1Chemical compound cinobufotalin potently induces FOXO1-stimulated cisplatin sensitivity by antagonizing its binding partner MYH9显示文摘In this study,we present novel molecular mechanisms by which FOXO1 functions as a tumor suppressor to prevent the pathogenesis of nasopharyngeal carcinoma(NPC).First,we observed that FOXO1 not only controlled tumor stemness and metastasis,but also sensitized NPC cells to cisplatin(DDP)in vitro and in vivo.Mechanistic studies demonstrated that FOXO1-induced miR-200b expression through the GSK3β/β-catenin/TCF4 network-mediated stimulation of ZEB1,which reduced tumor stemness and the epithelial–mesenchymal transition(EMT)signal.Furthermore,we observed FOXO1 interaction with MYH9 and suppression of MYH9 expression by modulating the PI3K/AKT/c-Myc/P53/miR-133a-3p pathway.Decreased MYH9 expression not only reduced its interactions with GSK3β,but also attenuated TRAF6 expression,which then decreased the ubiquitin-mediated degradation of GSK3βprotein.Increased GSK3βexpression stimulated theβ-catenin/TCF4/ZEB1/miR-200b network,which increased the downstream tumor stemness and EMT signals.Subsequently,we observed that chemically synthesized cinobufotalin(CB)strongly increased FOXO1-induced DDP chemosensitivity by reducing MYH9 expression,and the reduction in MYH9 modulated GSK3β/β-catenin and its downstream tumor stemness and EMT signal in NPC.In clinical samples,the combination of low FOXO1 expression and high MYH9 expression indicated the worst overall survival rates.Our studies demonstrated that CB potently induced FOXO1-mediated DDP sensitivity by antagonizing its binding partner MYH9 to modulate tumor stemness in NPC.YongHao Li Xiong Liu Xian Lin Menyang Zhao Yanyi Xiao Chen Liu Zixi Liang Zelong Lin Renhui Yi Zibo Tang Jiahao Liu Xin Li Qingping Jiang Libo Li Yinyin Xie Zhen Liu Weiyi Fang 2019Signal Transduction and Targeted Therapy2019,4,1:6
2Super enhancer inhibitors suppress MYC driven transcriptional amplification and tumor progression in osteosarcoma显示文摘Osteosarcoma is the most common primary bone sarcoma that mostly occurs in young adults. The causes of osteosarcoma are heterogeneous and still not fully understood. Identification of novel, important oncogenic factors in osteosarcoma and development of better, effective therapeutic approaches are in urgent need for better treatment of osteosarcoma patients. In this study, we uncovered that the oncogene MYC is significantly upregulated in metastastic osteosarcoma samples. In addition, high MYC expression is associated with poor survival of osteosarcoma patients. Analysis of MYC targets in osteosarcoma revealed that most of the osteosarcoma super enhancer genes are bound by MYC. Treatment of osteosarcoma cells with super enhancer inhibitors THZ1 and JQ1 effectively suppresses the proliferation, migration, and invasion of osteosarcoma cells. Mechanistically,THZ1 treatment suppresses a large group of super enhancer containing MYC target genes including CDK6 and TGFB2. 8Manganese and chromium doping in atomically thin MoS2显示文摘Recently,two-dimensional materials have been attracting increasing attention because of their novel properties and promising applications.However,the impurity doping remains a significant challenge owing to the lack of the doping strategy in the atomically thin layers.Here we report on the chromium(Cr) and manganese(Mn)doping in atomically-thin MoS_2 crystals grown by chemical vapor deposition.The Cr/Mn doped MoS_2 samples are characterized by a peak at 1.76 and 1.79 eV in photoluminescence spectra,respectively,compared with the undoped one at 1.85 eV.The field-effect transistor(FET) devices based on the Mn doping show a higher threshold voltage than that of the pure MoS_2 while the Cr doping exhibits the opposite behavior.Importantly,the carrier concentration in these samples displays a remarkable difference arising from the doping effect,consistent with the evolution of the FET performance.The temperature-dependent conductivity measurements further demonstrate a large variation in activation energy.The successful incorporation of the Mn and Cr impurities into the monolayer MoS_2 paves the way towards the high Curie temperature two-dimensional dilute magnetic semiconductors.Ce Huang Yibo Jin Weiyi Wang Lei Tang Chaoyu Song Faxian Xiu 2017Journal of Semiconductors2017,38,3:1

19Theoretical perspective on synthetic man-made life:Learning from the origin of life显示文摘Creating a man-made life in the laboratory is one of life science's most intriguing yet challenging problems.Advances in synthetic biology and related theories,particularly those related to the origin of life,have laid the groundwork for further exploration and understanding in this field of artificial life or man-made life.But there remains a wealth of quantitative mathematical models and tools that have yet to be applied to this area.In this paper,we review the two main approaches often employed in the field of man-made life:the top-down approach that reduces the complexity of extant and existing living systems and the bottom-up approach that integrates welldefined components,by introducing the theoretical basis,recent advances,and their limitations.We then argue for another possible approach,namely'bottom-up from the origin of life':Starting with the establishment of autocatalytic chemical reaction networks that employ physical boundaries as the initial compartments,then designing directed evolutionary systems,with the expectation that independent compartments will eventually emerge so that the system becomes free-living.This approach is actually analogous to the process of how life originated.With this paper,we aim to stimulate the interest of synthetic biologists and experimentalists to consider a more theoretical perspective,and to promote the communication between the origin of life community and the synthetic man-made life community.Lu Peng Zecheng Zhang Xianyi Wang Weiyi Qiu Liqian Zhou Hui Xiao Chunxiuzi Liu Shaohua Tang Zhiwei Qin Jiakun Jiang Zengru Di Yu Liu 2023Quantitative Biology2023,11,4:0
20Prediction of protected band edge states and dielectric tunable quasiparticle and excitonic properties of monolayer MoSi_(2)N_(4)显示文摘The electronic structure of two-dimensional(2D)materials are inherently prone to environmental perturbations,which may pose significant challenges to their applications in electronic or optoelectronic devices.A 2D material couples with its environment through two mechanisms:local chemical coupling and nonlocal dielectric screening effects.The local chemical coupling is often difficult to predict or control experimentally.Nonlocal dielectric screening,on the other hand,can be tuned by choosing the substrates or layer thickness in a controllable manner.Therefore,a compelling 2D electronic material should offer band edge states that are robust against local chemical coupling effects.Here it is demonstrated that the recently synthesized MoSi_(2)N_(4)is an ideal 2D semiconductor with robust band edge states protected from capricious environmental chemical coupling effects.Detailed many-body perturbation theory calculations are carried out to illustrate how the band edge states of MoSi_(2)N_(4)are shielded from the direct chemical coupling effects,but its quasiparticle and excitonic properties can be modulated through the nonlocal dielectric screening effects.This unique property,together with the moderate band gap and the thermodynamic and mechanical stability of this material,paves the way for a range of applications of MoSi_(2)N_(4)in areas including energy,2D electronics,and optoelectronics.Yabei Wu Zhao Tang Weiyi Xia Weiwei Gao Fanhao Jia Yubo Zhang Wenguang Zhu Wenqing Zhang Peihong Zhang 2022npj Computational Materials2022,,1:0
