1Numerical simulation of sandy bed erosion by 2D vertical jet显示文摘A two-dimensional numerical model of vertical jet scour was developed based on the turbulent flow theory and jet scour mechanism. In this model,drag force acts as the main reference variable and the critical Shields number acts as the incipient motion criteria of sediment. The morphological change in the bed caused by bed-load transport is simulated using the moving mesh method and the suspended-load is not considered. An experiment investigating vertical clear water jet scour was conducted in the laboratory,and some effective experimental results,such as flow patterns and distribution characteristics of scour pits,were obtained. Numerical simulation of the 2D jet scour was conducted using the same parameters as were used in the flume experiment. The evolution process of the jet scour observed in the experiment was simulated by the new model;validation of the numerical model and the algorithm was conducted. 3An antibody against Siglec-15 promotes bone formation and fracture healing by increasing TRAP^(+)mononuclear cells and PDGF-BB secretion显示文摘Osteoporosis(OP)is a common age-related disease characterized by a deterioration of bone mass and structure that predisposes patients to fragility fractures.Pharmaceutical therapies that promote anabolic bone formation in OP patients and OP-induced fracture are needed.We investigated whether a neutralizing antibody against Siglec-15 can simultaneously inhibit bone resorption and stimulate bone formation.We found that the multinucleation of osteoclasts was inhibited in SIGLEC-15 conditional knockout mice and mice undergoing Siglec-15 neutralizing antibody treatment.The secretion of platelet-derived growth factor-BB(PDGF-BB),the number of tartrate-resistant acid phosphatase-positive(TRAP+)mononuclear cells,and bone formation were significantly increased in the SIGLEC-15 conditional knockout mice and antibody-treated mice.The anabolic effect of the Siglec-15 neutralizing antibody on bone formation was blunted in mice with Pdgfb deleted in TRAP-1'cells.These findings showed that the anabolic effect of the Siglec-15 neutralizing antibody was mediated by elevating PDGF-BB production of TRAP4 mononuclear cells.To test the therapeutic potential of the Siglec-15 neutralizing antibody,we injected the antibody in an ovariectomy-induced osteoporotic mouse model,which mimics postmenopausal osteoporosis in women,and in two fracture healing models because fracture is the most serious health consequence of osteoporosis.The Siglec-15 neutralizing antibody effectively reduced bone resorption and stimulated bone formation in estrogen deficiency-induced osteoporosis.Of note,the Siglec-15 neutralizing antibody promoted intramembranous and endochondral ossification at the damaged area of cortical bone in fracture healing mouse models.Thus,the Siglec-15 neutralizing antibody shows significant translational potential as a novel therapy for OP and bone fracture.Gehua Zhen Yang Dan Ruomei Wang Ce Dou Qiaoyue Guo Melissa Zarr Linda N.Liu Lieping Chen Ruoxian Deng Yusheng Li Zengwu Shao Xu Cao 2021Bone Research2021,9,4:6

5Comparison of lymph node number and prognosis in gastric cancer patients with perigastric lymph nodes retrieved by surgeons and pathologists显示文摘Objective: To compare the numbers of positive and total lymph nodes and prognosis in gastric cancer patients whose perigastric lymph node retrieval was performed by surgeons and pathologists.Methods: We conducted a retrospective analysis of clinical and follow-up data from 1,056 patients who underwent gastric cancer D2 radical lymph node resection between January 2008 and December 2010 in the Gastrointestinal Surgery Department of Yantai Yuhuangding Hospital.The follow-up ended in December 2015.Patients were divided into two groups according to the specialty of physicians who performed the postoperative perigastric lymph node retrieval: the surgeon group(475 cases) and the pathologist group(581 cases).The numbers of positive and total perigastric lymph nodes and the 3- and 5-year survival were compared between gastric cancer patients in the two groups overall and stratified by TNM stage(AJCC 7th Edition).Results: Overall, the numbers of positive and total lymph nodes were significantly higher in the surgeon group than in the pathologist group(6.53±4.07 vs.4.09±3.70, P=0.021; 29.64±11.50 vs.20.71±8.56, P<0.001).Further analysis showed that the total number of lymph nodes in stage Ⅰ patients(19.40±9.62 vs.15.45±8.59, P=0.011) and the numbers of positive and total lymph nodes in stage Ⅱ(1.38±1.08 