1Bph30confers resistance to brown planthopperby fortifying sclerenchyma in rice leaf sheaths显示文摘Phloem-feeding insects cause massive losses in agriculture and horticulture.Host plant resistance to phloem-feeding insects is often mediated by changes in phloem composition,which deter insect settling and feeding and decrease viability.Here,we report that rice plant resistance to the phloem-feeding brown planthopper(BPH)is associated with fortification of the sclerenchyma tissue,which is located just beneath the epidermis and a cell layer or two away from the vascular bundle in the rice leaf sheath.We found that BPHs prefer to feed on the smooth and soft region on the surface of rice leaf sheaths called the long-cell block.We identified Bph30 as a rice BPH resistance gene that prevents BPH stylets from reaching the phloem due to the fortified sclerenchyma.Bph30 is strongly expressed in sclerenchyma cells and enhances cellulose and hemicellulose synthesis,making the cell walls stiffer and sclerenchyma thicker.The structurally fortified sclerenchyma is a formidable barrier preventing BPH stylets from penetrating the leaf sheath tissues and arriving at the phloem to feed.Bph30 belongs to a novel gene family,encoding a protein with two leucine-rich domains.Another member of the family,Bph40,also conferred resistance to BPH.Collectively,the fortified sclerenchyma-mediated resistance mechanism revealed in this study expands our understanding of plant-insect interactions and opens a new path for controlling planthoppers in rice.Shaojie Shi Huiying Wang Lingyun Nie Di Tan Cong Zhou Qian Zhang YiLi Bo Du Jianping Guo Jin Huang DiWu Xiaohong Zheng Wei Guan Junhan Shan Lili Zhu Rongzhi Chen Longjian Xue Linda L.Walling Guangcun He 2021Molecular Plant2021,14,10:7
2Investigating different bio-responses of the upper ocean to Typhoon Haitang using Argo and satellite data显示文摘The responses of the upper ocean to Typhoon Haitang in July 2005 are investigated using Argo float and multiplatform satellite data.The results show decreasing sea surface temperature(SST),a deepening of the mixed layer depth(MLD),and enhanced Chlorophyll-a(Chl-a)concentration.Two extreme cool regions are identified.While the magnitude of SST cooling in the two regions is similar,the biological response(Chla enhancement)differs.To facilitate comparisons,the region to the northeast of Taiwan is defined as region A and the region east of Taiwan as region B.Ekman pumping and the intrusion of the Kuroshio play an important role in the enhancement of Chl-a in region A.Cold eddies provide the material source for the formation of the cold center in region B,where mixing is dominant.Because of the relatively high translation speed(5 m/s)in region B,Ekman pumping has little influence on the cooling and Chl-a enhancement processes.Moreover,the MLD is shallower than the nutricline,which means that mixing does not result in a marked increase in nutrients in the euphotic layer(where the nutrient concentration is uniformly depleted).Sea temperatures,in contrast,gradually decrease with depth below the bottom of the mixed layer.In contrast to region A,region B showed no significant enhancement of Chl-a but strong SST cooling.Haixia Shan Yuping Guan Jianping Huang 2014Chinese Science Bulletin2014,59,8:6
3Deep origin of Cenozoic volcanoes in Northeast China revealed by 3-D electrical structure显示文摘The three-dimensional(3-D)electrical structure of the upper-mantle was used to examine the deep origins of and relationship among the Cenozoic volcanoes located in Northeast China(NEC).High-quality,long-period magnetotelluric(LMT)full-impedance tensor data were collected in NEC and subjected to 3-D Gauss-Newton inversion in order to construct a resistivity model.The resulting model reveals the presence of multiple localized low-resistivity anomalies(LRAs)within the high resistivity lithosphere beneath NEC.These LRAs partially coincide with Cenozoic volcanoes on the surface.Three LRAs that form a larger,annular LRA were observed in the deep upper mantle beneath the Songliao Basin,whereas vein-like LRAs were found in the asthenosphere that connect the lithosphere and deep upper mantle.Petrophysical analyses suggest that the LRAs may have been caused by fluid-induced melting.Based on our electrical model,we propose that,following dehydration of the subducted Western Pacific slab into the mantle transition zone(MTZ)beneath NEC,the released water migrated