| 1 | m^6A-dependent biogenesis of circular RNAs in male germ cells显示文摘The majority of circular RNAs(circRNAs)spliced from coding genes contain open reading frames(ORFs)and thus,have protein coding potential.However,it remains unknown what regulates the biogenesis of these ORF-containing circRNAs,whether they are actually translated into proteins and what functions they play in specific physiological contexts.Here,we report that a large number of circRNAs are synthesized with increasing abundance when late pachytene spermatocytes develop into round and then elongating spermatids during murine spermatogenesis.For a subset of circRNAs,the back splicing appears to occur mostly at m^6A-enriched sites,which are usually located around the start and stop codons in linear mRNAs.Consequently,approximately a half of these male germ cell circRNAs contain large ORFs with m^6A-modified start codons in their junctions,features that have been recently shown to be associated with protein-coding potential.Hundreds of peptides encoded by the junction sequences of these circRNAs were detected using liquid chromatography coupled with mass spectrometry,suggesting that these circRNAs can indeed be translated into proteins in both developing(spermatocytes and spermatids)and mature(spermatozoa)male germ cells.The present study discovered not only a novel role of m^6A in the biogenesis of coding circRNAs,but also a potential mechanism to ensure stable and long-lasting protein production in the absence of linear mRNAs,i.e.,through production of circRNAs containing large ORFs and m^6A-modified start codons in junction sequences. | Chong Tang Yeming Xie Tian Yu Na Liu Zhuqing Wang Rebekah J.Woolsey Yunge Tang Xinzong Zhang Weibing Qin Ying Zhang Ge Song Weiwei Zheng Juan Wang Weitian Chen Xiongyi Wei Zhe Xie Rachel Klukovich Huili Zheng David R.Quilici Wei Yan | 2020 | Cell Research2020,30,3: | 12 |

| 4 | LRRK2, GBA and their interaction in the regulation of autophagy: implications on therapeutics in Parkinson’s disease显示文摘Mutations in leucine-rich repeat kinase 2 (LRRK2) and glucocerebrosidase (GBA) represent two most common genetic causes of Parkinson’s disease (PD). Both genes are important in the autophagic-lysosomal pathway (ALP), defects of which are associated with α-synuclein (α-syn) accumulation. LRRK2 regulates macroautophagy via activation of the mitogen activated protein kinase/extracellular signal regulated protein kinase (MAPK/ERK) kinase (MEK) and the calcium-dependent adenosine monophosphate (AMP)-activated protein kinase (AMPK) pathways. Phosphorylation of Rab GTPases by LRRK2 regulates lysosomal homeostasis and endosomal trafficking. Mutant LRRK2 impairs chaperone-mediated autophagy, resulting in α-syn binding and oligomerization on lysosomal membranes. Mutations in GBA reduce glucocerebrosidase (GCase) activity, leading to glucosylceramide accumulation, α-syn aggregation and broad autophagic abnormalities. LRRK2 and GBA influence each other: GCase activity is reduced in LRRK2 mutant cells, and LRRK2 kinase inhibition can alter GCase activity in GBA mutant cells. Clinically, LRRK2 G2019S mutation seems to modify the effects of GBA mutation, resulting in milder symptoms than those resulting from GBA mutation alone. However, dual mutation carriers have an increased risk of PD and earlier age of onset compared with single mutation carriers, suggesting an additive deleterious effect on the initiation of PD pathogenic processes. Crosstalk between LRRK2 and GBA in PD exists, but its exact mechanism is unclear. Drugs that inhibit LRRK2 kinase or activate GCase are showing efficacy in pre-clinical models. | 5 | Regulating the morphology of fluorinated non-fullerene acceptor and polymer donor via binary solvent mixture for high efficiency polymer solar cells显示文摘Fluorinated non-fullerene acceptors(NFAs) usually have planar backbone and a higher tendency to crystallize compared to their non-fluorinated counterparts, which leads to enhanced charge mobility in organic solar cells(OSCs). However, this selforganization behavior may result in excessive phase separation with electron donors and thereby deteriorate device efficiency.Herein, we demonstrate an effective approach to tune the molecular organization of a fluorinated NFA(INPIC-4 F), and its phase separation with the donor PBDB-T, by varying the casting solvent. A prolonged film drying time encourages the crystallization of INPIC-4 F into spherulites and consequently results in excessive phase separation, leading to a low device power conversion efficiency(PCE) of 8.1%. Contrarily, a drying time leads to fine mixed domains with inefficient charge transport properties,resulting in a moderate device PCE of 11.4%. An intermediate film drying time results in the formation of face-on π-π stacked PBDB-T and INPIC-4 F domains with continuous phase-separated networks, which facilitates light absorption, exciton dissociation as well as balanced charge transport towards the electrode, and achieves a remarkable PCE of 13.1%. | 19 | Three X-ray flares near primary eclipse of the RS CVn binary XY UMa显示文摘We report on an archival X-ray observation of the eclipsing RS CVn binary XY UMa(P_(orb) ≈0.48 d). In two Chandra ACIS observations spanning 200 ks and almost five orbital periods, three flares occurred. We find no evidence for eclipses in the X-ray flux. The flares took place around times of primary eclipse, with one flare occurring shortly(< 0.125 P_(orb)) after a primary eclipse, and the other two happening shortly(< 0.05 P_(orb)) before a primary eclipse. Two flares occurred within roughly one orbital period(?φ≈1.024 P_(orb)) of each other. We analyze the light curve and spectra of the system, and investigate coronal length scales during both quiescence and flares, as well as the timing of the flares.