vs.0.87±1.55, P=0.031; 25.35±10.80 vs.16.75±8.56, P<0.001) and stage Ⅲ patients(8.11±6.91 vs.6.66±5.12, P=0.026; 32.34±12.55 vs.25.45±8.31, P<0.001)were significantly higher in the surgeon group than in the pathologist group.The survival analysis showed that the3- and 5-year survival of stage Ⅱ and Ⅲ patients was significantly higher in the surgeon group than in the pathologist group(82.0% vs.73.1%, 69.5% vs.61.2%, P=0.038; 49.2% vs.38.9%, 36.3% vs.28.0%; P=0.045).Conclusions: Compared with retrieval performed by pathologists, postoperative perigastric lymph node retrieval performed by surgeons was associated with significant increase in the total lymph node number of stage Ⅰ patients, the numbers of positive and total lymph nodes of stage Ⅱ and Ⅲ patients, and the survival of stage Ⅱ and stage Ⅲ gastric cancer patients.Lixin Jiang Zengwu Yao Yifei Zhang Jinchen Hu Dawei Zhao Huiyuan Zhai Xixun Wang Zhenbin Zhang Dong Wang 2016Chinese Journal of Cancer Research2016,28,5:5

7Retrieval of Aerosol Optical Depth for Chongqing Using the HJ-1 Satellite Data显示文摘Aerosol optical depth(AOD) is a common indicator applied in monitoring aerosols in the atmosphere. The hilly landscape and rapid economic growth of the megacity Chongqing have facilitated increased aerosol concentration,and it is meaningful to accurately retrieve AOD over Chongqing. The HJ-1A/B satellite of China carries a sensor/camera called the Charge Coupled Device(CCD), the spatial resolution of which meets the requirement for retrieving high resolution AOD. In this paper, analysis of the AOD retrievals from different methods using the HJ-1satellite data revealed the most suitable algorithm. Through comparison with the AOD product of Moderate Resolution Imaging Spectroradiometer(MODIS), the AOD retrieval results using enhanced vegetation index(EVI) to estimate dark pixels showed the highest correlation. The continental aerosol model was used to build a lookup table that was able to facilitate a good AOD retrieval for both city and rural areas. Finally, the algorithm that combined dark pixels, buffer areas, and the deep blue algorithm was found to be most suitable for AOD retrieval. The AOD retrieval results based on the HJ-1 data were consistent with MODIS products, and our algorithm yields reasonable results in most cases. The results were also compared with ground-based PM10 measurements synchronized with the overpass time of the HJ-1 satellite, and high correlation was found. 9Influence on lacustrine source rock by hydrothermal fluid: a case study of the Chang 7 oil shale, southern Ordos Basin显示文摘Hydrothermal fluid activity during sedimentation of the Triassic Yanchang Formation in the Ordos Basin and the impact of said activity on formation and preservation conditions of source rocks have received little attention. Oil yield, major element, trace element, rare earth element, and total sulfur(TS) data from the oil shale within the Yanchang are here presented and discussed in the context of hydrothermal influence. Oil shale samples returned relatively high total organic carbon(TOC), in the range of 4.69%–25.48%. A high correlation between TS and TOC suggests TS in the oil shale is dominated by organic sulfur and affected by organic matter. The low Al/Si ratio of oil shale samples implies quartz is a major mineralogical component. Si/(Si + Al + Fe) values suggest close proximity of the oil shale to a terrigenous source.d Eu; Fe versus Mn versus(Cu + Co + Ni) 9 10; and Si O2/(K2 O + Na2 O) versus Mn O/Ti O2, Fe/Ti, and(Fe + Mn)/Ti are evidence of hydrothermal fluid activity during oil shale sedimentation, and d U and U/Th of the oil shale indicate reducing conditions. The Sr/Ba of oil shale samples suggests fresh-water deposition. The high correlations of Fe/Ti and(Fe + Mn)/Ti with d U, U/Th, and TS demonstrate that hydrothermal fluid activity promotes reducing conditions. Sr/Ba ratios had low correlation with Fe/Ti and(Fe + Mn)/Ti, implying that hydrothermal fluid activity had little impact on paleosalinity. 