6Extracellular vesicles from human urine-derived stem cells prevent osteoporosis by transferring CTHRC1 and OPG显示文摘Osteoporosis is a debilitating bone disease affecting millions of people. Here, we used human urine-derived stem cells(USCs),which were noninvasively harvested from unlimited and easily available urine, as a 'factory' to obtain extracellular vesicles(USCEVs) and demonstrated that the systemic injection of USC-EVs effectively alleviates bone loss and maintains bone strength in osteoporotic mice by enhancing osteoblastic bone formation and suppressing osteoclastic bone resorption. More importantly, the anti-osteoporotic properties of USC-EVs are not notably disrupted by the age, gender, or health condition(with or without osteoporosis) of the USC donor. Mechanistic studies determined that collagen triple-helix repeat containing 1(CTHRC1) and osteoprotegerin(OPG) proteins are enriched in USC-EVs and required for USC-EV-induced pro-osteogenic and anti-osteoclastic effects. 7A rapid reporter assay for recombinant human brain natriuretic peptide(rhBNP)by GloSensor technology显示文摘Accurate determination of biological activity is essential in quality control of recombinant human brain natriuretic peptide(rhBNP). In previous study, we successfully developed a genetically modified cell line 293GCAC3-based ELISA assay for rhBNP. But ELISA procedure is still tedious, so this study was aimed to develop a rapid and simple bioassay for rhBNP using GloSensor technology, which provides a platform of flexible luciferase-based biosensors for real-time detection of signaling events in live cells, including cGMP production. A reporter cell line 293GCAGlo-G1 was constructed by transfecting pGloSensor^(TM) 40 F plasmid into 293GCAC3. The reporter assay based on 293GCAGlo-G1 showed high precision with intraassay CV being 8.3% and inter-assay CV being 14.1%; high accuracy with 80%, 100% and 120% recovery rate being 99.2%, 102.4% and 99.0% respectively; and great linearity with R^2 of linear fitting equation being 0.99. Besides, no significant difference was found in test results of reporter assay and 293GCAC3-based ELISA assay(paired t test, p=0.630). 8Spatial transmission and meteorological determinants of tuberculosis incidence in Qinghai Province,China:a spatial clustering panel analysis显示文摘Background:Tuberculosis(TB)is the notifiable infectious disease with the second highest incidence in the Qinghai province,a province with poor primary health care infrastructure.Understanding the spatial distribution of TB and related environmental factors is necessary for developing effective strategies to control and further eliminate TB.Methods:Our TB incidence data and meteorological data were extracted from the China Information System of Disease Control and Prevention and statistical yearbooks,respectively.We calculated the global and local Moran’s I by using spatial autocorrelation analysis to detect the spatial clustering of TB incidence each year.A spatial panel data model was applied to examine the associations of meteorological factors with TB incidence after adjustment of spatial individual effects and spatial autocorrelation.Results:The Local Moran’s I method detected 11 counties with a significantly high-high spatial clustering(average annual incidence:294/100000)and 17 counties with a significantly low-low spatial clustering(average annual incidence:68/100000)of TB annual incidence within the examined five-year period;the global Moran’s I values ranged from 0.40 to 0.58(all P-values<0.05).The TB incidence was positively associated with the temperature,precipitation,and wind speed(all P-values<0.05),which were confirmed by the spatial panel data model.Each 10°C,2 cm,and 1 m/s increase in temperature,precipitation,and wind speed associated with 9%and 3%decrements and a 7%increment in the TB incidence,respectively.Conclusions:High TB incidence areas were mainly concentrated in south-western Qinghai,while low TB incidence areas clustered in eastern and north-western Qinghai.Areas with low temperature and precipitation and with strong wind speeds tended to have higher TB incidences.Hua-Xiang Rao Xi Zhang Lei Zhao Juan Yu Wen Ren Xue-Lei Zhang Yong-Cheng Ma Yan Shi Bin-Zhong Ma Xiang Wang Zhen Wei Hua-Fang Wang Li-Xia Qiu 2016Infectious Diseases of Poverty2016,5,1:10
9Macro-and Microstructure Evolution of 5CrNiMo Steel Ingots during Electroslag Remelting Process显示文摘A comprehensive mathematical model was established and used to simulate the macro-and microstructure evolution during the production process of 5CrNiMo steel ingot by electroslag remelting(ESR)method.Along the ingot height,the macrostructure distribution characteristics changed from vertical,fine columnar grains to tilted,coarse columnar grains,and this transformation process occurred at the very beginning of ESR.In the cross section of the ingot,there were three grain morphology regions and two grain type transition regions from the outside to the center of the ingot.These regions were the fine columnar grain region,columnar competitive growth transition region,coarse columnar grain region,columnar to equiaxed grain transition(CET)region,and