2The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 3Effect of temperature,salinity and irradiance on growth and photosynthesis of Ulva prolifera显示文摘Intensive Pyropia aquaculture in the coast of southwestern Yellow Sea and its subsequent waste, including disposed Ulva prolifera, was speculated to be one of the major sources for the large-scale green tide proceeding in the Yellow Sea since 2007. It was, however, unclear how the detached U. prolifera responded and resumed growing after they detached from its original habitat. In this study, we investigated the growth and photosynthetic response of the detached U. prolifera to various temperature, salinity and irradiance in the laboratory. The photosynthetic rate of the detached U. prolifera was significantly higher at moderate temperature levels(14–27°C)and high salinity(26–32), with optimum at 23°C and 32. Both low(<14°C) and highest temperature(40°C), as well as low salinity(8) had adverse effects on the photosynthesis. Compared with the other Ulva species, U. prolifera showed higher saturated irradiance and no significant photoinhibition at high irradiance, indicating the great tolerance of U. prolifera to the high irradiance. The dense branch and complex structure of floating mats could help protect the thalli and reduce photoinhibition in field. Furthermore, temperature exerted a stronger influence on the growth rate of the detached U. prolifera compared to salinity. Overall, the high growth rate of this detached U. prolifera(10.6%–16.7% d^(–1)) at a wide range of temperature(5–32°C) and salinity(14–32) implied its blooming tendency with fluctuated salinity and temperature during floating. 4Parentage Analysis of Dongfang No.2, a Hybrid of a Female Gametophyte Clone of Laminaria japonica (Laminariales, Phaeophyta) and a Male Clone of L. longissima显示文摘The cultivation of the first filial generation of monoploid gametophyte clones of different Laminaria species (hybrid Laminaria) is an effective way of utilizing heterozygous vigor (heterosis). A female gametophyte clone of L. japonica and a male gametophyte clone of L. longissima were hybridized, generating Dongfang No.2 hybrid Laminaria. The parentage of this hybrid Laminaria was determined using AFLP of total DNA, SNP of the ITS region of ribosomal RNA transcription unit and microsatellite DNA variation at two loci. In addition to 167 AFLP bands shared by Dongfang No.2, L. japonica and L. longissima, Dongfang No.2 hybrid Laminaria shared another 70 and 55 bands with L. japonica and L. longissima, respectively, which were obviously more than 11 bands shared by L. japonica and L. longissima. Dongfang No.2 held both ‘T’ and ‘C’ at position 847 of the ITS region, while ‘T’ at this position was specific for L. japonica and ‘C’ for L. longissima, respectively. Dongfang No.2 also held the microsatellite DNA alleles of two parents together at two microsatellite DNA marker loci. These observations clearly proved that Dongfang No.2 is a true hybrid of L. japonica and L. longissiuma. Unfortunately, the origin of the chloroplast of Dongfang No.2 was not determined based on the variation of RuBisCo spacer. More sequence variants of both ITS region and RuBisCo spacer were identified in Dongfang No.2 and most of them did not exist in either L. japonica or L. longissima. 5Optimization of Protectant, Salinity and Freezing Condition for Freeze-Drying Preservation of Edwardsiella tarda显示文摘Novel preservation condition without ultra-low temperature is needed for the study of pathogen in marine fishes. Freeze-drying is such a method usually used for preservation of terrigenous bacteria. However, studies using freeze-drying method to preserving marine microorganisms remain very limited. In this study, we optimized the composition of protectants during the freeze-drying of Edwardsiella tarda, a fish pathogen that causes systemic infection in marine fishes. We found that the optimal composition of protectant mixture contained trehalose(8.0%), skim milk(12.0%), sodium citrate(2.0%), serum(12.0%) and PVP(2.0%). Orthogonal and interaction analyses demonstrated the interaction between serum and skim milk or sodium citrate. The highest survival rate of E. tarda was observed when the concentration of Na Cl was 10.0, 30.0 and between 5.0 and 10.0 g L^(-1) for preparing TSB medium, E. tarda suspension and protectant mixture, respectively. When E. tarda was frozen at-80℃ or-40℃ for 6 h, its survival rate was higher than that under other tested conditions. Under the optimized conditions, when the protectant mixture was used during freeze-drying process, the survival rate(79.63%–82.30%) of E. tarda was significantly higher than that obtained using single protectant. Scanning electron microscopy(SEM) image indicated that E. tarda was embedded in thick matrix with detectable aggregation. 