1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Genome-wide analysis of heat shock transcription factor families in rice and Arabidopsis显示文摘The heat shock transcription factors (HSFs) are the major heat shock factors regulating the heat stress response. They participate in regulating the expression of heat shock proteins (HSPs), which are critical in the protection against stress damage and many other important biological processes. Study of the HSF gene family is important for understanding the mechanism by which plants respond to stress. The completed genome sequences of rice (Oryza sativa) and Arabidopsis (Arabidopsis thaliana) constitute a valuable resource for comparative genomic analysis, as they are representatives of the two major evolutionary lineages within the angiosperms: the monocotyledons and the dicotyledons. The identification of phylogenetic relationships among HSF proteins in these species is a fundamental step to unravel the functionality of new and yet uncharacterized genes belonging to this family.In this study, the full complement of HSF genes in rice and Arabidopsis has probably been identified through the genome-wide scan. Phylogenetic analyses resulted in the identification of three major clusters of orthologous genes that contain members belonging to both species, which must have been represented in their common ancestor before the taxonomic splitting of the angiosperms. Further analysis of the phylogenetic tree reveals a possible dicot specific gene group. We also identified nine pairs of paralogs, as evidence for studies on the evolution history of rice HSF family and rice genome evolution. Expression data analysis indicates that HSF proteins are widely expressed in plants. 4Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. 6EXPERIMENTAL AND NUMERICAL STUDIES ON DYNAMIC COMPRESSIVE BEHAVIOR OF REACTIVE POWDER CONCRETES显示文摘Split Hopkinson pressure bar (SHPB) technique is used to determine the dynamic strength of reactive powder concretes (RPCs) with different steel-fiber contents. Two types of pulse shapers with different thicknesses are considered to reduce the high-frequency-oscillation effect and achieve a nearly constant strain rate over a certain deformation range. It is known that the compressive strength of concrete-like materials is hydrostatic-stress-dependent, and the apparent dynamic strength enhancement comes from both the effects of the hydrostatic stress and strain rate. In order to differentiate them, numerical method is used to calculate the contribution of the hydrostatic stress, and then the genuine strain-rate effect on dynamic compressive strength of RPCs is determined. 9Spatial-temporal Dynamics and Driving Forces of Land Development Intensity in the Western China from 2000 to 2015显示文摘The change in land development intensity is an important perspective to reflect the variation in regional social and economic development and spatial differentiation.In this paper,spatial statistical analysis,Ordinary Least Squares(OLS),and Geographically weighted regression(GWR)methods are used to systematically analyse the spatial-temporal characteristics and driving forces of land development intensity for 131 spatial units in the western China from 2000 to 2015.The findings of the study are as follows:1)The land development intensity in the western China has been increasing rapidly.From 2000 to 2015,land development intensity increased by 3.4 times on average.2)The hotspot areas have shifted from central Inner Mongolia,northern Shaanxi and the Beibu Gulf of Guangxi to the Guanzhong Plain and the Chengdu-Chongqing urban agglomeration.The areas of cold spots were mainly concentrated in the Qinghai-Tibet Plateau,Yunnan,and Xinjiang.3)Investment intensity and the natural environment have always been the main drivers of land development intensity in the western China.Investment played a powerful role in promoting land development intensity,while the natural and ecological environment distinctly constrained such development.The effect of the economic factors on land development intensity in the western China has changed,which is reflected in the driving factor of construction land development shifting from economic growth in 2000 to economic structure,especially industrial structure,in 2015.HUANG Xin HUANG Xiaojun LIU Mengmeng WANG Bo ZHAO Yonghua 2020Chinese Geographical Science2020,30,1:12
