| 4 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 5 | Relationship between composite structures and compressive properties in CuZr-based bulk metallic glass system显示文摘Bulk metallic glass (BMG) composites with the austenite B2 phase as reinforcement macroscopically showed strain hardening behavior due to the plasticity induced by martensitic transformation during deformation. Relationship between characteristics of the B2-CuZr reinforcing phase and uniaxial compressive properties of CuZr-based BMG composites was studied. Mechanical properties of these BMG composites were found to depend on not only the reinforced phases but also the amorphous matrix,and the yield and fracture strength can be roughly estimated by the rule of mixture principle. | 7 | Biodegradation of indole by a newly isolated Cupriavidus sp. SHE显示文摘Indole, a typical nitrogen heterocyclic aromatic pollutant, is extensively spread in industrial wastewater. Microbial degradation has been proven to be a feasible approach to remove indole, whereas the microbial resources are fairly limited. A bacterial strain designated as SHE was isolated and found to be an efficient indole degrader. It was identified as Cupriavidus sp. according to 16 SrRNA gene analysis. Strain SHE could utilize indole as the sole carbon source and almost completely degrade 100 mg/L of indole within 24 hr. It still harbored relatively high indole degradation capacity within p H 4–9 and temperature 25°C–35°C. Experiments also showed that some heavy metals such as Mn2+, Pb2+and Co2+did not pose severe inhibition on indole degradation. Based on high performance liquid chromatography–mass spectrum analysis, isatin was identified as a minor intermediate during the process of indole biodegradation. A major yellow product with m/z 265.0605(C15H8N2O3) was generated and accumulated, suggesting a novel indole conversion pathway existed. Genome analysis of strain SHE indicated that there existed a rich set of oxidoreductases, which might be the key reason for the efficient degradation of indole. | 8 | Design and application of 60mer oligonucleotide microarray in SARS coronavirus detection显示文摘The 60mer oligonucleotide microarray was designed and applied to detecting of SARS (severe acute res-piratory syndrome) coronavirus. Thirty 60mer specific oligos were designed to cover the whole genome of the first submit-ted coronavirus strain, according to the sequence of TOR2 (GENEBANK Accession: AY274119). These primers were synthesized and printed into a microarray with 12×12 spots. RNAs were extracted from the throat swab and gargling fluid of SARS patients and reverse-transcripted into the double strand cDNAs. The cDNAs were prepared as re-stricted cDNA fragments by the restriction display (RD) technique and labeled by PCR with the Cy5-universal primer. The labeled samples were then applied to the oligo microar-ray for hybridization. The diagnostic capability of the mi-croarray was evaluated after the washing and scanning steps. The scanning result showed that samples of SARS patients were hybridized with multiple SARS probes on the microar-ray, and there is no signal on the negative and blank controls. These results indicate that the genome of SARS coronavirus can be detected in parallel by the 60mer oligonucleotide mi-croarray, which can improve the positive ratio of the diagno-sis. | 9 | Direct electrochemistry of glucose oxidase on the hydroxyapatite/Nafion composite film modified electrode and its application for glucose biosensing显示文摘A novel glucose biosensor was constructed by immobilizing the glucose oxidase(GOD) on a hydroxyapatite(HAp)/Nafion composite film modified glassy carbon electrode(GCE) and applied to the highly selective and sensitive determination of glucose.With the cooperation of HAp and Nafion,the composite film played an important role in enhancing the stability and sensitivity of the biosensor.The results demonstrate that the GOD adsorbed onto the HAp/Nafion composite film exhibits a pair of welldefined nearly reversible redox peaks and fine catalysis to the oxidation of glucose companied with the consumption of dissolved oxygen.On the basis of the decrease of the reduction current of dissolved oxygen at the applied potential of -0.80 V(vs.SCE) upon the addition of glucose,the concentration of glucose could be detected sensitively and selectively.The decreased reduction current was linear with the concentration of glucose in the range of 0.12―2.16 mM.The detection limit and sensitivity were 0.02 mM(S/N=3) and 6.75 mA·M-1,respectively.All the results demonstrate that HAp/Nafion composite film provides a novel and efficient platform for the immobilization of enzymes and realizes the direct electrochemistry.The composite materials should have potential applications in the fabrication of third-generation biosensors. | MA RongNa WANG Bin LIU Yan LI Jing ZHAO Qian WANG GuoTao JIA WenLi WANG HuaiSheng | 2009 | Science China Chemistry2009,52,11: | 3 |

The problems of deciphering the signals of MFR and inferring the information of its capabilities have posed a great challenge to the Electronic intelligence(ELINT) society. In this paper, we propose a method to reconstruct MFR's search plan from its signals by drawing lessons from the bioinformatics approach'Multiple sequence alignment(MSA)', which is used extensively for gene analysis. In the method, the signal of MFR is represented as a structured task sequence, and is segmented into several blocks according to the inherent periodicity of the search plan. Then, these blocks are aligned optimally to distinguish the tasks relevant to the search function from the others. In this study, green synthesis of gold nanoparticles(AuNPs) was carried out by a newly isolated strain Trichoderma sp. WL-Go. UV–vis spectra of Au NPs showed a surface plasmon resonance peak at 550 nm, and transmission electron microscopy images revealed that the Au NPs were of varied shape with well dispersibility.The optimal conditions for Au NPs synthesis were HAuCl_4 1.0 mmol/L, biomass 0.5 g and pH 7–11. Moreover, the bio-Au NPs could efficiently catalyze the decolorization of various azo dyes.
