1Simultaneous virus identification and characterization of severe unexplained pneumonia cases using a metagenomics sequencing technique显示文摘Many viruses can cause respiratory diseases in humans.Although great advances have been achieved in methods of diagnosis,it remains challenging to identify pathogens in unexplained pneumonia(UP) cases.In this study,we applied next-generation sequencing(NGS) technology and a metagenomic approach to detect and characterize respiratory viruses in UP cases from Guizhou Province,China.A total of 33 oropharyngeal swabs were obtained from hospitalized UP patients and subjected to NGS.An unbiased metagenomic analysis pipeline identified 13 virus species in 16 samples.Human rhinovirus C was the virus most frequently detected and was identified in seven samples.Human measles virus,adenovirus B 55 and coxsackievirus A10 were also identified.Metagenomic sequencing also provided virus genomic sequences,which enabled genotype characterization and phylogenetic analysis.For cases of multiple infection,metagenomic sequencing afforded information regarding the quantity of each virus in the sample,which could be used to evaluate each viruses' role in the disease.Our study highlights the potential of metagenomic sequencing for pathogen identification in UP cases.Xiaohui Zou Guangpeng Tang Xiang Zhao Yan Huang Tao Chen Mingyu Lei Wenbing Chen Lei Yang Wenfei Zhu Li Zhuang Jing Yang Zhaomin Feng Dayan Wang Dingming Wang Yuelong Shu 2017Science China(Life Sciences)2017,60,3:14

3Advanced fabrication for electrospun three-dimensional nanofiber aerogels and scaffolds显示文摘Electrospinning is a versatile strategy for creating nanofiber materials with various structures,which has broad application for a myriad of areas ranging from tissue engineering,energy harvesting,filtration and has become one of the most important academic and technical activities in the field of material science in recent years.In addition to playing a significant role in the construction of two-dimensional(2D)nanomaterials,electrospinning holds great promise as a robust method for producing three-dimensional(3D)aerogels and scaffolds.This article reviews and summarizes the recent advanced methods for fabricating electrospun three-dimensional nanofiber aerogels and scaffolds,including gas foaming,direct electrospinning of 3D nanofibrous scaffold,short nanofibers assembling into 3D aerogels/scaffolds,3D printing,electrospray,origami and cell sheet engineering,centrifugal electrospinning,and other methods.Besides,intriguing formation process,crosslinking pathway,properties,and applications of 3D aerogels and scaffolds are also introduced.Taken together,these aerogels and scaffolds with various excellent features present tremendous potential in various fields.Yujie Chen Muhammad Shafiq Mingyue Liu Yosry Morsi Xiumei Mo 2020Bioactive Materials2020,5,4:12
4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 7Artificial intelligence in drug design显示文摘Thanks to the fast improvement of the computing power and the rapid development of the computational chemistry and biology,the computer-aided drug design techniques have been successfully applied in almost every stage of the drug discovery and development pipeline to speed up the process of research and reduce the cost and risk related to preclinical and clinical trials.Owing to the development of machine learning theory and the accumulation of pharmacological data, the artificial intelligence(AI) technology, as a powerful data mining tool, has cut a figure in various fields of the drug design, such as virtual screening,activity scoring, quantitative structure-activity relationship(QSAR) analysis, de novo drug design, and in silico evaluation of absorption, distribution, metabolism, excretion and toxicity(ADME/T) properties. 8Cloning and expression of two sterol C-24 methyltransferase genes from upland cotton(Gossypium hirsuturm L.)显示文摘Brassinosteroids (BRs) are an important class of plant steroidal hormones that are essential in a wide variety of physiological proc-esses. Two kinds of intermediates,sitosterol and campesterol,play a crucial role in cell elongation,cellulose biosynthesis,and accumula-tion. To illuminate the effects of sitosterol and campesterol on the development of cotton (Gossypium hirsuturm L.) fibers through screening cotton fiber EST database and contigging the candidate ESTs,two key genes GhSMT2-1 and GhSMT2-2 controlling the sitos-terol biosynthesis were cloned from developing fibers of upland cotton cv. Xuzhou 142. The full length of GhSMT2-1 was 1,151 bp,in-cluding an 8 bp 5′-untranslated region (UTR),a 1,086 bp open reading frame (ORF),and a 57 bp 3′-UTR. GhSMT2-1 gene encoded a polypeptide of 361 amino acid residues with a predicted molecular mass of 40 kDa. The full length of GhSMT2-2 was 1,166 bp,including an 18 bp 5′-UTR,a 1,086 bp ORF,and a 62 bp 3′-UTR. GhSMT2-2 gene encoded a polypeptide of 361 amino acid residues with