| 1 | The chimeric genes in the hybrid lineage of Carassius auratus cuvieri(♀)×Carassius auratus red var.(♂)显示文摘Hybridization can combine the genomes of different strains or species, which leads to changes of genotype and phenotype in the hybrids. In this study, we aimed to investigate the genetic variations of hybrids(WR-F1 and WR-F2) derived from the intraspecific hybridization of white crucian carp(Carassius auratus cuvieri, WCC, ♀) and red crucian carp(Carassius auratus red var., RCC, ♂). Here, we compared the orthologous genes in the liver transcriptomes of hybrids with those of WCC and RCC, and classified the orthologous genes into eight gene patterns within three categories(chimera, mutant, and biparental origin genes).The results revealed 19.04%, 4.17% chimeric genes and 6.90%, 5.05% mutations of orthologous genes in WR-F1 and WR-F2 respectively. Seventeen of twenty-three characterized genes(77%) were confirmed to be the chimeras at the genomic DNA level.The GO classification discovered that some chimeric and mutant genes were related to metabolic process, immune system and developmental process in WR-F1. Our results provide the new evidence that hybridization can combine the parental genomes,leading to changes in the genotype of the resultant hybrids. | 2 | Targeted disruption of tyrosinase causes melanin reduction in Carassius auratus cuvieri and its hybrid progeny显示文摘The white crucian carp(Carassius auratus cuvieri,WCC) not only is one of the most economically important fish in Asia,characterized by strong reproductive ability and rapid growth rates,but also represents a good germplasm to produce hybrid progenies with heterosis.Gene knockout technique provides a safe and acceptant way for fish breeding.Achieving gene knockout in WCC and its hybrid progeny will be of great importance for both genetic studies and hybridization breeding.Tyrosinase(TYR) is a key enzyme in melanin synthesis.Depletion of tyr in zebrafish and mice results in mosaic pigmentation or total albinism.Here,we successfully used CRISPR-Cas9 to target tyr in WCC and its hybrid progeny(WR) derived from the cross of WCC(♀) and red crucian carp(Carassius auratus red var.,RCC,♂).The level of TYR protein was significantly reduced in mutant WCC.Both the mutant WCC and the mutant WR showed different degrees of melanin reduction compared with the wild-type sibling control fish,resulting from different mutation efficiency ranging from 60% to 90%.In addition,the transcriptional expression profiles of a series of pivotal pigment synthesis genes,i.e.tyrp1,mitfa,mitfb,dct and sox10,were down-regulated in tyr-CRISPR WCC,which ultimately caused a reduction in melanin synthesis.These results demonstrated that tyr plays a key role in melanin synthesis in WCC and WR,and CRISPR-Cas9 is an effective tool for modifying the genome of economical fish.Furthermore,the tyr-CRISPR models could be valuable in understanding fundamental mechanisms of pigment formation in non-model fish. | Qingfeng Liu Yanhua Qi Qiuli Liang Jia Song Junmei Liu Wuhui Li Yuqin Shu Min Tao Chun Zhang Qinbo Qin Jing Wang Shaojun Liu | 2019 | Science China(Life Sciences)2019,62,9: | 4 |

The effect of the casting speed on the ingot surface and subsurface layer was studied by surface observation and subsurface structure analysis. It was found that increasing the casting speed results in the adding of segregation knots in the ingot surface. The thickness of the dendrite microstructure layer in the subsurface reduces with increasing the casting speed. | 4 | Hot-top direct chill casting assisted by a twin-cooling field:Improving the ingot quality of a large-size 2024 Al alloy显示文摘Microstructure inhomogeneity and negative segregation have long been challenges for large-size alloy ingots,directly affecting the downstream processing and final performance of products.Here,we used2024 aluminum alloy as a model alloy to propose a technique,named double-cooling field casting,i.e.,one 2024 Al alloy rod(Φ20 mm)at room temperature was introduced into the melt along the central axis of the hot-top with the protection of a thermal-insulation tube during the direct chill(DC)casting process of aΦ300 mm 2024 Al alloy ingot.The results show that the introduction of the same alloy solid insert has a remarkable influence on refining grains in the center region of the ingot,reducing negative centerline segregation and decreasing the depth of the center part of the sump.With the application of the 2024 Al insert,the mean size of equiaxed grains at the center part of the ingot decreased from1204±132μm to 721±69μm.The relative deviation of the Cu and Mg main solutes reduced from-0.062 and-0.054 to-0.03 and-0.024,respectively,and the sump depth decreased from 280 mm to242 mm.Moreover,the shape of the solidification front was changed from‘V’-shaped to‘W’-shaped.The ingot quality was thus improved,mainly arising from the dissolution of the cold 2024 Al insert at a proper position of the hot-top counteracting some latent heat of solidification of the ingot,dissipating the heat of the central part of the hot-top by conducting the 2024 Al insert to the outside,and providing extra-nuclei from the unmoltenα-Al particles of the insert. | Cheng Zhu Zhihao Zhao Qingfeng Zhu Gaosong Wang Yubo Zuo Qiangqiang Li Gaowu Qin | 2022 | Journal of Materials Science & Technology2022,,17: | 1 |
| 5 | A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. | 9 | Geochemistry of rare earth elements and yttrium in Late Permian coals from the Zhongliangshan coalfield, southwestern China显示文摘Rare earth elements and yttrium(REY)in coal deposits are considered promising alternative sources for these resources owing to their increasing global demand.This paper reports the geochemical characteristics of REY in the Late Permian coals from an underground K1a seam section of the Zhongliangshan coalfield in Chongqing,southwestern China.The mineralogy,degree of enrichment,distribution patterns,modes of occurrence,and sediment origin of REY were investigated.Compared with the average of world coals,the concentration of REY in the K1a coals were normal,dominated by light REY(LREY),with less medium and heavy REY(MREY,HREY).The fractionation degree of the MREY and HREY are higher than that of LREY in most K1a coal samples,deduced from the mixed enrichment type of REY,mainly including M-H-type,and a few L-M type and H-type.In addition,the combination of anomalies of Ce,Eu,Gd,and Al_(2)O_(3)/TiO_(2) parameters,the terrigenous materials in the K1a coal were derived from the felsic-intermediate rocks at the top of the Emeishan basalt sequence,and the samples were affected by seawater intrusion during early peat accumulation.Although the minerals primarily consist of kaolinite,illite,pyrite,and small amounts of quartz,calcite and anatase,REY are correlated with ash yield,SiO2,and Al_(2)O_(3),revealing that the REY mainly occur in aluminosilicate minerals,especially kaolinite and illite.Meanwhile,REY positively relate to P_(2)O_(5) and Zr,which may exist in phosphate-containing minerals or zircon.Furthermore,most samples in the K1a coal or ash do not reach the cut-off grade for the beneficial recovery of REY.With the exception of central Guizhou,southwestern Chongqing,and the junction of western Guizhou and northeastern Yunnan,the REY content in coals from southwestern China are high,and its by-products are suitable as potential REY sources. | Qingfeng LU Shenjun QIN Hongyang BAI Wenfeng WANG De’e QI Xin HE Bofei ZHANG | 2023 | Frontiers of Earth Science2023,17,1: | 0 |
