1Origin and Distribution of Groundwater Chemical Fields of the Oilfield in the Songliao Basin,NE China显示文摘There are many factors affecting the chemical characteristics of groundwater in the forming process of groundwater chemical fields, such as freshening due to meteoric water leaching downwards, freshening due to mudstone compaction and water release, concentration due to infiltration and freshening due to dehydration of clay minerals. As a result, the groundwater chemical fields are characterized by lengthwise stages and planar selectivity. The former arouses vertical chemical zonality of groundwater. Five units could be identified downwards in the Songliao basin: (1) freshening zone due to downward-leaching meteoric water, (2) concentration zone due to evaporation near the ground surface, (3) freshening zone due to mudstone compaction and water release, and concentration zone due to compaction and infiltration, (4) freshening zone due to dehydration of clay minerals, and (5) filtration-concentration zone; whereas the latter determines the planar division of groundwater chemical fields: (1) the freshening area due to meteoric water, asymmetrically on the margin of the basin, (2) the freshening area due to mudstone compaction and water release in the central part of the basin, (3) the leaky area, which is a transitional zone, and (4) leakage-evaporation area, which is a concentration zone. In the direction of centrifugal flows caused by mudstone compaction in the depression area, the mineralization degree, concentrations of Na+ and Cl?, and salinity coefficient (SC) increase, while concentrations of (CO32 ?+HCO3 ?) and SO42 ?, metamorphism coefficient (MC) and desulfuration coefficient (DSC) decrease. 3Severe human infection with a novel avian-origin influenza A(H7N4) virus显示文摘Human infections with influenza H7 subtypes, such as H7N9, have raised concerns worldwide. Here, we report a human infection with a novel influenza A(H7N4) virus. A 68 years-old woman with cardiovascular and cholecystic comorbidities developed rapidly progressed pneumonia with influenza-like-illness as initial symptom, recovered after 23 days-hospitalization including 8 days in ICU. Laboratory indicators for liver and blood coagulation dysfunction were observed. Oseltamivir phosphate, glucocorticoids and antibiotics were jointly implemented, with nasal catheterization of oxygen inhalation for this patient.We obtained the medical records and collected serial respiratory and blood specimens from her. We collected throat, cloacal and/or feces samples of poultry and wild birds from the patient's backyard, neighborhood, local live poultry markets(LPMs) and the nearest lake. All close contacts of the patient were followed up and sampled with throat swabs and sera. Influenza viruses and other respiratory pathogens were tested by real-time RT-PCR, viral culturing and/or sequencing for human respiratory and bird samples. Micro-neutralizing assay was performed for sera. A novel reassortant wild bird-origin H7N4 virus is identified from the patient and her backyard poultry(chickens and ducks) by sequencing, which is distinct from previously-reported avian H7N4 and H7N9 viruses. At least four folds increase of neutralizing antibodies to H7N4 was detected in her convalescent sera. 4Antibiotics-mediated intestinal microbiome perturbation aggravates tacrolimus-induced glucose disorders in mice显示文摘Both immunosuppressants and antibiotics (ABX) are indispensable for transplant patients. However, the former increases the risk of new-onset diabetes, whereas the latter impacts intestinal microbiota (IM). It is still unclear whether and how the interaction between immunosuppressants and ABX alters the IM and thus leads to glucose metabolism disorders. This study examined the alterations of glucose and lipid metabolism and IM in mice exposed to tacrolimus (TAC) with or without ABX. We found that ABX further aggravated TAC-induced glucose tolerance and increased insulin secretion. Combined treatment resulted in exacerbated lipid accumulation in the liver. TAC-altered microbial community was further amplified by ABX administration, as characterized by reductions in phylum Firmicutes, family Lachnospiraceae, and genus Copyococcus. Analyses based on the metagenomic profiles revealed that ABX augmented the effect of TAC on microbial metabolic function mostly related to lipid metabolism. The altered components of gut microbiome and predicted microbial functional profiles showed significant correlation with hepatic lipid accumulation and glucose disorders. In conclusion, ABX aggravated the effect of TAC on the microbiome and its metabolic capacities, which might contribute to hepatic lipid accumulation and glucose disorders. 