| 2 | Plant innate immunity in rice:a defense against pathogen infection显示文摘A large number of pathogenic microorganisms cause rice diseases that lead to enormous yield losses worldwide. Such losses are important because rice is a staple food for more than half of the world's population. Over the past two decades, the extensive study of the molecular interactions between rice and the fungal pathogen Magnaporthe oryzae and between rice and the bacterial pathogen Xanthomonas oryzae pv. oryzae has made rice a model for investigating plant–microbe interactions of monocotyledons.Impressive progress has been recently achieved in understanding the molecular basis of rice pathogen-associated molecular pattern-immunity and effector-triggered immunity. Here, we briefly summarize these recent advances, emphasizing the diverse functions of the structurally conserved fungal effectors, the regulatory mechanisms of the immune receptor complexes, and the novel strategies for breeding disease resistance. | 3 | Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy. | Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou | 2022 | Cell Research2022,32,6: | 10 |

| 8 | Plasmonic tip internally excited via an azimuthal vector beam for surface enhanced Raman spectroscopy显示文摘The synergy of a plasmonic tip and fiber-based structure light field excitation can provide a powerful tool for Raman examination. Here, we present a method of Raman spectrum enhancement with an Ag-nanoparticles(Ag-NPs)-coated fiber probe internally excited via an azimuthal vector beam(AVB), which is directly generated in a few-mode fiber by using an acoustically induced fiber grating. Theoretical analysis shows that gap mode can be effectively generated on the surface of the Ag-NPs-coated fiber probe excited via an AVB. | 10 | The ANIP1-OsWRKY62 module regulates both basal defense and Pi9-mediated immunity against Magnaporthe oryzae in rice显示文摘During effector-triggered immunity(ETI)against the devastating rice blast pathogen Magnaporthe oryzae,Pi9 functions as an intracellular resistance protein sensing the pathogen-secreted effector AvrPi9 in rice.Importantly,the underlying recognition mechanism(s)between Pi9 and AvrPi9 remains elusive.In this study,We identified a rice ubiquitin-like domain-containing protein(UDP),AVRPI9-INTERACTING PROTEIN 1(ANP1),which is directly targeted by AvrPi9 and also binds to Pi9 in plants.Phenotypic analysis of anip1 mu-tants and plants overexpressing ANIP1 revealed that ANIP1 negatively modulates rice basal defense against M.oryzae.ANiP1 undergoes 26S proteasome-mediated degradation,which can be blocked by both AvrPi9 and Pi9.Moreover,ANIP1 physically associates with the rice WRKY transcription factor OsWRKY62,which also interacts with AvrPi9 and Pi9 in plants.In the absence of Pi9,ANIP1 negatively regulates OsWRKY62 abundance,which can be promoted by AvrPi9.Accordingly,knocking out of OsWRKY62 in a non-Pi9 back-ground decreased immunity against M.oryzae.However,we also observed that OsWRKY62 plays negative roles in defense against a compatible M.oryzae strain in Pi9-harboring rice.Pi9 binds to ANiP1 and OsWRKY62 to form a complex,which may help to keep Pi9 in an inactive state and weaken rice immunity.Furthermore,using competitive binding assays,we showed that AvrPi9 promotes Pi9 dissociation from ANiP1,which could be an important step toward ETI activation.Taken together,our results reveal an immune strategy whereby a UDP-WRKY module,targeted by a fungal effector,modulates rice immunity in distinct ways in the presence or absence of the corresponding resistance protein. | Xuetao Shi Yehui Xiong Kai Zhang Yinshan Zhang Junqi Zhang Lili Zhang Yutao Xiao Guo-Liang Wang Wende Liu | 2023 | Molecular Plant2023,16,4: | 3 |
| 11 | Identification of New Resistance Loci Against Sheath Blight Disease in Rice Through Genome-Wide Association Study显示文摘Sheath blight(SB) caused by the soil borne pathogen Rhizoctonia solani is one of the most serious global rice diseases. Breeding resistant cultivar is the most economical and effective strategy to control the disease. However, no rice varieties are completely resistant to SB, and only a few reliable quantitative trait loci(QTLs) linked with SB resistance have been identified to date. In this study, we conducted a genome-wide association study(GWAS) of SB resistance using 299 varieties from the rice diversity panel 1(RDP1) that were genotyped using 44 000 high-density single nucleotide polymorphism(SNP) markers. Through artificial inoculation, we found that only 36.5% of the tested varieties displayed resistance or moderate resistance to SB. In particular, the aromatic and aus sub-populations displayed higher SB resistance than the tropical japonica(TRJ), indica and temperate japonica sub-populations. Seven varieties showed similar resistance levels to the resistant control YSBR1. GWAS identified at least 11 SNP loci significantly associated with SB resistance in the three independent trials, leading to the identification of two reliable QTLs, qSB-3 and qSB-6, on chromosomes 3 and 6. Using favorable alleles or haplotypes of significantly associated SNP loci, we estimated that both QTLs had obvious effects on reducing SB disease severity and can be used for enhancing SB resistance, especially in improving SB resistance of TRJ sub-population rice varieties.
