| 1 | A phase 3,double-blind,randomized placebo-controlled efficacy and safety study of abiraterone acetate in chemotherapynaı¨ve patients with mCRPC in China,Malaysia,Thailand and Russia显示文摘Objective:This double-blind,placebo-controlled phase 3 study was designed to compare efficacy and safety of abiraterone acetate+prednisone(abiraterone)to prednisone alone in chemotherapy-naı¨ve,asymptomatic or mildly symptomatic metastatic castrationresistant prostate cancer(mCRPC)patients from China,Malaysia,Thailand and Russia.Methods:Adult chemotherapy-naı¨ve patients with confirmed prostate adenocarcinoma,Eastern Cooperative Oncology Group(ECOG)performance status(PS)grade 0e1,ongoing androgen deprivation(serum testosterone<50 ng/dL)with prostate specific antigen(PSA)or radiographic progression were randomized to receive abiraterone acetate(1000 mg,QD)t prednisone(5 mg,BID)or placebo t prednisone(5 mg,BID),until disease progression,unacceptable toxicity or consent withdrawal.Primary endpoint was improvements in time to PSA progression(TTPP).Results:Totally,313 patients were randomized(abiraterone:n Z 157;prednisone:n Z 156);and baseline characteristics were balanced.At clinical cut-off(median follow-up time:3.9 months),80% patients received treatment(abiraterone:n Z 138,prednisone:n Z 112).Median time to PSA progression was not reached with abiraterone versus 3.8 months for prednisone,attaining 58%reduction in PSA progression risk(HR=0.418;p<0.0001).Abirateronetreated patients had higher confirmed PSA response rate(50%vs.21%;relative odds=2.4;p<0.0001)and were 5 times more likely to achieve radiographic response than prednisonetreated patients(22.9%vs.4.8%,p=0.0369).Median survival was not reached.Most common(≥10% abiraterone vs.prednisone-treated)adverse events:bone pain(7%vs.14%),pain in extremity(6%vs.12%),arthralgia(10%vs.8%),back pain(7%vs.11%),and hypertension(15%vs.14%).Conclusion:Interim analysis confirmed favorable benefit-to-risk ratio of abiraterone in chemotherapy-naı¨ve men with mCRPC,consistent with global study,thus supporting use of abiraterone in this patient population. | Dingwei Ye Yiran Huang Fangjian Zhou Keji Xie Vsevolod Matveev Changling Li Boris Alexeev Ye Tian Mingxing Qiu Hanzhong Li Tie Zhou Peter De Porre Margaret Yu Vahid Naini Hongchuan Liang Zhuli Wu Yinghao Sun | 2017 | Asian Journal of Urology2017,4,2: | 3 |

| 3 | High-quality reference genome and annotation aids understanding of berry development for evergreen blueberry(Vaccinium darrowii)显示文摘Vaccinium darrowii Camp(2 n=2×=24)is a native North American blueberry species and an important source oftraits such as low chill requirement in commercial southern highbush blueberry breeding(Vaccinium coymbosum,2n=4 ×=48).We present a chromosomal-scale genome of V.darrowii generated by the combination of PacBio sequencing and high throughput chromatin conformation capture(Hi-C)scaffolding technologies,yielding a total length of 1.06 Gigabases(Gb).Over 97.8%of the genome sequences are scaffolded into 24 chromosomes representing the two haplotypes.The primary haplotype assembly of V,darrowii contains 34,809 protein-coding genes.Comparison to a V.corymbosum haplotype assembly reveals high collinearity between the two genomes with small intrachromosomal rearrangements in eight chromosome pairs.With small RNA sequencing,the annotation was further expanded to include more than 200,000 small RNA loci and 638 microRNAs expressed in berry tissues.Transcriptome analysis across fruit development stages indicates that genes involved in photosynthesis are downregulated,while genes involved in flavonoid and anthocyanin biosynthesis are signi ficantly increased at the late stage of berry ripening.A high-quality reference genome and accompanying annotation of V.darrowii is a signi ficant new resource for assessing the evergreen blueberry contribution to the breeding of southern highbush blueberries. | Jiali Yu Amanda M.Hulse-Kemp Ebrahiem Babiker Margaret Staton | 2021 | Horticulture Research2021,8,1: | 2 |

| 19 | Indirect contact matters:Mid-flight external trunk perturbation increased unilateral anterior cruciate ligament loading variables during jump-landings显示文摘Purpose:To determine the effect of unanticipated mid-flight medial-lateral external perturbation of the upper or lower trunk on anterior cruciate ligament(ACL)loading variable s during jump-landings.Methods:Thirty-two participants performed double-leg vertical jump-landings while bilateral kinematics and kinetics were collected under 6conditions(upper or lower trunk perturbation locations;no,left,or right perturbation directions).Two customized catapult apparatuses were created to apply pushing perturbation to participants near the maximal jump height.Results:The ball contacted participants near the center of mass for the lower-trunk conditions and approximately 23 cm above the center of mass for the upper-trunk conditions.Under upper-trunk perturbation,the contralateral leg demonstrated significantly smaller knee flexion angles at initial contact and greater peak knee abduction angles,peak vertical ground reaction forces,peak knee extension moments,and peak knee adduction moments compared to other legs among all conditions.Under lower-trunk perturbation,the contralateral leg showed significantly smaller knee flexion angles at initial contact and increased peak vertical ground reaction forces and peak knee extension moments compared to legs in the no-perturbation conditions.Conclusion:Mid-flight external trunk pushing perturbation increased ACL loading variables for the leg contralateral to the perturbation.The uppertrunk perturbation resulted in greater changes in ACL loading variables compared to the lower-trunk perturbation,likely due to trunk and ipsilateral leg rotation and more laterally located center of mass relative to the contralateral leg.These findings may help us understand the mechanisms of indirect-contact ACL injuries and develop jump-landing training strategies under mid-flight trunk perturbation to better prevent ACL injury. | Yu Song Ling Li Jacob Layer Raychl Fairbanks Maddy Jenkins Gerwyn Hughes Derek Smith Margaret Wilson Qin Zhu Boyi Dai | 2023 | Journal of Sport and Health Science2023,12,4: | 0 |
