| 1 | Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. | 2 | Sedimentary Records of Mangrove Evolution During the Past one Hundred Years Based on Stable Carbon Isotope and Pollen Evidences in Maowei, SW China显示文摘Mangroves accumulate sedimentary sequences, where cores can provide historical records of mangrove evolution with past climate change and human activity. The study traced the history of mangrove evolution during the past one hundred years in a mangrove swamp of Maowei Sea, SW China. The sedimentation rates(0.38-0.95 cm yr^(-1)) were calculated on the basis of ln(^(210)Pb_(xs)/Al) and mass depth in the core sediments. Chemical tracers, such as δ^(13)C_(org) and C:N values, were utilized to trace the contribution of mangrove-derived organic matter using a ternary mixing model. Because of potential diagenetic alteration and / or overlap in the isotopic signatures of different components, simultaneous use of mangrove pollen diagrams can help to supplement some of these limitations. Combined with mangrove pollen, mangrove evolution was reconstructed and could be divided into three stages: flourishment(1886-1905 AD), slight degradation(1905-1949 AD) and rapid degradation period(1949-2007 AD), which was consistent with previous reports. | 3 | Organic carbon isotope and pollen evidence for mangrove development and response to human activity in Guangxi(Southwest China) over the last 140 years显示文摘Mangrove degradation must reduce carbon sequestration in recent years, thereby aggravating global warming.Thus, short-term impacts of human activity on mangrove ecosystems are cause for concern from local governments and scientists. Mangroves sediments can provide detailed records of mangrove species variation in the last one hundred years, based on detailed 210 Pb data. The study traced the history of mangrove development and its response to environmental change over the last 140 years in two mangrove swamps of Guangxi, Southwest China. Average sedimentation rates were calculated to be 0.48 cm/a and 0.56 cm/a in the Yingluo Bay and the Maowei Sea, respectively. Chemical indicators(δ13Corg and C:N) were utilized to trace the contribution of mangrove-derived organic matter(MOM) using a ternary mixing model. Simultaneous use of mangrove pollen can help to supplement some of these limitations in diagenetic/overlap of isotopic signatures. We found that vertical distribution of MOM was consistent with mangrove pollen, which could provide similar information for tracing mangrove ecosystems. Therefore, mangrove development was reconstructed and divided into three stages: flourishing, degradation and re-flourishing/re-degradation period. | 4 | Organic anion transporters also mediate the drug–drug interaction between imipenem and cilastatin显示文摘This study aimed to clarify that organic anion transporters(OATs)mediate the drug–drug interaction(DDI)between imipenem and cilastatin.After co-administration with imipenem,the plasma concentrations and the plasma concentration-time curve(AUC)of cilastatin were significantly increased,while renal clearance and cumulative urinary excretion of cilastatin were decreased.At the same time,imipenem significantly inhibited the uptake of cilastatin in rat kidney slices and in human OAT1(hOAT1)-HEK293 and human OAT3(hOAT3)-HEK293 cells.Probenecid,p-aminohippurate,and benzylpenicillin inhibited the uptake of imipenem and cilastatin in rat kidney slices and in hOAT1-and hOAT3-HEK 293 cells,respectively.The uptakes of imipenem and cilastatin in hOAT1-and hOAT3-HEK 293 cells were significantly higher than that in mock-HEK-293 cells.Moreover,the K m values of cilastatin were increased in the presence of imipenem with unchanged V max,indicating that imipenem inhibited the uptake of cilastatin in a competitive manner.When imipenem and cilastatin were co-administered,the level of imipenem was higher compared with imipenem alone both in vivo and in vitro.But,cilastatin significantly inhibited the uptake of imipenem when dehydropeptidase-1(DPEP1)was silenced by RNAi technology in hOAT1-and hOAT3-HEK 293 cells.In conclusion,imipenem and cilastatin are the substrates of OAT1 and OAT3.OAT1 and OAT3 mediate the DDI between imipenem and cilastatin.Meanwhile,cilastatin also reduces the hydrolysis of imipenem by inhibiting the uptake of imipenem mediated by OAT1 and OAT3 in the kidney as a complement. | Yanna Zhu Xiaokui Huo Changyuan Wang Qiang Meng Zhihao Liu Huijun Sun Aiping Tan Xiaodong Ma Jinyong Peng Kexin Liu | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,2: | 3 |
| 5 | ^(210)Pb Chronology and Trace Metal Geochemistry in the Intertidal Sediment of Qinjiang River Estuary, China显示文摘Historical records of metal inputs were studied by using a sediment core collected from a sand-rich mudflat in the Qinjiang River estuary, China. 