| 1 | Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. | 4 | Parametric Problems in Power System Analysis:Recent Applications of Polynomial Approximation Based on Galerkin Method显示文摘In power systems, there are many uncertainty factors such as power outputs of distributed generations and fluctuations of loads. It is very beneficial to power system analysis to acquire an explicit function describing the relationship between these factors(namely parameters) and power system states(or performances). This problem, termed as parametric problem(PP) in this paper, can be solved by Galerkin method,which is a powerful and mathematically rigorous method aiming to seek an accurate explicit approximate function by projection techniques. This paper provides a review of the applications of polynomial approximation based on Galerkin method in power system PPs as well as stochastic problems. First, the fundamentals of polynomial approximation and Galerkin method are introduced. Then, the process of solving three types of typical PPs by polynomial approximation based on Galerkin method is elaborated. | 5 | Serum macrophage migration inhibitory factor as a potential biomarker to evaluate therapeutic response in patients with allergic asthma:an exploratory study显示文摘Background:Previous studies have shown that macrophage migration inhibitory factor(MIF)is involved in the pathogenesis of asthma.This study aimed to investigate whether serum MIF reflects a therapeutic response in allergic asthma.Methods:We enrolled 30 asthmatic patients with mild-to-moderate exacerbations and 20 healthy controls,analyzing the parameter levels of serum MIF,serum total immunoglobulin E(tIgE),peripheral blood eosinophil percentage(EOS%),and fractional exhaled nitric oxide(FeNO).Lung function indices were used to identify disease severity and therapeutic response.Results:Our study showed that all measured parameters in patients were at higher levels than those of controls.After one week of treatment,most parameter levels decreased significantly except for serum tIgE.Furthermore,we found that serum MIF positively correlated with EOS%as well as FeNO,but negatively correlated with lung function indices.Receiver operator characteristic(ROC)curve analysis indicated that among the parameters,serum MIF exhibited a higher capacity to evaluate therapeutic response.The area under the curve(AUC)of MIF was 0.931,with a sensitivity of 0.967 and a specificity of 0.800.Conclusions:Our results suggested that serum MIF may serve as a potential biomarker for evaluating therapeutic response in allergic asthma with mild-to-moderate exacerbations. | Huiyuan ZHU Shaochun YAN Jingshuo WU Zhong ZHANG Xiaolin LI Zheng LIU Xing MA Lina ZHOU Lin ZHANG Mingming FENG Yiwei GENG Aixin ZHANG Sabina JANCIAUSKIENE Aiguo XU | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,6: | 3 |
| 6 | Confined Ni-In intermetallic alloy nanocatalyst with excellent coking resistance for methane dry reforming显示文摘Carbon dioxide and methane are two main greenhouse gases which are contributed to serious global warming.Fortunately,dry reforming of methane(DRM),a very important reaction developed decades ago,can convert these two major greenhouse gases into value-added syngas or hydrogen.The main problem retarding its industrialization is the seriously coking formation upon the nickel-based catalysts.Herein,a series of confined indium-nickel(In-Ni)intermetallic alloy nanocatalysts(In_(x)Ni@SiO_(2))have been prepared and displayed superior coking resistance for DRM reaction.The sample containing 0.5 wt.%of In loading(In_(0.5)Ni@SiO_(2))shows the best balance of carbon deposition resistance and DRM reactivity even after 430 h long term stability test.The boosted carbon resistance can be ascribed to the confinement of core–shell structure and to the transfer of electrons from Indium to Nickel in In-Ni intermetallic alloys due to the smaller electronegativity of In.Both the silica shell and the increase of electron cloud density on metallic Ni can weaken the ability of Ni to activate C–H bond and decrease the deep cracking process of methane.The reaction over the confined InNi intermetallic alloy nanocatalyst was conformed to the Langmuir-Hinshelwood(L-H)mechanism revealed by in situ diffuse reflectance infrared Fourier transform spectroscopy(in-situ DRIFTS).This work provides a guidance to design high performance coking resistance catalysts for methane dry reforming to efficiently utilize these two main greenhouse gases. | Wenming Liu Le Li Sixue Lin Yiwei Luo Zhenghong Bao Yiru Mao Kongzhai Li Daishe Wu Honggen Peng | 2022 | Journal of Energy Chemistry2022,31,2: | 3 |
