2Effects of GS3 and GL3.1 for Grain Size Editing by CRISPR/Cas9 in Rice显示文摘Grain size is one of key agronomic traits associated with grain yield and grain quality. Both major quantitative trait loci GS3 and GL3.1 play a predominant role in negative regulation of grain size. In this study, a CRISPR/Cas9-mediated multiplex genome editing system was used to simultaneously edit GS3 and GL3.1 in a typical japonica rice Nipponbare. In T1 generation, we found that gs3 formed slender grain with lower chalkiness percentage, while gs3gl3.1 produced larger grain with higher chalkiness percentage. In terms of other agronomic traits, flag leaf size, grain number and grain yield of both gs3 and gs3gl3.1 mutants were affected. It is noteworthy that gs3 and gs3gl3.1 mutants both led to dramatical reduction of grain number, thereby decreased grain yield. 3Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research. 4Easily prepared and stable functionalized magnetic ordered mesoporous silica for efficient uranium extraction显示文摘Functionalized magnetic Fe_3O_4@SiO_2 composite nanoparticles were prepared by simply embedding iron oxide nanoparticles into MCM-41 through one-step synthesis process, followed by aminopropyls grafting on the mesopore channels, aiming to efficiently and conveniently uptake U(VI) from aqueous solution. The resultant material possesses highly ordered mesoporous structure with large surface area, uniform pore size, excellent thermal stability, quick magnetic response, and desirable acids resistance, confirmed by Fourier transform infrared spectroscopy(FTIR), scanning electron microscopy(SEM), N_2 adsorption/desorption experiments, powder X-ray diffraction(PXRD), and thermogravimetric analysis(TGA). Detailed U(VI) sorption test indicated that this material is indeed an effective U(VI) sorbent with fast sorption kinetics of less than 2 h, large sorption capacity of 160 mg/g at p H 5.0±0.1, and desirable selectivity towards U(VI) ions over a range of competing metal ions. The absorbed U(VI) can be easily desorbed by 0.01 mol/L or more concentrated HNO_3 solution, and the reclaimed sorbent can be reused with no obvious decrease of sorption capacity even after 4 sorption-desorption cycles. 7Statistical method for mapping QTLs for complex traits based on two backcross populations显示文摘Most important agronomic and quality traits of crops are quantitative in nature.The genetic variations in such traits are usually controlled by sets of genes called quantitative trait loci (QTLs),and the interactions between QTLs and the environment.It is crucial to understand the genetic architecture of complex traits to design efficient strategies for plant breeding.In the present study,a new experimental design and the corresponding statistical method are presented for QTL mapping.The proposed mapping population is composed of double backcross populations derived from backcrossing both homozygous parents to DH (double haploid) or RI (recombinant inbreeding) lines separately.Such an immortal mapping population allows for across-environment replications,and can be used to estimate dominance effects,epistatic effects,and QTL-environment interactions,remedying the drawbacks of a single backcross population.In this method,the mixed linear model approach is used to estimate the positions of QTLs and their various effects including the QTL additive,dominance,and epistatic effects,and QTL-environment interaction effects (QE).Monte Carlo simulations were conducted to investigate the performance of the proposed method and to assess the accuracy and efficiency of its estimations.The results showed that the proposed method could estimate the positions and the genetic effects of QTLs with high efficiency.ZHU ZhiHong HAYART Yousaf YANG Jian CAO LiYong LOU XiangYang XU HaiMing 2012Chinese Science Bulletin2012,57,21:2