6Nicotinamide mononucleotide supplementation improves the quality of porcine oocytes under heat stress显示文摘Background:Elevated ambient temperature-caused heat stress is a major concern for livestock production due to its negative impact on animal feed intake,growth,reproduction,and health.Particularly,the germ cells are extremely sensitive to the heat stress.However,the effective approach and strategy regarding how to protect mammalian oocytes from heat stress-induced defects have not been determined.Methods:Germinal vesicle(GV)porcine oocytes were cultured at 41.5℃ for 24 h to induce heat stress,and then cultured at 38.5℃ to the specific developmental stage for subsequent analysis.Nicotinamide mononucleotide(NMN)was dissolved in water to 1 mol/L for a stock solution and further diluted with the maturation medium to the final concentrations of 10μmol/L,20μmol/L,50μmol/L or 100μmol/L,respectively,during heat stress.Immunostaining and fluorescence intensity quantification were applied to assess the effects of heat stress and NMN supplementation on the key processes during the oocyte meiotic maturation.Results:Here,we report that NMN supplementation improves the quality of porcine oocytes under heat stress.Specifically,we found that heat stress resulted in oocyte maturation failure by disturbing the dynamics of meiotic organelles,including the cytoskeleton assembly,cortical granule distribution and mitochondrial function.In addition,heat stress induced the production of excessive reactive oxygen species(ROS)and DNA damage,leading to the occurrence of apoptosis in oocytes and subsequent embryonic development arrest.More importantly,we validated that supplementation of NMN during heat stress restored the meiotic defects during porcine oocyte maturation.Conclusions:Taken together,our study documents that NMN supplementation is an effective approach to improve the quality of oocytes under heat stress by promoting both nuclear and cytoplasmic maturation.Meijie Song Yu Li Yihang Zhou Jiner Yan Xiaohua Zhou Qian Gao Yilong Miao Bo Xiong 2022Journal of Animal Science and Biotechnology2022,13,5:2
7Effects of Different Preservation Methods on Physicochemical Property of Marine Pathogen Vibrio anguillarum显示文摘Freeze-drying,continuous passage and ultra-low temperature cryopreservation are often used to preserve pathogens.In this study,Vibrio anguillarum was rejuvenated by intramuscular infection as the initial strain.The difference between cells preserved with different preservation methods and their initial strains were compared with physiological and biochemical methods and through antibiotics resistance analysis.The composition of protectants for freeze-drying V.anguillarum was optimized.We found that the optimal composition of protectants was 8% of trehalose,12% of skim milk,8.0% of lactose,2.0% of sodium citrate,12.0% of serum and 8.0% of mannitol.The indexes of lysine decarboxylase and urease changed after continuous passage.The urease reaction changed after freeze-drying and freeze-thawing,but the reaction can be restored to the initial after freeze-drying.Based on the antibiotics resistance analyses,the sensitivity of V.anguillarum to different drugs including rifampicin,erythrocin,furazolidone,ceftazidime,lomefloxacin,gentamycin,azithromycin,doxycycline,ampicillin,co-trimoxazole and cefoperazone changed after different treatments,and some of these changes can be restored to the original through activation culture.In sum,compared with cryopreservation and continuous passage,the freeze-drying is more sustainable for the long-term preservation of V.anguillarum,which showed a better effect in maintaining the original characteristics of pathogen.YU Yongxiang ZHANG Zheng WANG Yingeng LIAO Meijie RONG Xiaojun LI Bin ZHANG Hao 2019Journal of Ocean University of China2019,18,6:2
