| 1 | Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism. | Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu | 2019 | National Science Review2019,6,6: | 9 |

| 4 | Direct Effects of Activin A on the Activation of Mouse Macrophage RAW264.7 Cells显示文摘Macrophages play critical roles in innate immune and acquired immune via secreting pro-inflammatory mediators, phagocytosing microorganisms and presenting antigens. Activin A, a member of transforming growth factor β (TGF-β) superfamily, is produced by macrophages and microglia cells. In this study, we reported a direct effect of activin A as a pro-inflammatory factor on mouse macrophage cell line RAW264.7 cells. Our data revealed that activin A could not only increase IL-1β and IL-6 production from RAW264.7 cells, but also promote pinocytic and phagocytic activities of RAW264.7 cells. In addition, activin A obviously up-regulated MHC II expression on the surface of RAW264.7 cells, whereas did not influence MHC I expression. Activin A also enhanced CD80 expression, which is a marker of activated macrophages, but did not influence RAW264.7 cell proliferation. | 5 | The enhanced protective effects of salvianic acid A: A functionalized nanoparticles against ischemic stroke through increasing the permeability of the blood-brain barrier显示文摘Ischemic stroke is the leading cause of disability and death worldwide. Currently, the only proven treatment for ischemic stroke is restoring the cerebral blood supply. In addition, some of the tissue is damaged during the subsequent reperfusion because of the overproduction of reactive oxygen species (ROS). Furthermore, antioxidant therapies have shown promise in preclinical studies for the treatment of ischemia-reperfusion injury. However, their therapeutic efficacy has been limited because of their low bioavailability in brain. To resolve this issue, we synthesized ROS-responsive, fan-shaped dendrimer nanoparticles (NPs) and conjugated them with a blood-brain barrier (BBB)-targeting peptide, COG1410, and salvianic acid A (SA), which is an effective antioxidant in ischemic stroke. The BBB targeting peptide acts as a ligand of the nanocarrier system and penetrates the BBB through the endocytosis of the ligand receptor. The results showed that T-SA-NPs not only target and accumulate in the infarct area, they also reduce over 2 times of the infarct area and reverse the behavioral deficits in MCAO mice, which illustrates that these NPs have an effective therapeutic effect on the ischemic stroke. In addition, these NPs had no toxicity in any organs of the body. | 17 | High-Order Statistics of Temperature Fluctuations in an Unstable Atmospheric Surface Layer over Grassland显示文摘Skewness(S) and kurtosis(K) of temperature in the surface layer over a grassland are investigated under unstable thermal stratifications. We find that both skewness and kurtosis generally obey Monin–Obukhov similarity theory and tend to be constant values(1.5 and 5.3, respectively) when the stability parameter z/L <-2. Quantitative formulas of the similarity functions are proposed. The temperature probability density function(PDF) is close to Gaussian in near neutral stratification and non-Gaussian in unstable stratification. The influence of coherent motions on the PDF behavior is analyzed using the quadrant analysis technique. It shows that PDF behaviors are controlled by ejections and sweeps. | 18 | A Whole Plant Herbaceous Angiosperm from the Middle Jurassic of China显示文摘In contrast to woody habit with secondary growth,truthful herbaceous habit lacking secondary growth is restricted to angiosperms among seed plants.Although angiosperms might have occurred as early as in the Triassic and herbaceous habit theoretically may have been well adopted by pioneer angiosperms,pre-Cretaceous herbs are missing hitherto,leaving the origin of herbs and evolution of herbaceous angiosperms mysterious.Here we report Juraherba bodae gen.et sp.nov,a whole plant herbaceous angiosperm,from the Middle Jurassic(>164 Ma)at Daohugou Village,Inner Mongolia,China,a fossil Lagerstatten that is worldwide famous for various fossil finds.The angiospermous affinity of Juraherba is ensured by its enclosed ovules/seeds.The plant is small but complete,with physically connected hairy root,stem,leaves,and fructifications.The Middle Jurassic age recommends Juraherba as the earliest record of herbaceous seed plants,demanding a refresh look at the evolutionary history of angiosperms. | HAN Gang LIU Zhongjian LIU Xueling MAO Limi Frederic M.B.
