| 1 | miR-124 suppresses multiple steps of breast cancer metastasis by targeting a cohort of pro-metastatic genes in vitro显示文摘Metastasis is a multistep process involving modification of morphology to suit migration, reduction of tumor cell adhesion to the extracellular matrix, increase of cell mobility, tumor cell resistance to anoikis, and other steps. MicroRNAs are well-suited to regulate tumor metastasis due to their capacity to repress numerous target genes in a coordinated manner, thereby enabling their intervention at multiple steps of the invasion-metastasis cascade. In this study, we identified a microRNA exemplifying these attributes, miR-124, whose expression was reduced in aggressive MDA-MB-231 and SK-3rd breast cancer cells. Down-regulation of miR-124 expression in highly aggressive breast cancer cells contributed in part to DNA hypermethylation around the promoters of the three genes encoding miR-124. Ectopic expression of miR-124 in MDA-MB-231 cells suppressed metastasis-related traits including formation of spindle-like morphology, migratory capacity, adhesion to fibronectin, and anoikis. | 4 | The study of selective primary culture and determination of a breast cancer cell line in vitro显示文摘Objective The successful establishment of a tumor cell bank is based on the premise that the target cells can be cultured by a legitimate approach.In this experiment,we used primary culture to select and detect breast cancer cells in vitro,which can provide experimental ideas and methods for the establishment of a living tumor tissue cell bank.Methods Fifty-two specimens were collected over a two-year period from people with breast cancer who needed surgical treatment in our hospital.Cells were isolated and used to establish successful cell culture.Cell activity and cell purity were measured before liquid nitrogen cryopreservation.Results(1)At the initial culture stage,cells grew with adherence.Cell multiplication could be seen after the cell medium was exchanged three times.Cell viability was above 86%,while the viability of the target cells was above 75%,as detected by hematoxylin and eosin(HE)staining.(2)The number of breast cancer cells decreased,while the number of fibroblasts increased after five rounds of passage.(3)The success rate was 73.08%,which did not include polluted cells and those that were not successfully cryopreserved.Conclusion(1)breast cancer cells could be selected from primary culture in vitro through an appropriate method.(2)Exchange of the cell medium and further cell passage improved cell multiplication.(3)The experimental results could be monitored using trypan blue and HE staining.(4)The success of breast cancer cell culture in vitro could be used as a reference for other cell culture,so as to establish a tumor tissue cell bank. | Meng Ren Huixia Xu Xiangji Lu Bingping Wang Rina Su Hao Zhang Song Jiang Fengying Gao Yanwei Gao | 2020 | Oncology and Translational Medicine2020,6,2: | 1 |

| 6 | Comprehensive and deep profiling of the plasma proteome with protein corona on zeolite NaY显示文摘Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically,zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins,minimizing the“masking”effect caused by high-abundance proteins.The relative abundance of middleand low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 highabundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approximately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins identified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states.In summary,this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications. | Congcong Ma Yanwei Li Jie Li Lei Song Liangyu Chen Na Zhao Xueping Li Ning Chen Lixia Long Jin Zhao Xin Hou Li Ren Xubo Yuan | 2023 | Journal of Pharmaceutical Analysis2023,13,5: | 0 |
| 7 | Role of BraRGL1 in regulation of Brassica rapa bolting and flowering显示文摘Gibberellin(GA)plays a major role in controlling Brassica rapa stalk development.As an essential negative regulator of GA signal transduction,DELLA proteins may exert significant effects on stalk development.However,the regulatory mechanisms underlying this regulation remain unclear.In this study,we report highly efficient and inheritable mutagenesis using the CRISPR/Cas9 gene editing system in BraPDS(phytoene desaturase)and BraRGL1(key DELLA protein)genes.We observed a loss-of-function mutation in BraRGL1 due to two amino acids in GRAS domain.The flower bud differentiation and bolting time of BraRGL1 mutants were significantly advanced.The expression of GA-regulatory protein(BraGASA6),flowering related genes(BraSOC1,BraLFY),expansion protein(BraEXPA11)and xyloglucan endotransferase(BraXTH3)genes was also significantly upregulated in these mutants.BraRGL1-overexpressing plants displayed the contrasting phenotypes.BraRGL1 mutants were more sensitive to GA signaling.BraRGL1 interacted with BraSOC1,and the interaction intensity decreased after GA3 treatment.In addition,BraRGL1 inhibited the transcription-activation ability of BraSOC1 for BraXTH3 and BraLFY genes,but the presence of GA3 enhanced the activation ability of BraSOC1,suggesting that the BraRGL1-BraSOC1 module regulates bolting and flowering of B.rapa through GA signal transduction.Thus,we hypothesized that BraRGL1 is degraded,and BraSOC1 is released in the presence of GA3,which promotes the expression of BraXTH3 and BraLFY,thereby inducing stalk development in B.rapa.Further,the BraRGL1-M mutant promoted the flower bud differentiation without affecting the stalk quality.Thus,BraRGL1 can serve as a valuable target for the molecular breeding of early maturing varieties. | Yudan Wang Shiwei Song Yanwei Hao Changming Chen Xi Ou Bin He Jiewen Zhang Zhehao Jiang Chengming Li Shuaiwei Zhang Wei Su Riyuan Chen | 2023 | Horticulture Research2023,10,8: | 0 |

