Passive seismic experiment and ScS wave splitting in the southwestern subbasin of South China Sea显示文摘The seismic experiment of 3D array of OBS in the southwestern sub-basin of the South China Sea(SCS) is briefly introduced in this paper. The data analysis of broadband OBS shows that totally 93 earthquakes with magnitude of Ms 6.0-6.9 and 10 earthquakes with magnitude above Ms 7.0 were recorded in high quality during this experiment, especially the catastrophic earthquake Ms 9.0 occurred in the east sea area of Japan on March 11,2011.The anisotropy parameters inversion of ScS wave of four events above Ms 7.0 indicates that the fast direction of shear wave is N58°E parallel to the ceased spreading ridge of the southwestern sub-basin of SCS(the slow direction is S35°E perpendicular to the spreading ridge),which means the spreading ridge is under compressing stress at present and the cessation of seafloor spreading is related to such stress field as well. | RUAN AiGuo LI JiaBiao LEE ChaoShing QIU XueLin PAN ShaoJun | 2012 | Chinese Science Bulletin2012,57,25: | 9

A pair of transposon-derived proteins function in a histone acetyltransferase complex for active DNA demethylation显示文摘Transposons 被 epigenetic 机制通常包括 DNA methylation 使沉默。 | Cheng-Guo Duan Xingang Wang Shaojun Xie Li Pan Daisuke Miki Kai Tang Chuan-Chih Hsu Mingguang Lei Yingli Zhong Yueh-Ju Hou ZhijuanWang Zhengjing Zhang Satendra K Mangrauthia Huawei Xu Heng Zhang Brian Dilkes W Andy Tao Jian-Kang Zhu | 2017 | Cell Research2017,27,2: | 6
Comparison of viral RNA-host protein interactomes across pathogenic RNA viruses informs rapid antiviral drug discovery for SARS-CoV-2显示文摘In contrast to the extensive research about viral protein–host protein interactions that has revealed major insights about how RNA viruses engage with host cells during infection, few studies have examined interactions between host factors and viral RNAs(vRNAs).Here,we profiled vRNA–host protein interactomes for three RNA virus pathogens(SARS-CoV-2,Zika,and Ebola viruses)using ChIRP-MS.Comparative interactome analyses discovered both common and virus-specific host responses and vRNA-associated proteins that variously promote or restrict viral infection. In particular,SARS-CoV-2 binds and hijacks the host factor IGF2BP1 to stabilize vRNA and augment viral translation. Our interactome-informed drug repurposing efforts identified several FDA-approved drugs(e.g.,Cepharanthine)as broad-spectrum antivirals in cells and hACE2 transgenic mice.A co-treatment comprising Cepharanthine and Trifluoperazine was highly potent against the newly emerged SARS-CoV-2 B.1.351 variant. Thus, our study illustrates the scientific and medical discovery utility of adopting a comparative vRNA-host protein interactome perspective. | Shaojun Zhang Wenze Huang Lili Ren Xiaohui Ju Mingli Gong Jian Rao Lei Sun Pan Li Qiang Ding Jianwei Wang Qiangfeng Cliff Zhang | 2022 | Cell Research2022,32,1: | 5
GSH-triggered sequential catalysis for tumor imaging and eradication based on star-like Au/Pt enzyme carrier system显示文摘Distinctively different metabolism between tumor cells and normal cells endows tumor tissues unique microenvironment. In this regard,we have successfully prepared a sequential catalytic platform based on Au/Pt star for tumor theragnostic. The multifunctional probes consisted of a gold/platinum star-shaped core(Au/Pt star)conjugated with a GSH-sensitive disulfide bond(S–S),a targeting ligand(rHSA-FA),a near-infrared fluorophore(IR780)and glucose oxidase(GOx).When systemically administered in a xenografted murine model, the probes specifically targeted the tumor sites. As the disulfide linker was cleaved by intracellular GSH,the IR780 molecules could be released for photo-thermal therapy&photodynamic therapy(PTT&PDT)and imaging.Subsequently, the Pt nanolayer of the Au/Pt star and the GOx formed a sequential catalytic system:GOx effectively catalyzed intracellular glucose by consuming oxygen to generate H2O2 and enhance the local acidity, and the Pt layer exhibited peroxidase-like property to catalyze H2O2 producing toxic·OH for tumor oxidative damage. Here we demonstrated that our probes simultaneously possessed a GSH-sensitive release,real-time imaging ability, and synergetic cancer starving-like therapy/enzyme oxidative therapy/PTT/PDT features, which provides a potential strategy for effective tumor theragnostic. | Amin Zhang Qian Zhang Gabriel Alfranca Shaojun Pan Zhicheng Huang Jin Cheng Qiang Ma Jie Song Yunxiang Pan Jian Ni Lijun Ma Daxiang Cui | 2020 | Nano Research2020,13,1: | 4
Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified, natural variations in these genes that affect grain size are poorly characterized. Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription, results in shorter grain length, lower grain weight and delayed flowering in gl10 plants. The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages. Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape. | Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang | 2022 | Journal of Genetics and Genomics2022,49,5: | 3