8Influences of immersion bathing in Bacillus velezensis DY-6 on growth performance,non-specific immune enzyme activities and gut microbiota of Apostichopus japonicus显示文摘In this study, the influences of immersion bathing in different concentrations of Bacillus velezensis DY-6 on the body weight gain rate and non-specific immune enzyme activities of the coelom fluid of sea cucumber (Apostichopus japonicus) were determined in order to obtain the optimum bacterial concentration. The gut microbiota change in A. japonicus was then analyzed through high-throughput sequencing during the immersion bathing in B. velezensis DY-6 at the optimum concentration for 49 d. The results illustrate that the body weight growth rate of all bathing groups was higher than that of the control. The highest growth rate (25.3%) was achieved when the bacterial concentration was 1×10^3 CFU/mL. The activities of non-specific immune enzymes (ACP, AKP, SOD and LZM) of all bathing groups increased, and the activities of the enzymes of groups bathed with the bacterium at 1×10^3 and 1×10^4 CFU/mL reached the highest on day 21 and day 28. Taking the growth rate and economic cost into consideration, the optimum concentration of B. velezensis DY-6 was 1×10^3 CFU/mL. The influences of immersion bathing in B. velezensis DY-6 at 1×10^3 CFU/mL on the gut microbiota of A. japonicus were then evaluated through 16S rDNA sequencing analysis. Results showed that the gut microbiota changed with the addition of B. velezensis DY-6, and the richness and diversity of the gut microbiota peaked twice on day 14 and day 21, respectively. In association with the non-specific immune enzyme activities and if day 28 was selected as the dividing point, the community structure of the gut microbiota could be obviously divided into two types. The correlation analysis revealed that the non-specific immune enzyme activities were correlated significantly to some gut bacteria (in the phyla Firmicutes, Proteobacteria, and Bacteroidetes) after immersion bathing in B. velezensis DY-6. 9Low-loading gold in situ doped with sulfur by biomolecule-assisted approach for promoted electrochemical carbon dioxide reduction显示文摘For electrochemical carbon dioxide reduction(CO_(2)RR),CO_(2)-to-CO conversion is considered an ideal route towards carbon neutrality for practical applications.Gold(Au)is known as a promising catalyst with high selectivity for CO;however,it suffers from high cost and low mass-specific activity.In this study,we design and prepare a catalyst featuring uniform S-doped Au nanoparticles on N-doped carbon support(denoted as S-Au/NC)by an in situ synthesis strategy using biomolecules.The S-Au/NC displays high activity and selectivity for CO in CO_(2)RR with a Au loading as low as 0.4 wt.%.The Faradaic efficiency of CO(FECO)for S-Au/NC is above 95%at−0.75 V(vs.RHE);by contrast,the FECO of Au/NC(without S)is only 58%.The Tafel slope is 77.4 mV·dec−1,revealing a favorable kinetics process.Furthermore,S-Au/NC exhibits an excellent long-term stability for CO_(2)RR.Density functional theory(DFT)calculations reveal that the S dopant can boost the activity by reducing the free energy change of the potential-limiting step(formation of the*COOH intermediate).This work not only demonstrates a model catalyst featuring significantly reduced use of noble metals,but also establishes an in situ synthesis strategy for preparing high-performance catalysts.Meijie Tan Xiaoqian Han Sen Ru Chao Zhang Zhouru Ji Zhaolin Shi Guomeng Qiao Yunying Wang Ruixue Cui Qiquan Luo Jiqing Jiao Yaguang Li Tongbu Lu 2023Nano Research2023,16,2:1
10The slope creep law for a soft rock in an open-pit mine in the Gobi region of Xinjiang, China显示文摘The lithology of the strata in the Gobi region of the Xinjiang autonomous region of China is mainly composed of mudstone,silty mudstone,and other soft rocks.Because of the low strength of the rock mass and the serious effects of physical weathering in this area,the slope stability in open-pit mines is poor,and creep deformation and instability can readily occur.Taking the Dananhu No.2 open-pit mine as a typical example,the creep test of a mudstone sample under different stress levels was studied.Then,based on a bottom friction experiment and a FLAC3D numerical simulation,the deformation and failure processes of the slope were analyzed.The stress–displacement curve and the displacement–time curve for the monitoring points were plotted to obtain the relationship between the stress and displacement for the slope of the soft rock.The results showed that the long-term strength of the mudstone was between 8.0 and 8.8 MPa,and that stable creep occurred when the slope was under low stress.The potential failure mode for this type of slope is that the front edge creeps along the weak layer and then a crack is formed at the trailing edge of the slope.When the crack penetrates the weak layer,cutting bedding and bedding sliding occur.The deformation process of the stable creep slope includes an initial deformation stage,an initial creep stage,a constant velocity creep stage and a deceleration creep stage.Hongze Zhao Yu Tian Qingyong Guo Meijie Li Jianwen Wu 2020International Journal of Coal Science & Technology2020,7,2:1
