2Speciation and fractionation of heavy metals in soil experimentally contaminated with Pb, Cd, Cu and Zn together and effects on soil negative surface charge显示文摘Speciation and fractionation of heavy metals in soil subsamples experimentally loaded with Pb, Cd, Cu and Zn in orthogonal design was investigated by sequential extraction, and operationally defined as water soluble and exchangeable(SE), weakly specific adsorbed(WSA), Fe and Mn oxides bound(OX) and organic bound(ORG). The results showed that fractions of heavy metals in the soil subsamples depended on their speciation. About 90% of Cd and 75% of Zn existed in soil subsamples in the SE fraction. Lead and Cu existed in soil subsamples as SE, WSA and OX fractions simultaneously, although SE was still the major fraction. Organic bound heavy metals were not clearly apparent in all the soil subsamples. The concentration of some heavy metal fractions in soil subsamples showed the good correlation with ionic impulsion of soil, especially for the SE fraction. Continuous saturation of soil subsamples with 0.20 mol/L NH 4Cl, which is the first step for determination of the negative surface charge of soil by the ion retention method, resulted in desorption of certain heavy metals from the soil. It was found that the percentage desorption of heavy metals from soil subsamples depended greatly on pH, the composition and original heavy metal content of the soil subsamples. However, most of the heavy metals in the soil subsamples were still be retained after multiple saturation. Compared with the parent soil, the negative surface charge of soil subsamples loaded with heavy metals did not show difference significantly from that of the parent one by statistical analysis. 3OsSPL18 controls grain weight and grain number in rice显示文摘Grain weight and grain number are two important traits directly determining grain yield in rice. To date,a lot of genes related to grain weight and grain number have been identified; however, the regulatory mechanism underlying these genes remains largely unknown. In this study, we studied the biological function of OsSPL18 during grain and panicle development in rice. Knockout (KO) mutants of OsSPL18exhibited reduced grain width and thickness, panicle length and grain number, but increased tiller number. Cytological analysis showed that OsSPL18 regulates the development of spikelet hulls by affecting cell proliferation. qRT-PCR and GUS staining analyses showed that OsSPL18 was highly expressed in developing young panicles and young spikelet hulls, in agreement with its function in regulating grain and panicle development. Transcriptional activation experiments indicated that OsSPL18is a functional transcription factor with activation domains in both the N-terminus and C-terminus, and both activation domains are indispensable for its biological functions. Quantitative expression analysis showed that DEP1, a major grain number regulator, was significantly down-regulated in OsSPL18 KO lines.Both yeast one-hybrid and dual-luciferase (LUC) assays showed that OsSPL18 could bind to the DEP1promoter, suggesting that OsSPL18 regulates panicle development by positively regulating the expression of DEP1. Sequence analysis showed that OsSPL18 contains the OsmiR156k complementary sequence in the third exon; 5?RLM-RACE experiments indicated that OsSPL18 could be cleaved by OsmiR156k. 4Correlations between low thyroid function and incidence of atrial fibrillation in hypertrophic obstructive cardiomyopathy显示文摘Background::Atrial fibrillation (AF) is the most common arrhythmia in patients with hypertrophic obstructive cardiomyopathy (HOCM). Data regarding the correlations of thyroid dysfunction and the incidence of AF in HOCM are quite limited. This study aimed to reveal the correlations between different thyroid status and the corresponding incidence of AF in a large HOCM cohort.Methods::A total of 806 HOCM patients with complete information on thyroid function tests and comprehensive cardiac evaluations were recruited. The participants were divided into the AF group (n = 159) and non-AF group (n = 647) according to established medical history and results of Holter monitoring. The thyroid status of the study population and the corresponding incidence of AF were assessed and analyzed.Results::Hypothyroidism accounted for the greatest proportion of thyroid dysfunction in HOCM patients. The incidence of AF significantly increased in individuals with both overt ( P = 0.022) and subclinical ( P = 0.007) hypothyroidism. Compared with participants in the non-AF group, those with positive AF episodes presented with lower free triiodothyronine (FT3) (2.86 ± 0.52 pg/mL vs. 3.01 ± 0.42 pg/mL, P = 0.001), higher free thyroxine (FT4) (1.24 ± 0.25 ng/dL vs. 1.15 ± 0.16 ng/dL, P < 0.001), and remarkably increased levels of thyrotropin (TSH) (12.6% vs. 5.3%, P = 0.001). Multivariable analyses demonstrated that the concentrations of FT3 (odds ratio [OR] = 0.470, 95% confidence interval [CI]: 0.272-0.813, P = 0.007) and FT4 (OR = 17.992, 95% CI: 5.750-56.296, P < 0.001), as well as TSH levels above normal ranges (OR = 2.276, 95% CI: 1.113-4.652, P = 0.024) were independently associated with the occurrence of AF in the large HOCM cohort. Conclusions::This study indicated a strong link between low thyroid function and the presence of AF in HOCM. 7Evaluation of the Global and Regional Assimilation and Prediction System for Predicting Sea Fog over the South China Sea显示文摘In the South China Sea, sea fog brings severe disasters every year, but forecasters have yet to implement an effective seafog forecast. To address this issue, we test a liquid-water-content-only(LWC-only) operational sea-fog prediction method based on a regional mesoscale numerical model with a horizontal resolution of about 3 km, the Global and Regional Assimilation and Prediction System(GRAPES), hereafter GRAPES-3 km. GRAPES-3 km models the LWC over the sea, from which we infer the visibility that is then used to identify fog. We test the GRAPES-3 km here against measurements in 2016 and 2017 from coastal-station observations, as well as from buoy data, data from the Integrated Observation Platform for Marine Meteorology, and retrieved fog and cloud patterns from Himawari-8 satellite data. For two cases that we examine in detail, the forecast region of sea fog overlaps well with the multi-observational data within 72 h. Considering forecasting for0–24 h, GRAPES-3 km has a 2-year-average equitable threat score(ETS) of 0.20 and a Heidke skill score(HSS) of 0.335,which is about 5.6%(ETS) and 6.4%(HSS) better than our previous method(GRAPES-MOS). 8Effect of warm rolling process on microstructures and tensile properties of 10 Mn steel显示文摘The influence of warm rolling processes on the microstructures and tensile properties of 100 Mn steel was studied.Strength appeared to increase with the rolling temperature but strengthening mechanisms varied.The increase of warm rolling temperature from 250℃ to 600℃ leads to enhanced recrystallization in martensite during the intercritical annealing(IA) at 620℃ for 5 h.As a result,both ultimate tensile strength(UTS) and total elongation(TE) increase.However,the size of relatively coarse recrystallized austenite grains and the resultant yield strength(YS) remain almost constant in this temperature range.The further increase of rolling temperature to 700-800℃ causes a considerable amount of pearlite to be formed during the IA,and then martensite is formed after the IA,resulting in dramatical increases in both YS and UTS but at the great loss of ductility.The warm rolling at 600℃ with 63% thickness reduction can produce the steel with the best mechanical combination of 1.2 GPa UTS and 35% TE,due to the formation of many ultrafine austenite grains and strain-induced cementite precipitates.This demonstrates that the mechanical combination of non-V-alloyed medium Mn steel can be improved to an equivalent level of 0.7% V alloyed 10 Mn steel just via the economic strain-induced cementite precipitation.Bin Hu Xin Tu Haiwen Luo Xinping Mao 2020Journal of Materials Science & Technology2020,47,12:6
9Platinum-group elements for the mantle peridotites in the Dazhuka ophiolite,Tibet,China显示文摘The total PGE amounts of mantle peridotites in the Dazhuka ophiolite, Tibet, are 28.37-50.67 ng/g, slightly higher than those of mantle peridotites in the primitive mantle, and typical ophiolites in the world, and the Alps-type mantle peridotites. The PGE distribution patterns in the Dazhuka mantle peridotites are also different from those of the mantle peridotites of partial melting relict origin. The Dazhuka mantle peridotites have relatively high total PGE amounts and are enriched in Pt, Pd, and Ru. Their PGE distribution patterns belong to the positively inclined-or swallow-type patterns. The PGE distribution patterns in the mantle peridotites of partial melting relict origin belong to the negative-slope patterns or flat patterns. This reflects the unique features of the upper mantle in this region. 