10Internet of Things to network smart devices for ecosystem monitoring显示文摘Smart, real-time, low-cost, and distributed ecosystem monitoring is essential for understanding and managing rapidly changing ecosystems. However, new techniques in the big data era have rarely been introduced into operational ecosystem monitoring, particularly for fragile ecosystems in remote areas.We introduce the Internet of Things(IoT) techniques to establish a prototype ecosystem monitoring system by developing innovative smart devices and using IoT technologies for ecosystem monitoring in isolated environments. The developed smart devices include four categories: large-scale and nonintrusive instruments to measure evapotranspiration and soil moisture, in situ observing systems for CO2 and d13 C associated with soil respiration, portable and distributed devices for monitoring vegetation variables, and Bi-CMOS cameras and pressure trigger sensors for terrestrial vertebrate monitoring. These new devices outperform conventional devices and are connected to each other via wireless communication networks. The breakthroughs in the ecosystem monitoring IoT include new data loggers and longdistance wireless sensor network technology that supports the rapid transmission of data from devices to wireless networks. The applicability of this ecosystem monitoring IoT is verified in three fragile ecosystems, including a karst rocky desertification area, the National Park for Amur Tigers, and the oasis-desert ecotone in China. By integrating these devices and technologies with an ecosystem monitoring information system, a seamless data acquisition, transmission, processing, and application IoT is created. 14Surrogate production of genome-edited sperm from a different subfamily by spermatogonial stem cell transplantation显示文摘The surrogate reproduction technique,such as inter-specific spermatogonial stem cells(SSCs)transplantation(SSCT),provides a powerful tool for production of gametes derived from endangered species or those with desirable traits.However,generation of genome-edited gametes from a different species or production of gametes from a phylogenetically distant species such as from a different subfamily,by SSCT,has not succeeded.Here,using two small cyprinid fishes from different subfamilies,Chinese rare minnow(gobiocypris rarus,for brief:Gr)and zebrafish(danio rerio),we successfully obtained Gr-derived genome-edited sperm in zebrafish by an optimized SSCT procedure.The transplanted Gr SSCs supported the host gonadal development and underwent normal spermatogenesis,resulting in a reconstructed fertile testis containing Gr spermatids and zebrafish testicular somatic cells.Interestingly,the surrogate spermatozoa resembled those of host zebrafish but not donor Gr in morphology and swimming behavior.When pou5f3 and chd knockout Gr SSCs were transplanted,Gr-derived genome-edited sperm was successfully produced in zebrafish.This is the first report demonstrating surrogate production of gametes from a different subfamily by SSCT,and surrogate production of genome-edited gametes from another species as well.This method is feasible to be applied to future breeding of commercial fish and livestock.Fenghua Zhang Yongkang Hao Xianmei Li Yi Li Ding Ye Ru Zhang Xiaosi Wang Mudan He Houpeng Wang Zuoyan Zhu Yonghua Sun 2022Science China(Life Sciences)2022,65,5:7
15Growth ages of ferromanganese crusts from the western and central Pacific:Comparison between nannofossil analysis and ^(10)Be dating显示文摘Based on results of nannofossil analysis and 10Be dating in ferromanganese crusts M1-1 and A1-1 (no nannofossils were found in it),from the western and central Pacific respectively,it is found that the crust growth ages from nannofossil biostrati-graphy agree well with those based on 10Be isotope analysis. Both crusts have three growth layers,and the oldest layer was deposited in Miocene at about 12.80 Ma. 17Experimental investigation of thermal effect on cavitation characteristics in a liquid rocket engine turbopump inducer显示文摘The objective of this paper is to investigate the effect of water temperature on cavitation characteristics in a turbopump inducer,a series of experiments at different temperatures have been conducted in a newly developed visualization test facility.It is found that higher temperature shows little influence on the non-cavitation performance and breakdown characteristic in the investigated range.The relationship between cavitation development and pressure fluctuation has been discussed in detail.Higher temperature displays a remarkable stabilization effect on the cavitation excited pressure.In particular,the inception cavitation numbers of both the super-synchronous rotating cavitation and synchronous rotating cavitation are decreased at higher temperatures,and the corresponding frequencies are not affected,while the amplitudes are distinctly reduced,and the occurrence range of synchronous rotating cavitation is significantly narrowed.A generalized RayleighPlesset equation has been employed to account for the thermal effect on the bubble development,which may provide a deep insight in understanding the experimental results.Thermal effect is found to act as a remarkable dissipation mechanism to suppress the bubble growth,smooth the collapse.In particular,the excited pressure during collapse is smaller at higher temperatures,which may lead to the stabilization effect of high temperature in this study.Chunmin WANG Le XIANG Yonghua TAN Hui CHEN Kaifu XU 2021Chinese Journal of Aeronautics2021,34,8:7