11Effect of esculetin on bone metabolism in ovariectomized rats显示文摘OBJECTIVE: To determine the effect of an esculetin formulation(at 97.4% purity) on osteoporosis, and to investigate the potential underlying molecular mechanism(s).METHODS: Sixty specific pathogen free-grade female Wistar rats were randomly assigned to three groups: blank control(n = 12), sham(n = 12), and model(n = 36). The model group were bilaterally ovariectomized. The sham group had the tissue surrounding the ovaries removed, while the ovaries were retained. After 3 months, the model group was randomly divided into three subgroups: OVX(n = 12), positive control(n = 12), and esculetin(n =12). The positive control group and the esculetin group were intragastrically administered diethylstilbestrol(0.046 mgkd^(-1)), respec·kg^(-1)tively,·d^(-1)) or esculetin(384 mgg^(-1) once per day for 6 consecu··-tive days; medication administration was then stopped for 1 d, before being administered for another 6 consecutive days. All rats were treated for 3months. Samples were collected at the end of the treatment period. An Osteocore3 Digital 2D bone densitometer was used to test the bone mineral density, and histomorphometric analysis was performed to measure bone mass, bone formation,and bone resorption. Enzyme-linked immunosorbent assay analysis was used to measure the serum concentrations of interleukin-6(IL-6), osteoprotegerin(OPG), and receptor activator of nuclear factor-kappa B ligand(RANKL). Immunohistochemistry and in situ hybridization were performed to detect the protein and mRNA expressions of OPG and RANKL in osteoblasts and bone marrow stromal cells.RESULTS: Compared with the OVX group, the esculetin group had significantly greater femoral bone mineral density and tibial trabecular bone volume,and significantly smaller trabecular resorption surface. The percentage of trabecular formation surface, average osteoid width, trabecular bone mineralization rate, and cortical bone mineralization rate did not significantly differ between groups. Compared with the sham group, the esculetin group had significantly decreased serum levels of IL-6and RANKL, and significant downregulation of RANKL protein and mRNA expression levels in osteoblasts and bone marrow stromal cells; however, there was no significant difference between groups in OPG.CONCLUSION: Esculetin can increase bone mass by upregulating RANKL expression in osteoblasts and bone marrow stromal cells, and decreasing serum IL-6 concentration. 12Highly efficient multiplex base editing:One-shot deactivation of eight genes in Shewanella oneidensis MR-1显示文摘Obtaining electroactive microbes capable of efficient extracellular electron transfer is a large undertaking for the scalability of bio-electrochemical systems.Inevitably,researchers need to pursue the co-modification of multiple genes rather than expecting that modification of a single gene would make a significant contribution to improving extracellular electron transfer rates.Base editing has enabled highly-efficient gene deactivation in model electroactive microbe Shewanella oneidensis MR-1.Since multiplexed application of base editing is still limited by its low throughput procedure,we thus here develop a rapid and efficient multiplex base editing system in S.oneidensis.Four approaches to express multiple gRNAs were assessed firstly,and transcription of each gRNA cassette into a monocistronic unit was validated as a more favorable option than transcription of multiple gRNAs into a polycistronic cluster.Then,a smart scheme was designed to deliver one-pot assembly of multiple gRNAs.3,5,and 8 genes were deactivated using this system with editing efficiency of 83.3%,100%and 12.5%,respectively.To offer some nonrepetitive components as alternatives genetic parts of sgRNA cassette,different