coarse equiaxed grain region.The influence of the remelting rate on the macrostructure and microstructure was investigated using a series of experiments and simulations.The results showed that a low remelting rate could produce a small grain growth angle(GGA);the average secondary dendrite arm spacing(SDAS)firstly decreased and then increased as the remelting rate increased.An excessively high or low remelting rate can increase the GGA and average SDAS in ingots.Thus,the remelting rate should be controlled within a suitable range to reduce composition microsegregation and microshrinkage in the ingot to produce an ESR ingot with satisfactory hot forging performance.Lei RAO Jian-hua ZHAO Zhan-xi ZHAO Gang DING Mao-peng GENG 2014Journal of Iron and Steel Research(International)2014,21,7:10
10CD30 expression in extranodal natural killer/T-cell lymphoma,nasal type among 622 cases of mature T-cell and natural killer-cell lymphoma at a single institution in South China显示文摘Background:Mature T-cell and natural killer(NK)-cell lymphomas compose a heterogeneous group of non-Hodgkin lymphomas,and extranodal NK/T-cell lymphoma,nasal type(ENKTL) is an aggressive subtype with sporadic CD30 expression.However,the significance of CD30 expression in ENKTL is controversial.We aimed to classify a large cohort of patients with mature T-cell and NK-cell lymphomas according to the 2016 World Health Organization(WHO) classification guidelines and to study the association between CD30 expression and prognosis of patients with ENKTL.Methods:We selected consecutive patients with mature T-cell and NK-cell lymphomas who attended our institution between September 1,2009 and August 31,2013.We classified the lymphomas according to the 2016 revision of the WHO classification of lymphoid neoplasms,analyzed the associations between CD30 expression and clinicopathologic features of ENKTL patients,and evaluated the prognostic implications of CD30 expression.Results:We identified 622 consecutive patients with mature T-cell and NK-cell lymphomas,including 317(51.0%)patients with ENKTL.In addition,CD30 expression was detected in 43(47.3%) of a subset of 91 patients with ENKTL.No clinicopathologic features were associated with CD30 expression,and CD30 positivity showed no prognostic significance in patients with ENKTL.Conclusions:ENKTL is the most common type of mature T-cell and NK-cell lymphoma diagnosed at our institution.CD30 is frequently expressed in ENKTL and represents a therapeutic target;however,it may not be a prognostic marker.Yanfen Feng Huilan Rao Yiyan Lei Yuhua Huang Fang Wang Yu Zhang Shaoyan Xi Qiuliang Wu Jianyong Shao 2017Chinese Journal of Cancer2017,36,6:10

12Magnetic properties and large magnetocaloric effects of GdPd intermetallic compound显示文摘With the intention to explore excellent magnetocaloric materials, the intermetallic compound GdPd was synthesized by arc melting and heat treatment. The microstructure, magnetic and magnetocaloric properties of the intermetallic compound of GdPd were investigated by X-ray diffraction(XRD), scanning electron microscopy(SEM) and the physical property measurement system(PPMS). A large reversible magnetocaloric effect is observed in GdPd accompanied by a second order magnetic phase transition from paramagnetism to ferromagnetism at ~39 K. The paramagnetic Curie temperature(θp) and the effective magnetic moment(μ_(eff))are determined to be 34.7 K and 8.12 μ_B/Gd,respectively. The maximum entropy change(|△S_M^(Max)|) and the relative cooling power(RCP) under a field change of 5 T are estimated to be 20.14 J/(kg·K) and 433 J/kg, respectively. 17Determination of estrogens and estrogenic activities in water from three rivers in Tianjin, China显示文摘Studies on estrogenic disrupting compounds(EDCs) occurrence and identification of main responsible compounds in river water discharged into the sea are of significance.In the present research,we screened estrogenic activities of 10 river water samples from 3 main rivers discharged into Bohai Sea in Tianjin using a recombinant two-hybrid yeast assay and chemical analysis by gas chromatography-mass spectrometry.All sample extracts induced significant estrogenic activity,with 17β-estradiol equivalents(EEQ) of raw water ranging from 5.72 to 59.06 ng/L.Six most concerned EDCs in the river water samples including estrone,17β-estradiol,17α-ethinylestradiol,estriol,diethylstilbestrol and estradiol valerate were determined,with their concentrations up to 50.70,31.40,24.40,37.20,2.56,and 8.47 ng/L,respectively.Through causality analysis by comparing the EEQ values of yeast assay and chemical analysis,17α-ethinylestradiol and 17β-estradiol were identified as the main contributors to the estrogenic effects of the river samples,accounting for the whole estrogenic activities(62.99% to 185.66%),and estrogen antagonistic compounds might presented in the heavy polluted water samples.The proposed approach using both chemical analysis and bioassay could be used for identification and evaluation of the estrogenic activity of EDCs in river water.Kaifeng Rao Bingli Lei Na Li Mei Ma Zijian Wang 2013Journal of Environmental Sciences2013,25,6:6
