2KIF2C:a novel link between Wnt/β-catenin and mTORCI signaling in the pathogenesis of hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is the most common primary liver malignancy and is the fourth-leading cause of cancer-related deaths worldwide.HCC is refractory to many standard cancer treatments and the prognosis is often poor,highlighting a pressing need to identify biomarkers of aggressiveness and potential targets for future treatments.Kinesin family member 2C(KIF2C)is reported to be highly expressed in several human tumors.Nevertheless,the molecular mechanisms underlying the role of KIF2C in tumor development and progression have not been investigated.In this study,we found that KIF2C expression was significantly upregulated in HCC,and that KIF2C up-regulation was associated with a poor prognosis.Utilizing both gain and loss of function assays,we showed that KIF2C promoted HCC cell proliferation,migration,invasion,and metastasis both in vitro and in vivo.Mechanistically,we identified TBC1D7 as a binding partner of KIF2C,and this interaction disrupts the formation of the TSC complex,resulting in the enhancement of mammalian target of rapamycin complexl(mTORCI)signal transduction.Additionally,we found that KIF2C is a direct target of the Wnt/β-catenin pathway,and acts as a key factor in mediating the crosstalk between Wnt/β-catenin and mTORCI signaling.Thus,the results of our study establish a link between Wnt/β-catenin and mTORCI signaling,which highlights the potential of KIF2C as a therapeutic target for the treatment of HCC.Shi Wei Miaomiao Dai Chi Zhang Kai Teng Fengwei Wang Hongbo Li Weipeng Sun Zihao Feng Tiebang Kang Xinyuan Guan Ruihua Xu Muyan Cai Dan Xie 2021Protein & Cell2021,12,10:8

4Color spaces of safflower (Carthamus tinctorius L.) for quality assessment显示文摘Objective:In this study,safflower(Carthamus tinctorius L.)was taken as a representative example to examine the application of color characteristics to evaluate quality.Methods:A computer vision system was established for the objective and nondestructive assessment of color using image processing algorithms.Color parameters were investigated based on the RGB,L*a*b and HSV color spaces.The content of hydroxysafflor yellow A(HSYA),a major bioactive constituent of safflower,was determined by high-performance liquid chromatography.The relationship between HSYA content and color values was investigated by Pearson correlation analysis.A multiple linear regression model was established to predict the HSYA content from color values.Results:The red color and lightness of safflower were found to be significantly related to HSYA content.The prediction equation obtained by multiple regression was reliable with an R2 value of 0.805(P<.01).Conclusion:The results suggest that the computer vision technique could be used as a promising and non-destructive technology for color measurement and quality evaluation of CHM.Manfei Xu Chenzhao Du Na Zhang Xinyuan Shi Zhisheng Wu Yanjiang Qiao 2016Journal of Traditional Chinese Medical Sciences2016,3,3:5
5Magnetic properties enhancement of hot-deformed NdFeB magnets by two different methods of CeNdCu diffusion显示文摘Two different ways were used to control the distribution of cerium for the enhancement of coercivity.One was by coating CeNdCu in the prepared magnet and annealing to make CeNdCu diffuse into the grain boundary to increase the coercivity,the other was by mixing CeNdCu with initial magnetic powders and then preparing the hot-deformed magnet.The SEM-EDS result indicates that cerium diffuses more easily into the main phase by the mixing way,while cerium is mainly distributed in the grain boundary via the coating way.The heat-treatment process may be one of the dominant influencing factors for the distribution of Ce,Multi-steps heat treatment in the mixing way,consisting of hot-pressing,hot-deforming and post heat process,easily introduces Ce into the main phase Nd2 Fe14B,which forms the Ce2 Fe14B shell resulting in the decrease of HA.But the coating way can ensure uniform dispersion of Ce in the grain boundary,which leads to the high coercivity.Guibing Shi Zilong Wang Xinyuan Bai Wenlong Yan Yang Luo Dunbo Yu 2020Journal of Rare Earths2020,38,12:3
6Amphiphilic drug-drug conjugate for cancer therapy with combination of chemotherapeutic and antiangiogenesis drugs显示文摘The progression and metastasis of solid tumors strongly rely on the process of forming nascent blood vessels.However,using angiogenesis inhibitors alone does not meet the cancer treatment needs.Herein,we used the amphiphilic drug-drug conjugate(ADDC)strategy to fabricate a new drug conjugate with the combination of chemotherapeutic drug and antiangiogenesis drug together.With one-step esterification of hydrophilic floxuridine(FUDR)and hydrophobic pseudolaric acid B(PAB),the conjugate was synthesized.The amphiphilic property of FUDR-PAB conjugate induced the self-assembly to form nanoparticles in water.From further in vitro and in vivo experiments,this FUDR-PAB conjugate does not only have a high antitumor effect,but also shows efficient antianiogenesis property.These results offer a promising ADDC strategy for designing drugs with combination of chemotherapeutic drug and antiangiogenesis drug together.Mo Sun Qiuhui Qian Leilei Shi Li Xu Qunfang Liu Linzhu Zhou Xinyuan Zhu Jian-Min Yue Deyue Yan 2020Science China Chemistry2020,63,1:3
