2Microstructure of fibrils separated from polyacrylonitrile fibers by ultrasonic etching显示文摘Polyacrylonitrile (PAN) fiber is an important precursor fiber for high performance carbon fiber.The properties of the final carbon fiber depend strongly on the nature of the PAN fibers.The PAN fibrils were separated successfully from fibers by ultrasonic etching and were systematically investigated by field emission scanning electron microscopy (FESEM) and high resolution transmission electron microscopy (HRTEM).It is found that in certain ultrasonic etching conditions (at 75±2oC for 6 h with a frequency of 40 kHz) the PAN fibers are dissolved in the 95 wt.% aqueous dimethylsulphoxide (DMSO) solution;the fibrils consisting of numerous periodic lamellae with thickness of 30-45 nm and perpendicular to the fiber axis are separated in the 90 wt.% aqueous DMSO solution;and the fibrils with smooth surface exfoliated from the PAN fibers are obtained in the 70-90 wt.% aqueous DMSO solutions.Inner periodical structure of fibrils was observed in HRTEM,which indicates that there are different densities and two phases in fibrils.The PAN fibers are dissolved layer by layer with increasing ultrasonic etching time.The fiber surface experiences ultrasonic cleaning,selective etching,excessive etching and dissolution,and then the sublayer experiences the same process.There are numerous periodic lamellae in fibrils of nascent fibers.This means that the fibrils with lamellae are formed by orientation and crystallization in shearing field of spinning pipe and drawing stress field of coagulation bath.WANG QiFen,WANG ChengGuo,YU MeiJie,MA Jie,HU XiuYing & ZHU Bo Carbon Fiber Engineering Research Center,Key Laboratory for Liquid-Solid Structural Evolution and Processing of Materials (Ministry of Education),Shandong University,Jinan 250061,China 2010Science China(Technological Sciences)2010,53,6:3
3Optimization of Protectant, Salinity and Freezing Condition for Freeze-Drying Preservation of Edwardsiella tarda显示文摘Novel preservation condition without ultra-low temperature is needed for the study of pathogen in marine fishes. Freeze-drying is such a method usually used for preservation of terrigenous bacteria. However, studies using freeze-drying method to preserving marine microorganisms remain very limited. In this study, we optimized the composition of protectants during the freeze-drying of Edwardsiella tarda, a fish pathogen that causes systemic infection in marine fishes. We found that the optimal composition of protectant mixture contained trehalose(8.0%), skim milk(12.0%), sodium citrate(2.0%), serum(12.0%) and PVP(2.0%). Orthogonal and interaction analyses demonstrated the interaction between serum and skim milk or sodium citrate. The highest survival rate of E. tarda was observed when the concentration of Na Cl was 10.0, 30.0 and between 5.0 and 10.0 g L^(-1) for preparing TSB medium, E. tarda suspension and protectant mixture, respectively. When E. tarda was frozen at-80℃ or-40℃ for 6 h, its survival rate was higher than that under other tested conditions. Under the optimized conditions, when the protectant mixture was used during freeze-drying process, the survival rate(79.63%–82.30%) of E. tarda was significantly higher than that obtained using single protectant. Scanning electron microscopy(SEM) image indicated that E. tarda was embedded in thick matrix with detectable aggregation. 4Nicotinamide mononucleotide supplementation improves the quality of porcine oocytes under heat stress显示文摘Background:Elevated ambient temperature-caused heat stress is a major concern for livestock production due to its negative impact on animal feed intake,growth,reproduction,and health.Particularly,the germ cells are extremely sensitive to the heat stress.However,the effective approach and strategy regarding how to protect mammalian oocytes from heat stress-induced defects have not been determined.Methods:Germinal vesicle(GV)porcine oocytes were cultured at 41.5℃ for 24 h to induce heat stress,and then cultured at 38.5℃ to the specific developmental stage for subsequent analysis.Nicotinamide mononucleotide(NMN)was dissolved in water to 1 mol/L for a stock solution and further diluted with the maturation medium to the final concentrations of 10μmol/L,20μmol/L,50μmol/L or 100μmol/L,respectively,during heat stress.Immunostaining and fluorescence intensity quantification were applied to assess the effects of heat stress and NMN supplementation on the key processes during the oocyte meiotic maturation.Results:Here,we report that NMN supplementation improves the quality of porcine oocytes under heat stress.Specifically,we found that heat stress resulted in oocyte maturation failure by disturbing the dynamics of meiotic organelles,including the cytoskeleton assembly,cortical granule distribution and mitochondrial function.In addition,heat stress induced the production of excessive reactive oxygen species(ROS)and DNA damage,leading to the occurrence of apoptosis in oocytes and subsequent embryonic development arrest.More importantly,we validated that supplementation of NMN during heat stress restored the meiotic defects during porcine oocyte maturation.Conclusions:Taken together,our study documents that NMN supplementation is an effective approach to improve the quality of oocytes under heat stress by promoting both nuclear and cytoplasmic maturation.Meijie Song Yu Li Yihang Zhou Jiner Yan Xiaohua Zhou Qian Gao Yilong Miao Bo Xiong 2022Journal of Animal Science and Biotechnology2022,13,5:2
