| 1 | Effect of age on aortic atherosclerosis显示文摘Objective To examine the association of atherosclerosis burden in the survivors of an asymptomatic elderly cohort study and its relationship to other coronary risk factors (specifically, age) by evaluating aortic atherosclerotic wall burden by magnetic resonance imaging (MRI). Methods A total of 312 participants in an ongoing observational cohort study underwent cardiac and descending thoracic aorta imaging by MRI. Maximum wall thickness was measured and the mean wall thickness calculated.Wall/outer wall ratio was used as a normalized wall index (NWI) adjusted for artery size difference among participants. Percent wall volume (PWV) was calculated as NWI ×100.Results In this asymptomatic cohort (mean age: 76 years), the mean (SD) aortic wall area andwall thicknesswere 222 ±45 mm2 and 2.7 ±0.4 mm, respectively. Maximum wall thickness was 3.4 ±0.6 mm, and PWV was 32% ±4%. Women appeared to have smaller wall area,but after correcting for their smaller artery size, had significantly higher PWV than men (P = 0.03). Older age was associatedwith larger wall area (P = 0.04 for trend) with similar PWVs. However, there were no statistically significant associations between standard risk factors,Framingham global risk, or metabolic syndrome status, therapy for cholesterol or hypertension, coronary or aortic calcium score, and the aortic wall burden. Aortic calcificationwas associated with coronary calcification. Conclusions Asymptomatic elderly in this cohort had a greater descending thoracic aortic wall volume that correlated with age, andwomen had a significantly increased PWV compared to men. In these survivors, the atherosclerotic aortic wall burden was not significantly associated with traditional risk factors or with coronary or aortic calcium scores or coronary calcium progression. | 2 | ^(125)I-labeled anti-b FGF monoclonal antibody inhibits growth of hepatocellular carcinoma显示文摘AIM: To investigate the inhibitory efficacy of ^(125)I-labeled anti-basic fibroblast growth factor(b FGF) monoclonal antibody(m Ab) in hepatocellular carcinoma(HCC).METHODS: b FGF m Ab was prepared by using the 1G9B9 hybridoma cell line with hybridization technology and extracted from ascites fluid through a Protein G Sepharose affinity column. After labeling with ^(125)I through the chloramine-T method, b FGF m Ab was further purified by a Sephadex G-25 column. Gamma radiation counter GC-1200 detected radioactivity of ^(125)I-b FGF m Ab. The murine H22 HCC xenograft model was established and randomized to interventions with control(phosphate-buffered saline), ^(125)I-b FGF m Ab,^(125)I plus b FGF m Ab, b FGF m Ab, or ^(125)I. The ratios of tumor inhibition were then calculated. Expression of b FGF, fibroblast growth factor receptor(FGFR), plateletderived growth factor, and vascular endothelial growth factor(VEGF) m RNA was determined by quantitative reverse transcriptase real-time polymerase chain reaction. RESULTS: The purified b FGF m Ab solution was 8.145 mg/m L with a titer of 1:2560000 and was stored at-20 ℃. After coupling, ^(125)I-b FGF m Ab was used at a 1: 1280000 dilution, stored at 4 ℃, and its specific radioactivity was 37 MBq/mg. The corresponding tumor weight in the control, ^(125)I, b FGF m Ab, ^(125)I plus b FGF m Ab, and ^(125)I-b FGF m Ab groups was 1.88 ± 0.25, 1.625 ± 0.21, 1.5 ± 0.18, 1.41 ± 0.16, and 0.98 ± 0.11 g, respectively. The tumor inhibition ratio in the ^(125)I, b FGF m Ab, ^(125)I plus b FGF m Ab, and ^(125)I-b FGF m Ab groups was 13.6%, 20.2%, 25.1%, and 47.9%, respectively. Growth of HCC xenografts was inhibited significantly more in the ^(125)I-b FGF m Ab group than in the other groups(P < 0.05). Expression of b FGF and FGFR m RNA in the ^(125)I-b FGF m Ab group was significantly decreased in comparison with other groups(P < 0.05). Groups under interventions revealed increased expression of VEGF m RNA(except for ^(125)I group) compared with the control group.CONCLUSION: ^(125)I-b FGF m Ab inhibits growth of HCC xenografts. | 3 | Laser pattern-induced unidirectional lubricant flow for lubrication track replenishment显示文摘Effective oil replenishment to the lubrication track of a running bearing is crucial to its sustainable operation.Reliable practical solutions are rare despite numerous theoretical studies were conducted in the last few decades.This paper proposes the use of surface effect,wettability gradient,to achieve the goal.This method is simple and can be nicely implemented using femtosecond laser ablation.A periodic comb-tooth-shaped pattern with anisotropic wetting capability is devised and its effect on the anisotropic spreading behaviour of an oil droplet is studied.Results show that the comb-tooth-shaped pattern enables the rearrangement of oil distribution,thereby escalating oil replenishment to the lubrication track.The effect is due to the unbalanced interfacial force created by the surface pattern.The influence of the shape and the pitch of teeth,which are the two governing factors,on oil transport is also reported.The effects of the newly devised surface pattern on lubrication are experimentally evaluated under the conditions of limited lubricant supply.These results are promising,demonstrating the reduction in bearing friction and the increase in lubricating film thickness. | Chenglong LIU Feng GUO Patrick WONG Xinming LI | 2022 | Friction2022,10,8: | 4 |
