 ng/L for the four estrogens, with the RSD ranging from 7% to 11%. The method was successfully applied to determine E2 and EE2 in simulated natural water, which found that about 70% of E2 was degraded (with a half-life of about 30 hr) within 48 hr and about 55% of EE2 was degraded (with a half-life of about 36 hr). 19High-efficiency acetaldehyde removal during solid-state polycondensation of poly(ethylene terephthalate) assisted by supercritical carbon dioxide显示文摘The concentration of acetaldehyde(AA) is the main quality index of poly(ethylene terephthalate)(PET) used in food and drink packaging.A new method for AA removal has been developed by using supercritical carbon dioxide(sc CO2) during the solid-state polycondensation of PET.The influence factors of AA removal including the temperature,pressure,reaction time and the size of pre-polymer particles are systematically studied in this work.The results indicate that it is a highly efficient way to obtain high molecular weight PET with relative low concentration of AA.Correspondingly,the polymerization degree of PET could increase from 27.9 to 85.6 while the concentration of AA reduces from 0.229 × 10^(-6) to 0.055 × 10^(-6) under the optimal operation conditions of 230 °C,8 MPa and size of 0.30–0.45 mm.Thermodynamic performance tests show the increasing extent of PET crystallinity due to the fact that the plasticization of sc CO_2 is not obvious with extended reaction time,therefore the increasing crystallinity has no significant influence on AA removal.SEM observations reveal that the effects of sc CO_(2-) induced plasticization and swelling on PET increase significantly with the decrease of prepolymer size,and the surface of PET becomes more loose and porous in favor of the AA removal.Zhenhao Xi Tian Liu Wei Si Fenglei Bi Zhimei Xu Ling Zhao 2018Chinese Journal of Chemical Engineering2018,26,6:2
20The initiation and connection of vessel elements in Arabi-dopsis thaliana (Brassicaceae) seedlings显示文摘This research used confocal laser scanning mi-croscopy to examine the initiation and connection of vessel elements in Arabidopsis thaliana (L.) Heynh seedlings. The results indicated that vessel elements differentiated first in the lower portion of cotyledon-node zone (CNZ) 2 h after germination, and then extended downwards to the hypocotyl and root and upwardly to the middle portion of CNZ. 10 h after germination, vessel elements developed in the lower portion of cotyledon blade, the second initiating site, con-necting gradually with that initiated in cotyledon node and forming pinnate-marginal venation. The connection of vessel elements occurred between the upper portions of CNZ and epicotyl-shoot zone 7 d after germination.
