1Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 3Simultaneous virus identification and characterization of severe unexplained pneumonia cases using a metagenomics sequencing technique显示文摘Many viruses can cause respiratory diseases in humans.Although great advances have been achieved in methods of diagnosis,it remains challenging to identify pathogens in unexplained pneumonia(UP) cases.In this study,we applied next-generation sequencing(NGS) technology and a metagenomic approach to detect and characterize respiratory viruses in UP cases from Guizhou Province,China.A total of 33 oropharyngeal swabs were obtained from hospitalized UP patients and subjected to NGS.An unbiased metagenomic analysis pipeline identified 13 virus species in 16 samples.Human rhinovirus C was the virus most frequently detected and was identified in seven samples.Human measles virus,adenovirus B 55 and coxsackievirus A10 were also identified.Metagenomic sequencing also provided virus genomic sequences,which enabled genotype characterization and phylogenetic analysis.For cases of multiple infection,metagenomic sequencing afforded information regarding the quantity of each virus in the sample,which could be used to evaluate each viruses' role in the disease.Our study highlights the potential of metagenomic sequencing for pathogen identification in UP cases.Xiaohui Zou Guangpeng Tang Xiang Zhao Yan Huang Tao Chen Mingyu Lei Wenbing Chen Lei Yang Wenfei Zhu Li Zhuang Jing Yang Zhaomin Feng Dayan Wang Dingming Wang Yuelong Shu 2017Science China(Life Sciences)2017,60,3:14
4Research progress in human infection with avian influenza H7N9 virus显示文摘Since the identification of the novel reassortant avian influenza A(H7N9) virus in China in 2013, until Jun 30, 2017, the virus has caused five epidemic waves leading to a total of 1,552 human infections, with a fatality rate of about 40%. In the spring of2017, highly pathogenic avian influenza(HPAI) H7N9 virus emerged and has caused 25 human infections. The HPAI H7N9 virus has some biological differences from the LPAI one, such as its multiple basic amino acid residues on HA leading to its independence on trypsin for replication. The pathogenicity of the HPAI H7N9 virus to experimental animals or humans is still unclear. A(H7N9) vaccine development for pandemic preparedness is ongoing, including the reassortment(H7N9/PR8)reverse genetic based vaccine, the virus like particle(VLP) vaccine, the intranasal live attenuated influenza vaccine(LAIV),the non-adjuvant Vero cell culture-derived inactivated whole-virus vaccine, the MDCK culture-derived vaccine, the H7 DNA vaccine and the recombinant replicative H7N9 virus \(H7N9-53 TM\) vaccine. Five neuramidinase resistant sites of A(H7N9)virus isolated from patients have been reported. Some alternative drugs have been studied, such as DAS181(Fludase), ribavirin,troglitazone and minocycline. 9Epidemiological and Virological Surveillance of Seasonal Influenza Viruses—China,2020–2021显示文摘Introduction:During the coronavirus disease 2019(COVID-19)pandemic,the circulation of seasonal influenza virus declined globally and remained below previous seasonal levels.We analyzed the results of the epidemiology,antigenic,and genetic characteristics,and antiviral susceptibilities of seasonal influenza viruses isolated from the mainland of China during October 5,2020 through September 5,2021,to better assess the risk of influenza during subsequent influenza season in 2021–2022.Methods:Positive rates of influenza virus detection during this period were based on real-time polymerase chain reaction(PCR)detection by the Chinese National Influenza Surveillance Network laboratories,and isolated viruses from influenza positive samples were submitted to the Chinese National Influenza Center.Antigenic analyses for influenza viruses were conducted using the hemagglutination inhibition assay.Next-generation sequencing was used for genetic analyses.Viruses were tested for resistance to antiviral medications using a phenotypic assay and next-generation sequencing.Results:In southern China,the influenza positivity rate was elevated especially after March 2021 and was higher than the same period the previous year with the COVID-19 pandemic.In northern China,influenza positive rate peaked at Week 18 in 2021 and has declined since then.Nearly all isolated viruses were B/Victoria lineage viruses during the study period,and 37.3%of these viruses are antigenically similar to the reference viruses representing the vaccine components for the 2020–2021 and 2021–2022 Northern Hemisphere influenza season.All seasonal influenza viruses were susceptible to neuraminidase inhibitors and endonuclease inhibitors.Conclusions:Influenza activity has gradually increased in the mainland of China in 2021,although the intensity of activity is still lower than before the COVID-19 pandemic.The diversity of circulating influenza types/subtypes decreased,with the vast majority being B/Victoria lineage viruses.The surveillance data from this study suggest that we should strengthen influenza surveillance during the upcoming traditional influenza season.It also provided evidence for vaccine recommendations and prevention and control of influenza and clinical use of antiviral drugs.Weijuan Huang Xiyan Li Minju Tan Yanhui Cheng Tao Chen Hejiang Wei Xiaoxu Zeng Yiran Xie Jia Liu Ning Xiao Lei Yang Dayan Wang 2021China CDC weekly2021,3,44:4