| 11 | Effect of ketamine on aquaporin-4 expression and neuronal apoptosis in brain tissues following brain injury in rats显示文摘BACKGROUND: Aquaporin-4 (AQP-4) is closely related to the formation of brain edema. Neuronal apoptosis plays an important part in the conversion of swelled neuron following traumatic brain injury. At present, the studies on the protective effect of ketamine on brain have involved in its effect on aquaporin-4 expression and neuronal apoptosis in the brain tissues following brain injury in rats. OBJECTIVE: To observe the effect of ketamine on AQP-4 expression and neuronal apoptosis in the brain tissue following rat brain injury, and analyze the time-dependence of ketamine in the treatment of brain injury. DESIGN: Randomized grouping design, controlled animal trial.SETTING: Department of Anesthesiology, the Medical School Hospital of Qingdao University.MATERIALS: Totally 150 rats of clean grade, aged 3 months, were involved and randomized into control group and ketamine-treated group, with 75 rats in each. Each group was divided into 5 subgroups separately at 6,12, 24, 48 and 72 hours after injury, with 15 rats at each time point. Main instruments and reagents: homemade beat machine, ketamine hydrochloride (Hengrui Pharmaceutical Factory, Jiangsu), rabbit anti-rat AQP-4 polyclonal antibody, SABC immunohistochemical reagent kit and TUNEL reagent kit (Boster Co.,Ltd., Wuhan). METHODS: This trial was carried out in the Institute of Cerebrovascular Disease, Medical College of Qingdao University during March 2005 to February 2006. A weight-dropping rat model of brain injury was created with Feeney method. The rats in the ketamine-treated group were intraperitoneally administered with 50 g/L ketamine (120 mg/kg) one hour after injury, but ketamine was replaced by normal saline in the control group. In each subgroup, the water content of cerebral hemisphere was measured in 5 rats chosen randomly. The left 10 rats in each subgroup were transcardiacally perfused with ketamine, then the brain tissue was made into paraffin sections and stained by haematoxylin and eosin. Neuronal morphology was observed. AQP-4 expression and neuronal apoptosis were measured with immunohistochemical method and TUNEL method respectively. MAIN OUTCOME MEASURES: Water content in brain tissue, neuronal morphology, the number of AQP-4 positive neurons and TUNEL positive neurons in rats of two groups at each time point after injury.RESULTS: Totally 150 rats entered the stage of result analysis. ① Water content of brain tissue: The water content of brain tissue at each time point after injury in the ketamine-treated group was lower than that in the control group. There were very significant differences in water content at 12 and 24 hours after injury respectively between ketamine-treated group and control group [(77.34±2.35)% vs.(82.31±1.48)%;(78.01±2.21)%vs.(83.86±2.37)%,t=4.001 6,4.036 7,both P < 0.01]. ② Neuronal morphology: Pathological changes in traumatic region and peripheral region of injury in the ketamine-treated group were significantly lessened, and necrotic and apoptotic cells in the ketamine-treated group were also significantly reduced as compared with control group. ③ AQP-4 expression: AQP-4 positive neurons at each time point in the ketamine-treated group were significantly less than those in the control group. There were very significant differences in AQP-4 expression at 12 and 24 hours after injury between ketamine-treated group and control group [(34.17±4.74) /visual field vs. (43.42±5.65) /visual field;(40.83±3.17) /visual field vs. (58.88±6.23) /visual field,t =3.966 3,8.165 7, both P < 0.01]. ④ Neuronal apoptosis: TUNEL positive neurons at each time point in the ketamine-treated group were less than those in the control group. | 13 | The SlTPL3–SlWUS module regulates multi-locule formation in tomato by modulating auxin and gibberellin levels in the shoot apical meristem显示文摘Malformed fruits depreciate a plant’s market value.In tomato(Solanum lycopersicum),fruit malformation is associated with the multi-locule trait,which involves genes regulating shoot apical meristem(SAM)development.The expression pattern of TOPLESS3(SITPL3)throughout SAM development prompted us to investigate its functional significance via RNA interference(RNAi)and clustered regularly interspaced short palindromic repeats/CRISPR-associated nuclease 9(Cas9)-mediated gene editing.Lower SITPL3 transcript levels resulted in larger fruits with more locules and larger SAMs at the 5 d after germination(DAG5)stage.Differentially expressed genes in the SAM of wild-type(WT)and SITPL3-RNAi plants,identified by transcriptome deep sequencing(RNA-seq),were enriched in the gibberellin(GA)biosynthesis and plant hormone signaling pathways.Moreover,exogenous auxin and paclobutrazol treatments rescued the multi-locule phenotype,indicating that SITPL3 affects SAM size by mediating auxin and GA levels in the SAM.Furthermore,SITPL3 interacted with WUSCHEL(SIWUS),which plays an important role in SAM size maintenance.We conducted RNA-seq and DNA affinity purification followed by sequencing(DAP-seq)analyses to identify the genes regulated by SITPL3 and SIWUS in the SAM and to determine how they regulate SAM size.We detected24 overlapping genes regulated by SITPL3 and SIWUS and harboring an SIWUS-binding motif in their promoters.Furthermore,functional annotation revealed a notable enrichment for functions in auxin transport,auxin signal transduction,and GA biosynthesis.Dual-luciferase assays also revealed that SITPL3 enhances SIWUS-mediated regulation(repression and activation)of SIPIN3 and SIGA2 ox4 transcription,indicating that the SITPL3-SIWUS module regulates SAM size by mediating auxin distribution and GA levels,and perturbations of this module result in enlarged SAM.These results provide novel insights into the molecular mechanism of SAM maintenance and locule formation in tomato and highlight the SITPL3-SIWUS module as a key regulator. | Shiwei Song Binbin Huang Zanlin Pan Qiuxiang Zhong Yinghua Yang Da Chen Lisha Zhu Guojian Hu Mi He Caiyu Wu Mohammed Zouine Riyuan Chen Mondher Bouzayen Yanwei Hao | 2022 | Journal of Integrative Plant Biology2022,64,11: | 0 |
