| 2 | Fe_3O_4@C nanoparticles as high-performance Fenton-like catalyst for dye decoloration显示文摘Water pollution has become serious environmental problem nowadays. Advanced oxidation processes(AOP) have been widely applied in water treatment.However, traditional Fenton reaction based on Fe2﹢-H2O2 system has obvious drawbacks, which limit its applications In this study, magnetic Fe3O4core-C shell nanoparticles(Fe3O4@C NPs) were prepared for the decoloration of methylene blue(MB) via the co-precipitation followed by the hydrothermal dehydrogenation of glucose. Fe3O4@C NPs showed high catalytic activity of the decoloration of MB through the decomposition of H2O2 in Fenton-like reactions. Fe3O4@C NPs had much higher activity than bare Fe3O4 cores, suggesting the coating of carbon enhanced the catalytic activity. The performance of Fe3O4@C NPs was better at lower pH and higher temperature, but was significantly inhibited in the presence of radical scavenger tertiary butanol. Fe3O4@C NPs could be magnetic separated and regenerated, and maintained with very good catalytic activity. | 3 | Chromosome analysis of esophageal squamous cell carcinoma cell line KYSE 410-4 by repetitive multicolor fluorescence in situ hybridization显示文摘Chromosome aberrations are distinctive features of human malignant tumors. Analysis of chromosomal changes can illuminate the molecular mechanisms underlying the development and progression of cancer. To establish the technique of multicolor fluorescence in situ hybridization (M-FISH) for identifying chromosome aberrations in esophageal carcinoma cell line KYSE 410-4, four pools of 6-color whole-chromosome painting probes have been designed and hybridized on the same metaphase spread by four rounds of repetitive FISH. Repetitive 6-color M-FISH was successfully established and the cytogenetic abnormalities in KYSE 410-4 cells were characterized. Chromosome gains occurred at 2q, 3, 8, 17p, and X. An isochromosome 3q was visualized in the cell line, which might be one intermedi- ate mechanism leading to 3p losses and/or 3q gains. Furthermore, 16 structural arrangements were detected, including four derivative chromosomes. The rearrangement of the centromeric regions accounted for approximately 44% of all rearrangements. The results added a more complete and accurate information of the genetic alterations to the classical cytogenetic description of KYSE 410-4 and provided a detailed cytogenetic background data for appropriate use of the cell line. | 5 | Inhibition of cyclooxygenase-2 activity in subchondral bone modifies a subtype of osteoarthritis显示文摘Osteoarthritis(OA) causes the destruction of joints. Its pathogenesis is still under investigation, and there is no effective diseasemodifying therapy. Here, we report that elevated cyclooxygenase-2(COX-2) expression in the osteocytes of subchondral bone causes both spontaneous OA and rheumatoid arthritis(RA). The knockout of COX-2 in osteocytes or treatment with a COX-2 inhibitor effectively rescues the structure of subchondral bone and attenuates cartilage degeneration in spontaneous OA(STR/Ort)mice and tumor necrosis factor-α transgenic RA mice. Thus, elevated COX-2 expression in subchondral bone induces both OAassociated and RA-associated joint cartilage degeneration. | 6 | Bamboo charcoal enhances cellulase and urease activities during chicken manure composting:Roles of the bacterial community and metabolic functions显示文摘Microbial enzymes are crucial for material biotransformation during the composting process.In this study,we investigated the effects of adding bamboo charcoal(BC)(i.e.,at 5%,10%,and 20%corresponding to BC5,BC10,and BC20,respectively)on the enzyme activity levels during chicken manure composting.The results showed that BC10 could increase the cellulose and urease activities by 56%and 96%,respectively.The bacterial community structure in BC10 differed from those in the other treatments,and Luteivirga,Lactobacillus,Paenalcaligenes,Ulvibacter,Bacillus,Facklamia,Pelagibacterium,Sporosarcina,Cellvibrio,and Corynebacterium had the most important roles in composting.Compared with other treatments,BC10 significantly enhanced the average rates of degradation of carbohydrates(Dxylose(40%)andα-D-lactose(44%))and amino acids(L-arginine(16%),L-asparagine(14%),and L-threonine(52%)).We also explored the associations among the bacterial community and their metabolic functions with the changes in the activities of enzymes.Network analysis demonstrated that BC10 altered the co-occurrence patterns of the bacterial communities,where Ulvibacter and class Bacilli were the keystone bacterial taxa with high capacities for degrading carbon source,and they were related to increases in the activities of cellulase and urease,respectively.The results obtained in this study may help to further enhance the efficiency of composting. | Yanan Yin Chao Yang Jingrui Tang Jie Gu Haichao Li Manli Duan Xiaochang Wang Rong Chen | 2021 | Journal of Environmental Sciences2021,33,10: | 4 |

