PI3K/Akt/mTOR signaling orchestrates the phenotypic transition and chemo-resistance of small cell lung cancer显示文摘Small cell lung cancer(SCLC)is a phenotypically heterogeneous disease with an extremely poor prognosis, which is mainly attributed to the rapid development of resistance to chemotherapy. However, the relation between the growth phenotypes and chemo-resistance of SCLC remains largely unclear.Through comprehensive bioinformatic analyses,we found that the heterogeneity of SCLC phenotype was significantly associated with different sensitivity to chemotherapy.Adherent or semiadherent SCLC cells were enriched with activation of the PI3K/Akt/mTOR pathway and were highly chemoresistant.Mechanistically, activation of the PI3K/Akt/mTOR pathway promotes the phenotypic transition from suspension to adhesion growth pattern and confers SCLC cells with chemo-resistance.Such chemo-resistance could be largely overcome by combining chemotherapy with PI3K/Akt/mTOR pathway inhibitors. Our findings support that the PI3K/Akt/mTOR pathway plays an important role in SCLC phenotype transition and chemo-resistance,which holds important clinical implications for improving SCLC treatment. | Xuefeng Li Cheng Li Chenchen Guo Qiqi Zhao Jiayu Cao Hsin-Yi Huang Meiting Yue Yun Xue Yujuan Jin Liang Hu Hongbin Ji | 2021 | Journal of Genetics and Genomics2021,48,7: | 5
Characterization of spoilage bacterial communities in chilled duck meat treated by kojic acid显示文摘Chilled duck meat is a popularly consumed meat in China but easily perishes at room/low temperature. Kojic acid is usually used as an antimicrobial. This study investigated the sensory scores, total bacteria number, and total volatile base nitrogen (TVB-N) value of chilled duck meat under kojic acid treatment during storage. The results presented that kojic acid suppressed the growth of spoilage bacteria and prolonged the shelf-life of chilled duck meat by at least two days. In further investigation, the bacterial community composition was explored through high-throughput sequencing. Pseudomonas spp. were the predominant spoilage bacteria after 6-day storage and were significantly suppressed by kojic acid. Besides, Brochothrix showed drug resistance and became the dominant bacteria in the kojic acid treated sample after 6-day storage. In conclusion, kojic acid maintained the sensorial, chemical, and microbial quality of duck meat stored at low temperatures. Meanwhile, kojic acid enriched the bacteria composition and showed strong antimicrobial activity against Pseudomonas spp. | Wenfu Hou Qiqi Yue Wen Liu Ji Wu Yang Yi Hongxun Wang | 2021 | Food Science and Human Wellness2021,10,1: | 3
Glycosylation of dentin matrix protein 1 is critical for fracture healing via promoting chondrogenesis显示文摘Fractures are frequently occurring diseases that endanger human health. Crucial to fracture healing is cartilage formation, which provides a bone-regeneration environment. Cartilage consists of both chondrocytes and extracellular matrix (ECM). The ECM of cartilage includes collagens and various types of proteoglycans (PGs), which play important roles in maintaining primary stability in fracture healing. The PG form of dentin matrix protein 1 (DMP1-PG) is involved in maintaining the health of articular cartilage and bone. Our previous data have shown that DMP1-PG is richly expressed in the cartilaginous calluses of fracture sites. However, the possible significant role of DMP1-PG in chondrogenesis and fracture healing is unknown. To further detect the potential role of DMP1-PG in fracture repair, we established a mouse fracture model by using a glycosylation site mutant DMP1 mouse (S89G-DMP1 mouse). Upon inspection, fewer cartilaginous calluses and down-regulated expression levels of chondrogenesis genes were observed in the fracture sites of S89G-DMP1 mice. Given the deficiency of DMP1-PG, the impaired IL-6/JAK/STAT signaling pathway was observed to affect the chondrogenesis of fracture healing. Overall, these results suggest that DMP1-PG is an indispensable proteoglycan in chondrogenesis during fracture healing. | Hui Xue Dike Tao Yuteng Weng Qiqi Fan Shuang Zhou Ruilin Zhang Han Zhang Rui Yue Xiaogang Wang Zuolin Wang Yao Sun | 2019 | Frontiers of Medicine2019,13,5: | 2
The First High-quality Reference Genome of Sika Deer Provides Insights into High-tannin Adaptation显示文摘Sika deer are known to prefer oak leaves, which are rich in tannins and toxic to most mammals;however, the genetic mechanisms underlying their unique ability to adapt to living in the jungle are still unclear. In identifying the mechanism responsible for the tolerance of a highly toxic diet,we have made a major advancement by explaining the genome of sika deer.We generated the first high-quality,chromosome-level genome assembly of sika deer and measured the correlation between tannin intake and RNA expression in 15 tissues through 180 experiments.Comparative genome analyses showed that the UGT and CYP gene families are functionally involved in the adaptation