2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 4Application of machine learning in predicting the rate-dependent compressive strength of rocks显示文摘Accurate prediction of compressive strength of rocks relies on the rate-dependent behaviors of rocks, and correlation among the geometrical, physical, and mechanical properties of rocks. However, these properties may not be easy to control in laboratory experiments, particularly in dynamic compression experiments. By training three machine learning models based on the support vector machine(SVM), backpropagation neural network(BPNN), and random forest(RF) algorithms, we isolated different input parameters, such as static compressive strength, P-wave velocity, specimen dimension, grain size, bulk density, and strain rate, to identify their importance in the strength prediction. Our results demonstrated that the RF algorithm shows a better performance than the other two algorithms. The strain rate is a key input parameter influencing the performance of these models, while the others(e.g. static compressive strength and P-wave velocity) are less important as their roles can be compensated by alternative parameters. The results also revealed that the effect of specimen dimension on the rock strength can be overshadowed at high strain rates, while the effect on the dynamic increase factor(i.e. the ratio of dynamic to static compressive strength) becomes significant. The dynamic increase factors for different specimen dimensions bifurcate when the strain rate reaches a relatively high value, a clue to improve our understanding of the transitional behaviors of rocks from low to high strain rates.Mingdong Wei Wenzhao Meng Feng Dai Wei Wu 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,5:2
5Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 6Two Ultraviolet Radiation Datasets that Cover China显示文摘Ultraviolet(UV) radiation has significant effects on ecosystems, environments, and human health, as well as atmospheric processes and climate change. Two ultraviolet radiation datasets are described in this paper. One contains hourly observations of UV radiation measured at 40 Chinese Ecosystem Research Network stations from 2005 to 2015. CUV3 broadband radiometers were used to observe the UV radiation, with an accuracy of 5%, which meets the World Meteorology Organization's measurement standards. The extremum method was used to control the quality of the measured datasets. The other dataset contains daily cumulative UV radiation estimates that were calculated using an all-sky estimation model combined with a hybrid model. The reconstructed daily UV radiation data span from 1961 to 2014. The mean absolute bias error and root-mean-square error are smaller than 30% at most stations, and most of the mean bias error values are negative, which indicates underestimation of the UV radiation intensity. These datasets can improve our basic knowledge of the spatial and temporal variations in UV radiation. 10Construction and heterologous expression of the di-AFN A_(1) biosynthetic gene cluster in Streptomyces model strains显示文摘Natural cyclohexapeptide AFN A_(1) from Streptomyces alboflavus 313 has moderate antibacterial and antitumor activities.An artificial designed AFN A_(1) homodimer,di-AFN A_(1),is an antibiotic exhibiting 10 to 150 fold higher biological activities,compared with the monomer.Unfortunately,the yield of di-AFN A_(1) is very low(0.09±0.03 mg·L^(−1))in the engineered strain Streptomyces alboflavus 313_hmtS(S.albo/313_hmtS),which is not friendly to be genetically engineered for titer improvement of di-AFN A_(1) production.In this study,we constructed a biosynthetic gene cluster for di-AFN A_(1) and increased its production through heterologous expression.During the collection of di-AFN A_(1) biosynthetic genes,the afn genes were located at three sites of S.alboflavus 313 genome.The di-AFN A_(1) biosynthetic gene cluster(BGC)was first assembled on one plasmid and introduced into the model strain Streptomyces lividans TK24,which produced di-AFN A_(1) at a titer of 0.43±0.01 mg·L^(−1).To further increase the yield of di-AFN A_(1),the di-AFN A_(1) BGC was multiplied and split to mimic the natural afn biosynthetic genes,and the production of di-AFN A_(1) increased to 0.62±0.11 mg·L^(−1) in S.lividans TK24 by the later strategy.Finally,different Streptomyces hosts were tested and the titer of di-AFN A_(1) increased to 0.81±0.17 mg·L^(−1),about 8.0-fold higher than that in S.albo/313_hmtS.Successful heterologous expression of di-AFN A_(1) with a remarkable increased titer will greatly facilitate the following synthetic biological study and drug development of this dimeric cyclohexapeptide.WEI Weijia WANG Wenzhao LI Chao TANG Yue GUO Zhengyan CHEN Yihua 2022Chinese Journal of Natural Medicines2022,20,11:0

14The earlier,the better?A review of neoadjuvant immunotherapy in resectable non-small-cell lung cancer显示文摘Immune checkpoint inhibitors(ICIs)have revolutionized the approach to advanced and locally advanced non-small-cell lung cancer(NSCLC).Antibodies blocking inhibitory immune checkpoints,such as programmed death 1(PD-1)and its ligand(PD-L1),have remarkable antitumor efficacy and have been approved as a standard first-or second-line treatment in non-oncogene-addicted advanced NSCLC.The successful application of immunotherapy in advanced lung cancer has motivated researchers to further evaluate its clinical role as a neoadjuvant setting for resectable NSCLC and for improved long-term overall survival and curative rates.In this review,we discuss the efforts that incorporate ICIs into the treatment paradigm for surgically resectable lung cancer.We reviewed the early-phase results from neoadjuvant clinical trials,the landscape of the majority of ongoing phase III trials,and discuss the prospects of ICIs as a curative therapy for resectable lung cancer.We also summarized the potential biomarkers and beneficiaries involved in the current study,as well as the remaining unresolved challenges for neoadjuvant immunotherapy.Fajiu Li Ying Chen Juanjuan Wu Chenghong Li Shi Chen Ziyang Zhu Wei Qin Min Liu Bingzhu Hu Shuang Liu Wenzhao Zhong 2022Chronic Diseases and Translational Medicine2022,8,2:0