210Pb chronology was used to reconstruct the fluxes of Hg, Cu, Pb, Zn, Cd, Cr and As to the core site during the last 86 years. Based on the constant initial concentration model, the sedimentation rates are 1.18 cm year-1 in the top 30 cm sandy layer and 0.92 cm year-1 in the muddy bottom layer. To compensate for grain-size and mineralogy effects on metal concentra-tions, aluminum was used as the normalizing element. The enrichment factors (EF) indicate that the natural inputs had prevailed up to the early 1980s. After this period, the intensity of human activities has resulted in continual increasing trend of metals towards the surface. Recent sediment samples from the Qinjiang River estuary are found moderately enriched by Cd (EF>1.5) and slightly enriched by other metals (EF<1.5). Therefore, the method of computer simulation on astronaut extravehicular activity is broadly promoted. However, validations and verifications for these simulations stated in related literatures are incomplete such as comparing with the limits of human body movements or reconstructing a three-dimensional movement for some parts of EVA video. Novel modeling and verification methods for the task of an astronaut handling large-mass payload during EVA were revealed. A simplified model of an astronaut was constructed, and the astronaut motion was conceived as a planar movement of a multi-body system which includes seven segments with six revolute joints in the human body sagittal plane. The inverse kinematics method was used to calculate joint angles, joint velocities, and joint accelerations in time domain. The solution of joint torques using the inverse recursive dynamics was achieved. Furthermore, a virtual model with the ADAMSTM software was developed and implemented to verify the results by adding the kinematical data calculated to joints in order to achieve the trace of the center of mass of the hand. Additionally, the joints kinematics and kinetics data with time in the virtual model were obtained and compared with the corresponding calculated data. This result indicates that the modeling methods proposed can be employed as a solid algorithm to conduct the simulation of astronaut’s tasks in EVA, and verification using the virtual model can be easily operated and has a good accuracy. | 16 | Role of the STAT3/survivin signaling pathway in the EML4-ALK-positive lung adenocarcinoma cell line H2228 before and after crizotinib-induced resistance显示文摘Objective This study investigated the role of the STAT3/survivin signaling pathway in the EML4-ALK–positive lung adenocarcinoma cell line H2228 before and after crizotinib-induced resistance. The mechanism of resistance was studied. Methods Cell viability was determined using the MTT assay. Crizotinib-induced apoptosis in H2228 and H2228 crizotinib-resistant cells treated with the indicated doses of crizotinib was measured at different times(24 h, 48 h, 72 h) using flow cytometry. The levels of p-ALK, ALK, p-STAT3, STAT3, and survivin after treatment of cells with 0, 0.3, and 1 μM crizotinib for 72 h were determined using Western blot analysis. DNA sequencing was used to identify mutations in H2228 crizotinib-resistant cells. Results The crizotinib IC50 values in H2228 and H2228 crizotinib-resistant cells at 72 h were 334.5 n M and 3418 n M, respectively. The resistance index of H2228 crizotinib-resistant cells was 10.20. Crizotinib induced apoptosis in H2228 cells and reduced the levels of p-ALK, p-STAT3, and survivin. In contrast, no changes in the levels of p-ALK, p-STAT3, and survivin were observed in H2228 crizotinib-resistant cells. The mutations 2067G→A and 2182G→C in EML4-ALK were present in the H2228 crizotinib-resistant cells. Conclusion Crizotinib decreased the viability of H2228 cells in a dose- and time-dependent manner. In the STAT3/survivin pathway, downregulation of p-ALK, p-STAT3, and survivin might contribute to crizotinib-induced apoptosis in H2228 cells. However, the STAT3/survivin pathway in H2228 crizotinib-resistant cells was unaffected by crizotinib treatment. | 18 | Dynamic emission characteristics and control strategies of air pollutants from motor vehicles in downtown Beijing,China显示文摘This study combined the real-time monitoring and investigation of traffic flows to comprehensively analyze the road traffic flow and vehicle structure in downtown Beijing.A dynamic emission inventory of motor vehicle air pollutants in downtown Beijing in 2021 was established,and the impact of these emissions on air quality was simulated and quantified,and different emission reduction control scenarios were proposed to evaluate their environmental improvement effects and explore measures to mitigate the impact of pollution emissions.The results show that the high traffic flow and the structure of the motor vehicle emissions in downtown Beijing are the main causes of severe motor vehicle pollution.Monitoring data shows that traffic flow in central Beijing is dominated by small passenger vehicles,while the vehicle mix is better than in other regions,with 72.0%of vehicles meeting'National V'or higher emission standards.However,to achieve higher air quality goals,further reducing vehicle emissions is necessary.Based on dynamic traffic flow,the average daily emissions of nitrogen oxides(NOX),particulate matters(PM_(2.5))and volatile organic compounds(VOC)from motor vehicles in central Beijing are 17.7 tons,0.6 tons and 14.0 tons,respectively,accounting for 23.0%of the city’s average daily motor vehicle emissions.If a zero-emission zone for motor vehicles were implemented in central Beijing,the annual average emission level of pollutants will be reduced by 10.4%to 21.0%.The designation of ultra-low emission zones for motor vehicles could be effective in improving the air quality in the center of Beijing. | Yan Shen Tongran Wu Aiping Lian Jie Gao Fei Peng Guohua Song Xuefang Wu Yangyang Cui Xinyu Liu Yunfei Wan Jing Yan Yifeng Xue | 2024 | Journal of Environmental Sciences2024,,2: | 0 |
