| 1 | Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers. | 2 | Inheritance of Perpetual Blooming in Rosa chinensis ‘Old Blush'显示文摘Perpetual blooming is one of the most important biological and economical traits in modern rose, while the genetic basis underlining the control of this trait is poorly investigated. With an aim in dissecting the genetic determinism of perpetual blooming, we developed six rose populations(OB, W, F1, F2, BC1 OB and BC1W) derived from a WOB population [interspecific diploid hybridization between Rosa chinensis ‘Old Blush'(OB) and R. wichuriana ‘Basye's Thornless'(W)]. Perpetual blooming is absent both in a F1 population with 296 individuals and a BC1 W population(W as the backcross parent) with 150 individuals. However, the perpetual blooming trait showed a typical 3︰1 segregation in a backcross population BC1 OB with OB as the backcross parent. In this population with 300 individuals, 83 plants had the perpetual blooming phenotype while the other 217 featured non-perpetual blooming, indicating that the perpetual blooming trait is very likely controlled by two recessive genes in R. chinensis(rpb1 and rpb2). | 5 | Mechanism of Deca-BDE-induced apoptosis in Neuro-2a cells:Role of death-receptor pathway and reactive oxygen species-mediated mitochondrial pathway显示文摘Decabromodiphenyl ether(BDE-209) is a prevalent polybrominated diphenyl ether(PBDE)congener known to have neurotoxicity. Effects of BDE-209 on Neuro-2a cells were performed in the present study and the possible apoptotic pathway was discussed. Results indicated that BDE-209 induced Neuro-2a cell apoptosis, increased the protein expression of Fas and Fas-associated death domain-containing protein(FADD) and activated the caspase-8 and-3activities in a concentration-dependent manner, inferring the death-receptor pathway was involved in the apoptotic process. Meanwhile, BDE-209 exposure increased the Bax/Bcl-2 ratio and decreased the cellular mitochondrial membrane potential(MMP) which led to cytochrome C released to the cytoplasm. The intracellular caspase-9 was elevated simultaneously,which caused downstream caspase cascade and triggered cell apoptosis. Moreover, BDE-209 exposure increased cellular reactive oxygen species(ROS) level in a concentration-dependent manner and the addition of N-acetyl-L-cysteine(NAC), known as ROS scavengers, obviously reduced the apoptotic rate and a positive relationship was observed between the degree of apoptosis blocking and the loss of MMP and ROS production. We thus concluded that BDE-209 induced Neuro-2a cell apoptosis via the combination of the death-receptor signaling pathway and the mitochondrial signaling pathway. | 7 | Using DGT to Assess Cadmium Bioavailability to Ryegrass as Influenced by Soil Properties显示文摘A generally accepted method for predicting the bioavailability and transfer of cadmium(Cd) from soil to plants has not yet been established. In this study, the diffusive gradients in thin films(DGT) and conventional extraction methods for metal fractions were used to investigate how effective these methods were at predicting Cd bioavailability to ryegrass(Lolium perenne) and to assess whether these holistic variables were independent of the soil property variables. The influences of soil properties on the predictive capabilities of the different methods were evaluated using multivariate statistics. The Cd concentrations in the shoots and roots of ryegrass correlated more closely with the effective Cd concentration measured by DGT(CE), compared with those with soil total Cd, pore water Cd, and ethylenediaminetetraacetic acid disodium salt(Na2EDTA)- and acetic acid(HOAc)-extractable Cd. When soil properties were included in a stepwise multiple linear regression, Cd transfer to L. perenne was negatively influenced by p H and positively influenced by organic matter(OM). The multivariate statistics showed that the adjusted correlation coefficients for the plots involving soil total Cd, pore water Cd, and Na2EDTA- and HOAc-extractable Cd had been significantly improved after considering the influences of soil properties, which suggested that