1Preparation and evaluation of poly(L-histidine) based pH-sensitive micelles for intracellular delivery of doxorubicin against MCF-7/ADR cells显示文摘In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intracellular doxorubicin(Dox) delivery for cancer chemotherapy. Dox was loaded into the micelles by thin-film hydration method and a Box–Behnken design for three factors at three levels was used to optimize the preparations. The optimized mPEG-PLA-Phis/Dox micelles exhibited good encapsulation efficiency of 91.12%,a mean diameter of 45 nm and narrow size distribution with polydispersity index of 0.256.In vitro drug release studies demonstrated that Dox was released from the micelles in a p Hdependent manner. Furthermore, the cellular evaluation of Dox loaded micelles displayed that the micelles possessed high antitumor activity in vitro with an IC50 of 35.30 μg/ml against MCF-7/ADR cells. The confocal microscopy and flow cytometry experiments indicated that m PEG-PLA-Phis micelles mediated efficient cytoplasmic delivery of Dox with the aid of poly(Lhistidine) mediated endosomal escape. In addition, blank m PEG-PLA-Phis micelles were shown to be nontoxic to MCF-7/ADR cells even at a high concentration of 200 μg/ml. 2Co-delivery of resveratrol and docetaxel via polymeric micelles to improve the treatment of drug-resistant tumors显示文摘Co-delivery of anti-cancer drugs is promising to improve the efficacy of cancer treatment.This study was aiming to investigate the potential of concurrent delivery of resveratrol(RES)and docetaxel(DTX)via polymeric nanocarriers to treat breast cancer.To this end,methoxyl poly(ethylene glycol)-poly(D,L-lactide)copolymer(mPEG-PDLA)was prepared and characterized using FTIR and 1H NMR,and their molecular weights were determined by GPC.Isobologram analysis and combination index calculation were performed to find the optimal ratio between RES and DTX to against human breast adenocarcinoma cell line(MCF-7 cells).Subsequently,RES and DTX were loaded in the mPEG-PDLA micelles simultaneously,and the morphology,particle size distribution,in vitro release,pharmacokinetic profiles,as well as cytotoxicity to the MCF-7 cells were characterized.IC50 of RES and DTX in MCF-7 cells were determined to be 23.0μg/ml and 10.4μg/ml,respectively,while a lower IC50 of 4.8μg/ml of the combination of RES and DTX was obtained.The combination of RES and DTX at a ratio of 1:1(w/w)generated stronger synergistic effect than other ratios in the MCF-7 cells.RES and DTX loaded mPEG-PDLA micelles exhibited prolonged release profiles,and enhanced cytotoxicity in vitro against MCF-7 cells.The AUC(0→t)of DTX and RES in mPEG-PDLA micelles after i.v.administration to rats were 3.0-fold and 1.6-fold higher than that of i.v.injections of the individual drugs.These findings indicated that the co-delivery of RES and DTX using mPEG-PDLA micelles could have better treatment of tumors.Xiong Guo Zhiyue Zhao Dawei Chen Mingxi Qiao Feng Wan Dongmei Cun Yi Sun Mingshi Yang 2019Asian Journal of Pharmaceutical Sciences2019,14,1:4
3Engineering Cu^(+)/CeZrO_(x) interfaces to promote CO_(2) hydrogenation to methanol显示文摘Cu-based catalysts are widely employed for CO_(2) hydrogenation to methanol,which is expected as a promising process to achieving carbon neutrality.However,most Cu-based catalysts still suffer from low methanol yield with a passable CO_(2) conversion and lack insight into its reaction mechanism for guiding the design of catalysts.In this work,Cu^(+)/CeZrO_(x) interfaces are engineered by employing a series of ceria-zirconia solid solution catalysts with various Ce/Zr ratios,forming a Cu^(+)-O_(v)-Ce^(3+)structure where Cu^(+)atoms are bonded to the oxygen vacancies(O_(v))of ceria.Compared to Cu/CeO_(2) and Cu/ZrO_(2),the optimized catalyst(i.e.,Cu_(0.3)Ce_(0.3)Zr_(0.7))exhibits a much higher mass-specific methanol formation rate(192g_(MeOH)/kg_(cat)/h)at 240℃and 3 MPa.Through a series of in-situ and ex-situ characterization,it is revealed that oxygen vacancies in solid solutions can effectively assist the activation of CO_(2) and tune the electronic state of copper to promote the formation of Cu^(+)/CeZrO_(x) interfaces,which stabilizes the key*CO intermediate,inhibits its desorption and facilitates its further hydrogenation to methanol via the reverse watergas-shift(RWGS)+CO-Hydro pathway.Therefore,the concentration of*CO or the apparent Cu^(+)/(Cu^(+)+Cu^(0))ratio could be employed as a quantitative descriptor of the methanol formation rate.This work is expected to give a deep insight into the mechanism of metal/support interfaces in CO_(2) hydrogenation to methanol,offering an effective strategy to develop new catalysts with high performance.Jingpeng Zhang Xiaohang Sun Congyi Wu Wenquan Hang Xu Hu Dawei Qiao Binhang Yan 2023Journal of Energy Chemistry2023,,2:3