6FLT3 tyrosine kinase inhibitors synergize with BCL-2 inhibition to eliminate FLT3/ITD acute leukemia cells through BIM activation显示文摘Tyrosine kinase inhibitors(TKIs)targeting FLT3 have shown activity but when used alone have achieved limited success in clinical trials,suggesting the need for combination with other drugs.We investigated the combination of FLT3 TKIs(Gilteritinib or Sorafenib),with Venetodax,a BCL-2 selective inhibitor(BCL-2i),on FLT3/ITD leukemia cells.The combination of a FLT3 TKI and a BCL-2i synergistically reduced cell proliferation and enhanced apoptosis/cell death in FLT3/ITD cell lines and primary AML samples.Venetoclax also re-sensitized FLT3 TKI-resistant cells to Gilteritinib or Sorafenib treatment,mediated through MAPK pathway inhibition.Gilteritinib treatment alone dissociated BIM from MCL-1 but increased the binding of BIM to BCL-2.Venetoclax treatment enhanced the binding of BIM to MCL-1 but dissociated BIM from BCL-2.Treatment with the drugs together resulted in dissociation of BIM from both BCL-2 and MCL-1,with an increased binding of BIM to the cell death mediator BAX,leading to increased apoptosis.These findings suggest that Venetoclax mitigates the unintended pro-survival effects of FLT3 TKI mainly through the dissociation of BIM and BCL-2 and also decreased BIM expression.This study provides evidence that the addition of BCL-2i enhances the effect of FLT3 TKI therapy in FLT3/ITD AML treatment.Ruiqi Zhu Li Li Bao Nguyen Jaesung Seo Min Wu Tessa Seale Mark Levis Amy Duffield Yu Hu Donald Small 2021Signal Transduction and Targeted Therapy2021,6,6:2

8Rabies Virus Pseudotyped with CVS-N2C Glycoprotein as a Powerful Tool for Retrograde Neuronal Network Tracing显示文摘Efficient viral vectors for mapping and manipulating long-projection neuronal circuits are crucial in structural and functional studies of the brain. The SAD strain rabies virus with the glycoprotein gene deleted pseudotyped with the N2 C glycoprotein(SAD-RV(DG)-N2 C(G)) shows strong neuro-tropism in cell culture, but its in vivo efficiency for retrograde gene transduction and neuro-tropism have not been systematically characterized.We compared these features in different mouse brain regions for SAD-RV-N2 C(G) and two other widely-used retrograde tracers, SAD-RV(DG)-B19(G) and r AAV2-retro. We found that SAD-RV(DG)-N2 C(G) enhanced the infection efficiency of long-projecting neurons by^10 times but with very similar neuro-tropism, compared with SAD-RV(DG)-B19(G). On the other hand, SAD-RV(DG)-N2 C(G) had an infection efficiency comparable with r AAV2-retro, but a more restricted diffusion range, and broader tropism to different types and regions of longprojecting neuronal populations. 9The C/EBPβ-Dependent Induction of TFDP2 Facilitates Porcine Reproductive and Respiratory Syndrome Virus Proliferation显示文摘Porcine reproductive and respiratory syndrome(PRRS) is an important infectious disease caused by porcine reproductive and respiratory syndrome virus(PRRSV), leading to significant economic losses in swine industry worldwide. Although several studies have shown that PRRSV can affect the cell cycle of infected cells, it is still unclear how it manipulates the cell cycle to facilitate its proliferation. In this study, we analyzed the mRNA expression profiles of transcription factors in PRRSV-infected 3D4/21 cells by RNA-sequencing. The result shows that the expression of transcription factor DP2(TFDP2) is remarkably upregulated in PRRSV-infected cells. Further studies show that TFDP2 contributes to PRRSV proliferation and the PRRSV nucleocapsid(N) protein induces TFDP2 expression by activating C/EBPb. TFDP2 positively regulates cyclin A expression and triggers a less proportion of cells in the S phase, which contributes to PRRSV proliferation. This study proposes a novel mechanism by which PRRSV utilizes host protein to regulate the cell cycle to favor its infection.