10An Explicit-Implicit Predictor-Corrector Domain Decomposition Method for Time Dependent Multi-Dimensional Convection Diffusion Equations显示文摘The numerical solution of large scale multi-dimensional convection diffusion equations often requires efficient parallel algorithms.In this work,we consider the extension of a recently proposed non-overlapping domain decomposition method for two dimensional time dependent convection diffusion equations with variable coefficients. 12Breeding dominant genic male sterility restorer line of Brassica napus L.显示文摘To facilate breeding process of Brassica napus,a microspore culture and molecular marker-assisted screening combined system were proposed in this research.At early flowering stage,F1 offspring of hybridized combination HY15 A×HF06 was used as donor for microspore culture to analyze effects of colchicine concentration on embryogenic and diploid rates of microspore.Treatment with 50 mg/L colchicine resulted in embryogenic rate of3.56 embryos/bud,which was substantially higher than control(0.78 embryos/bud).A total of 1,387 embryos and 862 single plants were obtained after induction culture.Ploidy detection was performed for the regenerated plants by flow cytometry.Diploid rates of microspores treated with 50 mg/L and 70 mg/L colchicine were 17.2%and 21.0%respectively,which was significantly higher than control(10.5%).Totally 108 single plants that doubled successfully were randomly selected and screened using molecular marker BE10.Approximately 54 of108 plants generated a 305 bp amplification product,whereas the other 54 plants showed a 398 bp band,thereby satisfying 1:1 separation ratio(x0.052=0.0093).These coincided with field identification results.Findings of this study indicated that homozygous breeding material could be obtained by microspore culture in a short time,thereby remarkably accelerate breeding.Junying Zhang Jianxia Jiang Liyong Yang Meiyan Jiang Yanli Li Weirong Wang Chaocai Sun Jifeng Zhu Xirong Zhou 2020Oil Crop Science2020,5,2:0
13The telomere-to-telomere gap-free reference genome of wild blueberry (Vaccinium duclouxii) provides its high soluble sugar and anthocyanin accumulation显示文摘Vaccinium duclouxii,endemic to southwestern China,is a berry-producing shrub or small tree belonging to the Ericaceae family,with high nutritive,medicinal,and ornamental value,abundant germplasm resources,and good edible properties.In addition,V.duclouxii exhibits strong tolerance to adverse environmental conditions,making it a promising candidate for research and offering wide-ranging possibilities for utilization.However,the lack of V.duclouxii genome sequence has hampered its development and utilization.Here,a high-quality telomere-to-telomere genome sequence of V.duclouxii was de novo assembled and annotated.All of 12 chromosomes were assembled into gap-free single contigs,providing the highest integrity and quality assembly reported so far for blueberry.The V.duclouxii genome is 573.67 Mb,which encodes 41953 protein-coding genes.Combining transcriptomics and metabolomics analyses,we have uncovered the molecular mechanisms involved in sugar and acid accumulation and anthocyanin biosynthesis in V.duclouxii.This provides essentialmolecular information for further research on the quality of V.duclouxii.Moreover,the high-quality telomere-to-telomere assembly of the V.duclouxii genome will provide insights into the genomic evolution of Vaccinium and support advancements in blueberry genetics and molecular breeding.Tuo Zeng Zhijiao He Jiefang He Wei Lv Shixiang Huang Jiawen Li Liyong Zhu Shuang Wan Wanfei Zhou Zhengsong Yang Yatao Zhang Chong Luo Jiawei He Caiyun Wang Liangsheng Wang 2023Horticulture Research2023,10,11:0

15内脏脂肪组织对于Roux-en-Y胃旁路手术后胰岛素敏感性改善的影响:一项对伴有轻度中心性肥胖的中国糖尿病患者的研究显示文摘背景:大多中国2型糖尿病(T2DM)患者都有一个轻度、中心性肥胖。中心性肥胖常合并胰岛素抵抗。本研究目的是评估腹部脂肪组织对合并轻度中心性肥胖的中国糖尿病患者Roux-en-Y胃旁路手术(RYGB)后胰岛素敏感性改善的影响。方法:17例T2DM患者拟行腹腔镜RYGB手术,平均体质指数30.3 kg/m^(2)。术前和术后3个月行高胰岛素-正葡萄糖钳夹和双能X线吸收检测。主要结局指标是胰岛素敏感性与腹部脂肪组织的关系,后者包括术前和术后3个月检测的内脏脂肪组织(VAT)和皮下脂肪组织(SAT)。结果:外周胰岛素敏感指标,包括葡萄糖代谢清除率(M值)和葡萄糖输注率,均于RYGB术后显著增加,人体脂肪总量、VAT重量和SAT重量则显著下降。术前M值与VAT重量呈显著负相关(r=-0.57,P=0.02),但与SAT重量无关。RYGB术后M值的变化值与VAT变化值(r=-0.59,P=0.01)、VAT重量占比(r=-0.66,P<0.01)、VAT面积(r=-0.56,P=0.02)及VAT面积占比(r=-0.57,P=0.02)均呈显著负相关。结论:合并轻度中心性肥胖的中国T2DM患者RYGB手术后,其外周胰岛素敏感性的增加与VAT的减少具有显著关联。RYGB术后,VAT和SAT均显著下降,同时胰岛素敏感明显改善。VAT重量或可作为胃旁路术的一个适应证,用于手术病例的选择。Lei Zhao Liyong Zhu Zhihong Su Weizheng Li Pengzhou Li Yong Liu Shengping Liu Shaihong Zhu 2018Gastroenterology Report2018,6,4:0

19Efficient and Stable Exponential Runge-Kutta Methods for Parabolic Equations显示文摘In this paper we develop explicit fast exponential Runge-Kutta methods for the numerical solutions of a class of parabolic equations.By incorporating the linear splitting technique into the explicit exponential Runge-Kutta schemes,we are able to greatly improve the numerical stability.The proposed numerical methods could be fast implemented through use of decompositions of compact spatial difference operators on a regular mesh together with discrete fast Fourier transform techniques.The exponential Runge-Kutta schemes are easy to be adopted in adaptive temporal approximations with variable time step sizes,as well as applied to stiff nonlinearity and boundary conditions of different types.Linear stabilities of the proposed schemes and their comparison with other schemes are presented.We also numerically demonstrate accuracy,stability and robustness of the proposed method through some typical model problems.Liyong Zhu 2017Advances in Applied Mathematics and Mechanics2017,9,1:0
