| 1 | Clinical impact of plasma TGF-β1 and circulating TGF-β1 mRNA in diagnosis of hepatocellular carcinoma显示文摘BACKGROUND:Transforming growth factor-β(TGF-β) plays an important role in the regulation of cell growth and differentiation,angiogenesis,extracellular matrix formation,immunosuppression and cancer development. In this study,we investigated the levels of TGF-β1 and TGF-β1 mRNA expression,their relationship with HBV replication,and their diagnostic value for hepatocellular carcinoma(HCC). METHODS:Total RNAs were extracted from HCC samples and matched non-tumor tissues,and from peripheral blood mononuclear cells in HCC patients.TGF-β1 mRNA was amplified by RT-PCR and confirmed by DNA sequencing. The distribution of TGF-β1 expression was assessed by immunohistochemistry.The clinical characteristics were analyzed between TGF-β1 and HBV replication.The diagnostic value of circulating TGF-β1 and TGF-β1 mRNA levels were investigated in HCC patients. RESULTS:The incidence of hepatic TGF-β1 expression was 83.3%in HCC samples,43.3%in the surrounding tissues, 94.7%in the HBV DNA-positive group,and 63.6%in the HBV DNA-negative group.Liver TGF-β1 expression was associated with the degree of HCC differentiation and the status of HBV replication,but not with the size or number of tumors.Circulating TGF-β1 level and incidence of TGF-β1 mRNA were significantly higher in the HCC groupthan in any group of patients with benign liver disease, with a higher sensitivity of 89.5%and a specificity of 94.0% for HCC diagnosis when circulating TGF-β1 levels were >1.2μg/L.No significant correlation was found between TGF-β1 expression and AFP level or tumor size.Combining TGF-β1 level and serum AFP raised the detection rate to 97.4%. | 2 | In-situ U-Pb dating of uraninite by fs-LA-ICP-MS显示文摘In this study,the Pb/U fractionation between zircon and uraninite during femtosecond Laser Ablation Inductively Coupled Plasma Mass Spectrometry(fs-LA-ICP-MS)analysis was studied in detail.The results show significant Pb/U fractionation between zircon and uraninite during fs-LA-ICP-MS analysis that when calibrated against the zircon standard M257,the obtained U-Pb age of the Chinese national uraninite standard GBW04420 is 17%older than the recommended value.Thus,the accurate in-situ U-Pb dating of uraninite by LA-ICP-MS requires matrix-matched external standards for calibration.Uraninite in thin sections of two U-mineralized leucogranite from the Gaudeanmus in Namibia was analyzed by a fs-LA-ICP-MS equipped with a Signal Smooth Device(SSD),using laser spot and frequency of 10μm and 1 Hz,respectively.When calibrated using GBW04420 as the external standard,two samples give weighted mean 206Pb/238U ages of 504±3 Ma(2σ,n=21)and503±3 Ma(2σ,n=22),and only one of two samples yields a concordia U-Pb age of 507±1 Ma(2σ,n=21).These results are consistent with ID-TIMS U-Pb ages of 509±1 and 508±12 Ma and are also indistinguishable from zircon U-Pb upper intercept ages of 506±33 Ma(2σ,n=29)and 501±51 Ma(2σ,n=29).The present study shows that in-situ U-Pb dating of uraninite can deliver more reliable formation ages of the deposit than dating coeval high-U zircon because the latter commonly suffer significant Pb loss after formation.Our results confirm that GBW04420 is an ideal matrix matching standard for in-situ U-Pb dating of uraninite. | ZONG KeQing CHEN JinYong HU ZhaoChu LIU YongSheng LI Ming FAN HongHai MENG YanNing | 2015 | Science China Earth Sciences2015，58，10： | 22 |
| 3 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 4 | Cerebral small vessel disease or intracranial large vessel atherosclerosis may carry different risk for future strokes显示文摘Background The effect of cerebral small vessel disease(CSVD)and intracranial arterial stenosis(ICAS)on stroke outcomes remains unclear.Methods Data of 1045 patients with minor stroke or transient ischaemic attack(TIA)were obtained from 45 sites of the Clopidogrel in High-Risk Patients with Acute Non-disabling Cerebrovascular Events(CHANCE)trial.We assessed the associations of burdens of CSVD and ICAS with new strokes and bleeding events using multivariate Cox regression models and those with modified Rankin Scale(mRS)scores using ordinal logistic regression models.Results Among the 1045 patients,CSVD was present in 830 cases(79.4%)and ICAS in 460(44.0%).Patients with>1 ICAS segment showed the highest risk of new strokes(HR 2.03,95%CI 1.15 to 3.56,p=0.01).No association between CSVD and the occurrence of new strokes was found.The presence of severe CSVD(common OR(cOR)2.01,95%CI 1.40 to 