7Rapid analysis of dyed safflowers by color objectification and pattern recognition methods显示文摘Objective:Rapid discrimination of three classes of safflowers,dyed safflower,adulterated safflower,and pure safflower using computer vision and image processing algorithms.Methods:A low cost computer vision system(CVS)was designed to measure the color of safflowers in the RGB(red,green,blue),L^*a^*b^*,and HSV(hue,saturation,vale)color spaces.The color moments in these three color spaces were extracted from the acquired images as color features of safflower.In addition,five kinds of pigments that are commonly used to dye safflowers were identified by high-performance liquid chromatography as a reference.Pattern recognition methods were investigated for rapid discrimination,including an unsupervised principal component analysis(PCA)algorithm and a supervised partial least squares discriminant analysis(PLS-DA)algorithm.Results:The mean error(e)between color values measured with the colorimeter and calculated with the CVS was 2.4%,with a high correlation coefficient(r)of 0.9905.This result indicated that the established CVS was reliable for color estimation of safflowers.The PLS-DA model,which had a total accuracy of 91.89%,outperformed the PCA model in classifying pure,adulterated,and dyed safflowers.Conclusion:The color objectification is a promising tool for rapid identification of dyed and adulterated safflowers.Manfei Xu Shengyun Dai Zhisheng Wu Xinyuan Shi Yanjiang Qiao 2016Journal of Traditional Chinese Medical Sciences2016,3,4:2

9Controllable synthesis of a hollow Cr2O3 electrocatalyst for enhanced nitrogen reduction toward ammonia synthesis显示文摘As a fascinating alternative to the energy-intensive Haber-Bosch process,the electrochemically-driven N_(2) reduction reaction(NRR)utilizing the N_(2) and H_(2)O for the production of NH3 has received enormous attention.The development and preparation of promising electrocatalysts are requisite to realize an efficient N_(2) conversion for NH3 production.In this research,we propose a template-assisted strategy to construct the hollow electrocatalyst with controllable morphology.As a paradigm,the hollow Cr_(2)O_(3) nanocatalyst with a uniform size(~170 nm),small cavity and ultrathin shell(~15 nm)is successfully fabricated with this strategy.This promising hollow structure is favourable to trap N_(2) into the cavity,provides abundant active sites to accelerate the three-phase interactions,and facilitates the reactant transfer across the shell.Attributed to these synergetic effects,the designed catalyst displays an outstanding behaviour in N_(2) fixation for NH3 production in ambient condition.In the neutral electrolyte of 0.1 mol·L^(-1) Na_(2)SO_(4),an impressive electrocatalytic performance with the NH3 generation rate of 2.72μg·h^(-1)·cm^(-2) and a high FE of 5.31%is acquired respectively at-0.85 V with the hollow Cr_(2)O_(3) catalyst.Inspired by this work,it is highly expected that this approach could be applied as a universal strategy and extended to fabricating other promising electrocatalysts for realizing highly efficient nitrogen reduction reaction(NRR).Lei Shi Yu Yin Hong Wu Rajan Arjan Kalyan Hirani Xinyuan Xu Jinqiang Zhang Nasir Rafique Abdul Hannan Asif Shu Zhang Hongqi Sun 2022Chinese Journal of Chemical Engineering2022,35,1:1
10Effect of Kushen(Radix Sophorae flavescentis) extract on laryngeal neoplasm Hep2 cells显示文摘OBJECTIVE:To evaluate the effect of fermented extract of Kushen(Radix Sophorae Flavescentis) or non-fermented ESF on laryngeal neoplasms Hep2 cells.METHODS:Use 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay to explore the effect of fermented ESF and non-fermented ESF on Hep2 cells,and detect the mRNA and protein expression level of Bcl-2,Bax and Caspase-3 with reverse transcription polymerase chain reaction(RT-PCR) andWestern blot.RESULTS:Both fermented ESF and non-fermented ESF could inhibit laryngeal neoplasm's Hep2 cells,but and the cells did not response to the dilution 1:320 of fermented ESF,nor to the 1:1280 dilution of non-fermented ESF.As time progressed,the dilution 1:80 of fermented ESF and 1:320 dilution of non-fermented ESF could significantly reduce Bcl-2 mRNA and protein expression and down-regulate Caspase-3 mRNA and protein expression.Bax mRNA and protein were not expressed in Hep2 cells.CONCLUSIONS:Both fermented ESF and non-fermented ESF could inhibit the proliferation of Hep2 cells,and the effect of non-fermented ESF was significantly better than that of the fermented.Yingzi Wang Chunchao Han Xinsheng Fang Xinyuan Shi Ailing Feng Kuibang He Shenghai Zhang Xiuyu Sun 2013Journal of Traditional Chinese Medicine2013,33,2:1