upward and caused partial melting at the top of the MTZ beneath the Songliao Basin.Under the effect of buoyancy,the melted mantle formed a thermal upwelling that caused melting of asthenosphere before diapiring at the base of the dry lithosphere.The magma then penetrated structural boundaries(such as thinner,weaker,or activated suture zones)and finally reached the Earth's surface.This melting and upwelling of hot mantle materials may have resulted in large-scale volcanism in the region throughout the Cenozoic,including the eruption of Changbai Mountain and Halaha Volcanoes.Our results suggest that the Cenozoic NEC volcanoes may all share a similar mode of genesis,and probably originated from the annular LRA in the deep upper mantle.Shiwen LI Aihua WENG Jianping LI Xuanlong SHAN Jiangtao HAN Yu TANG Yanhui ZHANG Xueqiu WANG 2020Science China Earth Sciences2020,63,4:4

5Origin and circulation of saline springs in the Kuqa Basin of the Tarim Basin,Northwest China显示文摘It is widely accepted that hydrogeochemistry of saline springs is extremely important to understand the water circulation and evolution of saline basins and to evaluate the potential of potassium-rich evaporites.The Kuqa Basin,located in the northern part of the Tarim Basin in Northwest China,is a saline basin regarded as the most potential potash-seeking area.However,the origin and water circulation processes of saline springs have yet to be fully characterized in this saline basin.In this study,a total of 30 saline spring samples and 11 river water samples were collected from the Qiulitage Structural Belt(QSB)of the Kuqa Basin.They were analyzed for major(K^+,Ca2^+,Na^+,Mg2^+,SO42-,Cl-and HCO3-)and trace(Sr2^+and Br-)ion concentrations,stable H-O-Sr isotopes and tritium concentrations in combination with previously published hydrogeochemical and isotopic(H-O)data in the same area.It is found that the water chemical type of saline springs in the study area belonged to the Na-Cl type,and that of river water belonged to the Ca-Mg-HCO3-SO4 type.The total dissolved solid(TDS)of saline springs in the QSB ranged from 117.77 to 314.92 g/L,reaching the brine level.On the basis of the general chemical compositions and the characteristics of the stable H-O-Sr isotopes of saline springs,we infer that those saline springs mainly originated from precipitation following river water recharging.In addition,we found that saline springs were not formed by evapo-concentration because it is unlikely that the high chloride concentration of saline springs resulted in evapo-concentration and high salinity.Therefore,we conclude that saline spring water may have experienced intense evapo-concentration before dissolving the salty minerals or after returning to the surface.The results show that the origin of salinity was mainly dominated by dissolving salty minerals due to the river water and/or precipitation that passed through the halite-rich stratum.Moreover,there are two possible origins of saline springs in the QSB:one is the infiltration of the meteoric water(river water),which then circulates deep into the earth,wherein it dissolves salty minerals,travels along the fault and returns to the surface;another is the mixture of formation water,or the mixture of seawater or marine evaporate sources and its subsequent discharge to the surface under fault conditions.Our findings provide new insight into the possible saltwater circulation and evolution of saline basins in the Tarim Basin.SHAN Junjie WANG Jianping SHAN Fashou TENG Xueming FAN Qishun LIQingkuan QIN Zhanjie ZHANG Xiangru 2020Journal of Arid Land2020,12,2:2

13Lipidomic analyses reveal enhanced lipolysis in planthoppers feeding on resistant host plants显示文摘The brown planthopper(BPH)(Nilaparvata lugens St?l)is a highly destructive pest that seriously damages rice(Oryza sativa L.)and causes severe yield losses.To better understand the physiological and metabolic mechanisms through which BPHs respond to resistant rice,we combined mass-spectrometry-based lipidomics with transcriptomic analysis and gene knockdown techniques to compare the lipidomes of BPHs feeding on either of the two resistant(NIL-Bph6 and NIL-Bph9)plants or a wild-type,BPH susceptible(9311)plant.Insects that were fed on resistant rice transformed triglyceride(TG)to phosphatidylcholine(PC)and digalactosyldiacylglycerol(DGDG),with these lipid classes showing significant alterations in fatty acid composition.Moreover,the