4Combining immune checkpoint blockade with ATP-based immunogenic cell death amplifier for cancer chemo-immunotherapy显示文摘Amplifying“eat me signal”during tumor immunogenic cell death(ICD)cascade is crucial for tumor immunotherapy.Inspired by the indispensable role of adenosine triphosphate(ATP,a necessary“eat me signal”for ICD),a versatile ICD amplifier was developed for chemotherapy-sensitized immunotherapy.Doxorubicin(DOX),ATP and ferrous ions(Fe^(2+))were co-assembled into nanosized amplifier(ADO-Fe)throughπ‒πstacking and coordination effect.Meanwhile,phenylboric acid-polyethylene glycol-phenylboric acid(PBA-PEG-PBA)was modified on the surface of ADO-Fe(denoted as PADO-Fe)by the virtue of d-ribose unit of ATP.PADO-Fe could display active targetability against tumor cells via sialic acid/PBA interaction.In acidic microenvironment,PBA-PEG-PBA would dissociate from amplifier.Moreover,high H_(2)O_(2)concentration would induce hydroxyl radical(·OH)and oxygen(O_(2))generation through Fenton reaction by Fe^(2+).DOX and ATP would be released from the amplifier,which could induce ICD effect and“ICD adjuvant”to amplify this process.Together with programmed death ligands 1(PD-L1)checkpoint blockade immunotherapy,PADO-Fe could not only activate immune response against primary tumor,but also strong abscopal effect against distant tumor.Our simple and multifunctional ICD amplifier opens a new window for enhancing ICD effect and immune checkpoint blockade therapy.Jiulong Zhang Xiaoyan Sun Xiufeng Zhao Chunrong Yang Menghao Shi Benzhuo Zhang Haiyang Hu Mingxi Qiao Dawei Chen Xiuli Zhao 2022Acta Pharmaceutica Sinica B2022,12,9:2

6Effect of surface ligand density on cytotoxicity and pharmacokinetic profile of docetaxel loaded liposomes显示文摘Various biotin-modified liposomes incorporated with docetaxel(DTX) were prepared to study the effect of surface biotin density on the pharmacokinetic profile of the liposome. Four types of liposomes such as PEG modified liposome(PDL), 0.5%(mol) biotin modified liposome(0.5 BDL), 1%(mol) biotin modified liposome(1 BDL) and 2%(mol) biotin modified liposome(2 BDL) were prepared using thin film dispersion method. The prepared liposomes were characterized by measuring encapsulation efficiency(EE), particle size, Zeta-potential, physical stability and drug release profiles in vitro. MTT assay was performed to elevate the cytotoxicity of liposomes on MCF-7 cells. In vivo evaluation was further performed to investigate the effect of biotin surface density on the pharmacokinetic profiles. All the prepared liposomes exhibited high encapsulation efficiency, small particle size, narrow particle distribution and sustained release profiles in vitro. In MTT assay, 0.5 BDL showed largest tumor cell toxicity, compared with DTX solution. All liposomes containing DTX showed prolonged blood circulation in vivo, and 0.5 BDL showed the longest circulation time among the biotin modified liposome. Surface modification of liposome had a negative impact on the circulation of liposomes in the blood, which needs to be considered when designing the ligand mediated targeting delivery systems. 9Is there a great unconformity between Xiamaling and Longshan formations in the North China Craton?显示文摘The existence of a 0.3 Ga'great unconformity'between the Mesoproterozoic'Undefined System'Xiamaling Formation and the'Neoproterozoic'Longshan Formation has long been controversial.In this study,stratigraphy,sedimentology,detrital zircon dating,lithium isotope,and major and trace elements were applied to systematically analyze their relationship.Results demonstrate that coarse to fine sandstone-mudstone normal cycles with different grades and thicknesses exist from the Xiamaling to the Longshan formations.Sedimentary succession formed from a restricted platform with lagoon-shallow marine shelf-tidal flat-beach and tidal shoal developing in ascending order.The pebbly sandstone at the bottom of the Long shan Formation does not have the properties of basal conglomerate with tectonic significance.The youngest detrital zircon is older than 1.6 Ga for the Long shan Formation and overlying Jing'eryu Formation sandstones.In addition,no(zircon)provenance has been found from the large igneous province of 1.32-1.2 Ga in Xiamaling Formation.Theδ^(7)Li isotope values exhibit a gradually increasing trend.The Li content and CIA(Chemical index of alteration)gradually decreases from the top of the Xiamaling Formation to the lower part of the Long shan Formation.Simultaneously,as K_(2)O/Al_(2)O_(3)and FeO/Fe_(2)O_(3)decreases,Na_(2)O/Al_(2)O_(3)increases,and Li/Al,V/Cr,and V/(V+Ni)fluctuates slightly.At the boundary,the Ce/Ce^(*)and Eu/Eu^(*)show positive and negative anomalies,respectively.Further,the Rb/Sr