| 1 | Effect of Bcl-2 and Bax on survival of side population cells from hepatocellular carcinoma cells显示文摘AIM: To understand the role and significance of side population (SP) cells from hepatocellular carcinoma (HCC) in hepatocarcinogenesis, development, relapse and metastasis, we simulated the denutrition conditions that cancer cells experience in clinical therapy, observed the different anti-apoptosis ability of SP cells and non-SP cells under such conditions, and established the possible effects of P53, Bcl-2 and Bax on survival of SP cells. METHODS: We used ? ow cytometry to analyze and sort the SP and non-SP cells in established HCC lines MHCC97 and hHCC. We evaluated cell proliferation by methyl thiazolyl tetrazolium (MTT) assay and investigated the expression of p53, bcl-2 and bax genes during denutrition, by RT-PCR and immuno? uorescence staining. RESULTS: The percentage of SP cells in the two established HCC lines was 0.25% and 0.5%, respectively. SP cells had greater anti-apoptosis and proliferation ability than non-SP cells. Expression of Bcl-2 and Bax in SP and non-SP cells differed during denutrition. The former was up-regulated in SP cells, and the latter was up-regulated in non-SP cells. CONCLUSION: It may be that different upstream molecules acted and led to different expression levels of Bcl-2 and Bax in these two cell lines. There was a direct relationship between up-regulation of Bcl-2 and down-regulation of Bax and higher anti-apoptosis ability in SP cells. It may be that the existence and activity of SP cells are partly responsible for some of the clinical phenomena which are seen in HCC, such as relapse or metastasis. | 2 | Size distribution of chemical elements and their source apportionment in ambient coarse, fine, and ultrafine particles in Shanghai urban summer atmosphere显示文摘Ambient coarse particles (diameter 1.8-10 μm), fine particles (diameter 0.1-1.8 μm), and ultrafine particles (diameter < 0.1 μm) in the atmosphere of the city of Shanghai were sampled during the summer of 2008 (from Aug 27 to Sep 08). Microscopic characterization of the particles was investigated by scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM/EDX). Mass concentrations of Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Rb, Sr, and Pb in the size-resolved particles were quantified by using synchrotron radiation X-ray fluorescence (SRXRF). Source apportionment of the chemical elements was analyzed by means of an enrichment factor method. Our results showed that the average mass concentrations of coarse particles, fine particles and ultrafine particles in the summer air were 9.38 ± 2.18, 8.82 ± 3.52, and 2.02 ± 0.41 μg/m3, respectively. The mass percentage of the fine particles accounted for 51.47% in the total mass of PM10, indicating that fine particles are the major component in the Shanghai ambient particles. SEM/EDX results showed that the coarse particles were dominated by minerals, fine particles by soot aggregates and fly ashes, and ultrafine particles by soot particles and unidentified particles. SRXRF results demonstrated that crustal elements were mainly distributed in the coarse particles, while heavy metals were in higher proportions in the fine particles. Source apportionment revealed that Si, K, Ca, Fe, Mn, Rb, and Sr were from crustal sources, and S, Cl, Cu, Zn, As, Se, Br, and Pb from anthropogenic sources. The disadvantage of CS is that the number of iterations in a greedy algorithm such as Orthogonal Matching Pursuit(OMP) is fixed, thus limiting reconstruction precision.Therefore, in this study, we present a novel Reducing Iteration Orthogonal Matching Pursuit(RIOMP) algorithm that calculates the correlation of the residual value and measurement matrix to reduce the number of iterations.The conditions for successful signal reconstruction are derived on the basis of detailed mathematical analyses.When compared with the OMP algorithm, the RIOMP algorithm has a smaller reconstruction error. | 9 | SbHKT1;4, a member of the high‐affinity potassium transporter gene family from Sorghum bicolor, functions to maintain optimal Na^+/K^+ balance under Na^+ stress显示文摘In halophytic plants, the high‐affinity potassium transporter HKT gene family can selectively uptake Kt in the presence of toxic concentrations of Nat. This has so far not been well examined in glycophytic crops. Here, we report the characterization of SbHKT1;4, a member of the HKT gene family from Sorghum bicolor. Upon Nat stress, SbHKT1;4 expression was more strongly upregulated in salt‐tolerant sorghum accession, correlating with a better balanced Nat/ Kt ratio and enhanced plant growth. Heterogeneous expression analyses in mutants of Saccharomyces cerevisiae and Arabidopsis thaliana indicated that overexpressing SbHKT1;4 resulted in hypersensitivity to Nat stress, and such hypersensitivity could be alleviated with the supply of elevated levels of Kt, implicating that SbHKT1;4 may mediate Kt uptake in the presence of excessive Nat. Further electrophysiological evidence demonstrated that SbHKT1;4 could transport Nat and Kt when expressed in Xenopus laevis oocytes. | 10 | Spatial and temporal variation of haze in China from 1961 to 2012显示文摘The purpose of this study is to analyze the climatic characteristics and long-term spatial and temporal variations of haze occurrence in China. The impact factors of haze trends are also discussed. Meteorological data from 1961 to 2012 and daily PM10 concentrations from2003 to 2012 were employed in this study. The results indicate that the annual-average hazy days at all stations have been increasing rapidly from 4 days in 1961 to 18 days in 2012. The maximum number of haze days occur in