18Deformation Temperature Dependence of Mechanical Properties and Microstructures for a Novel Quenching—Partitioning—Tempering Steel显示文摘The present investigation on a designed high strength Fe-0.25C-1.48Mn-1.20Si-1.51Ni-0.05Nb(wt%) steel treated by a novel quenching—partitioning—tempering(Q—P—T) process was focused on deformation temperature dependence of mechanical properties and microstructures.The results indicate that the Q—P—T steel deformed at various deformation temperatures from -70 to 300℃exhibits superior mechanical properties due to excellent thermal stability of retained austenite.The microstructural characterization by transmission electron microscopy(TEM) reveals that the high strength of the Q—P-T steel results from dislocation-type martensite laths and fcc NbC carbides or/and hcpε-carbides precipitated dispersively in martensite matrix,while excellent ductility is attributed to the significant transformation induced plasticity (TRIP) effect produced by considerable amount of retained austenite.The relationship between mechanical properties and microstructures at different deformation temperatures was clarified.Ying Wang Zhenghong Guo Nailu Chen Yonghua Rong 2013Journal of Materials Science & Technology2013,29,5:5
19Preparation and Characterization of Waterborne Polyurethane/Cellulose Nanocrystal Composite Membrane from Recycling Waste Paper显示文摘Cellulose plays a key role in abundant organic natural materials meeting the increasing demand for green and biocompatible products.The highly crystalline nanoscale component of cellulose nanocrystals has recently attracted great attention due to the versatile performance as filler or matrix in producing functional materials.In this work,we prepared the waterborne polyurethane via a prepolymer process,and obtained cellulose and cellulose nanocrystals from waste paper via a facile acid hydrolysis process.After that,the cellulose nanocrystals were assembled into film and mixed with polyurethane to prepare flexible polyurethane/cellulose nanocrystals composite membrane with different soaking time.The correlation between the bulk structure and applied properties including thermal resistance and mechanical property was investigated by using Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS),scanning electron microscopy(SEM),thermogravimetric analysis(TGA),differential scanning calorimetry(DSC)and folding test.The structure analysis indicates that cellulose nanocrystals prepared from used paper have a quality similar to that of commercial cellulose.Meanwhile,the cellulose nanocrystals have been mixed with polyurethane uniformly.Polyurethane can significantly benefit to the thermal resistance and mechanical property of the cellulose nanocrystals film.The polyurethane/cellulose nanocrystals composite membrane present good flexibility and may hold a significantly potential application as visual and flexible material.Xing Zhou Xin Zhang Dong Wang Changqing Fang Wanqing Lei Zhigang Huang Yonghua Song Xinyu He Yingwei Huang 2020Journal of Renewable Materials2020,8,6:5
20Self-crosslinkable chitosan-hyaluronic acid dialdehyde nanoparticles for CD44-targeted siRNA delivery to treat bladder cancer显示文摘Bladder cancer is one of the concerning malignancies worldwide,which is lacking effective targeted therapy.Gene therapy is a potential approach for bladder cancer treatment.While,a safe and effective targeted gene delivery system is urgently needed for prompting the bladder cancer treatment in vivo.In this study,we confirmed that the bladder cancer had CD44 overexpression and small interfering RNAs(siRNA)with high interfere to Bcl2 oncogene were designed and screened.Then hyaluronic acid dialdehyde(HAD)was prepared in an ethanol-water mixture and covalently conjugated to the chitosan nanoparticles(CS-HAD NPs)to achieve CD44 targeted siRNA delivery.The in vitro and in vivo evaluations indicated that the siRNA-loaded CS-HAD NPs(siRNA@CS-HAD NPs)were approximately 100 nm in size,with improved stability,high siRNA encapsulation efficiency and low cytotoxicity.CS-HAD NPs could target to CD44 receptor and deliver the therapeutic siRNA into T24 bladder cancer cells through a ligand-receptor-mediated targeting mechanism and had a specific accumulation capacity in vivo to interfere the targeted oncogene Bcl2 in bladder cancer.Overall,a CD44 targeted gene delivery system based on natural macromolecules was developed for effective bladder cancer treatment,which could be more conducive to clinical application due to its simple preparation and high biological safety.Ye Liang Yonghua Wang Liping Wang Zhijuan Liang Dan Li Xiaoyu Xu Yuanbin Chen Xuecheng Yang Hongbo Zhang Haitao Niu 2021Bioactive Materials2021,6,2:5