and Al/Si values decrease,and U/Th values increase.This indicates that the geochemistry of the weathering crust at the interface of the Xiamaling Formation-Long shan Formation is not characteristic.However,it indicates a weakening of continental weathering intensity from the bottom to the top of the interface.These results do not support the existence of a large unconformity between the Xiamaling and the Longshan formations.Therefore,it is suggested that the Long shan Formation-Jing'eryu Formation should be moved to the lower level in the stratigraphic chart,where it should form part of the Mesoproterozoic'Undefined System'along with the Xiamaling Formation.On this basis,the paleogeography of the Xiamaling Formation-Longshan Formation-Jing'eryu Formation sedimentary period in the Yanliao area of North China has been reclassified.Hongwei KUANG Nan PENG Yongqing LIU Yuchong WANG Mingming CUI Xiaoshuai CHEN Dawei QIAO Keining QI 2023Science China Earth Sciences2023,66,5:1

13Anomalous-plasmoid-ejection-induced secondary magnetic reconnection: modeling solar flares and coronal mass ejections by laser–plasma experiments显示文摘The driving mechanism of solar flares and coronal mass ejections is a topic of ongoing debate, apart from the consensus that magnetic reconnection plays a key role during the impulsive process. While present solar research mostly depends on observations and theoretical models, laboratory experiments based on high-energy density facilities provide the third method for quantitatively comparing astrophysical observations and models with data achieved in experimental settings.In this article, we show laboratory modeling of solar flares and coronal mass ejections by constructing the magnetic reconnection system with two mutually approaching laser-produced plasmas circumfused of self-generated megagauss magnetic fields. 14A hierarchical bilayer architecture for complex tissue regeneration显示文摘Engineering a complete,physiologically functional,periodontal complex structure remains a great clinical challenge due to the highly hierarchical architecture of the periodontium and coordinated regulation of multiple growth factors required to induce stem cell multilineage differentiation.Using biomimetic self-assembly and microstamping techniques,we construct a hierarchical bilayer architecture consisting of intrafibrillarly mineralized collagen resembling bone and cementum,and unmineralized parallel-aligned fibrils mimicking periodontal ligament.The prepared biphasic scaffold possesses unique micro/nano structure,differential mechanical properties,and growth factor-rich microenvironment between the two phases,realizing a perfect simulation of natural periodontal hard/soft tissue interface.The interconnected porous hard compartment with a Young’s modulus of 1409.00±160.83 MPa could induce cross-arrangement and osteogenic differentiation of stem cells in vitro,whereas the micropatterned soft compartment with a Young’s modulus of 42.62±4.58 MPa containing abundant endogenous growth factors,could guide parallel arrangement and fibrogenic differentiation of stem cells in vitro.After implantation in critical-sized complete periodontal tissue defect,the biomimetic bilayer architecture potently reconstructs native periodontium with the insertion of periodontal ligament fibers into newly formed cementum and alveolar bone by recruiting host mesenchymal stem cells and activating the transforming growth factor beta 1/Smad3 signaling pathway.Taken together,integration of self-assembly and microstamping strategies could successfully fabricate a hierarchical bilayer architecture,which exhibits great potential for recruiting and regulating host stem cells to promote synergistic regeneration of hard/soft tissues.Min Yu Dan Luo Jing Qiao Jiusi Guo Danqing He Shanshan Jin Lin Tang Yu Wang Xin Shi Jing Mao Shengjie Cui Yu Fu Zixin Li Dawei Liu Ting Zhang Chi Zhang Zhou Li Yongsheng Zhou Yan Liu 2022Bioactive Materials2022,7,4:0
15Reversing ferroptosis resistance by MOFs through regulation intracellular redox homeostasis显示文摘As a non-apoptotic cell death form,ferroptosis offers an alternative approach to overcome cancer chemotherapy resistance.However,accumulating evidence indicates cancer cells can develop ferroptosis resistance by evolving antioxidative defense mechanisms.To address this issue,we prepared a Buthionine-(S,R)-sulfoximine(BSO)loaded metal organic framework(MOF)of BSO-MOF-HA(BMH)with the combination effect of boosting oxidative damage and inhibiting antioxidative defense.MOF nanoparticle was constructed by the photosensitizer of[4,4,4,4-(porphine-5,10,15,20-tetrayl)tetrakis(benzoic acid)](TCPP)and the metal ion of Zr6,which was further decorated with hyaluronic acid(HA)in order to impart active targeting to CD44 