10Loss of Gn1a/OsCKX2 confers heavy-panicle rice with excellent lodging resistance显示文摘Significant achievements have been made in breeding programs for the heavy-panicle-type(HPT)rice(Oryza sativa) in Southwest China. The HPT varieties now exhibit excellent lodging resistance,allowing them to overcome the greater pressures caused by heavy panicles. However, the genetic mechanism of this lodging resistance remains elusive. Here, we isolated a major quantitative trait locus, Panicle Neck Diameter 1(PND1), andidentified the causal gene as GRAIN NUMBER 1 A/CYTOKININ OXIDASE 2(Gn1 A/Os CKX2). The null gn1 a allele from rice line R498(gn1 aR498) improved lodging resistance through increasing the culm diameter and promoting crown root development.Loss-of-function of Gn1 a/Os CKX2 led to cytokinin accumulation in the crown root tip and accelerated the development of adventitious roots. Gene pyramiding between the null gn1 aR498 allele with two gain-of-function alleles, STRONG CULM 2(SCM2)and SCM3, further improved lodging resistance.Moreover, Gn1 a/Os CKX2 had minimal influence on overall rice quality. 12Correlation between miR-564, TGF-β1, and radiation-induced lung injury显示文摘Objective Our study aimed to analyze the expression of miR-564 and TGF-β1 in cancer tissues and the serum of patients with radiation-induced lung injury,and to investigate the relationship between them and radiation-induced lung injury.Methods In situ hybridization and real-time fluorescence quantitative method were used to detect the expression of miR-564.Additionally,immunohistochemistry and enzyme-linked immunosorbent assay(ELISA)were performed to detect the expression of TGF-β1.Results The overall incidence of acute radiation pneumonia was 55.9%(100/179).The incidence of≥grade 2 radioactive pneumonia was 24.0%(43/179)and that of grade 1 was 31.8%(57/179).The expression of miR-564 in grade≥2 was slightly higher than that in patients without or with grade 1,but there was no statistical difference(P=0.86).The serum level and ratio of miR-564 in patients with grade≥2 were significantly higher than those without or with grade 1(P=0.005,P=0.025,respectively).The expression of TGF-β1 in grade≥2 was significantly higher than that of patients without or with grade 1(P=0.017).The serum levels of TGF-β1 in grade≥2 were significantly higher than those in patients without or with grade 1(P=0.038).Although the ratio of TGF-β1 in radiation pneumonia of grade≥2 was significantly higher than that of without or with grade 1,there was no significant difference(P=0.24).Moreover,patients with higher expression of miR-564 and lower expression of TGF-β1 had better prognosis.Conclusion MiR-564 and TGF-β1 are predictors of radiation-induced lung injury.Monitoring its changing trend can improve the accuracy of predicting radiation-induced lung injury.The levels and ratio of serum miR-564 and TGF-β1 in patients with radiation-induced lung injury are related to the severity of radiationinduced lung injury.Yunzhang Ge Tao Xie Bin Yang Qianxia Li Qingrong Ren Xiaoyi Zhou Desheng Hu Zhongshu Tu 2019Oncology and Translational Medicine2019,5,5:4

17The Brassica napus GATA transcription factor BnA5.ZML1 is a stigma compatibility factor显示文摘Self-incompatibility(SI)is a genetic mechanism that rejects self-pollen and thus prevents inbreeding in some hermaphroditic angiosperms.In the Brassicaceae,SI involves a pollen-stigma recognition system controlled by a single locus known as the S locus,which consists of two highly polymorphic genes that encode S-locus cysteine-rich protein(SCR)and S-receptor kinase(SRK).When self-pollen lands on the stigma,the S-haplotype-specific interaction between SCR and SRK triggers SI.Here,we show that the GATA transcription factor BnA5.ZML1 suppresses SI responses in Brassica napus and is induced after compatible pollination.The loss-of-function mutant bna5.zml1 displays reduced self-compatibility.In contrast,overexpression of BnA5.ZML1 in self-incompatible stigmas leads to a partial breakdown of SI responses,suggesting that BnA5.ZML1 is a stigmatic compatibility factor.Furthermore,the expression levels of SRK and ARC1 are up-regulated in bna5.zml1 mutants,and they are down-regulated in BnA5.ZML1 overexpressing lines.SRK affects the cellular localization of BnA5.ZML1 through direct protein–protein interaction.Overall,our findings highlight the fundamental role of BnA5.ZML1 in SI responses in B.napus,establishing a direct interaction between BnA5.ZML1 and SRK in this process.Zhiqiang Duan Yatao Zhang Jinxing Tu Jinxiong Shen Bin Yi Tingdong Fu Cheng Dai Chaozhi Ma 2020Journal of Integrative Plant Biology2020,62,8:2
