1Timing of the Wudangshan, Yaolinghe volcanic sequences and mafic sills in South Qinling: U-Pb zircon geochronology and tectonic implication显示文摘The Wudangshan, Yaolinghe volcanic-sedimentary sequences and doleritic-gabbroic sills comprise the largest exposed Precambrian basement in South Qinling. Zircons separated from 5 volcanic-pyroclas- tic samples of the Wudangshan Group, 2 volcanic samples of the Yaolinghe Group and one sample for the mafic sills were used for U-Pb dating by laser ablation-inductively coupled plasma mass spec- trometry (LA-ICPMS). The results reveal that the Wudangshan volcanic sequence was formed at (755±3) Ma (a weighted mean from the 5 samples, MSWD=0.47), whereas the Yaolinghe volcanic suite and the mafic sill were crystallized at (685±5) (2 samples, MSWD=0.36) and (679±3) Ma (MSWD=1.6), respectively, which are equal to each other within analysis errors. These ages are markedly younger than those previously documented for the rocks. The newly obtained ages for the Wudangshan and Yaolinghe Groups are identical to those of the bottom Liantuo and slightly older than those of the Nantuo Forma- tions, respectively, lower strata of the Nanhua (middle to late Neoproterozoic) stratotype section in eastern Three Gorges, Yangtze craton. A range of inherited magmatic zircons was recognized with ages of 830 to 780 Ma, which are typical of Neoprotzrozoic magmatisms recorded along the margins and interior of the Yangtze craton. Thus, there is Neoproterozoic basement comprising 830―780 Ma igne- ous suites in South Qinling; the inherited zircons were detrital sediments derived from the northern margin of the Yangtze craton. 2U-Pb dating of detrital zircons from the Wudangshan Group in the South Qinling and its geological significance显示文摘U-Pb dating was made by the LA-ICP-MS method for detrital zircons from the Wudangshan volcanic-sedimentary succession in the South Qinling. Samples comprise quartz sandstones of the Wudangshan Group collected from the base of the Yangping For-mation and an upper layer of the Shuangtai Formation overlain its volcanic sequence, and two river-sand collections from the drainage systems cutting across the two formations, respectively. The results show that the Yangping detrital zircons are domi-nated by 830–780 Ma grains with a minor population of ~1.0–0.84 Ga and sporadic grains of ~2.6, ~2.4 and 2.0 Ga, whereas the Shuangtai zircons yield an upper intercept age of 763±33 Ma, identical to the timing of the Wudangshan volcanism within error, with few concordant grains of 1.9 and 0.86 Ga. Age spectra for the two river-sand samples are similar to those of the Yangping and Shuangtai Formations, respectively. It thus suggests that the Wudangshan strata are less than 780 Ma, whereas their major detrital zircon populations of 1.0–0.85 Ga and 830–780 Ma are consistent with timing of the Hannan magmatic activities along the northwestern margin of the South China Block. This suggests a Hannan or adjacent area provenance for the Wudangshan strata. The Wudang area is characterized by rift-related igneous events at ~755 and ~680 Ma, respectively, pointing to a tectono-magmatic history different from the Hannan area. 3The recognition of adakite-type gneisses in the North Dabie Mountain and its implication to ultrahigh pressure metamorphic geology显示文摘Adakite is an igneous rock generated by the partial melting of subducting ocean slab in an island arc setting. Meanwhile, residuals of the slab-melting are transformed to eclogite or garnet-amphibolite during a slab subduction process. This note reports the adakite-type grey gneisses discovered in the North Dabie Mountain. These adakitic rocks are characterized by high AI2O3(14.97%-17.56%), Sr (545-941 μg/g), Sr/Y (44.71-218.98), La/Yb (16.70-97.50), and low Y (3.32-12.19μg/g), Yb (0.31-1.34 μg/g), Sc (0.92-4.2μg/g), as well as positive Sr anomaly but absent or positive Eu anomaly. They are different from the low aluminum grey gneisses with low AI2O3 (13.73%-14.38%), Sr (262-409 μg/g), Sr/Y (6.46-15.97) and La/Yb (1.90-22.60), high Y(21.79-36.24 μg/g), Yb (2.00-5.59 μg/g) , Sc (8.30%-12.70%), and negative Sr, Eu anomaly in the North Dabie Mountain. 