13A multiscale study of the penetrationenhancing mechanism of menthol显示文摘Objective:Transdermal drug delivery systems represent a critical focus in the pharmaceutics field;however,their use is limited by the fact that many drugs usually pass through the skin with low permeability.Menthol is a common penetration enhancer because of its high penetration-enhancing efficiency and safety.Our research aimed to reveal the penetrationenhancing mechanisms of menthol via a multiscale study.Methods:First,the interaction of menthol with the stratum corneum was studied using vertical Franz diffusion cells obtained from the abdominal skin of rats as a model.Then,the skin samples were observed via transmission electron microscopy.Finally,the interaction of different concentrations of menthol with a mixed lipid model of the stratum corneum was investigated via molecular dynamics simulation using the GROMOS 54A7 force field on a microcosmic level.Results:At concentrations of 3.5%or lower,menthol changed the original structure of the stratum corneum to varying degrees,which increased its fluidity and facilitated the permeation and storage of menthol.Menthol increased the fluidity of the stratum corneum mainly via two mechanisms.First,menthol had strong hydrogen-bonding capability,and it could compete for the lipidelipid hydrogen bonding sites,thereby weakening the stability of the hydrogenbonding network connecting the skin lipids.In addition,menthol had strong affinity for cholesterol,probably due to their similar molecular structures,suggesting that the incorporation of menthol would increase the fluidity of the lipid membrane similarly to cholesterol.Conclusion:The penetration-enhancing mechanism of menthol was explained using in vitro and molecular dynamics simulation methods.These findings may advance the basic research of transdermal drug delivery systems and facilitate the discoveries of novel penetration enhancers.Liping Chen Lina Ma Shufang Yang Xiaowen Wu Xingxing Dai Shifeng Wang Xinyuan Shi 2019Journal of Traditional Chinese Medical Sciences2019,6,4:1

15Endogenous nucleotide as drug carrier:base-paired guanosine-5’-monophosphate:pemetrexed vesicles with enhanced anticancer capability显示文摘Endogenous substance such as nucleotide as a drug carrier has been proposed as a novel drug delivery system.The nucleotide guanosine-5’-monophosphate(GMP) is used to transport an anticancer drug pemetrexed disodium heptahydrate(PMX) via specific base pairing.The endogenous nature of GMP helps to avoid biocompatibility issues that are generally accompanied with nanocarriers including cytotoxicity,immunogenicity and blood compatibility.Furthermore,the low-molecular weight of the GMP nucleotide carrier significantly boosts the drug loading capacity compared to traditional liposomes and high-molecular weight carriers.Hydrogen-bonding interaction between the carrier and drug realizes the controlled release of loaded drug,and also facilitates large scale manufacture since no additional chemical synthesis is required.More importantly,in vivo experiments reveal that the base-paired GMP:PMX nanovesicles improve the target specificity and pharmacokinetic properties of PMX,and exhibit remarkably enhanced anticancer abilities compared to standalone PMX without any carriers.We envision that this strategy could be extended to other endogenous substances and drugs bearing functional groups capable of specific interaction,and promote the construction of drug delivery systems with inherent biocompatibility,enhanced drug delivery efficacy,and a simplified preparation method.Lei Xu Chunyang Yu Dali Wang Ji Pang Leilei Shi Yue Su Lidong Gong Deyue Yan Xinyuan Zhu 2020Science China Chemistry2020,63,2:0
16Study on Hydraulic Transmission of Offshore Low-speed Wind Energy Generator显示文摘For offshore hydraulic drive wind turbines,the problems of unsatisfactory speed control and low efficiency at low wind speeds are targeted.A low-speed high-torque radial piston pump is designed to replace the traditional fixed pump with a particular focus on its low-speed performance.The pump is characterized by small internal leakage at low wind speeds and high volumetric efficiency,which is beneficial to improve the power generation efficiency of the system.A new linear control method based on the PID algorithm and feedforward compensation was proposed to obtain the constant speed output control of variable motor at low wind speed.With the model for wind turbine and fixed pump-variable motor main drive system,the system was simulated and experimentally proved to verify the feasibility and anti-interference performance of the system control method at low wind speeds.A promising outcome was obtained on the response characteristics of system power and efficiency at low wind speeds.This can be a powerful technical support for the normal ustility of hydraulic drive wind turbines.SHI Qingyi DENG Xiaogang XIAO Xinyuan HU Yushuang WANG Rui 2021International Journal of Plant Engineering and Management2021,26,1:0