| 7 | Early monitoring of response to antimetabolic treatment of gemcitabine:A comparison of ^(18)F-FLT and ^(18)F-FDG uptake in Patu 8988 human pancreatic carcinoma cells显示文摘It is essential to predict the treatment efficacy of pancreatic carcinoma early.The purpose of this study was to examine whether ^(18)F-FDG(2'-deoxy-2'-[^(18)F]fiuoro-D-glucose) or ^(18)F-FLT(3'-deoxy-3'-^(18)F-fluorothymidine) PET can be used for chemosensitivity testing by investigating the binding characteristic of ^(18)F-FDG or ^(18)F-FLT with Patu 8988 human pancreatic carcinoma cell and the influence of gemcitabine in the uptake of ^(18)F-FDG or ^(18)F-FLT on Patu 8988.Under the conditions of 1×10~6 cells,3.7 kBq ^(18)F-FDG or ^(18)F-FLT,and incubation at 37℃for 100 min,the cell uptake of ^(18)F-FDG and ^(18)F-FLT was(60.60±3.05)%and(50.57±2.81)%,respectively.There was a significant decrease in TKl-LI(thymidine kinase 1 labeling index) 24 h after administration of gemcitabine.The uptakes of ^(18)F-FDG and ^(18)F-FLT were negatively correlated with the doses of gemcitabine(r= -0.928 for ^(18)F-FDG,r= -0.876 for ^(18)F-FLT,P<0.01).When same doses of gemcitabine were administered,the ^(18)F-FLT uptake inhibition rate was significantly higher than that of ^(18)F-FDG(P<0.01).These results indicate that the response to gemcitabine could be predicted as early as 24 h by ^(18)F-FDG or ^(18)F-FLT PET scans.^(18)F-FLT is more sensitive than ^(18)F-FDG to predict the response to therapy. | DENG Shengming ZHANG Bin WU Yiwei ZHANG Wei | 2011 | Nuclear Science and Techniques2011,22,5: | 2 |

| 10 | A simple fabricated thickness-based stiffness gradient for cell studies显示文摘In this work, we developed a simple method to fabricate a thickness-based continuous stiffness gradient for biological studies. It was made by glass slides, polydimethylsiloxane(PDMS) pre-polymer, spacer and clips only, without any sophisticated equipment. It is easy to fabricate in any general biological and pharmaceutical laboratories. The stiffness gradient was characterized in terms of apparent Young's modulus by atomic force microscopy(AFM) and the Young's modulus along the gradient was found to be 8.5–120 k Pa, which is within the physiological relevant range. He La-C3 cells were cultured on the gradient to study their morphological behavior according to the substrate stiffness. Furthermore, the drug efficiency of etoposide, an anti-cancer drug, was studied along the substrate stiffness gradient. It was found that He La-C3 cells cultured on the soft region of the gradient(8.5–11 k Pa) are more sensitive to etoposide. | 11 | Correlation between antioxidant depletion kinetic model and ageing behaviour of cross-linked polyethylene cable insulation显示文摘A concise kinetic model catering for the antioxidant reaction of thermally aged crosslinked polyethylene(XLPE)cable insulation before its abrupt performance deterioration is proposed for life prediction and condition assessment.The alternating current breakdown strength,elongation at break,carbonyl index,and pyrolysis activation energy of XLPE insulation are found to consistently display an abrupt change at a certain point of ageing time.This is strongly correlated to the depletion of the antioxidant whose content gradually decreases with ageing time and drops to a critical value,after which the abrupt degradation of the above macroscopic performances happens.Based on the simplified chain reactions of XLPE during thermal-oxidative ageing,a concise kinetic model about antioxidant prediction is accordingly established by solving the chemical kinetics equations.It indicates that the antioxidant content decays with the reciprocal of time before the abrupt degradation occurs.Results of antioxidant content predicted by the kinetic model at different temperatures matches well with the experimental results,and its reliability can also be proved by the Arrhenius relationship of the obtained results.The kinetic model proposed herein demonstrates satisfying conciseness and thus can be highly feasible in ageing condition assessment and life predicting of XLPE insulation. | Yingying Zheng Yiwei Long Delun Yi Haoran Zhang Jian Gao Kangning Wu Jianying Li | 2023 | High Voltage2023,8,2: | 2 |
| 12 | Composition engineering of Sb_2S_3 film enabling high performance solar cells显示文摘Sb_2S_3 is a kind of stable light absorption materials with suitable band gap, promising for practical applications. Here we demonstrate that the engineering on the composition ratio enables significant improvement in the device performance. We found that the co-evaporation of sulfur or antimony with Sb_2S_3 is able to generate sulfur-or antimony-rich Sb_2S_3. This composition does not generate essential influence on the crystal structure, optical band and film formability, while the carrier concentration and transport dynamics are considerably changed. The device investigations show that sulfur-rich Sb_2S_3 film is favorable for efficient energy conversion, while antimony-rich Sb_2S_3 leads to greatly decreased device performance. | 18 | Experiment and numerical simulation on the characteristics of fluid–structure interactions of non-rigid airships显示文摘Fluid–structure interaction is an important issue for non-rigid airships with inflated envelopes. In this study, a wind tunnel test is conducted, and a loosely coupled procedure is correspondingly established for numerical simulation based on computational fluid dynamics and nonlinear finite element analysis methods. The typical results of the numerical simulation and wind tunnel experiment, including the overall lift and deformation, are in good agreement with each other. The results obtained indicate that the effect of fluid–structure interaction is noticeable and should be considered for non-rigid airships. Flowinduced deformation can further intensify the upward lift force and pitching moment, which can lead to a large deformation.