receptors overexpressed cancer cells.BMH exhibited a negative charge and spherical shape with average particle size about 162.5nm.BMH was found to restore the susceptibility of 4T1 cells to ferroptosis under irradiation.This was attributed to the combination of photodynamic therapy(PDT)andγ-glutamylcysteine synthetase inhibitor of BSO,shifting the redox balance to oxidative stress.Enhanced ferroptosis also induced the release of damage associated molecular patterns(DAMPs)to maturate dendritic cells and activated T lymphocytes,leading to superior anti-tumor performance in vivo.Taken together,our findings demonstrated that boosting oxidative damage with photosensitizer serves as an effective strategy to reverse ferroptosis resistance.Chengcheng Wang JiaoWang Xue Pan Shuang Yu Meiqi Chen Yan Gao Zilin Song Haiyang Hu Xiuli Zhao Dawei Chen Fei Han Mingxi Qiao 2023Asian Journal of Pharmaceutical Sciences2023,18,1:0

17Overlay CR-NOMA Assisted Intelligent Transportation System Networks with Imperfect SIC and CEEs显示文摘With the development of the mobile communication and intelligent information technologies,the intelligent transportation systems driven by the sixth generation(6G)has many opportunities to achieve ultralow latency and higher data transmission rate.Nonetheless,it also faces the great challenges of spectral resource shortage and large-scale connection.To solve the above problems,non-orthogonal multiple access(NOMA)and cognitive radio(CR)technologies have been proposed.In this regard,we study the reliable and ergodic performance of CR-NOMA assisted intelligent transportation system networks in the presence of imperfect successive interference cancellation(SIC)and non-ideal channel state information.Specifically,the analytical expressions of the outage probability(OP)and ergodic sum rate(ESR)are derived through a string of calculations.In order to gain more insights,the asymptotic expressions for OP and ESR at high signal-to-noise ratio(SNR)regimes are discussed.We verify the accuracy of the analysis by Monte Carlo simulations,and the results show:i)Imperfect SIC and channel estimation errors(CEEs)have negative impacts on the OP and ESR;ii)The OP decreases with the SNR increasing until convergence to a fixed constant at high SNR regions;iii)The ESR increases with increasing SNR and there exists a ceiling in the high SNR region.LI Xingwang GAO Xuesong LIU Yingting HUANG Gaojian ZENG Ming QIAO Dawei 2023Chinese Journal of Electronics2023,32,6:0
18Laboratory study of astrophysical collisionless shock at SG-Ⅱ laser facility显示文摘Astrophysical collisionless shocks are amazing phenomena in space and astrophysical plasmas, where supersonic flows generate electromagnetic fields through instabilities and particles can be accelerated to high energy cosmic rays. Until now, understanding these micro-processes is still a challenge despite rich astrophysical observation data have been obtained. Laboratory astrophysics, a new route to study the astrophysics, allows us to investigate them at similar extreme physical conditions in laboratory. Here we will review the recent progress of the collisionless shock experiments performed at SG-Ⅱ laser facility in China. The evolution of the electrostatic shocks and Weibel-type/filamentation instabilities are observed. 19Pitx2 suppression at meiotic stages associates with seasonal inhibition of testis development in Rattus norvegicus caraco显示文摘The bicoid-related transcription factor 2(Pitx2)plays a crucial role in the development of many organs and tissues by affecting the mitotic cell cycle.Postnatal testis development is related to mitosis and meiosis in mul-tiple cell types,but the role of Pitx2 gene in seasonal inhibition of testicular development remains unknown in rodents.We analyzed PITX2 protein and Pitx2 mRNA expression features using both laboratory and wild male Rattus norvegicus caraco.In postnatal testicle of laboratory colony,we found that PITX2 was expressed in Leydig cells,pachytene spermatocytes,round spermatids,and elongating spermatids rather than spermatogonia and lep-totene/zygotene spermatocytes.Pitx2b expression significantly increased along with the occurrence of pachytene spermatocytes and round spermatids,while decreased along with the processes of elongated spermatids.In wild male rats with similar testes weight,a significantly suppressed Pitx2b expression occurred with an active meiotic stage in the inhibited testes in autumn and winter,compared with the normally developing testes in spring and summer.These results indicate that Pitx2b expression suppression plays a crucial role in the seasonal inhibition of testis development.Lin TIAN Xixuan LI Yaqi YING Lewen WANG Yanting QIAO Dawei WANG Ying SONG Ning LI Xiaohui LIU 2023Integrative Zoology2023,18,3:0