14Adenovirus-mediated tissue inhibitor of metalloproteinase-3 gene transfection inhibits rabbit intervertebral disc degeneration in vivo显示文摘The aim of this study was to investigate the inhibitory effects of recombinant adenovirus vector carrying tissue inhibitor of metalloproteinase-3(RAd-TIMP-3)against degeneration of rabbit intervertebral disc.Thirty Japanese white rabbits of 4 months old were randomly divided into 5 groups.Mild or moderate rabbit lumbar disc degeneration model was constructed with the controllable axial loading device by imposing 98 N pressure at the discs for 2 weeks.Various doses of virus were injected into the degenerated discs as follows:20μL of normal saline in group 1;20μL of RAd66(an empty adenovirus vector,1.0�1010 OPU/mL)in group 2;and 20,10,and 5μL of RAdTIMP-3(1.0�1010 OPU/mL)in groups 3,4,and 5,respectively.Two weeks after the injection,the discs were collected for investigations,including assessment of degeneration degrees according to the Thompson’s grading system,reverse-transcription polymerase chain reaction(RT-PCR)assay for TIMP-3 gene,Safranin O-Fast green staining,and immunohisto-chemical staining for TIMP-3 and type II collagen.According to Thompson’s criteria,the degeneration of groups 3,4,and 5,especially group 3,was alleviated as compared with groups 1 and 2.RT-PCR revealed that the expression of TIMP-3 in groups 3,4,and 5,especially in group 3,was significantly enhanced as compared with group 1(P<0.01).Both Safranin O-Fast green staining and type II collagen staining demonstrated better reserved integrity of disc matrix in groups 3,4,and 5 than in groups 1 and 2.TIMP-3 staining exhibited an obvious increase of positive-staining rate in groups 3,4,and 5 as compared with group 1.The positive-staining rate in group 3(79.42%�1.35%)was about 3 times that of group 1(25.47%�5.46%,P<0.01).RAdTIMP-3 can effectively protect the matrix of rabbit intervertebral disc against overloading-induced degeneration in a dose-dependent manner,resulting in the alleviation of disc degeneration.Xudong YU Zengwu SHAO Liming XIONG Weiwei XU Hezhong WANG Huifa XU 2009Frontiers of Medicine2009,3,4:1

16Characterizing the tumor microenvironment at the single-cell level reveals a novel immune evasion mechanism in osteosarcoma显示文摘The immune microenvironment extensively participates in tumorigenesis as well as progression in osteosarcoma(OS).However,the landscape and dynamics of immune cells in OS are poorly characterized.By analyzing single-cell RNA sequencing(sc RNA-seq)data,which characterize the transcription state at single-cell resolution,we produced an atlas of the immune microenvironment in OS.The results suggested that a cluster of regulatory dendritic cells(DCs)might shape the immunosuppressive microenvironment in OS by recruiting regulatory T cells.We also found that major histocompatibility complex class I(MHC-I)molecules were downregulated in cancer cells.The findings indicated a reduction in tumor immunogenicity in OS,which can be a potential mechanism of tumor immune escape.Of note,CD24 was identified as a novel“don’t eat me”signal that contributed to the immune evasion of OS cells.Altogether,our findings provide insights into the immune landscape of OS,suggesting that myeloid-targeted immunotherapy could be a promising approach to treat OS.Weijian Liu Hongzhi Hu Zengwu Shao Xiao Lv Zhicai Zhang Xiangtian Deng Qingcheng Song Yong Han Tao Guo Liming Xiong Baichuan Wang Yingze Zhang 2023Bone Research2023,11,1:1
17Traumatic brain injury stimulates sympathetic tone-mediated bone marrow myelopoiesis to favor fracture healing显示文摘Traumatic brain injury(TBI)accelerates fracture healing,but the underlying mechanism remains largely unknown.Accumulating evidence indicates that the central nervous system(CNS)plays a pivotal role in regulating immune system and skeletal homeostasis.However,the impact of CNS injury on hematopoiesis commitment was overlooked.Here,we found that the dramatically elevated sympathetic tone accompanied with TBI-accelerated fracture healing;chemical sympathectomy blocks TBIinduced fracture healing.TBI-induced hypersensitivity of adrenergic signaling promotes the proliferation of bone marrow hematopoietic stem cells(HSCs)and swiftly skews HSCs toward anti-inflammation myeloid cells within 14 days,which favor fracture healing.Knockout ofβ3-orβ2-adrenergic receptor(AR)eliminate TBI-mediated anti-inflammation macrophage expansion and TBIaccelerated fracture healing.RNA sequencing of bone marrow cells revealed that Adrb2 and Adrb3 maintain proliferation and commitment of immune cells.Importantly,flow cytometry confirmed that deletion ofβ2-AR inhibits M2 polarization of macrophages at 7th day and 14th day;and TBI-induced HSCs proliferation was impaired inβ3-AR knockout mice.Moreover,β3-andβ2-AR agonists synergistically promote infiltration of M2 macrophages in callus and accelerate bone healing process.Thus,we conclude that TBI accelerates bone formation during early stage of fracture healing process by shaping the anti-inflammation environment in the bone marrow.These results implicate that the adrenergic signals could serve as potential targets for fracture management.Weijian Liu Wei Chen Mao Xie Chao Chen Zengwu Shao Yiran Zhang Haiyue Zhao Qingcheng Song Hongzhi Hu Xin Xing Xianyi Cai Xiangtian Deng Xinyan Li Peng Wang Guohui Liu Liming Xiong Xiao Lv Yingze Zhang 2023Signal Transduction and Targeted Therapy2023,8,8:1
