| 1 | Properties of Supersulphated Phosphogysum-slag Cement (SSC) Concrete显示文摘Supersulphated phosphogysum-slag cement(SSC) is a newly developed non-burned cementitious material mainly composed of phosphogysum(PG) and ground granulated blast furnace slag(GGBFS), with small amount of steel slag(SS) and clinker(CL). SSC is a kind of environmentally-friendly cementitious material due to its energy-saving, low-carbon emission, and waste-utilization. We prepared concretes with supersulphated phosphogysum-slag cement, and studied the mechanical properties, microproperties and resistance to chloride penetration of concrete in comparison with those of portland slag cement(PSC) and ordinary portland cement(OPC) concrete. The test results show that the compressive strength of SSC concrete can reach 38.6 MPa after 28 d, close to PSC concrete and OPC concrete. Microanalyses indicate that large quantities of ettringite and C-S-H, and little amount of Ca(OH)2 are generated during the hydration of SSC. The dense cement paste structure of SSC is formed by ettringite and C-S-H, surrounded unreacted phosphogysum. | 2 | The essential oil components of Cinnamomum cassia: an analysis under different thinning models of plantation Pinus massoniana显示文摘Thinning is an important activity employed in forest management. To date, studies have mainly focused on the effects of thinning on the growth of trees during the same thinning period. In this study, plantation Pinus massoniana Lamb. near maturity were thinned at varying intensities and an economically important species, Cinnamomum cassia Presl., was planted beneath the thinned canopy. The aim of the study was to explore the effects of the extent of thinning on the essential oil content and its components of C. cassia in different parts of the plant, as well as the economic feasibility of the P. massoniana–C.cassia management model. Thinning significantly reduced the oil yield in the bark and branches of C. cassia, but hardly impacted the oil yield from C. cassia leaves compared with pure C. cassia forest(CK). Among the different thinning treatments, both light(T.4) and extensive(T.1)thinning reduced the oil yield of C. cassia bark and new branches. The concentrations of the main aldehydes differed in different parts of the plant and were affected by the extent of thinning. The influence on cinnamaldehyde in the bark was minor, but was much greater in the branches and leaves. Both the oil yield and content of cinnamaldehyde showed that moderate(T.3) thinning was more favorable than other thinning models. | 4 | Preparation of New Cementitious System using Fly Ash and Dehydrated Autoclaved Aerated Concrete显示文摘We experimentally studied the interaction between pozzolanic material(fly ash) and dehydrated autoclaved aerated concrete(DAAC). The DAAC powder was obtained by grinding aerated concrete waste to particles fi ner than 75μm and was then heated to temperatures up to 900 ℃. New cementitious material was prepared by proportioning fly ash and DAAC, named as AF. X-ray diffraction(XRD) was employed to identify the crystalline phases of DAAC before and after rehydration. The hydration process of AF was analyzed by the heat of hydration and non-evaporable water content(Wn). The experimental results show that the highest reactivity of DAAC can be obtained by calcining the powder at 700 ℃ and the dehydrated products are mainly β-C2 S and CaO. The cumulative heat of hydration and Wn was found to be strongly dependent on the replacement level of fl y ash, increasing the replacement level of fl y ash lowered them in AF. | 5 | Establishment and Application of a SYBR Green I Real-time Quantitative PCR Assay for the Detection of LAT Gene of Pseudorabies Virus in Bama Miniature Pigs显示文摘[Objective] This study aimed to establish a rapid,sensitive and specific SYBR Green I real-time quantitative PCR assay for the detection of latent pseudorabies virus( PRV) infection. [Method]SYBR Green I real-time quantitative PCR conditions and system for early detection of latent pseudorabies virus infection were optimized and compared with conventional PCR to investigate the sensitivity and specificity. Subsequently,the established assay was applied to detect different clinical samples. [Result]The sensitivity of SYBR Green I real-time quantitative PCR assay( 52 copies / μl) was 1 000 times higher than that of conventional PCR( 5. 2 × 104 copies / μl) and the detection time was shortened by 1 /2. The established assay could be used to detect PRV but could not be used to detect PCV2,PPV,CSFV or PRRSV. Various tissues were collected from Bama miniature pigs with latent PRV infection under sterile conditions for real-time PCR detection. Results showed that viral copy number in the brain,nasal swab,inguinal lymph node,liver,lung and spleen was above 20,while PRV was not detected in the kidney and heart tissues. | 6 | Expression of Pseudorabies Virus gE Core Epitopes in Escherichia coli Strain BL21 and Utilization of Indirect PRV gE-ELISA显示文摘Pseudorabies virus glycoprotein E( PRV gE) has been recognized as a suitable diagnostic antigen for pseudorabies. In order to produce gE antigen in large quantities and at low cost,a gene fragment encoding PRV gE core epitopes was expressed in E. coli BL21 expression system. SDS-PAGE and Western Blotting revealed that the expression product in culture supernatant of E. coli BL21 was a recombinant protein,approximately 51. 8 Kd. At 5 h post-induction,protein concentration assay showed that the expression product amounted to 1. 65 mg /ml,accounting for 24. 17% of total proteins in the culture supernatant. An indirect PRV gE-ELISA was established by using the recombinant expression product as a coating antigen. Cross-reactivity assay showed that this antigen was PRV specific. In addition,the assay was consistently reproducible. | 13 | In vitro Transcriptome Analysis of Two Chinese Isolates of Streptococcus suis Serotype 2显示文摘The Streptococcus suis serotype 2(S. suis 2) isolates 05ZYH33 and 98HAH33 have caused severe human infections in China. Using a strand-specific RNA-seq analysis, we compared the in vitro transcriptomes of these two Chinese isolates with that of a reference strain(P1/7). In the89 K genomic island that is specific to these Chinese isolates, a toxin–antitoxin system showed relatively high levels of transcription among the S. suis. The known virulence factors with high transcriptional activity in these two highly-pathogenic strains are mainly involved in adhesion, biofilm formation, hemolysis and the synthesis and transport of the outer membrane protein. Furthermore,our analysis of novel transcripts identified over 50 protein-coding genes with one of them encoding a toxin protein. We also predicted over 30 small RNAs(s RNAs) in each strain, and most of them are involved in riboswitches. We found that six s RNA candidates that are related to bacterial virulence, including csp A and rli38, are specific to Chinese isolates. ORF2 gene of porcine circovirus type2 cloned by PCR was ligated to the expression vector pEGFP-N1,which contains enhanced green fluorescent protein(EGFP) report gene,the recombinant eukaryotic expression plasmid pEGFP-N1-ORF2 was constructed successfully and was transfected into prepared duck myocardial cells(DMCs) by lipofectin. According to the result,the fluorescence expression was directly detected with fluorescence microscope,and the expression of ORF2 were analyzed by RT-PCR and indirect immunofluorescence assay(IFA) respectively. About 48 h after transfection,green fluorescent can be observed on transfected cells: T-PCR and IFA were positive.