5Single-cell analysis of cellular heterogeneity and interactions in the ischemia-reperfusion injured mouse intestine显示文摘Nine major cell populations among 46,716 cells were identified in mouse intestinal ischemia‒reperfusion(II/R)injury by single-cell RNA sequencing.For enterocyte cells,11 subclusters were found,in which enterocyte cluster 1(EC1),enterocyte cluster 3(EC3),and enterocyte cluster 8(EC8)were newly discovered cells in ischemia 45 min/reperfusion 720 min(I 45 min/R 720 min)group.EC1 and EC3 played roles in digestion and absorption,and EC8 played a role in cell junctions.For TA cells,after ischemia 45 min/reperfusion 90 min(I 45 min/R 90 min),many TA cells at the stage of proliferation were identified.For Paneth cells,Paneth cluster 3 was observed in the resting state of normal jejunum.After I 45 min/R 90 min,three new subsets were found,in which Paneth cluster 1 had good antigen presentation activity.The main functions of goblet cells were to synthesize and secrete mucus,and a novel subcluster(goblet cluster 5)with highly proliferative ability was discovered in I 45 min/R 90 min group.As a major part of immune system,the changes in T cells with important roles were clarified.Notably,enterocyte cells secreted Guca2b to interact with Gucy2c receptor on the membranes of stem cells,TA cells,Paneth cells,and goblet cells to elicit intercellular communication.One marker known as glutathione S-transferase mu 3(GSTM3)affected intestinal mucosal barrier function by adjusting mitogen-activated protein kinases(MAPK)signaling during II/R injury.The data on the heterogeneity of intestinal cells,cellular communication and the mechanism of GSTM3 provide a cellular basis for treating II/R injury.Lianhong Yin Meng Gao Lina Xu Yan Qi Lan Han Jinyong Peng 2023Journal of Pharmaceutical Analysis2023,13,7:1

73D disorganization and rearrangement of genome provide insights into pathogenesis of NAFLD by integrated Hi-C, Nanopore, and RNA sequencing显示文摘The three-dimensional(3D)conformation of chromatin is integral to the precise regulation of gene expression.The 3D genome and genomic variations in non-alcoholic fatty liver disease(NAFLD)are largely unknown,despite their key roles in cellular function and physiological processes.Highthroughput chromosome conformation capture(Hi-C),Nanopore sequencing,and RNA-sequencing(RNA-seq)assays were performed on the liver of normal and NAFLD mice.A high-resolution 3D chromatin interaction map was generated to examine different 3D genome hierarchies including A/B compartments,topologically associated domains(TADs),and chromatin loops by Hi-C,and whole genome sequencing identifying structural variations(SVs)and copy number variations(CNVs)by Nanopore sequencing.We identified variations in thousands of regions across the genome with respect to 3D chromatin organization and genomic rearrangements,between normal and NAFLD mice,and revealed gene dysregulation frequently accompanied by these variations.Candidate target genes were identified in NAFLD,impacted by genetic rearrangements and spatial organization disruption.Our data provide a high-resolution 3D genome interaction resource for NAFLD investigations,revealed the relationship among genetic rearrangements,spatial organization disruption,and gene regulation,and identified candidate genes associated with these variations implicated in the pathogenesis of NAFLD.The newly findings offer insights into novel mechanisms of NAFLD pathogenesis and can provide a new conceptual framework for NAFLD therapy.Lina Xu Lianhong Yin Yan Qi Xuemei Tan Meng Gao Jinyong Peng 2021Acta Pharmaceutica Sinica B2021,11,10:1