these methods were p H and OM dependent. For CE, the relationship was p H independent or only dependent in strongly acidic soils. These results suggested that the main soil factors affecting bioavailability had the lowest impact on the DGT technique. | 10 | Down-staging depth score could be a survival predictor for locally advanced gastric cancer patients after preoperative chemoradiotherapy显示文摘Objective:The predictive effect of preoperative chemoradiotherapy(CRT)is low and difficult in guiding individualized treatment.We examined a surrogate endpoint for long-term outcomes in locally advanced gastric cancer patients after preoperative CRT.Methods:From April 2012 to April 2019,95 patients with locally advanced gastric cancer who received preoperative concurrent CRT and who were enrolled in three prospective studies were included.All patients were stage T_(3/4) N_(+).Local control,distant metastasis-free survival(DMFS),disease-free survival(DFS)and overall survival(OS)were evaluated.Clinicopathological factors related to long-term prognosis were analyzed using univariate and multivariate analyses.The down-staging depth score(DDS),which is a novel method of evaluating CRT response,was used to predict long-term outcomes.Results:The median follow-up period for survivors was 30 months.The area under the curve(AUC)of the receiver operating characteristic(ROC)curve predicted by the DDS was 0.728,which was better than the pathological complete response(pCR),histological response and ypN0.Decision curve analysis further affirmed that DDS had the largest net benefit.The DDS cut-off value was 4.pCR and ypN0 were associated with OS(P=0.026 and 0.049).Surgery and DDS are correlated with DMFS,DFS and OS(surgery:P=0.001,<0.001 and<0.001,respectively;and DDS:P=0.009,0.013 and 0.032,respectively).Multivariate analysis showed that DDS was an independent prognostic factor of DFS(P=0.021).Conclusions:DDS is a simple,short-term indicator that was a better surrogate endpoint than pCR,histological response and ypN0 for DFS. | Ning Li Xin Wang Yuan Tang Dongbin Zhao Yihebali Chi Lin Yang Liming Jiang Jun Jiang Jinming Shi Wenyang Liu Hua Ren Hui Fang Yu Tang Bo Chen Ningning Lu Hao Jing Shunan Qi Shulian Wang Yueping Liu Yongwen Song Yexiong Li Jing Jin | 2021 | Chinese Journal of Cancer Research2021,33,4: | 1 |

| 14 | Complex granular flows of sticky-wet material on flip-flow screens:Calibration of discrete element simulations显示文摘Discrete element method(DEM)is an effective approach for studying the screening process of flip-flow screens.However,there have been few studies focusing on the thick layer of sticky-wet particles on flip-flow screens.To achieve accurate simulations of the thick layer of sticky-wet particles on a flip-flow screen,firstly,the movement law of particle flow was studied,and a multi-regime combination cali-bration method based on characteristics of particle flow regimes was proposed.Based on the Plackett-Burman experiment,the curse of dimensionality caused by multi-state and multi-contact parameters was overcome.Subsequently,the lifting cylinder,rotating drum,and trampoline tests were carried out to obtain macroscopic reference values under various granular flow regimes.The calibration results were then determined using the response surface method and climbing algorithm.Finally,the calibration results were tested at both macroscopic and mesoscopic scales and compared with a commonly used calibration method.The results demonstrate that the calibration method,which considers the multi-state characteristics,improves simulation accuracy by 2%-10%and reduces the simulation error to less than 10%,thus meeting the requirements for engineering optimization of flip-flow screens. | Ningning Xu Sen Tang Dongdong Lin Runhui Geng Xinwen Wang Xihui Liang | 2024 | Particuology2024,84,1: | 0 |