a pre-dicted molecular mass of 40 kDa. The two deduced amino acid sequences had high homology with the SMT2 from Arabidopsis thaliana and Nicotiana tabacum. Furthermore,the typical conserved structures characterized by the sterol C-24 methyltransferase,such as region I (LDVGCGVGGPMRAI),region Ⅱ (IEATCHAP),and region Ⅲ (YEWGWGQSFHF),were present in both deduced proteins. Southern blotting analysis indicated that GhSMT2-1 or GhSMT2-2 was a single copy in upland cotton genome. Quantitative real-time RT-PCR analysis revealed that the highest expression levels of both genes were detected in 10 DPA (day post anthesis) fibers,while the lowest levels were observed in cotyledon and leaves. The expression level of GhSMT2-1 was 10 times higher than that of GhSMT2-2 in all the organs and tissues detected. These results indicate that the homologue of sterol C-24 methyltransferase gene was cloned from upland cotton and both GhSMT2 genes play a crucial role in fiber elongation. 10Hydrolyzed seawater pearl tablet modulates the immunity via attenuating Th1/Th2 imbalance in an immunosuppressed mouse model显示文摘OBJECTIVE: To investigate whether Hydrolyzed Seawater Pearl tablet(HSPT) could modulate the Th1/Th2 imbalance in an immunosuppressed mouse model with Th1 to Th2 shift induced by Cyclosporine A(Cs A) which can be used in the clinical treatment of Th2 to Th1 shift diseases, and explore the possible mechanism for the adjuvant therapeutic efficacy of HSPT on recurrent respiratory infections(RRI) and acquired immune deficiency syndrome(AIDS).METHODS: The mice were randomly divided into six groups of five animals each, namely normal group, model group, lentinan polysaccharide tablet(LPT) group and three HPST treated groups. HPST treated groups were administered with HPST(0.51,1.02, 2.04 g/kg) via intragastric gavage(i.g) for 30 consecutive days. LPT used as reference drug for positive control, LPT group was administered with LPT(8.2 mg/kg) for 30 consecutive days. Normal group and model group were received distilled water. The animals in model group, LPT group and HPST treated groups were injected intraperitoneally with Cs A(50 mg/kg) to establish the immunosuppressed mice model with Th1 to Th2 shift on the20 th, 22 nd and 24 th day, one hour after the administration of the respective treatment. Animals were sacrificed one hour after the last administration to collect blood and splenic tissue. The proportion of T cells including CD8+ and CD4+ T cells, Th1 and Th2 in peripheral blood of experimental mice were measured by flow cytometric. The protein level in serum and m RNA level in splenic tissue of experimental mice for interleukin(IL)-2, IL-12, interferon-γ(IFN-γ), IL-4, IL-6, IL-10 and IL-13 were measured by enzyme linked immunosorbent assay and fluorescence quantitative polymerase chain reaction respectively.RESULTS: HSPT elevated the proportion of T cells including both CD8+ and CD4+ T cells, in which the proportion of Th1 and Th2 cells increased, while the ratio of Th1/Th2 cells decreased in peripheral blood of the immunosuppressed mouse model with Th1 to Th2 shift induced by Cs A. 11White-light emitting single-phase phosphor La_3Si_6N_(11):Dy^(3+),Tb^(3+):Color tunable emission, thermal stability and energy transfer process显示文摘A novel white-light emitting single-phase phosphor La_3 Si_6 N_(11):Dy^(3+),exhibiting two emission peaks centering at 475 and 575 nm, was prepared via conventional solid-state reactions. The structure and morphology of La_3 Si_6 N_(11):Dy^(3+)/Tb^(3+) were investigated by X-ray diffraction(XRD) and scanning electron microscopy(SEM). The emission colors can be tuned from white to yellow-green through increasing the Tb^(3+) concentration in La_3 Si_6 N_(11):Dy^(3+),Tb^(3+), The mechanism of energy transfer(ET) from Dy^(3+) to Tb^(3+) is confirmed according to the excitation,emission spectra and decay lifetimes curve. 12Performance of S-CO_2 Brayton Cycle and Organic Rankine Cycle(ORC) Combined System Considering the Diurnal Distribution of Solar Radiation显示文摘This paper researches the performance of a novel supercritical carbon dioxide(S-CO_2) Brayton cycle and organic Rankine cycle(ORC) combined system with a theoretical solar radiation diurnal distribution. The new system supplies all solar energy to a S-CO_2 Brayton cycle heater, where heat releasing from the S-CO_2 cooler is stored in the thermal storage system which is supplied to the ORC. Therefore, solar energy is kept at a high temperature, while at the same time the thermal storage system temperature is low. This paper builds a simple solar radiation diurnal distribution model. The maximum continuous working time, mass of thermal storage material, and parameter variations of the two cycles are simulated with the solar radiation diurnal distribution model. 