promoters,handles,and terminators were screened.This multiplex base editing tool was finally adopted to simultaneously deactivate eight genes that were identified as significantly downregulated targets in transcriptome analysis of riboflavin-overproducing strain and control strain.The maximum power density of the multiplex engineered strain HRF(8BE)in microbial fuel cells was 1108.1 mW/m2,which was 21.67 times higher than that of the wild-type strain.This highly efficient multiplexed base editing tool elevates our ability of genome manipulation and combinatorial engineering in Shewanella,and may provide valuable insights in fundamental and applied research of extracellular electron transfer.Yaru Chen Meijie Cheng Yan Li Lin Wang Lixia Fang Yingxiu Cao Hao Song 2023Synthetic and Systems Biotechnology2023,8,1:1

14A Bi-population Cooperative Optimization Algorithm Assisted by an Autoencoder for Medium-scale Expensive Problems显示文摘This study presents an autoencoder-embedded optimization(AEO)algorithm which involves a bi-population cooperative strategy for medium-scale expensive problems(MEPs).A huge search space can be compressed to an informative lowdimensional space by using an autoencoder as a dimension reduction tool.The search operation conducted in this low space facilitates the population with fast convergence towards the optima.To strike the balance between exploration and exploitation during optimization,two phases of a tailored teaching-learning-based optimization(TTLBO)are adopted to coevolve solutions in a distributed fashion,wherein one is assisted by an autoencoder and the other undergoes a regular evolutionary process.Also,a dynamic size adjustment scheme according to problem dimension and evolutionary progress is proposed to promote information exchange between these two phases and accelerate evolutionary convergence speed.The proposed algorithm is validated by testing benchmark functions with dimensions varying from 50 to 200.As indicated in our experiments,TTLBO is suitable for dealing with medium-scale problems and thus incorporated into the AEO framework as a base optimizer.Compared with the state-of-the-art algorithms for MEPs,AEO shows extraordinarily high efficiency for these challenging problems,t hus opening new directions for various evolutionary algorithms under AEO to tackle MEPs and greatly advancing the field of medium-scale computationally expensive optimization.Meiji Cui Li Li MengChu Zhou Jiankai Li Abdullah Abusorrah Khaled Sedraoui 2022IEEE/CAA Journal of Automatica Sinica2022,9,11:1
15Preparation,Performance and Slag Resistant Mechanism of Low Thermal Conductivity Microporous Corundum显示文摘The microporous corundum material was prepared using alumina micro-powder as the main raw material,alumina sol and starch as binders by a wet process,achieving the bulk density of 3. 05 g·cm^(-3),the apparent porosity of 9. 1%,the closed porosity of 12. 3%,the median pore diameter of 0. 43 μm,and the thermal conductivity of 6. 5 W·m^(-1)·K^(-1)at 800 ℃ which is41. 6% lower than that of common corundum. The slag resistance of the microporous corundum material was studied by immersion and compared with that of the common corundum aggregate, and the slag resistant mechanism of microporous corundum material was revealed. The results show that the slag resistance of the microporous corundum material is superior to that of the common corundum aggregate,the SEM and EDX show that on the reaction interface between microporous corundum and molten slag,a continuous isolation layer with a large quantity of CA_2 and CA_6columnar crystals is formed; while the common corundum aggregate reacts with the molten slag interface to form a discontinuous isolation layer of columnar crystals,through which a lot of molten slag corrodes or permeates into the aggregate. The mechanism is mainly that the microporous structure is more advantageous to nucleation and growth of CA_2 and CA_6 columnar crystals; in the reaction with the aggregate,the molten slag gets saturated and the critical solution thickness of the microporous corundum and the common corundum is 0. 16 μm and 0. 34 μm,respectively,this is caused by the smaller microporous corundum aggregate pores; and the smaller pores also increase the second phase ripening rate of microporous corundum,which is 9. 7 times of that of the common corundum.FU Lyuping HUANG Ao GU Huazhi ZHANG Meijie LI Zhengkun ZHAO Yi 2015China's Refractories2015,24,4:0