EB病毒膜潜伏蛋白LMP-1筛选鼻咽癌的初步结果(英文)显示文摘Objective: Early diagnosis of nasopharyngeal carcinoma (NPC) is an important method to improve the survival rate. However, the sensitivity and specificity of the screening protocols which was widely used in clinic now are considered to be unsatisfactory.Epstein-Barr virus (EBV)-encoded latent membrane protein-1 (LMP-1) is one of the proteins that have been suggested to be a classic oncogene with transformation properties. The current study set out to discuss the clinical significance of LMP-1 on the screening of NPC.Methods: Three hundred patients who visited our institution (Department of Radiation Oncology,Fujian Provincial Cancer Hospital,Fuzhou,China) with ENT symptoms between 2007 and 2008 were involved in this study, and all of them were agreed to be involved in this investigation. Not only did they undergo nasopharyngeal swab to obtain cells for the LMP-1 polymerase chain reaction (PCR) analysis, but also nasopharyngeal biopsy were taken to identify the diagnosis. Results: An amount of DNA that was sufficient for PCR was extracted from 243 (81%) swab samples, the positive rate of LMP-1 of those with non-nasopharyngeal carcinoma was 3.85% (4/108),which was much lower than those with nasopharyngeal carcinoma (P < 0.05).By detecting LMP-1 in nasopharyngeal swabs,NPC was diagnosed with a sensitivity of 88.15% (119 of 135 patients),specificity of 96.30% (104 of 108 patients),a positive predictive value of 95.2% (119 of 123 patients),a negative predictive value of 86.67% (104 of 120 patients),accuracy of 91.77%,and Youden index of 84.45%.Conclusion: The nasopharyngeal swab coupled with PCR-based EBV LMP-1 detection have high sensitivity and specificity, and also good repeatability,it could serve as part of the screening program for high-risk populations. | Shaojun Lin Qiaojuan Guo Jin Lin Jingfeng Zong Lu Han Jianji Pan | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,1: | 2

Effect of esculetin on bone metabolism in ovariectomized rats显示文摘OBJECTIVE: To determine the effect of an esculetin formulation(at 97.4% purity) on osteoporosis, and to investigate the potential underlying molecular mechanism(s).METHODS: Sixty specific pathogen free-grade female Wistar rats were randomly assigned to three groups: blank control(n = 12), sham(n = 12), and model(n = 36). The model group were bilaterally ovariectomized. The sham group had the tissue surrounding the ovaries removed, while the ovaries were retained. After 3 months, the model group was randomly divided into three subgroups: OVX(n = 12), positive control(n = 12), and esculetin(n =12). The positive control group and the esculetin group were intragastrically administered diethylstilbestrol(0.046 mgkd^(-1)), respec·kg^(-1)tively,·d^(-1)) or esculetin(384 mgg^(-1) once per day for 6 consecu··-tive days; medication administration was then stopped for 1 d, before being administered for another 6 consecutive days. All rats were treated for 3months. Samples were collected at the end of the treatment period. An Osteocore3 Digital 2D bone densitometer was used to test the bone mineral density, and histomorphometric analysis was performed to measure bone mass, bone formation, and bone resorption. Enzyme-linked immunosorbent assay analysis was used to measure the serum concentrations of interleukin-6(IL-6), osteoprotegerin(OPG), and receptor activator of nuclear factor-kappa B ligand(RANKL). Immunohistochemistry and in situ hybridization were performed to detect the protein and mRNA expressions of OPG and RANKL in osteoblasts and bone marrow stromal cells.RESULTS: Compared with the OVX group, the esculetin group had significantly greater femoral bone mineral density and tibial trabecular bone volume, and significantly smaller trabecular resorption surface. The percentage of trabecular formation surface, average osteoid width, trabecular bone mineralization rate, and cortical bone mineralization rate did not significantly differ between groups. Compared with the sham group, the esculetin group had significantly decreased serum levels of IL-6and RANKL, and significant downregulation of RANKL protein and mRNA expression levels in osteoblasts and bone marrow stromal cells; however, there was no significant difference between groups in OPG.CONCLUSION: Esculetin can increase bone mass by upregulating RANKL expression in osteoblasts and bone marrow stromal cells, and decreasing serum IL-6 concentration. This indicates that the therapeutic effect of esculetin on osteoporosis occurs via decreased bone resorption. | Liu Meijie Wang Ruihai Li Yan Bai Dong Pan Jinghua Liu Hong Wang Shaojun Wu Jiaying Sun Gang Miao Qing Ju Dahong Liu Limei | 2018 | Journal of Traditional Chinese Medicine2018,38,6: | 1

An ultra low-input method for global RNA structure probing uncovers Regnase-1-mediated regulation in macrophages显示文摘To enable diverse functions and precise regulation,an RNA sequence often folds into complex yet distinct structures in different cellular states.Probing RNA in its native environment is essential to uncovering RNA structures of biological contexts. However, current methods generally require large amounts of input RNA and are challenging for physiologically relevant use. Here,we report smartSHAPE,a new RNA structure probing method that requires very low amounts of RNA input due to the largely reduced artefact of probing signals and increased efficiency of library construction.Using smartSHAPE,we showcased the profiling of the RNA structure landscape of mouse intestinal macrophages upon inflammation, and provided evidence that RNA conformational changes regulate immune responses. These results demonstrate that smartSHAPE can greatly expand the scope of RNA structure-based investigations in practical biological systems, and also provide a research paradigm for the study of post-transcriptional regulation. | Meiling Piao Pan Li Xiaomin Zeng Xi-Wen Wang Lan Kang Jinsong Zhang Yifan Wei Shaojun Zhangas Lei Tang Jianghui Zhu Chun Kit Kwok Xiaoyu Hu Qiangfeng Cliff Zhang | 2022 | Fundamental Research2022,2,1: | 0