15Effects of kallikrein gene transfer on penumbral microvascular proliferation and on regional cerebral blood flow following cerebral ischemia/reperfusion injury显示文摘BACKGROUND: Recent findings have demonstrated that the kallikrein-kinin system (KKS) participates in the pathological process of cerebral ischemia/reperfusion injury. Kallikrein gene transfer exhibits neural protective effects following cerebral infarction. OBJECTIVE: To observe the effects of kallikrein gene transfer on vascular proliferation in the peripheral infarct focus and on regional cerebral blood flow (rCBF) following cerebral ischemia/reperfusion injury. DESIGN, TIME AND SETTING: The completely randomized, controlled experiment was performed at the Lin Baixin Laboratory Center, the Second Affiliated Hospital of Sun Yat-sun University between September 2007 and April 2008. MATERIALS: pUC19-HTK plasmid was constructed and maintained in the Laboratory for Neurology, the Second Affiliated Hospital of Sun Yat-sen University, China. Mouse anti-human kallikrein 1 monoclonal antibody was purchased from R&D Systems, USA. METHODS: Ninety healthy, male, Sprague Dawley rats were used. Middle cerebral artery occlusion (MCAO) was established in all rats to induce cerebral ischemia/reperfusion injury. Following MCAO establishment, all rats were randomly divided into three groups (n = 30): blank control, saline, and pAdCMV-HTK. The saline and pAdCMV-HTK groups were stereotactically micro-injected with 5 μL of physiological saline or with pAdCMV-HTK [multiplicity of infection (MOI) = 20], respectively, into the ischemic penumbra. In the blank control group, only sham injection was performed. MAIN OUTCOME MEASURES: At 12, 24, and 72 hours after treatment, cerebral infarction volume was measured by 2, 3, 5-triphenyltetrazolium chloride (TTC) staining. Exogenous HTK expression, as well as regional vascular endothelial growth factor (VEGF) expression, was detected by immunohistochemistry. rCBF was examined by 14C-iodoantipyrine micro tracing. In addition, neurological severity score (NSS) was performed. Higher scores indicated more severe neurological deficits. RESULTS: NSS results demonstrated that compared with the saline and the blank control groups, the pAdCMV-HTK group exhibited lower NSSs 24 hours after pAdCMV-HTK injection (P < 0.05). The NSSs were further decreased after 72 hours (P < 0.01). Cerebral infarction volume at 24 hours, and in particular at 72 hours after treatment, was significantly reduced in the pAdCMV-HTK group compared with the blank control and saline groups (P < 0.05). The rCBF in the area surrounding the infarction lesion was slightly decreased in all groups compared with the contralateral area. At 24 and 72 hours following treatment, the rCBF in the peripheral infarction lesion was significantly elevated in the pAdCMV-HTK group compared with the blank control and saline groups (P < 0.05). Immunohistochemistry results revealed that VEGF-positive cells were primarily found in the cortex and in some white matter surrounding the cerebral infarction lesion. In addition, the expression of VEGF in the pAdCMV-HTK group was significantly higher compared with that in the blank control and saline groups at 12, 24, and 72 hours following treatment (P < 0.05). 16Protective effects of dioscin against Parkinson's disease via regulating bile acid metabolism through remodeling gut microbiome/GLP-1 signaling显示文摘It is necessary to explore potent therapeutic agents via regulating gut microbiota and metabolism to combat Parkinson's disease(PD).Dioscin,a bioactive steroidal saponin,shows various activities.However,its effects and mechanisms against PD are limited.In this study,dioscin dramatically alleviated neuroinflammation and oxidative stress,and restored the disorders of mice induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).16 S rDNA sequencing assay demonstrated that dioscin reversed MPTP-induced gut dysbiosis to decrease Firmicutes-to-Bacteroidetes ratio and the abundances of Enterococcus,Streptococcus,Bacteroides and Lactobacillus genera,which further inhibited bile salt hydrolase(BSH)activity and blocked bile acid(BA)deconjugation.Fecal microbiome transplantation test showed that the anti-PD effect of dioscin was gut microbiota-dependent.In addition,non-targeted fecal metabolomics assays revealed many differential metabolites in adjusting steroid biosynthesis and primary bile acid biosynthesis.Moreover,targeted bile acid metabolomics assay indicated that dioscin increased the levels of ursodeoxycholic acid,tauroursodeoxycholic acid,taurodeoxycholic acid and bmuricholic acid in feces and serum.In addition,ursodeoxycholic acid administration markedly improved the protective effects of dioscin against