16Radiative-coupled evaporative cooling:Fundamentals,development,and applications显示文摘As global energy demand continues to rise and climate change accelerates,the need for sustainable and energy-efficient cooling solutions has reached a critical level.Conventional air conditioning systems heavily rely on energy-intensive mechanical cooling,which significantly contributes to both electricity demand and greenhouse gas emissions.Passive cooling strategies,particularly radiative cooling(RC)and evaporative cooling(EC),present an alternative approach by harnessing natural processes for temperature regulation.While standalone RC can be affected by weather conditions and EC relies on water availability,Radiative-coupled EC(REC)offers a versatile and sustainable cooling solution suitable for various applications.Here we summarize an overview of the theoretical foundations and mathematical models of REC,encompassing REC by bulk water(REC-BW),REC by perspiration(REC-P),and REC by sorbed water(REC-SW).Moreover,we explore a range of applications,spanning from industrial processes to personal thermal management,and examine the advantages and challenges associated with each REC approach.The significance of REC lies in its potential to revolutionize cooling technology,reduce energy consumption,and minimize the environmental impact.REC-BW can conserve water resources in industrial cooling processes,while REC-P offers innovative solutions for wearable electronics and textiles.REC-SW’s adaptability makes it suitable for food preservation and future potable cooling devices.By addressing the challenges posed by REC,including water consumption,textile design,and optimization of bilayer structures,we can unlock the transformative potential of REC and contribute to sustainable cooling technologies in a warming world.Li Yu Yimou Huang Weihong Li Changmin Shi Brian W.Sheldon Zhuo Chen Meijie Chen 2024Nano Research Energy2024,3,2:0
17Shifting the Paradigm:A Fresh Look at Physical Activity Frequency and Its Impact on Mental Health,Life Satisfaction,and Self-Rated Health in Adolescents显示文摘As adolescent mental health problems are becoming a more serious issue globally,this paper explores the relationship of physical activity in adolescents and its frequency on mental health as well as examines the mediating effects of life satisfaction and self-rated health in order to provide a reference for the promotion of mental health in adolescents.A sample of 3578 Chinese high school students completed questionnaires assessing their mental health,physical activity frequency,life satisfaction,and self-rated health.The mean SCL-90 value for adolescents was found to be 1.629%,and 24.73%of adolescents had varying degrees of mental health issue.Increased physical activity frequency is positively associated with improved mental health(p<0.001).Additionally,life satisfaction and self-rated health were found to play significant mediating roles in the relationship between physical activity frequency and mental health.Specifically,low-frequency physical activity had the most pronounced mediating effect on mental health through life satisfaction,while high-frequency physical activity exerted the most significant mediating effect on mental health through self-rated health.These findings underscore the importance of promoting physical activity among adolescents and highlight the distinct pathways through which physical activity frequency can influence mental health outcomes.Further research is needed to explore these relationships in diverse populations and settings,as well as to develop targeted intervention.Wenjie Li Yucheng Gao Guoqing Liu Rongkai Hao Meijie Zhang Xiaotian Li 2024International Journal of Mental Health Promotion2024,26,2:0