10 organic fluids and 5 representative thermal storage materials are compared in this paper, with the mass and volume of these materials being shown. The longer the continuous working time is, the lower the system thermal efficiency is. The maximum continuous working time can reach 19.1 hours if the system provides a constant power output. 13Discovering unusual structures from exception using big data and machine learning techniques显示文摘Recently, machine learning(ML) has become a widely used technique in materials science study. Most work focuses on predicting the rule and overall trend by building a machine learning model. However,new insights are often learnt from exceptions against the overall trend. In this work, we demonstrate that how unusual structures are discovered from exceptions when machine learning is used to get the relationship between atomic and electronic structures based on big data from high-throughput calculation database. For example, after training an ML model for the relationship between atomic and electronic structures of crystals, we find AgO2 F, an unusual structure with both Ag3+and O22à, from structures whose band gap deviates much from the prediction made by our model. 14Graphitizing N-doped mesoporous carbon nanospheres via facile single atom iron growth for highly efficient oxygen reduction reaction显示文摘Single atom catalyst is of great importance for the oxygen reduction reaction(ORR).However,facile preparation of single atom catalyst without using well-designed precursors or labor-intensive acid leaching remains an urgent challenge.Herein,a simple pyrolysis of Fe3+-loaded mesoporous phenolic resin(mPF)-melamine precursor is used to prepare the single atom iron-anchored N-doped mesoporous graphitic carbon nanospheres(Fe/N-MGN).Investigation of the synthesis reveals the appropriate Fe-assisted catalysis effect and mPF template effect,which not only spurs the highly graphitic porous framework of Fe/N-MGN with plentiful pyridinic N/graphitic N,but also assures the dispersed single atom Fe anchoring without elaborated procedures.As a result,the as-synthesized Fe/N-MGN demonstrates high catalytic activity,good durability and excellent methanol tolerance for ORR.This work promises a facile method to regulate the graphitic carbon growth and single atom Fe loading for the highly efficient electrocatalysis.Yunshi Xu Liping Zhu Xuexue Cui Mingyu Zhao Yaling Li Leilei Chen Weicun Jiang Ting Jiang Shuguang Yang Yi Wang 2020Nano Research2020,13,3:5
15Mechanical and thermal properties of RETaO4(RE=Yb,Lu,Sc)ceramics with monoclinic-prime phase显示文摘As candidate thermal/environmental barrier coatings(T/EBCs),the structure characteristics and comprehensive properties of monoclinic-prime(m')RETaO4(RE=Yb,Lu,Sc)with excellent Al2O3/SiO2 chemical compatibility are studied.Excellent thermal insulation protection will be provided by m'RETaO4 due to their low thermal conductivity(~1.6 Wm^-1 K^-1,900℃)and prominent thermal radiation resistance,which is much better than those of YSZ(~2.5 Wm^-1 K^-1,1000℃)and La-12 Zr2O7(~2.0 Wm^-1 K,900℃).The thermal expansion coefficients(TECs)are 3.0–8.0×10^-6 K^-1(200-1200℃),which is suitable for T/EBCs applications.Furthermore,absence of phase transition and extraordinary chemical compatibility with Al2O3/SiO2 up to 1500℃indicate the potential application prospect.The documented governing mechanisms of m'RETaO4 properties will enable researchers to promote their application in the future investigation.Lin Chen Mingyu Hu Jun Guo Xiaoyu Chong Jing Feng 2020Journal of Materials Science & Technology2020,52,17:4

17rAAV/ABAD-DP-6His attenuates oxidative stressinduced injury of PC12 cells显示文摘Our previous studies have revealed that amyloid β(Aβ)-binding alcohol dehydrogenase(ABAD) decoy peptide antagonizes Aβ42-induced neurotoxicity. However, whether it improves oxidative stress injury remains unclear. In this study, a recombinant adenovirus constitutively secreting and expressing Aβ-ABAD decoy peptide(rAAV/ABAD-DP-6His) was successfully constructed. Our results showed that rAAV/ABAD-DP-6His increased superoxide dismutase activity in hydrogen peroxide-induced oxidative stress-mediated injury of PC12 cells. Moreover, rAAV/ABADDP-6His decreased malondialdehyde content, intracellular Ca2+ concentration, and the level of reactive oxygen species. rAAV/ABAD-DP-6His maintained the stability of the mitochondrial membrane potential. In addition, the ATP level remained constant, and apoptosis was reduced. 182-D DOA Estimation Using Off-Grid Sparse Learning via Iterative Minimization with L-Parallel Coprime Array显示文摘An L-parallel coprime array is designed and an Off-grid sparse learning via iterative minimization(OGSLIM) algorithm is proposed in order to improve the performance of Two-dimensional direction-of-arrival(2-D DOA) estimation. The L-parallel coprime array consists of two parts, one is a parallel coprime array and the other one is a linear coprime array perpendicular to the parallel coprime array. The OGSLIM algorithm is based on sparse Bayesian framework and can learn the off-grid parameter.