insects that were fed on resistant rice were characterized by prominent expression changes in genes involved in lipid metabolism processes.Knockdown of the NlBmm gene,which encodes a lipase that regulates the mobilization of lipid reserves,significantly increased TG content and feeding performance of BPHs on resistant plants relative to dsGFP-injected BPHs.Our study provides the first detailed description of lipid changes in BPHs fed on resistant and susceptible rice genotypes.Results from BPHs fed on resistant rice plants reveal that these insects can accelerate TG mobilization to provide energy for cell proliferation,body maintenance,growth and oviposition.Xiaohong Zheng Yeyun Xin Yaxin Peng Junhan Shan Ning Zhang Di Wu Jianping Guo Jin Huang Wei Guan Shaojie Shi Cong Zhou Rongzhi Chen Bo Du Lili Zhu Fang Yang Xiqin Fu Longping Yuan Guangcun He 2021Science China(Life Sciences)2021,64,9:1

15A Rapid, Non-invasive Method for Anatomical Observations of Tadpole Vertebrae in Vivo显示文摘The tadpole is a critical stage in the amphibian life cycle and plays an important role during the transition from the aquatic to the terrestrial stage.However,there is a large gap in tadpole research,which represents a vital component of our understanding of the diversity and complexity of the life history traits of amphibians,especially their developmental biology.Some aspects of this gap are due to limited research approaches.To date,X-ray microcomputed tomography(micro-CT)has been widely used to conduct osteology research in adult amphibians and reptiles,but little is known about whether this tool can be applied in tadpole studies.Thus,we compared the results of two methods(the bone-cartilage double-staining technique and micro-CT)to study vertebrae in tadpole specimens.The results revealed no significant difference between the two methods in determining the number of vertebrae,and micro-CT represents a rapid,non-invasive,reliable method of studying tadpole vertebrae.When scanning tadpoles,voltage is the most critical of the scanning parameters(voltage,current and scan time),and moderate scanning parameters are recommended.In addition,micro-CT performed better using specimens stored in 70% ethanol than those preserved in 10% formalin.Finally,we suggest that micro-CT should be more widely applied in herpetological research to increase specimen utilization.Guocheng SHU Shan XIONG Wenyan ZHANG Jianping JIANG Cheng LI Feng XIE 2018Asian Herpetological Research2018,9,2:0
16Effects of Low Temperature Stress on the Morphology and hsp70 and hsp90 Gene Expression of Phascolosoma esculenta显示文摘Phascolosoma esculenta is an intertidal organism that has recently attracted attention because of its ability to survive at relatively low temperatures.However,the gene regulation in P.esculenta in relation to its response to low temperatures is unclear.To explore the low temperature adaptability of P.esculenta,this study analyzed the changes in the morphology and hsp70 and hsp90 gene expression of P.esculenta exposed to a low temperature gradient.At 5℃,P.esculenta stretched and softened,and some individuals moved apart from the group.Histological analysis revealed cuticle breaches,myofiber scattering,disruption of the body wall,and epithelial layer dispersion and muscle fiber rupturing in the nephridium.Furthermore,the mRNA expression levels of hsp70 and hsp90 increased under acute low temperature stress,suggesting that these genes function in low temperature tolerance.Overall,low temperature stress causes morphological changes and histological damage in P.esculenta,and hsp70 and hsp90 potentially function in the low temperature adaptability of P.esculenta.Our results provide new insights into the adaptive strategies of P.esculenta under low temperature environments.SHEN Weiliang LIU Cheng NI Jie GAO Xinming NI Junjie WANG Jianping JIN Shan HOU Congcong WU Xiongfei ZHU Junquan 2021Journal of Ocean University of China2021,20,1:0
