| 1 | OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. | 6 | Synergistic immunoreaction of acupuncture-like dissolving microneedles containing thymopentin at acupoints in immune-suppressed rats显示文摘Dissolving microneedles carried drug molecules can effectively penetrate the stratum corneum of skin to improve the transdermal drug delivery. The traditional Chinese medicine acupuncture is based on the needle stimulation at a specific location(acupoint) to generate and transmit biochemical and physiological signals which alter the pathophysiological state of patients. However, the pain associated with conventional acupuncture needles and the requirement of highly trained professionals limit the development of acupuncture in non-Asian countries. The purpose of this study is to investigate whether the dissolving microneedles can be utilized as a self-administered painless replacement for acupuncture and locally released drug molecules can achieve expected therapeutic outcomes. Immunosuppressive rats were treated with acupuncture at Zusanli(ST36) acupoint using microneedles containing thymopentin.The immune functions and psychological mood of the immunosuppressed animals were examined. The proliferation of splenocytes was examined by CCK-8 assay. CD4 and CD8 expression patterns in spleen cells were detected by flow cytometry. The current study showed that use of either microneedles containing thymopentin or conventional acupuncture both resulted in immune cell proliferation, which was confirmed by flow cytometry. | 9 | Inactivated Sendai Virus Induces ROS-dependent Apoptosis and Autophagy in Human Prostate Cancer Cells显示文摘Objective The current study aims to investigate the effect of Hemagglutinating virus of Japan envelope(HVJ-E) on induction of apoptosis and autophagy in human prostate cancer PC3 cells, and the underlying mechanisms. Methods PC3 cells were treated with HVJ-E at various multiplicity of infection(MOI), and the generated reactive oxygen species(ROS), cell viability, apoptosis, and autophagy were detected, respectively. Next, the role of ROS played in the regulation of HVJ-E-induced apoptosis and autuphagy in PC3 cells were analysed. In the end, the relationship between HVJ-E-induced apoptosis and autuophagy was investigated by using rapamycin and chloroquine. Results Flow cytometry assay revealed that HVJ-E treatment induced dose-dependent apoptosis and that the JNK and p38 MAPK signaling pathways were involved in HVJ-E-induced apoptosis in PC3 cells. In addition, HVJ-E was able to induce autophagy in PC3 cells via the class III PI3 K/beclin-1 pathway. The data also implyed that HVJ-E-triggered autophagy and apoptosis were ROS dependent. When ROS was blocked with N-acetylcysteine(NAC), HVJ-E-induced LC3-II conversion and apoptosis were reversed. Interestingly, HVJ-E-induced apoptosis was significantly increased by an inducer of autophagy, rapamycin pretreatment, both in vitro and in vivo. | 12 | Porous LaFeO3 nanofiber with oxygen vacancies as an efficient electrocatalyst for N2 conversion to NH3 under ambient conditions显示文摘Electrocatalytic N2 reduction to NH3 under ambient conditions is an eco-friendly and sustainable alternative to the traditional Haber-Bosch process. However, inhibited by the high activation barrier of N2, this process needs efficient electrocatalysts to adsorb and activate the N2, enabling the N2 reduction reaction(NRR). Herein, we report that porous LaFeO3 nanofiber with oxygen vacancies acts as an efficient NRR electrocatalyst with abundant active sites to enhance the adsorption and activation of N2. When tested in 0.1 M HCl, such electrocatalyst achieves a high Faradaic efficiency of 8.77% and a large NH3 yield rate of 18.59 μg h–1 mgcat–1.at-0.55 V versus reversible hydrogen electrode. This catalyst also shows high long-term electrochemical stability and excellent selectivity for NH3 formation. | 13 | Food Frequency Questionnaire for Chinese Children Aged 12-17 Years: Validity and Reliability显示文摘Objective The primary objective of this study was to examine the validity and reliability of a semi-quantitative food frequency questionnaire(FFQ) among Chinese children aged 12-17 years. Methods A semi-quantitative 72-food item FFQ was developed for children aged 12-17 