winter(41.1%) while the minimum occur in summer(10.4%). During 1961-2012, the high occurrence areas of haze shifted from central to south and east regions of China. The Beijing-Tianjin-Hebei(Jing-Jin-Ji) region, Shanxi,Shaanxi, and Henan Province are the high occurrence areas for haze, while the Yangtze River Delta(YRD) and the Pearl River Delta(PRD) have become regions with high haze occurrences in the last 25 years. Temperature and pressure are positively correlated with the number of haze days. However, wind, relative humidity, precipitation, and sunshine duration are negatively correlated with the number of haze days. The key meteorological factors affecting the formation and dissipation of haze vary for high and low altitudes, and are closely related to anthropogenic activities. | 11 | Using borehole core analysis to reveal Late Quaternary paleoearthquakes along the Nankou-Sunhe Fault,Beijing显示文摘The Nankou-Sunhe Fault is a buried active normal fault that traverses the urban area of Beijing.Its seismic risks have caused considerable concerns.This paper studies paleoearthquakes along this fault by analyzing and correlating bore-hole cores obtained from triple-tube coring,incorporating experience acquired from trenching.As a result,a model for identifying earthquake-derived colluvium by sediment-core analysis is proposed.Triple-tube coring technique is useful to collect continuous undis-turbed soil core near the Nankou-Sunhe Fault.By identifying fault-scarp colluviums,determining cumulative displacement,and analysing stratum thickening on the hanging wall,we are able to establish a preliminary paleoearthquake sequence consisting of 13 surface-rupturing events since 60 ka.The seismic history can be divided into three periods based on different recurrence intervals.Between 60 and 40 ka,three earthquakes occurred with recurrence interval of ~10 ka.From 40 to 25 ka,there were six earthquakes with the recurrence interval of about 2.5 ka.In the last 25 ka,four earthquakes have taken place with the recurrence interval varying considerably.The recurrence interval between the last three events is ~5 ka.Smaller recurrence intervals correspond to stages of faster fault slip.The coseismic displacement of a single event is 0.8 to 2.2 m,average 1.4 m,largely equivalent to moment magnitudes 6.7-7.1.This study demonstrates the feasibility of bore-hole drilling in investigating paleoearthquakes along normal faults.It also suggests that closely spaced boreholes with continuous undisturbed cores are essential for reconstructing the complete paleoearthquake sequence. | ZHANG ShiMin WANG DanDan LIU XuDong ZHANG GuoHong ZHAO JunXiang LUO MingHui REN JunJie WANG Rui ZHANG YingLi | 2008 | Science China Earth Sciences2008,51,8: | 11 |
| 12 | Fine Mapping and Candidate Gene Analysis of Resistance Gene R_(SC3Q) to Soybean mosaic virus in Qihuang 1显示文摘Soybean mosaic virus(SMV) disease is one of the most destructive viral diseases in soybean(Glycine max(L.) Merr.). SMV strain SC3 is the major prevalent strain in Huang-Huai and Yangtze valleys, China. The soybean cultivar Qihuang 1 is of a rich resistance spectrum and has a wide range of application in breeding programs in China. In this study, F1, F2 and F2:3 from Qihuang 1×Nannong 1138-2 were used to study inheritance and linkage mapping of the SC3 resistance gene in Qihuang 1. The secondary F2 population and near isogenic lines(NILs) derived from residual heterozygous lines(RHLs) of Qihuang 1×Nannong 1138-2 were separatively used in the fine mapping and candidate gene analysis of the resistance gene. Results indicated that a single dominant gene(designated RSC3Q) controls resistance, which was located on chromosome 13. Two genomic-simple sequence repeat(SSR) markers BARCSOYSSR_13_1114 and BARCSOYSSR_13_1136 were found flanking the two sides of the RSC3 Q. The interval between the two markers was 651 kb. Qu antitative real-time PCR analysis of the candidate genes sho wed that five genes(Glyma13g25730, 25750, 25950, 25970 and 26000) were likely involved in soybean SMV resistance. | 16 | Effect of heparin on apoptosis in human nasopharyngeal carcinoma CNE2 cells显示文摘In order to study the mechanism of the effect of heparin on apoptosis in carcinoma cells, the nasopharyngeal carcinoma cell line CNE2 was used to identify the effect of heparin on apoptosis associated with the expression of c-myc, bax, bcl-2 proteins by use of Hoechst 33258 staining, terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), agarose gel electrophoresis, and flow cytometry, as well as Western blot analysis. The results showed that heparin induced apoptosis of CNE2 cells including the morphologic changes such as reduction in the volume, and the nuclear chromatin condensation, as well as the “ladder pattern” revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner.The number of TUNEL-positive cells was dramatically increased to 33.6±1.2% from 2.8±0.3% by treat ment with heparin in different concentrations (10~40 kU/L). The apoptotic index was increased to 32.5% from 3.5% by detecting SubG1 peaks on flow cytometry. | 17 | Percent free prostate-specific antigen for prostate cancer diagnosis in Chinese men with a PSA of 4.0-10.0 ng/mL:Results from the Chinese Prostate Cancer Consortium显示文摘Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years. | Rui Chen Liping Xie Xiaobing Cai Yiran Huang Liqun Zhou Lulin Ma Xu Gao Chuanliang Xu Shancheng Ren Pengfei Shao Danfeng Xu Kexin Xu Zhangqun Ye Chunxiao Liu Dingwei Ye Li Lu Qiang Fu Jianquan Hou Jianlin Yuan Dalin He Tie Zhou Fubo Wang Biming He Yinghao Sun | 2015 | Asian Journal of Urology2015,2,2: | 8 |

| 19 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome.