of sika deer to high-tannin food, especially the expansion of the UGT family 2 subfamily B of UGT genes. The first chromosome-level assembly and genetic characterization of the tolerance to a highly toxic diet suggest that the sika deer genome may serve as an essential resource for understanding evolutionary events and tannin adaptation. Our study provides a paradigm of comparative expressive genomics that can be applied to the study of unique biological features in non-model animals. | Xiumei Xing Cheng Ai Tianjiao Wang Yang Li Huitao Liu Pengfei Hu Guiwu Wang Huamiao Liu Hongliang Wang Ranran Zhang Junjun Zheng Xiaobo Wang Lei Wang Yuxiao Chang Qian Qian Jinghua Yu Lixin Tang Shigang Wu Xiujuan Shao Alun Li Peng Cui Wei Zhan Sheng Zhao Zhichao Wu Xiqun Shao Yimeng Dong Min Rong Yihong Tan Xuezhe Cui Shuzhuo Chang Xingchao Song Tongao Yang Limin Sun Yan Ju Pei Zhao Huanhuan Fan Ying Liu Xinhui Wang Wanyun Yang Min Yang Tao Wei Shanshan Song Jiaping Xu Zhigang Yue Qiqi Liang Chunyi Li Jue Ruan Fuhe Yang | 2023 | Genomics, Proteomics & Bioinformatics2023,21,1: | 1
Constructing dual-functional porphyrin-based thorium metal-organic framework toward photocatalytic uranium(Ⅵ)reduction integrated with organic oxidation显示文摘The rational fabrication of photocatalysts with dual functions upon visible light, such as photocatalytic radioactive U(Ⅵ)reduction and value-added organic oxidation, is highly desirable but remains huge challenge. Here, we couple the photocatalytic U(Ⅵ) reduction with the oxidative organic synthesis to one system using novel extended π-conjugated framework(Cu@ThTCPP) without the expense of sacrificial reagents. Noticeably, the as-prepared Cu@Th-TCPP linked by tetratopic tetrakis(4-carboxyphenyl)porphyrin(TCPP) ligand and unique Th(μ-O)(HCOO)(HO)secondary building unit(SBU) exhibits significantly enhanced activity when the photocatalytic U(Ⅵ) reduction and thioanisole oxidation were integrated to one system.Further experimental characterizations demonstrate that the highly conjugated framework of Cu@Th-TCPP is good for the charge transfer and separation, while incorporating Cusite further accelerates the charge-carrier dynamics, thus giving rise to the dual-functional property. Apparently, this strategy conforms to atomic economy, opens a new horizon to address radioactive environmental pollution in natural water systems and soils, and simultaneously produces valuable chemicals. | Zhi Gao Yue Wang Yuzhi Lin Zhijian Zheng Yao Liu Qiqi Jing Feng Luo | 2022 | Science China Chemistry2022,65,8: | 1
Preparation and magnetic properties of anisotropic Nd2Fe14B/Sm2Co17 hybrid-bonded magnets显示文摘In the present work, anisotropic Nd2 Fe14 B/Sm2 Co17 hybrid-bonded magnets were prepared with different Nd-Fe-B contents. It is found that the particle distributions and ratios between the two magnetic phases have important roles in the magnetic properties, microstructures and thermal stability of the magnets. With increase of Nd-Fe-B content, the saturation magnetization of the anisotropic hybrid magnet increases significantly, however, coercivity decreases, and the demagnetization curves show magnetically single-phase behavior. The anisotropic Nd2 Fe14 B/Sm2 Co17 hybrid-bonded magnets exhibit a maximum energy product and remanence of 14.15 MGOe and 99.53 A·m2/kg, respectively, when the NdFe-B content is 70 wt% at room temperature. Furthermore, the hybrid magnets also have better thermal stability at elevated temperatures due to the interaction between the two magnetic particles. | Xueguo Yin Yue Sui Qiqi Yang Qian Shen Yilong Ma Bin Shao Jianchun Sun Donglin Guo | 2019 | Journal of Rare Earths2019,37,10: | 1
Coupling Mechanism between Tamm Plasmon Polaritons and Monolayer WS_(2) Embedded in Metal/Dielectric Bragg Reflector Hybrid Architecture显示文摘To reveal and utilize the interaction between Tamm plasmon polaritons(TPPs)and two-dimensional materials are promising for exploiting next-generation optoelectronic devices.Herein, the coupling mechanism between metal TPPs and monolayer WS_(2) along with its differences from that between metal TPPs and graphene was studied in detail by using the transfer matrix method. The experimental results show that it is difficult to excite TPPs at the boundary between monolayer WS_(2) and dielectric Bragg reflector(DBR)such that the strong coupling mainly stems from the interaction between metal TPPs and exciton in monolayer WS_(2).However,the coupling in graphene/DBR/metal hybrid structure derives from the interaction between two different TPP resonance modes. Thus, evolutions of Rabi splitting with various structural parameters including spacer thickness, incident angle and DBR period greatly differ from those observed in graphene/DBR/metal hybrid structure. In addition, the discrepancies induced via metal Ag and Au films as well as the possible influence mechanism were also discussed. | 贾光一 YUE Ke YANG Wenxin 黄珍献 LIANG Qiqi LI Yin | 2021 | Journal of Wuhan University of Technology(Materials Science)2021,36,6: | 0