years. The reliability and validity of this FFQ were evaluated against 24-h dietary recalls(24 h DRs) to measure the consumption of foods and nutrients. We administered two FFQs and three DRs to children(N = 160) over a period of 1 month to evaluate the reliability and validity. Reliability was examined by quartile agreement and intraclass correlation coefficients(ICCs), and validity was examined by quartile agreement, Bland-Altman plots and correlation with DRs. Results For reliability, the ICCs between the two FFQs ranged from 0.21 to 0.76 for foods and nutrients, and the quartile agreement ranged from 70.0% to 95.0% in the same or adjacent quartiles. Spearman’s correlation coefficients of foods and nutrients between the second FFQ and the 24 h DRs ranged from-0.04 to 0.59. The Bland-Altman plots demonstrated good agreement across the range of intakes among nutrients. The quartile agreement ranged from 50.0% to 100.0%, with infrequent misclassification. | 17 | The Impact of Relative Humidity and Atmospheric Pressure on Mortality in Guangzhou, China显示文摘Objective Although many studies have examined the effects of ambient temperatures on mortality, little evidence is on health impacts of atmospheric pressure and relative humidity. This study aimed to assess the impacts of atmospheric pressure and relative humidity on mortality in Guangzhou, China. Methods This study included 213,737 registered deaths during 2003-2011 in Guangzhou, China. A quasi-Poisson regression with a distributed lag non-linear model was used to assess the effects of atmospheric pressure/relative humidity. Results We found significant effect of low atmospheric pressure/relative humidity on mortality. There was a 1.79%(95% confidence interval: 0.38%-3.22%) increase in non-accidental mortality and a 2.27%(0.07%-4.51%) increase in cardiovascular mortality comparing the 5th and 25 th percentile of atmospheric pressure. A 3.97%(0.67%-7.39%) increase in cardiovascular mortality was also observed comparing the 5th and 25 th percentile of relative humidity. Women were more vulnerable to decrease in atmospheric pressure and relative humidity than men. Age and education attainment were also potential effect modifiers. Furthermore, low atmospheric pressure and relative humidity increased temperature-related mortality. Conclusion Both low atmospheric pressure and relative humidity are important risk factors of mortality. | 18 | Background-free latent fingerprint imaging based on nanocrystals with long-lived luminescence and pH-guided recognition显示文摘Latent fingerprints (LFPs)are highly specific to individuals,and LFP imaging has played an important role in areas such as forensic investigation and law enforcement.Presently,LFP imaging still faces considerable problems,including background interference and destructive and complex operations.Herein,we have designed a background-free,nondestructive,and easy-to-perform method for LFP imaging based on pH-mediated recognition of LFPs by carboxyl group- functionalized Zn2GeO4:Mn (ZGO:Mn-COOH)persistent luminescence nanorods (PLNRs).By simply adjusting the pH of the ZGO:Mn-COOH colloid dispersion to a certain acidic range,the negatively charged ZGO:Mn-COOH readily binds to protonated fingerprint ridges via electrostatic attraction.The ZGO:Mn-COOH colloid dispersion can be stored in portable commercial spray bottles,and the LFPs have been easily detected in situ by simply dropping the colloid dispersion on the LFPs.Moreover,since the ZGO:Mn-COOH can remain luminescent after excitation ceases,background color and background fluorescence interference were efficiently removed by simply capturing the luminescent LFP images after the excitation ceased.The entire LFP imaging process can be easily conducted without any destructive or complex operations.Due to the great versatility of the developed method for LFP imaging,clear LFP images with well-resolved ridge patterns were obtained.The designed background-free,nondestructive,and easy-to-perform LFP imaging strategy has great potential for future applications, such as forensic investigations and law enforcement. | Zhiheng Li Qian Wang Yingqian Wang Qinqin Ma Jie Wang Zhihao Li Yingxue Li Xiaobo Lv Wei Wei Lang Chen Quan Yuan | 2018 | Nano Research2018,11,12: | 5 |