18Neutralization of SARS-CoV-2 pseudovirus using ACE2-engineered extracellular vesicles显示文摘The spread of coronavirus disease 2019(COVID-19)throughout the world has resulted in stressful healthcare burdens and global health crises.Developing an effective measure to protect people from infection is an urgent need.The blockage of interaction between angiotensin-converting enzyme 2(ACE2)and S protein is considered an essential target for anti-severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)drugs.A full-length ACE2 protein could be a potential drug to block early entry of SARS-Co V-2 into host cells.In this study,a therapeutic strategy was developed by using extracellular vesicles(EVs)with decoy receptor ACE2 for neutralization of SARS-Co V-2.The EVs embedded with engineered ACE2(EVs-ACE2)were prepared;the EVs-ACE2 were derived from an engineered cell line with stable ACE2 expression.The potential effect of the EVs-ACE2 on anti-SARS-Co V-2 was demonstrated by both in vitro and in vivo neutralization experiments using the pseudovirus with the S protein(S-pseudovirus).EVs-ACE2 can inhibit the infection of S-pseudovirus in various cells,and importantly,the mice treated with intranasal administration of EVs-ACE2 can suppress the entry of S-pseudovirus into the mucosal epithelium.Therefore,the intranasal EVs-ACE2 could be a preventive medicine to protect from SARS-Co V-2 infection.This EVs-based strategy offers a potential route to COVID-19 drug development.Canhao Wu Qin Xu Huiyuan Wang Bin Tu Jiaxin Zeng Pengfei Zhao Mingjie Shi Hong Qiu Yongzhuo Huang 2022Acta Pharmaceutica Sinica B2022,12,3:2
19An in planta haploid induction system in Brassica napus显示文摘Doubled haploid technology is widely used to accelerate plant breeding,but its use in the important oilseed crop Brassica napus L.is limited because B.napus haploids could only be obtained through in vitro anther or microspore cultures.Recently,maize(Zea mays)lines containing mutations in Domain of unknown function 679 membrane protein(DMP)were used as haploid inducer lines.This new haploid induction mechanism has been extended to several other plants,including the dicots Arabidopsis thaliana,tomato(Solanum lycopersicum),and tobacco(Nicotiana tabacum).Here,we knocked out four BnaDMP genes in the B.napus cultivar Westar using a clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9 vector with an enhanced green fluorescent protein expression cassette.Plants with DMP mutations in B.napus in the T_(0),T_(1),and T_(2) generations exhibited a haploid induction rate up to 2.53%.These results suggest that targeting BnaDMP could be useful for haploid induction in B.napus.Yifan Li Dan Li Qing Xiao Huadong Wang Jing Wen Jinxing Tu Jinxiong Shen Tingdong Fu Bin Yi 2022Journal of Integrative Plant Biology2022,64,6:2
20Low‐intensity pulsed ultrasound ameliorates angiotensin II-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway显示文摘Objective:Cardiac hypertrophy and fibrosis are major pathological manifestations observed in left ventricular remodeling induced by angiotensin II(AngII).Low-intensity pulsed ultrasound(LIPUS)has been reported to ameliorate cardiac dysfunction and myocardial fibrosis in myocardial infarction(MI)through mechano-transduction and its downstream pathways.In this study,we aimed to investigate whether LIPUS could exert a protective effect by ameliorating AngII-induced cardiac hypertrophy and fibrosis and if so,to further elucidate the underlying molecular mechanisms.Methods:We used AngII to mimic animal and cell culture models of cardiac hypertrophy and fibrosis.LIPUS irradiation was applied in vivo for 20 min every 2 d from one week before mini-pump implantation to four weeks after mini-pump implantation,and in vitro for 20 min on each of two occasions 6 h apart.Cardiac hypertrophy and fibrosis levels were then evaluated by echocardiographic,histopathological,and molecular biological methods.Results:Our results showed that LIPUS could ameliorate left ventricular remodeling in vivo and cardiac fibrosis in vitro by reducing AngII-induced release of inflammatory cytokines,but the protective effects on cardiac hypertrophy were limited in vitro.Given that LIPUS increased the expression of caveolin-1 in response to mechanical stimulation,we inhibited caveolin-1 activity with pyrazolopyrimidine 2(pp2)in vivo and in vitro.LIPUS-induced downregulation of inflammation was reversed and the anti-fibrotic effects of LIPUS were absent.Conclusions:These results indicated that LIPUS could ameliorate AngII-induced cardiac fibrosis by alleviating inflammation via a caveolin-1-dependent pathway,providing new insights for the development of novel therapeutic apparatus in clinical practice.Kun ZHAO Jing ZHANG Tianhua XU Chuanxi YANG LiqingWENG TingtingWU XiaoguangWU Jiaming MIAO Xiasheng GUO Juan TU Dong ZHANG Bin ZHOU Wei SUN Xiangqing KONG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,10:2