2.89,p<0.001)and>1 ICAS segment(cOR 2.15,95%CI 1.57 to 2.93,p<0.001)was associated with higher mRS scores.Severe CSVD(HR 10.70,95%CI 1.16 to 99.04,p=0.04),but not ICAS,was associated with a higher risk of bleeding events.Six-point modified CSVD score improved the predictive power for bleeding events and disability.Interpretation CSVD is associated with more disability and bleeding events,and ICAS is associated with an increased risk of stroke and disability in patients with minor stroke and TIA at 3 months.CSVD and ICAS may represent different vascular pathologies and play distinct roles in stroke outcomes. | Huimin Chen Yuesong Pan Lixia Zong Jing Jing Xia Meng Yuyuan Xu Hongyi Yan Xingquan Zhao Liping Liu Hao Li S Claiborne Johnston Yongjun Wang Yilong Wang | 2020 | Stroke & Vascular Neurology2020，5，2： | 13 |

| 9 | H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak显示文摘AIM To investigate the protective efficacy ofH2 strain attenuated live hepatitis A vaccines(H2-strain vaccines)in hepatitis A(HA)outbreaks.METHODS With the permission of theirparents,5551 pre-school and grade 1-3 primaryschool children were inoculated with 1 dose(106.5TCID50)of H2-strain vaccines in anonrandomized,controlled trial conducted inFucheng County,Hebei Province in May 1997.Another 6485 children in the same grades andcompatible in gender and age were enrolled ascontrols.Epidemiological and serological surveywas conducted to evaluate the protectiveefficacy of the vaccines.ELISA was used todetect serum IgM anti-HAV.RESULTS HA outbreak started in early May1998,peaked in the middle of the same month,and lasted about 80 days.Overall 302 HA caseswere found,192(53.58%)were 5-9 years old.One vaccinee and 25 control cases were found tohave hepatitis A,which account for 0.28%(1/356)and 5.92%(25/422)of all vaccinees andcontrols in the 14 villages,respectively.The protective efficacy of vaccines was 95.27%(95% Cl:85.83%-104.72%).In subjects testedfor anti-HAV IgM from 13 villages,1(0.40%)overt and 11(4.06%)asymptomatic HAV caseswere found in 271 vaccinees,but 21(6.69%)ofovert and asymptomatic ones were found in 314controls.CONCLUSION H2 strain vaccines were excellent in preventing overt hepatitis A, but not so effective in preventing asymptomatic hepatitis A virus infection. | 16 | MAGI3 Suppresses Glioma Cell Proliferation via Upregulation of PTEN Expression显示文摘Objective To investigate the role and molecular mechanism of membrane-associated guanylate kinase inverted 3(MAGI3) in glioma cell proliferation. Methods The expression levels of MAGI3 and PTEN were assessed in glioma samples by Western blotting. MAGI3 was stably transfected into C6 glioma cells to obtain C6-MAGI3 cells. Then, the proliferation, the expression levels of MAGI3 and PTEN, and Akt phosphorylation were evaluated in C6 and C6-MAGI3 cells. Xenograft tumor models were established by subcutaneous injection of C6 and C6-MAGI3 cells into nude mice, and the growth rates of xenografts in the mice were compared. The potential role of MAGI3 expression in PI3K/Akt signaling activation was further investigated by examining the correlation between MAGI3 expression and the expression of PI3K/Akt signaling downstream target genes in a glioma dataset using gene set enrichment analysis(GSEA). Results Expression levels of MAGI3 and PTEN were significantly downregulated in gliomas. Overexpression of MAGI3 in the glioma C6 cell line upregulated PTEN protein expression, inhibited the phosphorylation of Akt, and suppressed cell proliferation. MAGI3 overexpression also inhibited the growth of C6 glioma tumor xenografts in nude mice. Analysis based on the GEO database confirmed the negative correlation between activation of PI3K/Akt pathway and MAGI3 mR NA levels in human glioma samples. Conclusion The loss of MAGI3 expression in glioma may enhance the proliferation of glioma cells via downregulation of PTEN expression, leading to the activation of the PI3K/Akt pathway. Both environmental and energy aspects should be considered to improve these chemical processes, favoring reaction conditions that involve room temperature and ambient O2 pressure. In the present work, hollow titanium dioxide nanospheres were fabricated via template-free method. The prepared samples were characterized by X-ray diffraction, N2 adsorption–desorption isotherms, transmission electron microscopy, and X-ray photoelectron spectroscopy. The photocatalytic activity was evaluated by photocatalytic oxidation of benzyl alcohol to benzaldehyde with visible light under atmospheric pressure at room temperature.