5Effects of Different Preservation Methods on Physicochemical Property of Marine Pathogen Vibrio anguillarum显示文摘Freeze-drying,continuous passage and ultra-low temperature cryopreservation are often used to preserve pathogens.In this study,Vibrio anguillarum was rejuvenated by intramuscular infection as the initial strain.The difference between cells preserved with different preservation methods and their initial strains were compared with physiological and biochemical methods and through antibiotics resistance analysis.The composition of protectants for freeze-drying V.anguillarum was optimized.We found that the optimal composition of protectants was 8% of trehalose,12% of skim milk,8.0% of lactose,2.0% of sodium citrate,12.0% of serum and 8.0% of mannitol.The indexes of lysine decarboxylase and urease changed after continuous passage.The urease reaction changed after freeze-drying and freeze-thawing,but the reaction can be restored to the initial after freeze-drying.Based on the antibiotics resistance analyses,the sensitivity of V.anguillarum to different drugs including rifampicin,erythrocin,furazolidone,ceftazidime,lomefloxacin,gentamycin,azithromycin,doxycycline,ampicillin,co-trimoxazole and cefoperazone changed after different treatments,and some of these changes can be restored to the original through activation culture.In sum,compared with cryopreservation and continuous passage,the freeze-drying is more sustainable for the long-term preservation of V.anguillarum,which showed a better effect in maintaining the original characteristics of pathogen.YU Yongxiang ZHANG Zheng WANG Yingeng LIAO Meijie RONG Xiaojun LI Bin ZHANG Hao 2019Journal of Ocean University of China2019,18,6:2
6Aggregation behavior of juvenile Neptunea cumingii and eff ects on seed production显示文摘Neptunea cumingii is an important nutrition-rich economic species in China.Juveniles of N.cumingii suff er from high mortality at low temperatures,which has proved a limiting factor in raising seedlings in artifi cial habitats.Previous research has shown that N.cumingii displays aggregation behavior in response to adverse environmental changes.Therefore,we determined the eff ects of temperature,food,size of juvenile snails,substratum type,and density of juvenile snails on the aggregation behavior of N.cumingii.Results show that,at a low(4°C)or a high(22°C)temperature,juvenile snails adjusted to the inhospitable environment by exhibiting increased aggregation behavior.However,their aggregation behaviors diff ered at these two temperatures.There was no signifi cant diff erence in the aggregation rate,but the typical aggregation size was larger at 4°C than at 22°C.At 10°C or 16°C,aggregation behavior of juvenile snails reduced.Aggregation increased in the satiation treatment at 10°C and 16°C.Small-sized juveniles tended to have higher aggregation rates(92.22%)and a larger typical aggregation size.More juveniles were distributed in the bottom of shaded substrata.A larger typical aggregation size or higher density signifi cantly reduced the mortality of juvenile snails at a low temperature(4°C).These results broaden our understanding of gastropod aggregation behavior and can be used to develop and improve commercial breeding strategies and resource recovery for N.cumingii.YU Zhenglin HU Zhi SONG Hao XU Tao YANG Meijie ZHOU Cong ZHANG Tao 2020Journal of Oceanology and Limnology2020,38,5:2

10The slope creep law for a soft rock in an open-pit mine in the Gobi region of Xinjiang, China显示文摘The lithology of the strata in the Gobi region of the Xinjiang autonomous region of China is mainly composed of mudstone,silty mudstone,and other soft rocks.Because of the low strength of the rock mass and the serious effects of physical weathering in this area,the slope stability in open-pit mines is poor,and creep deformation and instability can readily occur.Taking the Dananhu No.2 open-pit mine as a typical example,the creep test of a mudstone sample under different stress levels was studied.Then,based on a bottom friction experiment and a FLAC3D numerical simulation,the deformation and failure processes of the slope were analyzed.The stress–displacement curve and the displacement–time curve for the monitoring points were plotted to obtain the relationship between the stress and displacement for the slope of the soft rock.The results showed that the long-term strength of the mudstone was between 8.0 and 8.8 MPa,and that stable creep occurred when the slope was under low stress.The potential failure mode for this type of slope is that the front edge creeps along the weak layer and then a crack is formed at the trailing edge of the slope.When the crack penetrates the weak layer,cutting bedding and bedding sliding occur.The deformation process of the stable creep slope includes an initial deformation stage,an initial creep stage,a constant velocity creep stage and a deceleration creep stage.Hongze Zhao Yu Tian Qingyong Guo Meijie Li Jianwen Wu 2020International Journal of Coal Science & Technology2020,7,2:1