10Severe human infection with a novel avian-origin influenza A(H7N4) virus显示文摘Human infections with influenza H7 subtypes, such as H7N9, have raised concerns worldwide. Here, we report a human infection with a novel influenza A(H7N4) virus. A 68 years-old woman with cardiovascular and cholecystic comorbidities developed rapidly progressed pneumonia with influenza-like-illness as initial symptom, recovered after 23 days-hospitalization including 8 days in ICU. Laboratory indicators for liver and blood coagulation dysfunction were observed. Oseltamivir phosphate, glucocorticoids and antibiotics were jointly implemented, with nasal catheterization of oxygen inhalation for this patient.We obtained the medical records and collected serial respiratory and blood specimens from her. We collected throat, cloacal and/or feces samples of poultry and wild birds from the patient's backyard, neighborhood, local live poultry markets(LPMs) and the nearest lake. All close contacts of the patient were followed up and sampled with throat swabs and sera. Influenza viruses and other respiratory pathogens were tested by real-time RT-PCR, viral culturing and/or sequencing for human respiratory and bird samples. Micro-neutralizing assay was performed for sera. A novel reassortant wild bird-origin H7N4 virus is identified from the patient and her backyard poultry(chickens and ducks) by sequencing, which is distinct from previously-reported avian H7N4 and H7N9 viruses. At least four folds increase of neutralizing antibodies to H7N4 was detected in her convalescent sera. 11Genomic characterization of SARS-CoV-2 identified in a reemerging COVID-19 outbreak in Beijing's Xinfadi market in 2020显示文摘After 56 days without coronavirus disease 2019(COVID-19)cases,reemergent cases were reported in Beijing,China on June 11,2020.Here,we report the genetic characteristics of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)sequenced from the clinical specimens of 4 human cases and 2 environmental samples.The nucleotide similarity among six SARS-CoV-2 genomes ranged from 99.98%to 99.99%.Compared with the reference strain of SARS-CoV-2(GenBank No.NC_045512),all six genome sequences shared the same substitutions at nt241(C→T),nt3037(C→T),nt14408(C→T),nt23403(A→G),nt28881(G→A),nt28882(G→A),and nt28883(G→C),which are the characteristic nucleotide substitutions of L-lineage European branch I.This was also proved by themaximum likelihood phylogenetic tree based on the full-length genome of SARS-CoV-2.They also have a unique shared nucleotide substitution,nt6026(C→T),which is the characteristic nucleotide substitution of SARS-CoV-2 in Beijing's Xinfadi outbreak.It is noteworthy that there is an amino acid D614Gmutation caused by nt23403 substitution in all six genomes,which may enhance the virus's infectivity in humans and help it become the leading strain of the virus to spread around the world today.It is necessary to continuously monitor the genetic variation of SARS-CoV-2,focusing on the influence of key mutation sites of SARS-CoV-2 on viral transmission,clinical manifestations,severity,and course of disease.Yong Zhang Yang Pan Xiang Zhao Weifeng Shi Zhixiao Chen Sheng Zhang Peipei Liu Jinbo Xiao Wenjie Tan Dayan Wang William J.Liu Wenbo Xu Quanyi Wang Guizhen Wu 2020Biosafety and Health2020,2,4:3

17Increased urbanization reduced the efectiveness of school closures on seasonal infuenza epidemics in China显示文摘Background:School closure is a common mitigation strategy during severe infuenza epidemics and pandemics.However,the efectiveness of this strategy remains controversial.In this study,we aimed to explore the efectiveness of school closure on seasonal infuenza epidemics in provincial-level administrative divisions(PLADs)with varying urbanization rates in China.Methods:This study analyzed infuenza surveillance data between 2010 and 2019 provided by the Chinese National Infuenza Center.Taking into consideration the climate,this study included a region with 3 adjacent PLADs in Northern China and another region with 4 adjacent PLADs in Southern China.The efect of school closure on infuenza transmission was evaluated by the reduction of the efective reproductive number of seasonal infuenza during school winter breaks compared with that before school winter breaks.An age-structured Susceptible-Infected-Recovered-Susceptible(SIRS)model was built to model infuenza transmission in diferent levels of urbanization.Parameters were determined using the surveillance data via robust Bayesian method.Results:Between 2010 and 2019,in the less urbanized provinces:Hebei,Zhejiang,Jiangsu and Anhui,during school winter breaks,the efective reproductive number of seasonal infuenza epidemics reduced 14.6%[95%confdential interval(CI):6.2–22.9%],9.6%(95%CI:2.5–16.6%),7.3%(95%CI:0.1–14.4%)and 8.2%(95%CI:1.1–15.3%)respectively.However,in the highly urbanized cities:Beijing,Tianjin and Shanghai,it reduced only 5.2%(95%CI:-0.7–11.2%),4.1%(95%CI:-0.9–9.1%)and 3.9%(95%CI:-1.6–9.4%)respectively.In China,urbanization is associated with decreased proportion of children and increased social contact.According to the SIRS model,both factors could reduce the impact of school closure on seasonal infuenza epidemics,and the proportion of children in the population is thought to be the dominant infuencing factor.Conclusions:Efectiveness of school closure on the epidemics varies with the age structure in the population and social contact patterns.School closure should be recommended in the low urbanized regions in China in the infuenza seasons.Hao Lei Hangjin Jiang Nan Zhang Xiaoli Duan Tao Chen Lei Yang Dayan Wang Yuelong Shu 2021Infectious Diseases of Poverty2021,10,5:2