PD in mice.Mechanistic test indicated that dioscin significantly up-regulated the levels of takeda G protein-coupled receptor 5(TGR5),glucagon-like peptide-1 receptor(GLP-1R),GLP-1,superoxide dismutase(SOD),and down-regulated NADPH oxidases 2(NOX2)and nuclear factor-kappaB(NF-kB)levels.Our data indicated that dioscin ameliorated PD phenotype by restoring gut dysbiosis and regulating bile acid-mediated oxidative stress and neuroinflammation via targeting GLP-1 signal in MPTP-induced PD mice,suggesting that the compound should be considered as a prebiotic agent to treat PD in the future.Zhang Mao Haochen Hui Xuerong Zhao Lina Xu Yan Qi Lianhong Yin Liping Qu Lan Han Jinyong Peng 2023Journal of Pharmaceutical Analysis2023,13,10:0
17Stereotaxic microinjection of adenovirus-mediated human tissue Kallikrein gene reduces apoptosis in a rat model of middle cerebral artery occlusion显示文摘BACKGROUND:Several studies have demonstrated that Kallikrein gene transfer provides neuroprotection. Whether the neuroprotective effects of human tissue Kallikrein (HTK) are associated with apoptosis remains unclear. OBJECTIVE: To investigate the effects of HTK on apoptosis in the peripheral cerebral infarct region. DESIGN, TIME AND SETTING: The completely randomized grouping, gene engineered, controlled experiment was performed at the Lin Baixin Laboratory Center, the Second Affiliated Hospital of Sun Yat-sun University between September 2007 and April 2008. MATERIALS: Ninety clean, healthy, male, Sprague Dawley rats were included. pUC19-HTK plasmid was constructed in the Laboratory for Neurology, the Second Affiliated Hospital of Sun Yat-sen University, China. bcl-2, bax, caspase-3, and β-actin were designed and purified by Shanghai Shuiyuan Company, China. METHODS: Middle cerebral artery occlusion (MCAO) model was established in all rats. At 72 hours after MCAO model establishment, rats were randomly divided into 3 groups, with 30 rats per group: blank control, saline, and pAdCMV-HTK. The saline and pAdCMV-HTK groups were respectively stereotactically micro-injected with 5 μL physiological saline or pAdCMV-HTK at the area surrounding the cerebral infarction region. Only puncture was performed, without any injection, in the blank control group. MAIN OUTCOME MEASURES: At 72 hours after MCAO establishment, as well as at 24 hours, 72 hours, and 7 days subsequent to treatment, exogenous HTK expression was detected by immunohistochemistry. Apoptosis was examined by terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), while mRNA levels of bcl-2, bax, and caspase-3 were detected by reverse transcription-polymerase chain reaction. In addition, neurological severity scores were evaluated prior to and after treatments. RESULTS: Ninety rats were included in the final analysis. HTK was primarily detected in the cytoplasm at 24 hours after pAdCMV-HTK injection. Thereafter, HTK expression gradually increased and reached a peak level at 72 hours after injection, which was significantly different from the blank control and saline groups (P < 0.05). At 7 days, HTK expression began to decrease, but remained higher than the saline and blank control groups (P < 0.05). Apoptotic cells aggregated around the cerebral infarction region. Compared with the saline and blank control groups, the mean number of TUNEL-positive cells was notably decreased in the pAdCMV-HTK group at each time point after treatment (P < 0.05). mRNA levels of bcl-2, bax, and caspase-3 were elevated in all groups at 24 hours after treatment, peaked at 72 hours, and then gradually decreased again at 7 days. Compared with the saline and blank control groups, bcl-2 slightly increased, but was not significantly different from the pAdCMV-HTK group (P > 0.05). bax and caspase-3 mRNA levels were significantly reduced at 24 and 72 hours after treatment (P < 0.05). At 72 hours and, in particular, at 7 days after treatment, neurological severity scores were significantly less in the pAdCMV-HTK group compared with the saline and blank control groups (P < 0.05–0.01). CONCLUSION: HTK could protect neural cells in the peripheral cerebral infarction region from apoptosis, which resulted in a better outcome.