18Kindlin-2 regulates skeletal homeostasis by modulating PTH1R in mice显示文摘In vertebrates,the type 1 parathyroid hormone receptor(PTH1R)is a critical regulator of skeletal development and homeostasis;however,how it is modulated is incompletely understood.Here we report that deleting Kindlin-2 in osteoblastic cells using the mouse 10-kb Dmp1-Cre largely neutralizes the intermittent PTH-stimulated increasing of bone volume fraction and bone mineral density by impairing both osteoblast and osteoclast formation in murine adult bone.Single-cell profiling reveals that Kindlin-2 loss increases the proportion of osteoblasts,but not mesenchymal stem cells,chondrocytes and fibroblasts,in non-hematopoietic bone marrow cells,with concomitant depletion of osteoblasts on the bone surfaces,especially those stimulated by PTH.Furthermore,haploinsufficiency of Kindlin-2 and Pth1r genes,but not that of either gene,in mice significantly decreases basal and,to a larger extent,PTH-stimulated bone mass,supporting the notion that both factors function in the same genetic pathway.Mechanistically,Kindlin-2 interacts with the C-terminal cytoplasmic domain of PTH1R via aa 474–475 and Gsα.Kindlin-2 loss suppresses PTH induction of cAMP production and CREB phosphorylation in cultured osteoblasts and in bone.Interestingly,PTH promotes Kindlin-2 expression in vitro and in vivo,thus creating a positive feedback regulatory loop.Finally,estrogen deficiency induced by ovariectomy drastically decreases expression of Kindlin-2 protein in osteocytes embedded in the bone matrix and Kindlin-2 loss essentially abolishes the PTH anabolic activity in bone in ovariectomized mice.Thus,we demonstrate that Kindlin-2 functions as an intrinsic component of the PTH1R signaling pathway in osteoblastic cells to regulate bone mass accrual and homeostasis.Xuekun Fu Bo Zhou Qinnan Yan Chu Tao Lei Qin Xiaohao Wu Sixiong Lin Sheng Chen Yumei Lai Xuenong Zou Zengwu Shao Meiqing Wang Di Chen Wenfei Jin Youqiang Song Huiling Cao Ge Zhang Guozhi Xiao 2020Signal Transduction and Targeted Therapy2020,5,1:0

20Molten salt synthesis of neodynium oxyfluoride in various fluoride media with different fluoride ion activities显示文摘Neodymium oxyfluoride has received much attention in the fields of anionic solid electrolytes.luminescent,catalytic and magnetic materials because of its structure combined advantages of rareearth cations with F^(-)and O_(2)^(-)anions.In this work,neodynium oxyfluoride was synthesized by the reaction between neodymium oxide and four fluoride media with different fluoride ion activities.The synthesis processes in molten LiF-CaF_(2)-NdF_(3),LiF-NdF_(3),NaF-CaF_(2)-NdF_(3)and NaF-KF-NdF_(3)are observed in situ by a confocal scanning laser microscope.The expansion of neodymium oxide particle is observed in the LiF-CaF_(2)-NdF_(3),LiF-NdF_(3),and NaF-KF-NdF_(3)melts,and the growth of needle crystals on neodymium oxide particle is clearly observed in molten NaF-CaF_(2)-NdF_(3).Based on scanning electron microscopy(SEM)-energy dispersive X-ray spectroscopy(EDS)and X-ray diffraction(XRD)analyses of products,neodynium oxyfluoride was successfully synthesized in the four fluoride media.The neodynium oxyfluoride generated in the LiF-CaF_(2)-NdF_(3),LiF-NdF_(3),and NaF-KF-NdF_(3)melts is a tetragonal structure.However,in molten NaF-CaF_(2)-NdF_(3),neodynium oxyfluoride with a rhombohedral structure is formed.It is suggested that the substitution of Na(Ⅰ)and Ca(Ⅱ)for Nd(Ⅲ)can transform NdOF from tetragonal structure to rhombohedral structure.The growth rate of needle crystals generated in molten NaF-CaF_(2)-NdF_(3)was calculated based on the result of a confocal scanning laser microscope,and it is found that the reaction kinetics of crystal formation is zero-order reaction.The effect of fluoride media on the structure and morphology of formed NdOF were evaluated by XRD,X-ray photoelectron spectroscopy(XPS)and SEM.The neodymium oxyfluoride prepared in the fluoride media with high fluoride ion activity has low binding energy of F 1 s.The ratio of adsorbed oxygen to lattice oxygen for neodymium oxyfluoride prepared in molten LiF-NdF_(3)is larger than those in the other three fluoride media,so it can have better catalytic performance.Yusheng Yang Tuoyao Wang Ran Zhao Shuyan Zhang Zengwu Zhao 2022Journal of Rare Earths2022,40,12:0