| 15 | Comparative transcriptomic and metabonomic analysis revealed the relationships between biosynthesis of volatiles and flavonoid metabolites in Rosa rugosa显示文摘Rosa rugosa is not only cultivated as a landscaping plant,but also used in cosmetics,the medical and food industries.However,little information is currently available on the gene regulatory networks involved in its scent and color biosynthesis and metabolism.In this study,R.rugosa Thunb.f.rosea Rehd with red petals(RR)and its white petal variant(WR),were used to study the molecular mechanisms in flower color and scent.Sixty-five differential flavonoid metabolites and 15 volatiles were found to have significant differences between RR and WR.Correspondingly,the key regulators(MYB-bHLH-WD40)of anthocyanin synthesis pathway and their structural genes involved in flavonoid biosynthesis,benzenoid/phenylpropanoid biosynthesis,terpenoid biosynthesis pathways were also found to be differentially expressed by comparative transcriptome.Further,qPCR permitted the identification of some transcripts encoding proteins that were putatively associated with scent and color biosynthesis in roses.Particularly,the results showed that the ACT gene(encoding CoA geraniol/citronellol acetyltransferase,GeneID:112190420),which expressed lower in WR,was involved in three pathways:flavonoid biosynthesis,phenylpropanoid biosynthesis and terpenoid biosynthesis,however,GT5(anthocyanin glycosylation gene,GeneID:112186660),expressed higher in WR,was involved in both flavonoid and phenylpropanoid biosynthesis pathways.These results suggested that ACT and GT5 might play important roles in regulating the relationship of color pigmentation and volatile emission. | Dedang Feng Hao Zhang Xianqin Qiu Hongying Jian Qigang Wang Ningning Zhou Yu Ye Jun Lu Huijun Yan Kaixue Tang | 2021 | Ornamental Plant Research2021,1,1: | 0 |
| 16 | Genome-wide identification and evolutionary analysis of MLO gene family in Rosaceae plants显示文摘Mutants lacking wild-type MLO(Mildew resistance Locus O)proteins show broad-spectrum resistance to the powdery mildew fungus,and dysregulated cell death control,with spontaneous cell death in response to developmental or abiotic stimuli.In order to understand the evolution and divergence patterns of the MLO gene family in Rosaceae plants,we analysed systematically genome-wide data from Fragaria vesca,Prunus persica,Prunus mume,Malus domestica,Pyrus bretschneideri and Rubus occidentalis based on bioinformatics methods.Using three phylogenetic methods(the neighbour-joining,maximum likelihood,and Bayesian methods),we identified 117 MLO genes from 6 Rosaceae species.The results of all three phylogenetic analysis methods supported that these genes were divided into six clades.Conserved motif analysis found that only motif 2 was present in all MLO proteins and had 3 nearly invariant amino acid residues.The findings indicated that motif 2 might be shared by the MLO gene family.The structural features of these genes showed large variations in sequence length among different species,although the lengths and the numbers of exons exhibited high degrees of similarity.Selective pressure analysis showed extremely significant differences in all 6 clades,with 2,1,and 1 site(s)under significant positive selection detected in clades III,IV,and VI,respectively.These positive selection sites were important driving forces for the promotion of the functional differentiation of the MLO genes.Functional divergence analysis showed that the significantly divergent sites were located within the domains of the MLO genes.Functional distance analysis showed that the clade V had more conservative functions and might have retained more original functions during the evolutionary process.However,clade I may have undergone extensive altered functional constraints as a specialised functional role.Moreover,the most original function of the MLO genes in Rosaceae could be related to the evolution of their resistance to powdery mildew,which then gradually evolved into functions such as the regulation of flower development,the control of root morphology,and seed evolution due to the different evolutionary rates after gene duplication.These results provide a theoretical basis for further studies of the molecular evolutionary patterns of the plant MLO gene family. | Yongxian Tian Qigang Wang Hao Zhang Ningning Zhou Huijun Yan Hongying Jian Shubin Li Guisheng Xiang Kaixue Tang Xianqin Qiu | 2022 | Horticultural Plant Journal2022,8,1: | 0 |