17The synthesis of fluorescent nanoclusters based on the etching reaction显示文摘Thiolate-protected atomically precise nanoclusters(NCs)demonstrate a series of unique luminescent characteristics attributed to their various peculiar electronic structures.Therefore,fluorescent NCs present extraordinary practical values in biosensing and bioimaging research fields.Nevertheless,restricted by the types of fluorescent NCs,there are great difficulties in promoting the development of NCs in fluorescent research areas.As a result,it is of significant necessity for researchers to develop new synthetic pathways to produce high-quality fluorescent NCs.According to the analysis about the structural characteristics of fluorescent NCs,some general features like longer motif and higher ligand-to-metal ratio can be found,consistent to some presented regularities in etching reaction.Consequently,in this work,we used Au_(25)(MHA)_(18)(MHA=6-mercaptohexanoic acid)as a model nanocluster and utilized the etching reaction to systematically explore etching products and their corresponding luminescent properties.Moreover,we also identified three main reaction processes in the entire etching reaction process,which can generate new metal nanocluster species with various fluorescent properties.Hence,the etching reaction will provide a good platform to produce new luminescent metal NC species.Hongbin Lin Yitao Cao Qiaofeng Yao Tiankai Chen Huiting Shan Jianping Xie 2024Nano Research2024,17,5:0
18A secreted salivary effector from Riptortus pedestris impairs soybean defense through modulating phytohormone signaling pathways显示文摘Riptortus pedestris(Fabricius),one of the major piercing-sucking insects in soybeans,causes delayed plant senescence and abnormal pods,known as staygreen syndrome.Recent research has shown that direct feeding of this insect is the major cause of soybean staygreen syndrome.However,it remains unclear whether R.pedestris salivary proteins play vital roles in insect infestation.Here,we found that 4 secretory salivary proteins can induce cell death in Nicotiana benthamiana by transient heterologous expression.The cell death induced by Rp2155 relies on the nucleotide-binding leucine-rich repeat helper,HSP90.Tissue-specificity assays indicated that Rp2155 is specifically expressed in the salivary gland of R.pedestris and is significantly induced during insect feeding.The expression of salicylic acid(SA)-,jasmonic acid(JA)-related genes was increased in soybean when fed by Rp2155-silenced R.pedestris.More importantly,soybean staygreen symptoms caused by R.pedestris were significantly alleviated when Rp2155 was silenced.Together,these results suggest that the salivary effector Rp2155 is involved in promoting insect infestation by suppressing the JA and SA pathways,and it can be considered as a potential RNA interference target for insect control.Yue Huang Biao Hu Zhongyan Wei Shiqi Shan Chunyun Guo Hehong Zhang Yanjun Li Jianping Chen Xue Kang Haijian Huang Zongtao Sun 2023Insect Science2023,30,6:0
19Expression Characteristics and Putative Functions of KIF3A/KIF3B During Spermiogenesis of Phascolosoma esculenta显示文摘The microtubule(MT)-associated proteins KIF3A and KIF3B are ubiquitously expressed in a wide range of taxa.This study investigated the functions of these proteins in spermiogenesis,which involves various MT-dependent processes,in Phasco-losoma esculenta.We cloned the complete cDNA of Pe-KIF3A/3B.Structural predictions showed that Pe-KIF3A/3B are composed of a highly conserved motor domain,a coiled-coil domain,and a tail domain.Real-time quantitative PCR showed that Pe-kif3a/3b are expressed in all tissues evaluated,with the highest levels in sperm masses.Fluorescence in situ hybridization and immunofluo-rescence were employed to analyze the dynamic expression patterns of KIF3A/3B during spermiogenesis.Pe-KIF3A/3B consistently co-localized with MTs at all stages of spermiogenesis,indicating their potential functions in cargo trafficking.Pe-KIF3A/3B co-localized with mitochondria,suggesting that they may mediate mitochondrial movement.In late-stage spermatids and mature sperm,co-localization was detected in the midpiece,suggesting that Pe-KIF3A/3B could facilitate midpiece formation.The co-localization of Pe-KIF3A and Pe-KIF3B at all stages of spermiogenesis suggests their function as the heterodimeric KIF3AB.Basing on the ob-served temporal and spatial expression patterns of Pe-KIF3A/3B,MTs,and mitochondria in our study,we suggest that heterodimer KIF3AB has a potential role in acrosome biogenesis,sperm head remodeling,enflagellation,and mitochondrial migration during spermiogenesis in P.esculenta.In addition,our study on the morphological characteristics of spermatogenic cells provided fundamental data on the reproductive biology of P.esculenta.DU Chen MU Danli GAO Xinming LUO Shengyu WANG Jianping JIN Shan ZHU Junquan 2022Journal of Ocean University of China2022,21,4:0