| 11 | Numerical simulation of the behaviors of test square for prehistoric earthen sites during archaeological excavation显示文摘During the process of archaeological excavation in the regions of Southeast China, collapse of test square usually occurs due to poor site-specific conditions. In this paper, the fast Lagrangian analysis of continua in three dimensions(FLAC3D) is employed to reveal the behaviors of test square. Taking the archaeological works in Liangzhu prehistoric earthen sites as the research background, the paper first introduces the geological setting, excavation procedure and monitoring scheme of the studied test square. Then, the deformation of four sides of the test square is modeled using FLAC3D. By comparison, it shows that the numerical results are consistent with the results from field monitoring. | 17 | Genomics and proteomics of Apis mellifera filamentous virus isolated from honeybees in China显示文摘Apis mellifera filamentous virus(Am FV)is a large DNA virus that is endemic in honeybee colonies.The genome sequence of the Am FV Swiss isolate(Am FV CH–C05)has been reported,but so far very few molecular studies have been conducted on this virus.In this study,we isolated and purified Am FV(Am FV CN)from Chinese honeybee(Apis mellifera)colonies and elucidated its genomics and proteomics.Electron microscopy showed ovoid purified virions with dimensions of 300–500×210–285 nm,wrapping a 3165×40 nm filamentous nucleocapsid in three figure-eight loops.Unlike Am FV CH–C05,which was reported to have a circular genome,our data suggest that Am FV CN has a linear genome of approximately 493 kb.A total of 197 ORFs were identified,among which36 putative genes including 18 baculoviral homologs were annotated.The overall nucleotide similarity between the CN and CH–C05 isolates was 96.9%.Several ORFs were newly annotated in Am FV CN,including homologs of per os infectivity factor 4(PIF4)and a putative integrase.Phylogenomic analysis placed Am FVs on a separate branch within the newly proposed virus class Naldaviricetes.Proteomic analysis revealed 47 Am FV virionassociated proteins,of which 14 had over 50%sequence coverage,suggesting that they are likely to be main structural proteins.In addition,all six of the annotated PIFs(PIF-0–5)were identified by proteomics,suggesting that they may function as entry factors in Am FV infection.This study provides fundamental information regarding the molecular biology of Am FV. | Dahe Yang Jun Wang Xi Wang Fei Deng Qingyun Diao Manli Wang Zhihong Hu Chunsheng Hou | 2022 | Virologica Sinica2022,37,4: | 1 |
| 18 | The novel peptide Nb PPI1 identified from Nicotiana benthamiana triggers immune responses and enhances resistance against Phytophthora pathogens显示文摘In plants, recognition of small secreted peptides, such as damage/danger-associated molecular patterns(DAMPs), regulates diverse processes,includingstressandimmune responses. Here, we identified an SGPS(Ser-Gly-Pro-Ser) motif-containing peptide, Nicotiana tabacum NtPROPPI, and its two homologs in Nicotiana benthamiana, NbPROPPI1 and NbPROPPI2. Phytophthora parasitica infection and salicylic acid(SA) treatment induced NbPROPPI1/2 expression. Moreover, SignalP predicted that the 89-amino acid NtPROPPI includes a 24-amino acid N-terminal signal peptide and NbPROPPI1/2-GFP fusion proteins were mainly localized to the periplasm. Transient expression of NbPROPPI1/2 inhibited P.parasiticacolonization,andNbPROPPI1/2 knockdown rendered plants more susceptible to P. parasitica. An eight-amino-acid segment in the NbPROPPI1 C-terminus was essential for its immune function and a synthetic 20-residue peptide, NbPPI1, derived from the C-terminus of NbPROPPI1 provoked significant immune responses in N. benthamiana. These responses led to enhanced accumulation of reactive oxygen species, activation of mitogen-activated protein kinases, and up-regulation of the defense genes Flg22-induced receptor-like kinase(FRK) and WRKY DNA-binding protein 33(WRKY33). The NbPPI1-induced defense responses require Brassinosteroid insensitive1-associated receptor kinase 1(BAK1). | 19 | Comparative Studies on Preparation of Large Plant DNA Fragments by Pulse Field Gel Electrophoresis显示文摘The preparation of large plant DNA fragments is extremely important to the construction of large insert DNA libraries (YAC, BAC, PAC and TAC). Although several techniques have been developed in each step of large plant DNA fragments preparation, the whole processing remains complicated and difficult. Based on authors research experience and the recent worldwide development in this field, the following aspects are discussed in this paper: techniques of plant high molecular weight (HMW) DNA purification by pre-electrophoresis, the optimal conditions for the partial digestion of the HMW DNA by HindIII, the isolation effects of of large plant DNA fragments (100~400 kb) with different parameters of pulse field gel electrophoresis (PFGE), and the recovery of large DNA fragments. Through comparative studies, the advantages and disadvantages of each technique are discussed and some recommendations are proposed for preparing high quality large plant DNA fragments. The suggested techniques have been used in preparing the large DNA fragments of maize, rice, moss, laver, sea tangle and peach,and similar results are obtained among all the materials.