18Estimation of multivariate detection limits of four quality parameters in licorice using MEMS–NIR spectrometry coupled with two sampling accessories显示文摘In this work,multivariate detection limits(MDL)estimator was obtained based on the microelectro-mechanical systems–near infrared(MEMS–NIR)technology coupled with two sampling accessories to assess the detection capability of four quality parameters(glycyrrhizic acid,liquiritin,liquiritigenin and isoliquiritin)in licorice from di®erent geographical regions.112 licorice samples were divided into two parts(calibration set and prediction set)using Kennard–Stone(KS)method.Four quality parameters were measured using high-performance liquid chromatography(HPLC)method according to Chinese pharmacopoeia and previous studies.The MEMS–NIR spectra were acquired from¯ber optic probe(FOP)and integrating sphere,then the partial least squares(PLS)model was obtained using the optimum processing method.Chemometrics indicators have been utilized to assess the PLS model performance.Model assessment using chemometrics indicators is based on relative mean prediction error of all concentration levels,which indicated relatively low sensitivity for low-content analytes(below 1000 parts per million(ppm)).Therefore,MDL estimator was introduced with alpha error and beta error based on good prediction characteristic of low concentration levels.The result suggested that MEMS–NIR technology coupled with fiber optic probe(FOP)and integrating sphere was able to detect minor analytes.The result further demonstrated that integrating sphere mode(i.e.,MDL0:05;0:05,0.22%)was more robust than FOP mode(i.e.,MDL0:05;0:05,0.48%).In conclusion,this research proposed that MDL method was helpful to determine the detection capabilities of low-content analytes using MEMS–NIR technology and successful to compare two sampling accessories.Zhisheng Wu Xinyuan Shi Na Zhao Yanling Pei Manfei Xu Luwei Zhou Yang Li Yanjiang Qiao 2015Journal of Innovative Optical Health Sciences2015,8,5:0
19NIR assignment of isopsoralen by 2D-COS technology and model application in Yunkang Oral Liquid显示文摘Near infrared(NIR)assignment of Isopsoralen was performed using deuterated chloroform solvent and two-dimensional correlation spectroscopy(2D-COS)technology.Yunkang Oral Liquid was applied to study Isopsoralen,the characteristic bands by spectral assignment as well as the bands by interval partial least squares(iPLS)and synergy interval partial least squares(siPLS)were used to establish partial least squares(PLS)model.The coefficient of determination in calibration(R^(2)_(cal))were 0.9987,0.9970 and 0.9982.The coefficient of determination in cross validation(R^(2)_(val)) T were 0.9985,0.9921 and 0.9982.The coe±cient of determination in prediction(R^(2)_(pre)) T were 0.9987,0.9955 and 0.9988.The root mean square error of calibration(RMSEC)were 0.27,0.40 and 0.31 ppm.The root mean square error of cross validation(RMSECV)were 0.30,0.67 and 0.32 ppm.The root mean square error of prediction(RMSEP)were 0.23,0.43 and 0.22 ppm.The residual predictive deviation(RPD)were 31.00,16.58 and 32.41.It turned out that the characteristic bands by spectral assignment had the same results with the chemometrics methods in PLS model.It provided guidance for NIR spectral assignment of chemical compositions in Chinese Materia Medica(CMM).Yanling Pei Zhisheng Wu Xinyuan Shi Xiaoning Pan Yanfang Peng Yanjiang Qiao 2015Journal of Innovative Optical Health Sciences2015,8,6:0
20Rapid analysis of spatial distribution of PVPP and hardness of Yinhuang dispersible tablets by NIR-CI显示文摘The present study aimed at investigating the relationship between tablet hardness and homogeneity of different Yinhuang dispersible tablets by near-infrared chemical imaging(NIR-CI)technology.The regularity of best hardness was founded between tablet hardness and the spatial distribution uniformity of Yinhuang dispersible tablets.The ingredients homogeneity of Yinhuang dispersible tablets could be spatially determined using basic analysis of correlation between analysis(BACRA)method and binary image.Then different hardnesses of Yinhuang dispersible tablets were measured.Finally,the regularity between tablet hardness and the spatial distribution uniformity of Yinhuang dispersible tablets was illuminated by quantifying the agglomerate of polyvinyl poly pyrrolidone(PVPP).The result demonstrated that the distribution of PVPP was unstable when the hardness was too large or too small,while the agglomerate of PVPP was smaller and more stable when the best tablet hardness was 75 N.This paper provided a novel methodology for selecting the best hardness in the tabletting process of Chinese Medicine Tablet.Yifei Wang Zhisheng Wu Zhaoyi Wang Manfei Xu Xinyuan Shi Yanjiang Qiao 2016Journal of Innovative Optical Health Sciences2016,9,2:0