18Radiative-coupled evaporative cooling:Fundamentals,development,and applications显示文摘As global energy demand continues to rise and climate change accelerates,the need for sustainable and energy-efficient cooling solutions has reached a critical level.Conventional air conditioning systems heavily rely on energy-intensive mechanical cooling,which significantly contributes to both electricity demand and greenhouse gas emissions.Passive cooling strategies,particularly radiative cooling(RC)and evaporative cooling(EC),present an alternative approach by harnessing natural processes for temperature regulation.While standalone RC can be affected by weather conditions and EC relies on water availability,Radiative-coupled EC(REC)offers a versatile and sustainable cooling solution suitable for various applications.Here we summarize an overview of the theoretical foundations and mathematical models of REC,encompassing REC by bulk water(REC-BW),REC by perspiration(REC-P),and REC by sorbed water(REC-SW).Moreover,we explore a range of applications,spanning from industrial processes to personal thermal management,and examine the advantages and challenges associated with each REC approach.The significance of REC lies in its potential to revolutionize cooling technology,reduce energy consumption,and minimize the environmental impact.REC-BW can conserve water resources in industrial cooling processes,while REC-P offers innovative solutions for wearable electronics and textiles.REC-SW’s adaptability makes it suitable for food preservation and future potable cooling devices.By addressing the challenges posed by REC,including water consumption,textile design,and optimization of bilayer structures,we can unlock the transformative potential of REC and contribute to sustainable cooling technologies in a warming world.Li Yu Yimou Huang Weihong Li Changmin Shi Brian W.Sheldon Zhuo Chen Meijie Chen 2024Nano Research Energy2024,3,2:0
19Comparative proteomics reveals the response and adaptation mechanisms of white Hypsizygus marmoreus against the biological stress caused by Penicillium显示文摘White Hypsizygus marmoreus is a popular edible mushroom.Its mycelium is easy to be contaminated by Penicillium,which leads to a decrease in its quality and yield.Penicillium could compete for limited space and nutrients through rapid growth and produce a variety of harmful gases,such as benzene,aldehydes,phenols,etc.,to inhibit the growth of H.marmoreus mycelium.A series of changes occurred in H.marmoreus proteome after contamination when detected by the label-free tandem mass spectrometry(MS/MS)technique.Some proteins with up-regulated expression worked together to participate in some processes,such as the non-toxic transformation of harmful gases,glutathione metabolism,histone modification,nucleotide excision repair,clearing misfolded proteins,and synthesizing glutamine,which were mainly used in response to biological stress.The proteins with down-regulated expression are mainly related to the processes of ribosome function,protein processing,spliceosome,carbon metabolism,glycolysis,and gluconeogenesis.The reduction in the function of these proteins affected the production of the cell components,which might be an adjustment to adapt to growth retardation.This study further enhanced the understanding of the biological stress response and the growth restriction adaptation mechanisms in edible fungi.It also provided a theoretical basis for protein function exploration and edible mushroom food safety research.Xiuqing Yang Sizhu Li Xiaohang Li Chenxiao Zhang Meijie Liu Lizhong Guo Lin Liu Hao Yu 2024Food Science and Human Wellness2024,13,3:0
20Development of a Microfluidics-Based Quantitative Real-Time PCR to Rapidly Identify Photobacterium damselae subsp.damselae with Different Pathogenicity by Detecting the Presence of mcp or dly Gene显示文摘As a marine bacterial pathogen, Photobacterium damselae subsp. damselae(PDD) is distributed in seawater worldwide. It can infect different animals as well as humans, even cause deaths. The highly conserved regions of PDD mcp gene on chromosome and dly gene on plasmid were selected as the target fragments to design the specific primers. Recombinant plasmid standard was prepared based on the primers. With GENECHECKER UF-150 qRT-PCR instrument as the platform, a fluorescence-based quantitative real-time PCR(qRT-PCR) method was established for the detection of PDD. This method can specifically detect PDD and distinguish the highly virulent strains. Additionally, the test results can be qualitatively judged by visualization, while the quantitative detection can be achieved through the standard curve calculation. The minimum limit of detection was 1.0×101 copies μL-1, and the detection time was less than 20 min. In summary, this new method has outstanding advantages, such as strong specificity, high sensitivity, and low site requirements.