| 18 | Adaptive ultra-hypofractionated whole-pelvic radiotherapy in high-risk and very high-risk prostate cancer on 1.5-Tesla MR-Linac:Estimated delivered dose and early toxicity results显示文摘Background: Magnetic resonance(MR)-guided ultra-hypofractionated radiotherapy with whole-pelvic irradiation(UHF-WPRT)is a novel approach to radiotherapy for patients with high-risk(HR)and very high-risk(VHR)prostate cancer(PCa).However,the inherent complexity of adaptive UHF-WPRT might inevitably result in longer on-couch time.We aimed to estimate the delivered dose,study the feasibility and safety of adaptive UHF-WPRT on a 1.5-Tesla MR-Linac.Methods: Ten patients with clinical stage T3a-4N0-1M0-1c PCa,who consecutively received UHF-WPRT,were enrolled prospectively.The contours of the target and organ-at-risks on the position verification-MR(PV-MR),beam-on 3D-MR(Bn-MR),and post-MR(after radiotherapy delivery)were derived from the pre-MR data by deformable image registration.The physician then manually adjusted them,and dose recalculation was performed accordingly.GraphPad Prism 9(GraphPad Prism Software Inc.)was utilized for conducting statistical analyses.Results: In total,we collected 188 MR scans(50 pre-MR,50 PV-MR,44 Bn-MR,and 44 post-MR scans).With median 59 min,the mean prostate clinical target volume(CTV)-V_(100%)was 98.59%±2.74%,and the mean pelvic CTVp-V_(100%)relative percentages of all scans was 99.60%±1.18%.The median V29 Gy change in the rectal wall was−2%(−18%to 20%).With a median follow-up of 9 months,no patient had acute Common Terminology Criteria for Adverse Events(CTCAE)grade 2 or more severe genitourinary(GU)or gastrointestinal(GI)toxicities(0%).Conclusion: UHF-RT to the prostate and the whole pelvis with concomitant boost to positive nodes using an Adapt-To-Shape(ATS)workflow was technically feasible for patients with HR and VHR PCa,presenting only mild GU and GI toxicities.The estimated target dose during the beam-on phase was clinically acceptable based on the 3D-MR-based dosimetry analysis.Clinical trial registration Chinese Clinical Trial Registry ChiCTR2000033382. | Linrui Gao Ran Wei Shirui Qin Yuan Tian Wenlong Xia Yongwen Song Shulian Wang Hui Fang Yu Tang Hao Jing Yueping Liu Yuan Tang Shunan Qi Bo Chen Yexiong Li Nianzeng Xing Ningning Lu | 2024 | Chronic Diseases and Translational Medicine2024,10,1: | 0 |

| 20 | Low-content and highly effective zoned Rh and Pd three-way catalysts for gasoline particulate filter potentially meeting Euro 7显示文摘The next-generation Euro 7 standard proposed much lower pollutant limits from gasoline vehicles,specifically for CO and NO_(x),which would be challenging for the three-way catalysts(TWCs)utilized commercially to eliminate these pollutants.TWCs with reductive(Rh)and oxidative(Pd)active components on gasoline particulate filters(TWC on GPF)play importantly auxiliary roles in the remediation of CO and NO_(x)downstream the close coupled TWCs to meet their emission targets.Here,a low-content Rh-based TWC(0.17 wt%)zoned with a less expensive Pd-based TWC(0.29 wt%)for GPF applications(cGPF)is reported using improved colloidal deposition method.The supporting of Rh on Y-stabilized ZrO_(2)rather than on CeO_(2)-ZrO_(2)inhibits the formation of inactive Ce rhodate species,while Pd on CeO_(2)-ZrO_(2)not only guarantees the high oxygen storage capacity(OSC)but also enhances catalytic activity.The layout of the front one-fifth in volume being 0.29 wt%Pd on Ce_(0.43)Zr_(0.5)7O_(2)and the rear four-fifths being 0.17 wt%Rh on Zr_(0.85)Y_(0.15)O_(2)prevents the possible alloying of Rh with Pd.The highly effective zoned Rh and Pd TWCs show synergistic three-way activity before and after severe hydrothermal aging at 1000℃with 10%water for24 h,which could be potential choices for close coupled GPF application to satisfy the upcoming stringent emission standards,such as Euro 7 and China 6b. | Ying Xin Hanxue Zhang Pan Li Ningning Qu Ahui Tang Fuzhen Yang Dongxu Han Junxiu Jia Jin Wang Lirong Zheng Zhaoliang Zhang | 2023 | Journal of Rare Earths2023,41,6: | 0 |