18Preparation of Microporous Corundum and Its Effect on Properties of Al_2O_3-MgO Castables显示文摘Based on the pore size design of microporous aggregate by numerical simulation applying porous media model,α-Al2O3 micropowder,industrial Al2O3 fine powder,α-Al2O3 micropowder + Ca CO3 fine powder were used as starting materials,respectively,to prepare three microporous corundum aggregates specimens,named as A1,A2 and A3. Two lightweight Al2O3- Mg O castables were prepared by selecting A1 or A3 as aggregate with0. 6 μm of average pore size, then performances of castables with microporous corundum aggregates and castables with tabular corundum aggregates were compared. The results show that:( 1) the microporous corundum aggregates with bulk density of 3. 1- 3. 5g·cm- 3,apparent porosity of 5%,closed porosity of8%- 13%,and smaller thermal conductivity at 800 ℃than tabular corundum,can be prepared by sintering at above 1 800 ℃;( 2) compared to normal Al2O3- Mg O castables,the two lightweight Al2O3- Mg O castables have lower bulk density and higher apparent porosity;lower change rate in dimensions and higher strength firing at 1 500 ℃; and lower thermal conductivity at 600 and 800 ℃;( 3) compared to normal Al2O3- Mg O castables,Al2O3- Mg O castable with A1 has higher penetration index,and that with A3 has higher corrosion index and lower penetration index.GU Huazhi HUANG Ao ZHANG Meijie LI Zhengkun MA Lieying DU Bo ZOU Yang 2014China's Refractories2014,23,4:0
19Importin-αs are required for the nuclear localization and function of the Plasmopara viticola effector PvAVH53显示文摘Plant pathogenic oomycetes deliver a troop of effector proteins into the nucleus of host cells to manipulate plant cellular immunity and promote colonization.Recently,researchers have focused on identifying how effectors are transferred into the host cell nucleus,as well as the identity of the nuclear targets.In this study,we found that the RxLR effector PvAVH53 from the grapevine(Vitis vinifera)oomycete pathogen Plasmopara viticola physically interacts with grapevine nuclear import factor importin alphas(VvImpαand VvImpα4),localizes to the nucleus and triggers cell death when transiently expressed in tobacco(Nicotiana benthamiana)cells.Deletion of a nuclear localization signal(NLS)sequence from PvAVH53 or addition of a nuclear export signal(NES)sequence disrupted the nuclear localization of PvAVH53 and attenuated its ability to trigger cell death.Suppression of two tobacco importin-αgenes,namely,NbImp-α1 and NbImp-α2,by virus-induced gene silencing(VIGS)also disrupted the nuclear localization and ability of PvAVH53 to induce cell death.Likewise,we transiently silenced the expression of VvImpα/α4 in grape through CRISPR/Cas13a,which has been reported to target RNA in vivo.Finally,we found that attenuating the expression of the Importin-αs genes resulted in increased susceptibility to the oomycete pathogen Phytophthora capsici in N.benthamiana and P.viticola in V.vinifera.Our results demonstrate that importin-αs are required for the nuclear localization and function of PvAVH53 and are essential for host innate immunity.The findings provide insight into the functions of importin-αs in grapevine against downy mildew.Tingting Chen Jing Peng Xiao Yin Meijie Li Gaoqing Xiang Yuejin Wang Yan Lei Yan Xu 2021Horticulture Research2021,8,1:0
20Comparative proteomics reveals the response and adaptation mechanisms of white Hypsizygus marmoreus against the biological stress caused by Penicillium显示文摘White Hypsizygus marmoreus is a popular edible mushroom.Its mycelium is easy to be contaminated by Penicillium,which leads to a decrease in its quality and yield.Penicillium could compete for limited space and nutrients through rapid growth and produce a variety of harmful gases,such as benzene,aldehydes,phenols,etc.,to inhibit the growth of H.marmoreus mycelium.A series of changes occurred in H.marmoreus proteome after contamination when detected by the label-free tandem mass spectrometry(MS/MS)technique.Some proteins with up-regulated expression worked together to participate in some processes,such as the non-toxic transformation of harmful gases,glutathione metabolism,histone modification,nucleotide excision repair,clearing misfolded proteins,and synthesizing glutamine,which were mainly used in response to biological stress.The proteins with down-regulated expression are mainly related to the processes of ribosome function,protein processing,spliceosome,carbon metabolism,glycolysis,and gluconeogenesis.The reduction in the function of these proteins affected the production of the cell components,which might be an adjustment to adapt to growth retardation.This study further enhanced the understanding of the biological stress response and the growth restriction adaptation mechanisms in edible fungi.It also provided a theoretical basis for protein function exploration and edible mushroom food safety research.Xiuqing Yang Sizhu Li Xiaohang Li Chenxiao Zhang Meijie Liu Lizhong Guo Lin Liu Hao Yu 2024Food Science and Human Wellness2024,13,3:0
